1.Changes of sex hormone levels in male patients with type 2 diabetes complicated with non-alcoholic fatty liver disease

[Summary] The aim of this study was to investigate the changes of sex hormone in male patients with type 2 diabetes and complicated with non-alcoholi fattty liver disease (NAFLD),and to detect the characteristics of the metablic indexs.A total of 112 patients were enrolled and divided into NAFLD group (n =54) and non-NAFLD group (n =58).All patients were measured for waist circumference,body mass index,metablic index,sex hormone,and serum transaminase.Compared with non-NAFLD group,the levels of fasting blood glucose,2 hour postprandial blood glucose,HbA1C in NAFLD group were significantly elevated [(8.75 ± 2.58 vs 7.79 ± 1.89) mmol/L,(18.12 ± 3.95 vs 15.63 ± 3.89) mmol/L,(11.96 ±4.85 vs 10.05 ±4.15)%,all P<0.05].In addition,total cholesterol,triglyceride,body mass index,fasting insulin,and postprandial insulin in NAFLD group were significantly elevated [(4.97± 1.02 vs4.15±0.92) mmol/L,(2.74±2.25 vs 2.01± 1.45)mmol/L,(27.34±3.93 vs 22.38±3.39) kg/m2,(9.62 ± 5.80 vs 6.18 ± 4.21) μIU/ml,(72.71 ± 109.70 vs 31.72 ± 42.27) μIU/ml,all P<0.05];while the total testosterone and free testosterone were lowered dramatically[(14.18 ±6.39 vs 18.21 ±6.14) nmol/L,(0.32 ± ± 0.03 vs 0.39 ± 0.08) ng/dl,both P<0.05].Male patients with type 2 diabetes mellitus complicated with NAFLD are more prone to have disturbance in the lipid metabolism and had larger percentages of obesity and with lower level of testesterone.

Objective To study the impacts of variation of folate metabolic component including folic acid (FA), riboflavin (RF) and MTHFR C677T polymorphism on chromosome 8 and 17 segregation. Method RPMI1640 medium was modified with different combinations of low (LF) and high (HF) concentrations of folic acid (20, 200 nmol/L) and low (LR) and high (HR) concentrations of riboflavin (1 and 500 nmol/L). The lymphocytes with different MTFHR C677T genotype were cultured in the mentioned media for 9d. Cytokinesis was blocked by cytochalasin B and the slide was prepared as usual. 3.The Influence of Several Chief Parameters in Electrospinning on the Morphology and the Fiber Diameter of PLGA Fiber Scaffolds

To prepare PLGA fiber scaffolds by electrospinning process and investigate the influence of preparation parameters on the structure of the scaffolds. With the compound of THF and DMF as the solvent, the PLGA fiber scaffolds with different surface morphology were fabricated via altering PLGA solution concentration, flowing rate and applied electric field strength. The morphology and diameter of the fibers were observed using a scanning electron microscope (SEM). The biocompatibility of cell-scaffold complex was also evaluated by seeding human dermal fibroblasts onto the PLGA fiber scaffolds, including cell adhesion and proliferation. The results show that the diameter of fibers and the bound of distributing increase with the increase in the concentration of PLGA solution. As the flowing rate increases, the diameter of fibers increases. However, with the increase in the applied electric field strength, no significant difference in the diameter of the fiber can be observed. Furthermore, both the increase in the concentration of PLGA solution and applied electric field strength in the volume range of current investigation can lead to the reduction in the beads formation within the scaffold. The results of in vitro cell culture on the PLGA scaffolds also confirm that the PLGA fiber can support the adhesion and proliferation of huaman dermal fibroblasts.

5.Clinical Observation of Vesiculation Moxibustion for Allergic Rhinitis Due to Deficient Cold of Lung-qi

Objective To compare the clinical efficacies of vasiculation moxibustion with pastes prepared in different ways and at different acupoint groups in treating allergic rhinitis due to deficient cold of lung qi. Method Eighty subjects were randomized into 4 groups, 20 in each group. Raw Bai Jie Zi (Semen Brassicae) was used in the vesiculation paste for group 1 and 2;stir-baked Bai Jie Zi (Semen Brassicae) was used for group 3 and 4. In group 1 and group 3, bilateral Feishu (BL 13), Fengmen (BL12), and Gaohuang (BL 43) were selected; in group 2 and group 4, bilateral Dazhu (BL11), Geshu (BL17), and Shenshu (BL23) were selected. The symptom and sign scores were compared before and after intervention. Result There were significant differences in comparing the symptoms and signs among the four groups (P<0.05), and paired comparisons showed that the therapeutic efficacies of group 1 and group 3 were significantly higher than that of group 2 and group 4 (P<0.05), while the differences in the therapeutic efficacy between group 1 and group 3, group 2 and group 4 were statistically insignificant (P>0.05). Conclusion Vesiculation moxibustion at the point group of Fengmen (BL12), Feishu (BL13), and Gaohuang (BL43) can markedly improve the symptoms and signs of allergic rhinitis due to deficient cold of lung qi;raw and stir-baked Bai Jie Zi (Semen Brassicae) can work equally in vesiculation paste for allergic rhinitis.
7.New perspective in children and adolescents’ physical and mental health study: from positive youth development viewpoint

Objective Positive youth development (PYD) perspective provides a new paradigm for children and adolescents’ physical and mental health research. It emphasizes children and adolescents’ s strengths and plasticity, as well as the impact of the interaction between individual and environment on children and adoleseents’ physical and mental health. This perspective enriches the comprehensive understanding of children and adolescents’ physical and mental health, and provides theoretical support and practical guidance for physical and mental health prevention intervention and interdisciplinary research. It also has an important impact on public policy development. The present paper systematically reviewed the theoretical models, empirical research and future prospects regarding the relationship between PYD and children and adolescents' physical and mental health based on the connotation structure and measurement development.
8.Establishment and application of standard serum proteomic pattern for diagnosis of early liver cancer.

OBJECTIVETo establish standard serum proteomic pattern for early liver cancer and explore its value in early liver cancer diagnosis.
METHODSThe serum samples were obtained from 12 healthy subjects and 12 patients with early liver cancer. After removal of serum albumin and IgG, 300 µg serum sample was mixed with the hydrating solution for two-dimensional gel electrophoresis. Each experiment was repeated for 3 times. With silver staining and image analysis, the differentially expressed proteins were identified to establish the standard proteomic pattern for early liver cancer. The proteomics pattern was then tested in the diagnosis of 13 cases of chronic liver disease and 10 healthy individuals in a double-blind manner.
RESULTSThirty-eight differentially expressed protein spots were identified. Eleven protein spots with differential expressions by over 5 folds were analyzed to establish the standard proteomics pattern for early liver cancer, which showed a sensitivity of 92% and specificity of 90% in early diagnosis of liver cancer.
CONCLUSIONThe standard serum 2-DE proteomic pattern for early liver cancer shows distinct differences from that of healthy subjects. The differentially expressed proteins and their specific combination pattern may have the potential for diagnosis and prognostic evaluation of early liver cancer.

9.Detection of Helicobacter species related genes coding for 16S rRNA in the liver tissue of patients with chronic liver disease.

OBJECTIVETo investigate the infection of Helicobacter pylori (Hp) in the liver tissue of patients with chronic liver disease and the association between Hp and chronic liver disease.
METHODSLiver tissue samples were obtained by liver biopsy and surgical resection from 30 healthy subjects, 30 patients with chronic hepatitis, 30 with cirrhosis and 30 with liver cancer. All the samples were confirmed by pathological examination. The gene fragment coding for 16SrRNA were amplified by PCR with sequence analysis.
RESULTSThe PCR product of the 16SrRNA gene was 109 bp in length. Hp 16SrRNA was detected in 18 out of 30 liver biopsy samples from patients with primary cancer (60.0%), in 14 samples from patients with liver cirrhosis (47.0%), and in none of the samples from normal subjects or patients with chronic hepatitis. Sequencing analysis of Hp 16SrRNA gene in the liver tissue showed a 98.8% homology with the gene fragment encoding Hp 16SrRNA.
CONCLUSIONHp is identified in the liver tissue of patients with chronic liver disease, suggesting the possible correlation between Hp infection and hepatocellular carcinoma.

10.Inhibitory effect of dutasteride on the expressions of epididymal Claudin1 and β-catenin in male rats.

OBJECTIVETo explore the molecular mechanism of dutasteride inhibiting fertility by studying its effects on the expressions of the epididymal epithelial junction proteins Claudin1 and β-catenin in rats.
METHODSSixteen 3-month-old SD male rats were equally divided into an experimental and a negative control group to be treated intragastrically with dutasteride at 40 mg/kg per day and the same dose of solvent, respectively, for 14 consecutive days. Then, the sperm motility and morphology of the rats were detected by computer-assisted sperm analysis, the serum levels of testosterone (T) and dihydrotestosterone (DHT) measured by ELISA, changes in the tight junction of epididymal cells observed under the transmission electron microscope, the protein and gene expressions of Claudin1 and β-catenin determined by RT-PCR and immunohistochemistry, and the conception rate of the mated female rats calculated.
RESULTSDutasteride significantly suppressed the serum DHT level, sperm motility, and fertility of the rats (P <0.05). Interspaces between epididymal epithelial cell tight junctions were observed, the volume of epididymal fluid obviously increased, and the expressions of Claudin1 and β-catenin gene and protein remarkably downregulated in the experimental rats (P <0.05).
CONCLUSIONDutasteride can significantly inhibit the fertility of male rats by reducing the serum DHT level, suppressing Claudin1 and β-catenin expressions, and damaging epididymal epithelial cell junctions.
