Objective To explore the effect of DNA methylation inhibitor 5-aza-2'-deoxycytidine (5-Aza-CdR) on proliferation and tumor suppressor gene PRDM1α expression in Burkitt's lymphoma cell line NAMALWA. Methods MTT was used to study the impact of DNA methylation inhibitor on proliferation in NAMALWA cells,and comparative real-time reverse transcription-polymerase chain reaction (RQ-PCR) with SYBR Green assay was used to detect PRDM1α gene expression. Results MTT results showed that 5-AzaCdR inhibited proliferation of NAMALWA cells in a dose-dependent manner.After treated with 5-Aza-CdR for 72 h at 0.01,0.0625,0.1,0.125,0.25,0.5,1,2 and 4 μmol/L,the inhibition rates were 21.93 ％,39.23 ％,47.69 ％,50.37 ％,53.54 ％,57.72 ％,62.31％,65.68 ％,67.87 ％,respectively,and the differences of absorbance among the groups were also statistically significant (all P＜ 0.01). Meanwhile, 5-Aza-CdR could induce the re-expression of PRDM1α, and △Ct of 0.5, 1, 2 μmol/L groups showed significant differences compared with control group (all P ＜ 0.05). Conclusion DNA methylation inhibitor 5-Aza-CdR can significantly inhibit the proliferation of Burkitt' s lymphoma cell line NAM ALWA,and the possible mechanism of this inhibition may be induction of demethylation and re-expression of PRDM 1α.

Objective To investigate the impact of human adipose-derived stem cells(hASCs) -plastelet-rich plasma(PRP) composite on the wound healing of radiation skin injury of nude mice.Methods ASCs were isolated from healthy adults after liposuction, conducted primary culture and subculture.PRP was prepared by two-step ultracentrifugation.The ASCs-PRP composite was prepared with ASCs (1 ×107cells/ml) at passage 6 and PRP.Twenty-four nude mice were randomly divided into 4 groups (n=6).A round shaped full-thickness skin defect (D=1cm) was made on the mice back after 20Gy (60Co) round shaped exposure (D=2cm).Right after radiation, wounds were treated as follows,in group A,there was ASCs+PRP, in group B, PRP,in group C, ASCs,in group D, sham group.The wounds were observed at postoperative 7 and 14 days.HE and CD31 stain were observed at 14th days after surgery.Results The wound healing of group A healed completely at 14th days.In group D,the wound healing were slowest and still filled with granulation tissue at 14th days.The wound healing of group B and group C were faster than that of group D, but they haven′t healed completely at 14th days.The inflammatory cells were significantly lower and the blood vessel density positive rate were significantly higher in ACSs+PRP group than in ACSs group,PRP group and sham group (P<0.05).Conclusion ASCs-PRP Composite can accelerate the wound healing of radiation skin injury by promoting wound angiogenesis.

OBJECTIVE:To study the possible mechanism of Bushenshengsui I(BSSS I),a kind of compound Chinese herbs,in treatment of aplastic anaemiaMETHODS:The animal model of mice with aplastic anaemia was established by 60Co irradiationThe therapeutic effects of BSSS I on aplastic anaemia were observed with the changes of peripheral blood cell count,bone marrow GM-CFU-c and activities of blood IL-1,IL-2 and IL-6RESULTS:After treatment with BSSS I,the peripheral blood cell count,amount of bone marrow karyocyte and GM-CFU-c of bone marrow of the aplastic anaemia mice elevated markedly,and the blood IL-1 and IL-6 activities increased obviouslyThe levels of IL-6 increased to the level of normal and showed positive relationship with IL-1(r=0904)The levels of IL-2 of the treatment group showed a decrease but with no significant differencesCONCLUSION:The results showed that BSSS I may improve the function of hematopoiesis by elevating the peripheral blood cells and the amount of bone marrow karyocytes,accelerating GM-CFU-c of bone marrow,and through moderating the activities of IL-1,IL-2,IL-6

Objective To investigate the protection of panax notoginseng saponins(PNS) against ischemia reperfusion injury by liver graft in rats.Method The experimental rat model of orthotopic liver transplantation was set up according to Kamada's technique and the rats were randomly divided into three groups: normal saline control group(group N),PNS preconditioning group(group P) and sham-operated groups(group S).The expression levels of NF-?B and intercellular adhesion molecule-1(ICAM-1) were detected by immunohistochemistry and the filtration of polymorphonuclear neutrophils(PMNs) was measured with myeloperoxidase(MPO) enzyme method in 2h,6h and 24h after ischemia reperfusion respectively.The activities of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in the serum were also detected.Result The activation of NF-?B in group N increased significantly at 2h after reperfusion and then decreased.The expression level of ICAM-1 increased significantly at 2h after reperfusion,and reached its peak level at 6h in group N.The expression level of ICAM-1 was significantly lower in group P than that in group N(P

Objective To introduce the clinical experience and investigate the significance and safety of harmonic scalpel combined with meticulous capsular dissection technique in thyroidectomy.Methods A retrospective analysis was performed on 275 patients who underwent thyroidectomy among which 135 patients were treated by capsular dissection technique and harmonic scalpel (harmonic scalpel group),the other 140 patients were treated by electric knife and traditional ligation (traditional group).The thyroidectomy operative time,intraoperative bleeding volume,postoperative drainage volume and surgical complications were compared between two groups.Results In harmonic scalpel group,the thyroidectomy operative time of total thyroidectomy and subtotal thyroidectomy was significantly shorter than that in traditional group [(50.0±6.9) min vs.(82.0± 14.4) min,(39.9±6.6) min vs.(73.1 ± 14.5) min](P＜0.01).The intraoperative bleeding volume of total thyroidectomy and subtotal thyroidectomy in harmonic scalpel group was significantly lower than that in traditional group [(18.7 ± 10.9) ml vs.(41.9 ± 15.6) ml,(17.7 ± 9.3) ml vs.(42.2 ± 14.6) ml] (P ＜ 0.01).The postoperative drainage volume of total thyroidectomy and subtotal thyroidectomy in harmonic scalpel group was significantly lower than that in traditional group [(40.7 ± 11.7) ml vs.(53.9 ± 17.5) ml,(42.7 ± 14.5) ml vs.(61.8 ±22.9) ml](P＜0.01).The rate of temporary hypoparathyroidism and temporary recurrent laryngeal nerve palsy had no significant difference between two groups (P ＞ 0.05).Conclusions Application of harmonic scalpel combined with meticulous capsular dissection technique in thyroidectomy can significantly reduce thyroidectomy operative time,intraoperative bleeding volume,postoperative drainage volume and surgical complications.Harmonic scalpel combined with meticulous capsular dissection technique in thyroidectomy should be widely used.

Aim: To study the effects of berberine( Ber) on the rapidly activating component( Ⅰ_(Kr)), the slowly activating component(Ⅰ_(Ks)) of the delayed rectifier potassium current and the inward rectifier potassium current(Ⅰ_(K1)) in cardiomyopathic guinea pig ventricular myocytes. Methods: After guinea pigs were ip L-thyroxine 0. 5 mg/kg for 10 d, their hearts were cardiomyopathic. Then whole cell patch-clamp recording technique was used to observe the effect of 30 μmol/L Ber on the Ⅰ_(Kr), Ⅰ_(Ks) and Ⅰ_(K1) in cardiomyopathic guinea pig ventricular myocytes. Results: In cardiomyopathic guinea pig ventricular myocytes, Ber 30 μmol/L markedly inhibited Ⅰ_(Kr) and Ⅰ_(Ks) by 22. 8% and 29. 5% at + 10 mV and + 80 mV, respectively. The effect of Ber on Ⅰ_(Ks) was greater than that on Ⅰ_(Kr). Ber 30 μmol/L also inhibited the inward component of Ⅰ_(K1) by 29. 1% at + 120 mV, but the reverse potential of Ⅰ_(K1) was unaffected. Ber( 1-300 μmol/L) was shown to inhibit Ⅰ_(Kr) and Ⅰ_(Ks) in a concentration-dependent manner. Their IC_(50), were 76. 74 μmol/L and 55. 37 μmol/L, respectively. Conclusion: Ber inhibited Ⅰ_(Kr),Ⅰ_(Ks) and Ⅰ_(K1) in cardiomyopathic guinea pig ventricular myocytes, which may be important in understanding the antiarrhythmic effects of this drug.

Tuberculous meningitis is an important global medical problem which gives rise to high morbidity and mortality. It is the most severe form of extrapulmonary Mycobacterium tuberculosis. Comprehensive prevention effort, prompt diagnosis and rational treatment are all keys to improving treatment outcomes; yet many unsolved problems remain. On the other hand, the new problems, such as HIV co-infection and drug-resistance are posing important challenges. This review outlines the epidemiology, pathogenesis, diagnosis, management and prognosis of tuberculous meningitis. We mainly focus on research carried out in the recent decades, giving special attention to the work done among the Asian populations

Objective To examine the effects of nitroglycerin tolerance on the following myocardial ischemia reperfusion injury (MI/R). Methods Male Sprague-Dawley rats were rardomly subjected to either one of the following treatments: GTN 600 ?g/(kg/h) or saline infusion for 12 h. GTN tolerance was determined with the measurement of mean arterial blood pressure (MABP) and isolated arterial tension. All the GTN-tolerated and the control rats were subjected to 40 min of ischemia followed by 4 h of reperfusion. Myocardial apoptosis (TUNEL analysis), myocardial infarction size (Evans blue-TTC stain), plasma creatine kinase (CK) and lactate dehydrogenase (LDH) activity were determined. Results Nitrate tolerance aggravated MI/R injury as evidenced by increased myocyte apoptosis and infarction size and plasma CK, LDH activity. However, sole GTN tolerance without MI/R injury failed to show any deleterious effect on rat heart. Conclusion GTN tolerance may exert a “potential" deleterious effect on the ischemic hearts.

Objective To investigate the effectiveness of Ender nail fixation in the management of pediatric femoral shaft fractures. Methods A total of 24 children with femoral shaft fracture underwent small incision Ender nail internal fixation followed by 4 weeks of monolateral hip spica-cast immobilization. Results Follow-up observations for 6~24 months revealed no non-union or delayed union. The affected limbs were found within 1 cm shorter or longer than the contralateral in 4 cases. None became lame, and the functions of lower limbs were completely recovered. Conclusions So far as indications and contra-indications of Ender nail fixation are strictly followed, this treatment for femoral shaft fractures in children of age 5~10 years gives characters of small incision, anatomic reduction, unimpaired periosteum, short hospital stay, and quick functional recovery.

Objective To explore the diagnostic value of serum miRNA-182 in colorectal cancer(CRC).Methods A total of 53 CRC patients were enrolled in this study,serum miRNA-182 was detected by quantitative real time polymerase chain reaction(qRT-PCR).The results were compared with those of patients with benign intestinal diseases and healthy controls.Results The content of serum miRNA-182 in CRC patients was significantly higher than those in patients with benign intestinal diseases and healthy group(P<0.05).The ROC curve showed that the area under the curve of serum miRNA-182 was 0.846,the confidence interval was 0.768-0.925,the diagnostic sensitivity was 77.1%,and the specificity was 81.1%.Serum miRNA-182 levels were correlated with TNM stage(P=0.011) and distant metastasis(P=0.002) in patients with CRC,but no significant correlation with other clinical parameters(P>0.05).Serum miRNA-182 level in CRC patients after surgery was significantly lower than that before operation(P<0.01),and which was significantly higher in patients with recurrence/metastasis after surgery(P=0.019).Conclusion Serum miRNA-182 level increased significantly in CRC patients,and which was closely related to the disease and malignant progression,serum miRNA-182 could be used as a potential marker for clinical diagnosis and disease evaluation of CRC.
