1.Clinical analysis of amniotic membrane patches and grafts for acute ocular surface burn

AlM: To investigate the effect and value of amniotic membrane patches and grafts for acute ocular surface burn at different degrees.?METHODS: A retrospective analysis of 28 cases ( 28 eyes) affected by ocular chemical or thermal burn with different degree were included in our hospital from March 2007 to March 2012. Amniotic membrane patched was undergone in 13 eyes with fresh amnion that the patients corneal burns degree II or Ⅲ with partial limbal buns at degree Ⅳ. Amniotic membrane grafts was performed in 15 eyes with fresh amnion that the patients all corneal burns at degree Ⅲ with the whole limbal necrosis without severe eyelid defect. The follow-up time ranged 6 ~24mo. The postoperative visual acuity, the condition of amniotic membrane transplant, renovation of cornea and complications were observed.?RESULTS: Postoperative corrected visual acuity was improved in 20 eyes (71%), it was not changed in 5 eyes (18%), the visual acuity declined in 3 eyes (11%). The amniotic membrane survived in 23 eyes and the survival rate was up to 82%. The cornea of 4 eyes recovered to transparent, nebula emceed in 8 eyes eventually, corneal macula emerged in 10 eyes, 4 eyes ended up with leukoma, 2 eyes developed corneal melting after therapy, then received lamellar keratoplasty. Corneal surface become epithelization after amnion patches or grafts, but any of them have recurrent epithelial erosion, and become stable epithalization after repeat operation.?CONCLUSlON: Amniotic membrane patches and grafts is an effective method to deal with acute ocular surface burn.
2.Effects of Transgenic Biocontrol Bacterium 308R(pCPP430) on Tomato Rhizosphere Bacterial Community

The study aimed at investigation the effect of the transgenic biocontrol bacteria 308R(pCPP430) on the tomato rhizosphere bacterial communities. Strain 308R (Pantoea agglomerans), an adnascent bacterium isolated from tomato, was a biocontrol bacterium. The recombinant cosmid pCPP430 contained the hrp gene cluster of Erwinia amylovora was transformed into 308R Strain. Transgenic biocontrol bacterium 308R(pCPP430) produced Harpin protein which induce hypersensitive response and disease resistance in plants and, surprisingly, increase plant growth. Two complementary methods, sole-carbon-source utilization tests (SCSU) and ERIC-PCR, were used to compare the characterization of bacterial communities from three groups of tomato roots dipped in sterilized water, 308R suspended liquid and 308R(pCPP430) suspended liquid. The cluster analysis and principal components analysis of the number of colonies from SCSU indicated consistent differences in the rhizosphere of each group. Cluster analysis of SCSU data showed that there were consistent differences in the rhizosphere of each group. The rhizosphere bacteria communities of root dipped with 308R(pCPP 430) and 308R have a relative good replication, but the differences of corresponding replicates in dipped with distilled water was increased by degrees. Cluster analysis of ERIC-PCR fingerprints revealed that most of results (eight in ten) have overlaps between roots dipped in water and those dipped in 308R liquid. The methods used in this study may prove a useful approach for the comparison of bacterial communities.
3.Differentiation of xanthomonads causing the bacterial leaf spot of poinsettia in China from the pathotype strain of Xanthomonas axonopodis pv. poinsettiicola.

In October 2003, a new bacterial disease with symptoms similar to those caused by Xanthomonas axonopodis pv. poinsettiicola was observed on poinsettia leaves at a flower nursery in Zhejiang Province of China. Three Xanthomonas strains were isolated from infected plants and classified as X. axonopodis. They were differentiated from the pathotype strain LMG849 of X. axonopodis pv. poinsettiicola causing bacterial leaf spot of poinsettia by comparison of pathogenicity, substrate utilization and BOX-PCR genomic fingerprints.

4.Effect of Ginsenoside Rg1 on Apoptosis after Spinal Cord Ischemia-reperfusion Injury in Rats

Objective To investigate the effect of ginsenoside Rg1 on apoptosis after spinal cord ischemia-reperfusion injury (SCII) in rats. Methods Forty-eight adult Sprague-Dawley rats were randomly divided into sham group (n=8), ischemia group (n=8), ischemia-reper-fusion group (n=16) and drug group (n=16). Fogarty catheter was put in the thoracic aorta of the rats and the blood flow wasn't blocked in the sham group. The rats in the ischemia group were sacrificed 30 minutes after spinal cord ischemia. The drug group was injected with gin-senoside Rg1 30 mg/kg 30 minutes before and after SCII. The same volume of normal saline was injected in the ischemia-reperfusion group at the same time. The expression of Bcl-2 and survivin was detected with immunohistochemistry at six hours, 24 hours after reperfusion in the ischemia-reperfusion group and drug group, 30 minutes after ischemia in the ischemia group and in the sham group. The change of cells was observed in each group with HE staining. Results The cells were damaged in the ischemia group, the ischemia-reperfusion group and the drug group, in which the drug group was better than the other groups. The expression of survivin and Bcl-2 was higher in the ischemia group, the ischemia-reperfusion group and the drug group than in the sham group (t>3.896, P<0.01), and were significantly higher six hours and 24 hours after reperfusion in the drug group than in the reperfusion group (t>6.693, P<0.001). Conclusion Ginsenoside Rg1 can reduce the neurons damage and increase the expression of the Bcl-2 and survivin, that inhibit cells apoptosis after SCII in rats.
5.Preliminary application of the sentinel lymph node detection in early cervical cancer

Purpose:To assess the value of the sentinel lymph node by lymphoscintigraphy and gamma detector probe in early cervical cancer.Methods:22 patients with operable invasive early cervical cancer and clinically proven negative pelvic lymph nodes were included in the study. The 99m Tc-dextran ( 99m Tc-DX) of 74 MBq(2 mCi) was injected into the cervix at 2? and 10?. Lymphoscintigraphy and gamma probe detectors were used to detect the SLN. Results:The SLN was identified in 17 of the 22 patients. The sensitivity and the specificity of the SLN detection to predict the metastasis of the pelvic lymph node was 100% and 100% respectively. Conclusions:Identification of sentinel nodes using radionuclide is feasible and possible in women with early cervical cancer.
6.Culture of human retinal capillary endothelial cells and establishment of the two-dimensional vascular model

Objective To cultivate human retinal capillary endothelial cells (HRECs) and establish two-dimensional model of human retinal vessels in vitro. Methods In a fibronectin-coated raising pound, HRECs were cultured by non-serum human-endothelial-cells substrate and two-dimensional model of human retinal vessels was established. Horseradish peroxidase was used to detect the permeability. Some of the vascular models were cultivated with 5 ng/ml vascular endothelial growth factor (VEGF), whose changes of permeability was compared with which of the models without cultivation with VEGF. The effect of VEGF on vascular permeability was observed. Results Meshy vascular structure came into being due to the confluent HRECs after 2 to 4 days. Comparatively complete two-dimensional vascular model after about 6 days. VEGF increased vascular permeability and promoted the formation of blood vessels. Conclusion HRECs can be cultivated successfully with human-endothelial-cells substrate; standard retinal two-dimensional vascular model in vitro can be established by using cellular raising pound and non-serum human-endothelial-cells substrate.
7.Features and relevant influencing factor of ecological executive function in children with childhood absence epilepsy

Objective:To explore features and relevant influencing factor of ecological executive function in children with childhood absence epilepsy (CAE).Methods:Forty children with CAE (CAE group) and 40 healthy children with physical examination (control group) from April 2017 to July 2020 in Anhui Provincial Children′s Hospital were selected. The behavior rating inventory of executive function (BRIEF) parental questionnaire was used to evaluate the executive function of children. The differences of ecological executive function between 2 groups were compared.Results:The BRIEF total score, behavioral regulation index (BRI) and metacognition index (MI) in CAE group were significantly higher than those in control group: (52.03 ± 10.89) scores vs. (44.05±5.06) scores, (49.45 ± 9.93) scores vs. (43.85 ± 4.70) scores and (53.18 ± 11.24) scores vs. (44.95 ± 5.32) scores, and there were statistical differences ( P<0.01). Multiple stepwise regression analysis result showed that inhibit inhibition, shift, emotional control, initiation, working memory, planning, organization, monitoring, MI and total score were correlated with disease control ( P<0.01), and disease control had a negative predictive effect on them ( R2 = 0.174, 0.158, 0.234, 0.325, 0.383, 0.337, 0.378, 0.199, 0.463 and 0.435); BRI was correlated with onset age and disease control ( P<0.01 or <0.05), and onset age and disease control had a negative predictive effect on BRI ( R2 = 0.336). Conclusions:Children with CAE have ecological executive dysfunction. The control of the disease and the onset age are the main factors affecting the ecological executive function.
8.Interventional therapy of complications after liver transplantation:hepatic artery stricture

Hepatic artery stricture (HAS) after liver transplantation can lead directly to transplanted liver function exhaustion and complications of biliary system. The early diagnosis and treatment are crucial for better prognosis. Doppler ultrasound is the first method of choice, and angiography can give further clear dignosis. The balloon dilatation is still effective for hepatic arterial stenosis. With the more adaptable usage of oronary stent, if possible, would reveal more promising result especially for tortuous stenotic hepatic artery. The vascular reconstruction or repeated liver transplantation is still the effective therapeutic methods.
9.Subfoveal choroidal thickness measured by Cirrus HD optical coherence tomography in myopia

To measure the subfoveal choroidal thickness ( SFCT ) in myopia using Cirrus HD optical coherence tomography ( OCT ), and to explore the relationship between the SFCT, axial length and myopic refractive spherical equivalent.
● METHODS: One - hundred thirty - three eyes of 70 healthy volunteers were recruited, and were divided into emmetropia group, low- degree myopia, middle- degree myopia and high - degree myopia group. SFCT were measured by Cirrus HD OCT, and the relationship between the SFCT, axial length and myopic refractive spherical equivalent were evaluated.
●RESULTS: 1) Average SFCT was (275. 91±55. 74)μ m in normals, that in emmetropia group, low-degree myopia, middle- degree myopia and high - degree myopia group were (290. 03± 34. 82) μ m, (287. 64 ± 51. 51) μ m, (274. 95 ± 56. 83)μ m, (248. 37 ± 67. 98) μ m; 2) the SFCT of high -degree myopia group was significant thinner than that of emmetropia group (P< 0. 05); 3) There was a significant negatives correlation between the SFCT and axial length (P<0. 01), a significant positive correlation between SFCT and myopic refractive spherical equivalent (P<0. 01).
● CONCLUSlON: the SFCT is inversely correlated with increasing axial length and myopic refractive error.
10.Effect of isoflurane preconditioning on cell apoptosis during renal ischemia-reperfusion in rats

Objective To evaluate the effect of isoflurane preconditioning on cell apoptosis during renal ischemia-reperfusion (I/R) in rats.Methods Thirty male Sprague-Dawley rats, aged 12-14 weeks, weighing 300-320 g, were randomly divided into 3 groups (n =10 each) using a random number table: sham operation group (group S);I/R group;isoflurane preconditioning plus I/R group (group Iso+I/R).The rats were anesthetized with intraperitoneal pentobarbital sodium 30 mg/kg.To establish the model of renal I/R injury, the right kidney was removed, and the left renal pedicle was occluded for 30 min with atraumatic mini-clamp for 30 min, followed by 2 h reperfusion.In Iso+I/R group, 1.5% isoflurane was inhaled for 1 h, followed by 30 min of washout before I/R.In S and I/R groups only oxygen 2 L/min was inhaled for 1 h.Arterial blood samples were taken at 2 h of reperfusion to determine the concentrations of serum creatinine (Cr), blood urea nitrogen (BUN) and cystatin C (CysC).The animals were then sacrificed, and left kidneys were sampled for determination of the cell apoptosis (by TUNEL), expression of p53, Bax and Bcl-2 mRNA (using real-time fluorescent quantitative PCR), and expression of Bax, Bcl-2, and caspase-3 (by Western blot analysis).The Bcl-2/Bax ratio was calculated.Results Compared with group S, the serum Cr, BUN, and CysC concentrations were significantly increased, the Bcl-2/Bax ratio was decreased, Bcl-2 mRNA, Bax mRNA, Bcl-2, Bax and caspase-3 expression was up-regulated, and AI was increased in group I/R.Compared with group I/R, the serum Cr, BUN, and CysC concentrations were significantly decreased, the Bcl-2/Bax ratio was increased, Bcl-2 mRNA, Bax mRNA, Bax and caspase-3 expression was down-regulated, and AI was decreased in group Iso+I/R.There was no significant difference in p53 mRNA expression among the three groups.Conclusion Regulating the balance between Bcl-2 and Bax and inhibiting apoptosis in kidney cells are involved in the mechanism by which isoflurane preconditioning reduces renal I/R injury in rats.
