1.The clinical analysis of 12 patients with thrombotic Microangiopathy

Objective:To study the clinical characteristics and treatment of thrombotic microangiopathy(TMA),to improve recognition of TMA.Method:Types,clinical and laboratory characteristics,treatment and prognoses of 12 patients with thrombotic microangiopathy admitted to our hospital between October 2004 and December 2007 were analyzed retrospective1y.Result:The clinical manifestations were characterized by microangiopathic hemolytic anemia(12/12),thrombocytopenia(12/12),elevated serum LDH value(12/12),and renal lesion(12/12),fever(5/12),and neurological abnormalities or mental disorders(7/12).3 patients were treated by plasma exchange therapy.1 patient fully recovered,4 patients took a turn for the better,and 7 patients died.Conclusion:TMA is a kind of severe acute syndrome with complicated pathenogenesis and high mortality.Management should depend on the type of TMA in order to improve curative efficacy and reduce mortality.
2.Effects of anti-human augmenter of liver regeneration monoclonal antibody on proliferation of HepG2 cells in vitro

Objective:To detect the existence of expression of human augmenter of liver regeneration(hALR) in HepG2 cells,and to observe proliferation of HepG2 cells after neutralizing hALR with anti-hALR McAb.Methods:the expression of hALR in HepG2 cells was observed immunocytochemistry and reverse transcriptive PCR(RT-PCR).Proliferation of HepG2 cells after neutralization was detected by ~3?H-TdR incorporation approach.Results:① hALR was expressed by HepG2 cells.② Anti-hALR McAb inhibited partially the autonomous growth of HepG2 cells.Conclusion:Anti-hALR McAb inhibit the autonomous growth of hepatoma cells partially;moreover,hALR maintain the autonomous growth of hepatoma cells in vitro through autocrine mechanism.

4.Analysis of causes of extrauterine growth restriction in premature infants and the status of nutritional ;intake

Extrauterine growth restriction (EUGR) in premature infant is a common problem in the world. The reasons for high EUGR rate, such as declining gestational age and birth weight, getting more medical treatment and examination or interruption of nutritional support, had been profoundly studied. However, there are few reports about the limited ability of intakes of energy and various nutrients and the updating of the growth standard curve . Research suggested that the average time taken to reach the recommended fat intake in preterm infants is proximately day 6 after birth, protein day 4 after birth, carbohydrate one-week after birth. Vitamins and trace elements are usually not able to or take a long time to reach the recommended nutrient intake. Without enough intake of energy and nutrients to maintain the need of intrauterine growth rate and catch-up growth in preterm infants, EUGR cannot be improved. In this paper, the EUGR and the intake of important nutrients, such as energy, fat, protein, vitamins and minerals, etc., during hospitalization were reviewed in hope to achieve more rational and standardized management for preterm infant,and provide more reasonable advise to control EUGR.
5.Effect of mesenchymal stem cell in transplantation

Mesenchymal stem cell(MSC) is a kind of tissue stem cell having a characteristics of high self-renewal and multipotency. It is tended to be separated in vitro and play an important role in hematopoietic stem cell transplantation and transplantation of other systems. This article described mesenchymal stem cell's function and mechanism on supporting hematopoiesis, enhancing implantation,adjusting immunity and alleviating graft versus host disease (GVHD) in transplantation and also give some advices and experties about its further application.
6.Apoptosis of MDA-MB-231 cells induced by berberine alpha-hydroxy beta-decanoylethyl sulfonate.

To investigate the cell proliferation inhibition and apoptosis induced by berberine a-hydroxy f-decanoylethyl sulfonate (HB) on MDA-MB-231 cells in vitro, and the inhibitory effect of HB on the expression of poly adenosine diphosphate RNA polymerase (PARP), MTT assay was used to detect the viability of MDA-MB-231 cells and cell cycle was examined by flow cytometry. The results showed that HB could significantly inhibit the proliferation of MDA-MB-231 cells, and mildly arrested cell cycle progression at S phase. The IC50S for 24, 48 and 72 h treatment were 4.65, 1.46 and 0.75 mg.L-1 (7.55, 2.37 and 1.22 micromol.L-1), respectively. Annexin V-FITC/PI double staining assay showed that HB increased apoptotic ratio of MDA-MB-231 cells. Western blotting analysis showed the expressions of procaspase-3, procaspase-9 and PARP were decreased after HB treatment, while their fragment increased. The results suggest that HB can inhibit the growth and induce apoptosis of MDA-MB-231 cells, which may be associated with inhibition of the expression of procaspase-3, procaspase-9 and PARP.
7.STYK1 down-regulation suppresses colorectal cancer cell growth and invasion in vitro

Objective To determine the effects of down-regulating serine/threonine/tyrosine kinase 1 (STYK1) on the proliferation,apoptosis and invasion and on MEK/ERK and PI3K/AKT signaling pathways in human colorectal cancer cell line HCT-15.Methods The expression of STYK1 protein was downregulated by RNA interference technique in HCT-15 cells.Then,the STYK1 protein expression was detected by Western blotting.CCK-8 assay was used to measure the effect of STYK1 down-regulation on the proliferation of the cells,flow cytometry was employed to detect the cell apoptosis,and Transwell chamber assay was adopted for cell invasion ability.The expression levels of p-ERK1/2 and p-AKT were detected by Western blotting.Results Compared with the siRNA NC group,STYK1 down-regulation significantly inhibited the proliferation (P < 0.01) and invasion (P < 0.01),induced cell apoptosis (P < 0.01),and suppressed the expression levels of p-ERK1/2 and p-AKT in the HCT cells transfected with STYK1 siRNA (P <0.01).Conclusion STYK1 down-regulation inhibits the proliferation and invasion and induces the apoptosis in colorectal cancer cells,which may be related to its inhibition on the MEK/ERK and PI3K/AKT signaling pathways.
8.Rational Drug-use and Modernization of Medical and Health Work

Rational drug-use is an extremely important component part in morden medical and health work. And irrational drug-use will aggravate drug misadventure, increase overpreseribing, waste medicinal resources and harm the reputation of medical workers. The tendency of polypharmacy just for more hospital income, regardless of drug-related adverse effects, comes to be a serious problem and hard to control in recent years. No perfect management system and lack of information may be the major factors of the problem. In order to overcome the irrational drug-use characterized by polypharmacy, we should learn something from the Diagnosis Related Groups/Prepayment System (DRGS/PPS). Now, under the cooperation of The Chinese Pharmacological Society ,The Chinese Pharmaceutical Association and The Public Health Ministry of PLA, a Diagnosis Related Rational Drug-use Management System and a Computerized/Telecommunication Based Dynamic Knowledge Bank are able to be developed. We consider that it will contribute to the modernization of medical and health work in China in the near future.
9.Effects of LPS, IL-1? and IFN-? on morphological and functional changes of cultured human aortic endothelial cells

The significant changes in morphology and nitric oxide synthase (NOS) activity in cultured human aortic endothelial cells (HAEC) were observed when these cells were exposed to LPS, IL-1? and IFN-?. NOS activity increased at 6h of treatment, the time point that HAEC began to change and elongate. The cell shape became fibroblast like and the length to width ratio of the cells increased 2 times of that of the control at 20h treatment, the time point that NOS activity returned to control level.
10.Determination of Prodatin in Polygonum Cuspidatum Injection With HPLC

OBJECTIVE:To establish a HPLC method for determination of prodatin in Polygonum cuspidatum injection.M_ETHODS:The analytical column was Hypersil ODS column(100mm?2.1mm,5?m).The mobile phase consisted of methanol-water(25∶75).The flow rate was 1.0ml/min.The UV detection wavelength was 290nm.RESULTS:The linear range was 0.145~2.32?g/ml(r=0.9 999).The average recovery of prodatin was 98.8%(RSD=1.15%).CONCLUSION:The method is convenient,accurate and could be used for quality control in the production of Polygonum cuspidatum injection.
