1.STUDIES OF APOPTOSIS INDUCED IN MALIGNANT LYMPHOMA CELL LINES BY ARSENIC TRIOXIDE

Objective To investigate the effects of a wide range of concentrations of As2O3 malignant lymphoma cell lines. Methods Cell apoptosis situation ,cellular DNA contents and bcl-2 protein expression were determined by cell stain, TdT-mediated dUTP Nick-end Labeling (TUNEL) and flow cytometry. Results 0.5 μmol/L~2.0 μmol/L As2O3 could inhibit Raji cell growth accompanied by the appearance of morphologic characteristics of apoptosis. Number of sub-G1 cells significantly increased and bcl-2 protein expression in Raji cells was significantly down-regulated assayed by flow cytometry. The relationship between bcl-2 protein and the concentrations of As2O3 showed a dose-and time-dependent effect; so did the number of sub-G1 cells. However, there was no effect of 0.5 μmol/L~4.0 μmol/L As2O3 on Jurkat cell.Conclusion It is found that As2O3 could significantly inhibit the growth of B lymphoma cell lines and induce their apoptosis.
2.Relationship between abnormal activation of HGF/c-Met signaling pathway and EGFR-TKI acquired resistance in non-small cell lung cancer

Targeted therapy such as the epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI)has made huge progress in treatment of non-small cell lung cancer (NSCLC).However,the emergence of acquired drug resistance is an inevitable result of the targeted therapy.The hepatocyte growth fac-tor/c-mesenchymal-epithelial transition factor (HGF /c-Met)signaling pathway participates in cell formation, migration,angiogenesis and other important cellular processes of multiple tumors.The abnormal activation of this signaling pathway plays the pivotal role in the acquired resistance to EGFR-TKI.Recently,some clinic tri-als prove that HGF /c-Met inhibitors can make clinical benefit of some NSCLC patients with acquired drug re-sistance of EGFR-TKI.
3.Difference in dental arch form between Angle Ⅱ division 1 and normal occlusion assessed by Euclidean distance matrix analysis

Objective: To evaluate differences in dental arch forms between the subjects with Angle II division 1(AngleⅡ1 ) and those with normal occlusion. Methods:60 AngleⅡ1 patients and 60 normal occlusion subjects aged 13-17 years old were selected. Fourteen landmarks, corresponding to the cusp tips and incisor edges, were identified on the upper and lower dental casts of all subjects by YM-21115 three dimension measuring machine. Arch form differences between AngleⅡ1 and normal occlusion subjects were assessed by Euclidean distance matrix analysis (EDMA). 4.Analysis of missed detection in gonococci culture of male urethral secretions

Objective:
To investigate the undertected situation in gonococci culture of male urethral secretions.
Methods:
The 362 cases of male ureethral secreations were simultaneously detected with fluorescence quantitative PCR, germiculture and direct smear method. The results in three detections were statistically analyzed.
Results:
The fluorescence quantitative PCR positive rate was 29.56%, near to direct smear method (26.52%)(x2=0.83,P>0.05); the germiculture positive rate(4.42%) was significantly lower than the positive rates of fluorescence quantitative PCR(x2=81.10,P<0.01) and direct smear method(x2=67.60,P<0.01).
Conclusion
The simple gonococci culture has high missed detection rate(>80%) in detecting male cases. Ensuring the quality of samples before analysis and detecting with two methods or above are the effective measures to improve the gonococci detection rate.
5.Effect of Iodine Deficiency and Iodine Excess on Antioxidation of Rat Thyroid

Objective To study the effect of different iodine intake on the antioxidation of rat thyroid. Methods The rats were divided into 6 groups, low iodide (LI), normal iodine (NI), high iodide including five fold (5HI), ten fold (10HI), fifty fold (50HI), one hundred fold (100HI) and given potassium iodide (KI) at different dosages through food respectively. After 6 and 12 months of treatment, the rats were sacrificed respectively and the activity of glutathione peroxidase (GPx), superoxide dismutase (SOD), the content of malondialdehyde (MDA) in the thyroid were determined. Results The activity of GPx , SOD and the MDA content in LI group were significantly higher than those in NI group. There was no difference in the SOD and GPx activity and MDA content among NI, 5HI and 10HI groups. The MDA content in 50HI group was lower than that in NI group after treated for 12 months, but no difference was found between them after treated for 6 months. Compared with NI group, the GPx activity, SOD activity and MDA content in 100HI group increased after 6 months of treatment, however, decreased after 12 months of treatment. Conclusion Low iodine intake can induce oxidative damage of the thyroid gland in normal rats, high iodine (100 fold) intake for a long period (12 months) may decrease the activity of anti-oxidases in thyroid without obvious oxidative damage, that shows the anti-oxidative system of rat thyroid has a tolerance to high iodine intake.
6.Diagnostic value of string beads sign in peripheral small cell lung cancer

Objective To investigate the formation mechanism of string beads sign in peripheral small cell lung cancer (SCLC) and evaluate the significance of it in differential diagnosis among SCLC,peripheral lung adenocarcinoma and peripheral lung squa-mous cell carcinoma.Methods 78 cases of SCLC,69 cases of peripheral lung adenocarcinoma and 33 cases of peripheral lung squa-mous cell carcinoma,confirmed pathologically were included in this study.The positive rates of string beads sign,mediastinal lymph node metastasis and mediastinal lymph nodes larger than primary lung lesions were calculated and analyzed in these three groups.Results 10 out of SCLC cases (12.8%)have string beads sign,in which all mediastinal lymph nodes were larger than lung lesions.Mediasti-nal lymph node metastases were observed in 63(80.8%)of 78 cases,and 42 (53.8%)cases had larger mediastinal lymph nodes than lung lesions.No string beads sign was observed in patients with peripheral solid lung adenocarcinomas,but 25 of 69 cases (36.2%) have mediastinal lymph node metastasis and 2 cases (2.9%)had larger mediastinal lymph nodes than lung lesions.13 cases(39.4%) of 33 patients with peripheral lung squamous cell carcinomas had mediastinal lymph node metastasis,and 6 cases (16.7%)had larger mediastinal lymph nodes than lung lesions.The statistical results showed the positive rate of string beads sign was not significantly different between peripheral SCLC group and peripheral lung squamous cell carcinoma group,but that of mediastinal lymph node and larger mediastinal lymph nodes than lung lesions were statistically different among these three groups.Conclusion To some extent, string beads sign on CT could reflect the biologic character of SCLC.It played an important role in differential diagnosis of peripheral SCLC,peripheral lung adenocarcinoma and periph-eral lung squamous cell carcinoma,but it should be combined with mediastinal lymph node size.
7.Clinical study of Bryan prosthetic disc in the treatment of ossification of posterior longitudinal ligament of cervical spine

[Objective] To observe the clinical effect of Bryan cervical prosthetic disc in treating ossification of posterior longitudinal ligament(OPLL)of cervical spine.[Method]From October 2005 to June 2007,15 localized OPLL patients were treated surgically by cervical arthroplasty using Bryan disc prosthesis.JOA score and radiological study were performed before and after operation.The range of movement(ROM)of replaced level were investigated postoperatively.[Result]Neurological deficit was alleviated in all patients.All patients were followed up at least 6 months,JOA score increased from an average of 8.5 to 15.8.Replaced segment restored the normal ROM in flexion and extension.[Conclusion]Cervical arthroplasty using Bryan disc prosthesis can achieve good clinical outcome in treating localized OPLL of cervical spine;and the normal ROM can be retained in the replaced level.
8.Experimental study of motor and somatosensory evoked potentials on spinal cord injury

[Objective]To evaluate the validity and feasibility of somatosensory evoked potentials(SEP) and transcranial magnetic stimulation motor evoked potential(TMS-MEP) in early decompression of spinal cord injury(SCI). [Method]Thirty-two rabbits fell into four groups randomly. The static load model were used to simulate human SCI. According to the compression time, the animals were divided into 4 groups (control,5min, 15min,30min). In this study, somatosensory evoked potentials (SEP) and transcranial magnetic stimulation motor evoked potential (TMS-MEP) were used to assess neurologic function. The EPs were recorded at 5min, 15min, 30min, 60min, 6 hours, 24 hours, 3 days, 7 days post-decompression and before spinal cord injury in injury groups. Latency and amplitude were measured for each wave. Tarlov behaviour score was performed before surgery and after surgery at 1,3,7 days. Histologic studies were performed on part of animals. [Result]Spinal cord compression resulted in a gradual increment in the peak latency and significant decrement in the peak amplitude. After decompression, the recovery of the latency was more obvious than the amplitude in all injury groups, EPs recovered earlier than Tarlov score. [Conclusion]EPs may evaluate the neurologic function of SCI models sensitively and impartially. Dcompression early may promote SCI recovery.
9.Effect analysis of fertility preserving surgery for the treatment of primary ovarian mucinous carcinoma

Objective To analyze the clinical value of ovarian surgery in the treatment of primary ovarian mucinous carcinoma patients with fertility requirements.Methods 51 cases with fertility requirements with primary ovarian mucinous carcinoma who received surgical treatment were analyzed.The postoperative serum follicle stimulating hormone (FSH) and luteinizing hormone (LH),estradiol (E-2),anti Mullerian hormone (AMH) were deteced.The patients were followed up for 3 years,the survival rate and fertility were observed.Results Of 51 patients underwent surgery,postoperative 7d,14d,the FSH,LH levels in serum were higher than before operation (F=24.198,21.554,all P<0.05),E-2,AMH,PSV values were lower than before operation (F=24.407,35.028,11.064,all P<0.05).During the follow-up period to June 2015,there were 4 patients died of tumor recurrence,2 patients were lost to follow-up,45 patients survived,the survival rate was 88.23%.Of 45 patients survived,41 patients were normal menstrual cycle,menstruation,menstrual cycle 26-31 d,average (28.2±2.2)d;4 patients with irregular menstruation.Married women in the follow-up of 44 cases,including 28 patients with successful pregnancy,the pregnancy rate was 63.64%,there were 4 cases of spontaneous abortion,pregnancy in 3 patients because of abortion,pregnancy examination found 17 cases of abnormal pregnancy,4 cases of premature delivery.Conclusion In patients with primary ovarian mucinous carcinoma,it is feasible to use fertility preserving function,and the pregnancy rate is high,but the follow-up and monitoring should be strengthened to avoid the bad prognosis.
10.Expressions of estrogen receptor, progesterone receptor, human epidermal growth factor receptor 2 and relationships of these expressions with clinical pathology in breast cancer patients aged 70 years old and over

Objective To investigate the expressions of estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER-2) and relationships of these expressions with the clinicopathology in breast cancer patients aged 70 years old and over. Methods The expressions of ER, PR and HER-2 were detected by immunohistochemical staining from 124 elderly breast cancer patients, and the relationships of these expressions with pathological classification, tumor size, lymph node metastasis and TNM stage were analyzed. Results Among 124 patients, the positive expression rate of ER and PR was 75.0% (93/124) and 71.0% (88/124),respectively. They were not associated with pathological classification, tumor size, lymph node metastasis or TNM stage (all P>0. 05). The positive expression rate of Her-2(+++) and Her-2 (++) was 11.3% (14/124) and 24.2% (30/124), respectively. And the expression of Her-2 was significantly associated with tumor size, lymph node metastasis and TNM stage (P<0. 05) but not with pathological classification (P>0. 05). Conclusions The positive expressions of ER and PR are not correlated with the clinicopathology in elderly breast cancer patients. Her-2 overexpression is positively associated with tumor size, lymph node metastasis and TNM stage.
