1.Dynamic changes of osteopontin in the guinea pig model of cholesterol gallstone disease

Objective To investigate the dynamic changes of osteopontin (OPN) in the guinea pig model of cholesterol gallstone disease.Methods Forty-four guinea pigs were randomly assigned to cholesterol gallstone group and control group.The animals were sequentially killed on Day 7,14,28,42,56 and 70 after operation.The expressions of OPN mRNA in gallbladder and liver tissues were detected by real-time PCR.The changes of OPN,mucin,α1-acid glycoprotein (AAP) and apolipoprotein A1 (APOA1) in bile and blood plasma were detected by ELISA kits.Results The expression of OPN mRNA in gallbladder and liver tissues increased gradually and reached the peak in the 6th week,and then decreased.The increased expression of OPN in bile in the gallstone group started from the 1st week and reached the peak in the 6th week (P < 0.05),which gradually decreased to the baseline in the 10th week (P > 0.05).The expressions of OPN in bile and blood demonstrated similar trends,while the peak time in blood samples was much earlier (4th week).The changes of APOA1 in bile and blood were similar to OPN,although there was no advanced peak value in blood.The levels of mucin and AAP in bile and blood increased after operation,and were kept at high level throughout the study.Conclusions OPN is involved in the whole process of cholesterol gallstone formation,which may be associated with other nucleation-active factors.
2.Study on cytomegalovirus gene mutations conferring resistance to ganciclovir in heart and lung transplant patients

Objective To establish a rapid and convenient assay to screen for emerging ganciclovir(GCV) resistant mutations in the UL97 gene of human cytomegalovirus (HCMV) from transplant recipients. Methods Thirteen spefcific oligonucleotide probes were designed according to the target sequence encoding HCMV-UL97 gene. A nested polymerase chain reaction (PCR) amplifying UL97 gene was employed. Line probes assay (LiPA) was configured to detect relevant non-synonymous mutations at codon 460,520,594,595,603, and 607. In parallel, nested-PCR amplicons were subsequently sequenced directly. Results Four of 16 heart and lung transplant recipients had been infected with HCMV resistant strains during GCV therapy. The mutant codons of UL97 gene were at codon 520、595 and 603. All amplicons detected by LiPA were fully concordant with the nucleotide sequenced.Conclusions Long-term prophylaxis and treatment with GCV in heart and lung transplant recipients may develop HCMV resistant strains. HCMV-LiPA proves to be an alternative method for the resistance genotype analysis of HCMV.

Ranibizumab is a recombinant rat monoclonal antibody fragment of the second generation humanized VEGF. Ranibi-zumab can bind all of active VEGF-A to inhibit the bond between VEGF-A and VEGFR1 or VEGFR2. As the results, the vascular en-dothelial proliferation and vascular permeability can be reduced, and the new vessels angiogenesis can be prevented. The article sys-tematically searched and summarized the relative literatures to offer high-quality evidences for the recent clinical application of ranibi-zumab in ophthalmology.
4.Protocol of safety guidelines for workplace violence on health care sector using Delphi method

Objective To recommend the safety guidelines for workplace violence on health care sector according to the incidents of violence status on medical workplace.Methods A pilot study was conducted using a two-round Delphi method to study out the safety guidelines for hospital violence.Results In two subsequent rounds,the group discussed and screened out 50 entries from 51 items in the six modules as safety guidelines for hospital violence.Conclusions Establishment of safety guidelines for hospital violence on health care sector using Delphi method requires further clinical validation.
5.Study of the patients with vertebral artery insufficiency syndrome by use of TCD before and after external counterpulsation and traction therapy

Objective To investigate the value of external counterpulsation and traction therapy on patients with vertebral artery insufficiency syndrome by use of transcranial Doppler (TCD). Methods Based on the criteria of clinical diagnosis, 86 patients with vertebral artery insufficiency syndrome and 60 healthy persons were examined by EME 4004 type TCD. The peak velocity (Vp) and mean velocity (Vm) of blood flow of the vertebral artery (VA) and basilar artery (BA) were detected and analyzed before and after rehabilitation treatment. Results The blood flow velocities of the vertebrobasilar artery (VBA) of the patient group were much lower than that of the control group ( P 0.05). Conclusion TCD examination while rotating neck could provide valuable information for the diagnosis and rehabilitation assessment of patients with vertebral artery insufficiency syndrome. External counterpulsation and traction therapy are effective in the treatment of the vertebral artery insufficiency syndrome.
6.Comparison of SRAP and RAPD markers for genetic analysis of plants in Dendrobium Sw.

Objective This study was carried out in order to analyze genetic diversity in plants of Dendrobium Sw. and compare the marker index (MI) between SRAP and RAPD markers. Methods Using 40 SRAP primer combinations and 36 RAPD primers,the genetic diversity of the materials in Dendrobium Sw. was amplified. Results A total of 1 977 loci were generated by 40 SRAP primer combinations,of which 90.2% was polymorphic and the genetic similarity (GS) ranged from 0.330 2 to 0.789 2. As for RAPD,281 bands were obtained,in which 87.1% was polymorphic,and the GS was from 0.494 2 to 0.773 1. There were some differences existing in the two cluster results revealed by SRAP and RAPD markers,they were as follows:compared to that of RAPD,the result of SRAP could more comfortably reflect the kinship and geographical origin,simultaneously conform to the traditional classification. For the experiment materials,SRAP (16.46) got an MI value 4.5 folds higher than that of RAPD (3.67),which meant SRAP markers were more effective in detecting genomic polymorphisms in the plants of Dendrobium Sw. Conclusion These results show that SRAP and RAPD could be applied to detecting genetic diversity analysis of plants in Dendrobium Sw. SRAP Also shows great superiority and advantage in this study.
7.Effect of monoclonal antibody to CD47 molecule on dendritic cell differentiation and function

AIM: To explore the influence of CD47 molecules on the maturation and function of cultured dendritic cells (DCs). METHODS: Monocyte cell-derived DCs were propagated in granulocyte-macrophage colony-stimulating factor (GM-CSF) and lipopolysaccharide (LPS) plus interleukin (IL)-4, in the presence or absence of anti CD47 monoclonal antibodies (anti-CD47 mAbs). 8.An Experimental Study of the Protective Effects of Rosiglitazone on Lung Injury in Severe Acute Pancreatitis

Objective To investigate the protective effects and mechanisms of rosiglitazone on lung injury in severe acute pancreatitis (SAP). Methods Seventy-two SD rats were randomized into three groups: sham operation (SO) group, SAP group and rosiglitazone-pretreated group. The model of SAP was induced by retrograde injection of 5%sodium tauro-cholate into the bili-pancreatic duct in SD rats. Rats of rosiglitazone-pretreated group were given 10 mg/kg rosiglitazone in-traperitoneally 30 min before inducing SAP. The amylase plasma levels, levels of TNF-αand p(O2), the myeloperoxidase (MPO) and the wet/dry ratio of lung tissue were measured. The expression of NF-κB in pulmonary tissues was assayed by im-munohistochemistry. The expressions of TNF-αmRNA and intercellular adhesion molecule 1 (ICAM-1) mRNA in pulmo-nary tissues were detected by reverse transcript PCR (RT-PCR). The histopathological changes of pulmonary tissues were evaluated. Results Compared with SO group, the plasma levels of amylase and TNF-α, the intrapulmonary MPO were sig-nificantly increased in SAP group (P<0.05). The expression levels of NF-κB, TNF-αmRNA and ICAM-1 mRNA in lung tissue were also significantly increased in SAP group (P<0.01). There were significantly lower levels of the above indicator in rosiglitazone-pretreated group than those of SAP group (P<0.05). Compared with SO group, the lung wet/dryratio was sig-nificantly higher at 6 h and 12 h in SAP group (P<0.05). The values of lung wet/dry ratio were significantly lower at 6 h and 12 h in rosiglitazone-pretreated group than those of SAP group (P<0.05). The lung tissue damage was significantly reduced. The level of p(O2)was significantly decreased in SAP group than that of SO group (P<0.05). The level of p(O2)was signifi-cantly increased at 6 h and 12 h in rosiglitazone pretreated group than that of SAP group (P<0.05). Conclusion Rosigli-tazone might have protective effects on SAP-related lung injury by inhibiting NF-κB and decreasing the production of TNF-αand ICAM-1in the lung tissues.
9.Epidemiology and characteristics of acute poisoning treated at an emergency center

BACKGROUND:Acute poisoning is frequently encountered at emergency department. This study was to investigate the epidemiology and characteristics of patients with acute poisoning who were treated at the Emergency Center, Fujian Provincial Hospital, China. METHODS:We retrospectively analyzed the gender, age, causes of poisoning, types of poisons, poisoning route, emergency diagnoses, outcomes, and prognoses of these patients. RESULTS:Altogether 2867 patients with acute poisoning were treated from January 2004 to December 2009. The ratio of male to female was 1:1.04, and their average age was 33.8 years. Of the 2867 patients, 76.39% were between 18 and 40 years old. The incidence of acute poisoning was as high as 11.33% in January each year. The incidence of poisoning was in a descending order:alcohol poisoning (54.55%), medication poisoning (25.95%), pesticide poisoning (5.65%), and drug poisoning (4.88%). Most (56.44%) of the patients with drug poisoning were under 25 years and their mean age was significantly lower than that of patients with medication poisoning or alcohol poisoning (P < 0.01). Approximately 69.54% of the patients were followed up after emergency treatment, 30.39% were hospitalized, and four patients died. CONCLUSIONS:Acute poisoning is largely alcohol poisoning and medication poisoning in a city. The emergency green channel "pre-hospital emergency care-emergency department-hospital treatment"can significantly improve the survival rate of patients with acute poisoning.
10.Study on traceability system of genuine medicinal materials.

Genuine medicinal materials with special characteristics of Traditional Chinese Medicine (TCM), is recognized as high quality medicine. Both ancient records and modern research considered that the origin is an important reason for the formation of genuine medicinal materials. However, blindly transplanting of genuine medicinal materials has led to the quality decline and counterfeit medicines appeared in production or sale progress, which may increase the risk of accidents in TCM. Frequent accidents emerged in Chinese herbal affects its export. What's more, it is a great threat to the medication safety in TCM clinical. There is an urgent need to implement traceability systems of TCM, which could provide convenient information record and traceability of TCM circulation. This paper reviews a variety of technical methods for genuine medicinal materials traceability, and proposed the establishment of genuine medicinal materials traceability system based on two-dimensional code and network database.
