Chinese Health Economics 2014;(1):26-28
With the intensification of population floating in China, there has been a growing demand for ecdemic medical care. Because basic medical insurance in China conduct localized management, the differentiation between reimbursement policy and level in different areas result in the difficulty of immediate settlement. The great quantity of floating population are faced with rather heavy personal burden when need medical care at different locations, which to some extent restrict in-time healthcare for floating population. On the basis of analyzing the features of endemic medical care for floating population, it described three mode of typical immediate settlement on ecdemic medical care, discussed the main problems in restricting the flowing population get immediate settlement at different locations and put forward suggestions on improving the immediate settlement at different locations.
4.Improvement of the Optimum Temperature of Penicillium expansum Lipase by Site-directed Mutagenesis
Guo-Ren CHEN ; Lin LIN ;
Microbiology 1992;0(01):-
In order to improve the optimum temperature of lipases, The Penicillum expansum lipase (PEL) gene was mutated by site-directed mutagenesis using overlap extension PCR technique. The recombinant plasmid pPIC3.5K-lip-E83V containing matant gene was expressed in Pichia pastoris GS115. The comparison experiments of the mutant PEL-E83V-GS with the wild-type PEL-GS showed that: the optimum temperature of the mutant (45℃) was higher by 5℃ than that of the wild type. The thermostability of the mutant was similar to the wild type. The enzymatic activity of the mutant was 188 U/mL at 37 ℃, which was 80% that of the wild type at the same conditions. The hydrophobic interaction may be enhanced in the surface region by the hydrophilic amino acid, Glu substituted with the hydrophobic amino acid, Val, and be responsible for the improvement of optimum temperature.
5.Research on neuroprotective effects of mesenchymaI stem ceIIs in retinaI disease
International Eye Science 2015;(3):464-466
· Mesenchymal stem cells ( MSCs ) are a population of multipotent stem cells with various neurotrophins from bone marrow which are widely used in tissue, cell repair and alternative research.Recently, some researches have shown that MSCs could enhance the viability of neurons under a pathological circumstance by secreting some neurotrophins. So the neuroprotection of MSCs can provide a new method of the treatment on retinopathy that it possible to promote cells survial and functional recovery.Here we make a brief review on the secretory function of neurotrophin and neuroprotective effects of MSCs on retinal cells and its application in the treatment of retinal disease.
6.Activation of matrix metalloproteinase-13 in the osteoarthritis
International Journal of Surgery 2011;38(1):19-22
Objective To investigate the activation mechanism of matrix metalloproteinase-13 zymogen (pro-MMP-13) induced by nitric oxide (NO). Methods Human chondrosarcoma cells (SW1353) were grouped and treated with the NO donor S-nitroso-N-acetyl-penicillamine (SNAP), SNAP + NO scavenger oxyhemoglobin (OxyHb), and SNAP + tissue inhibitor of metalloproteinase material -2 (TIMP-2) respectively. After stimulation, matrix metalloproteinase -13 ( a-MMP-13 ) expression level was detected. Besides, the expression levels of MT1- MMP were detected after SW1353 cells stimulated with SNAP and SNAP + OxyHb, respectively. At last, the activities of the recombinant MMP-13 (r-MMP-13) were detected after r-MMP-13 stimulated with SNAP, recombinant MT1-MMP (r-MT1-MMP) and r-MT1-MMP + TIMP-2,respectively. Results SNAP increased human chondrocytes a-MMP-13 expression, OxyHb and TIMP-2 inhibited a- MMP- 13 expression. SNAP also increased the expression of MT1- MMP levels, OxyHb inhibited the expression of MT1-MMP. r-MT1-MMP activated the r-MMP-13, but SNAP did not, TIMP-2 inhibited the activity of r-MMP-13 induced by r-MT1-MMP. Conclusions NO can not directly activate pro-MMP13 by the role of S-nitroso-(S nitrosylation). MT1-MMP mediates the activiation of pro-MMP-13 induced by NO.

10.Patterns of Enterococcus faecalis in infected root canals: An in vitro study
Journal of Peking University(Health Sciences) 2009;41(6):699-701
Objective:To establish an in vitro root canal model infected by Enterococcus faecalis and to observe the morphology, distribution and relative position of Enterococcus faecalis in infected root canals.Methods: Ten human healthy premolars extracted for orthodontic reasons were collected. Following sterilization , a total of 5 specimens were aseptically transferred to separate Eppendorf tubes containing 1. 5 mL brain-heart infusion broth (BHI) inoculated with 0.1 mL Enterococcus faecalis suspension that had been adjusted to Mcfarland 5, and were incubated at 37℃ for 21 days. The other 5 specimens were as controls. The roots of all specimens were then split into two halves along the mesiodistal axis. One half was processed with light microscopic ( Brown & Brenn stain) to check the bacteria in dentinal tubules, and the other was observed with SEM to investigate the bacterial status in infected root canals. Results: Enterococcus faecalis could penetrate into the dentinal tubules about 330-1 000μm. A dense bacterial aggregation composed of Enterococcus faecalis and amorphous matrix was observed in the apical third of the root canals, whereas Enterococcus faecalis were seen free-floating or planktonic in the crown and middle third of the root canals . No microorganisms were found in the root canals of the controls. Conclusion:Enterococcus faecalis could form bacterial biofilm on the root canal walls and penetrate into the dentinal tubules. The in vitro model designed was simple, and had good practicability to make a further comparative evaluation of various antimicrobial methods in the reduction of intracanal bacteria.
