1.Establishment of transplantable rat prostate carcinomas and study of its relationship to hormone dependency

Objective To establish transplantable rat prostate carcinoma and study its relationship to hormone dependency. Methods Six transplantable rat prostate carcinoma lines were established from primary tumors developing in the dorsolateral prostate of F344 rats receiving a combined treatment with 3, 2’-dimethyl-4-aminobiphenyl (DMAB) and testosterone propionate (TP). Results Growth in the subcutis of nude mice (ICR-nu/nu) is rapid with a doubling time of approximately 10 days and transplantability is 100%. The histological appearance of the transplanted tumors is very similar to that of the primaries and their structures have not been altered by after 8 passages. Androgen receptor immunohistochemistry revealed all to be negative. Conclusions These data indicate that the model should prove useful for studies of invasion, metastasis, and hormone dependency of prostate carcinomas.
2.Implication of androgen receptor gene methylation in transformation of hormone-resistance in prostate carcinoma

Objective To study the mechanisms underlying this loss of AR expression in rat prostate cancer. Methods The methylation status of the AR gene promoter region in rat prostate cancer and cell lines (PLS10,20,and 30) was investigated by Southern blotting,methylation-specific polymerase chain reaction,and methylation-sensitive single-strand conformation analysis (MS-SSQA). Results AR mRNA expression was not detected in any of the rat prostate cancers or cancer cell lines tested by Northern blot analysis. Higher levels of methylated CpGs were observed in PLS20 than that in PLS10 or 30. Demethylation treatment by 5-aza-2’-deoxycytidine restored AR mRNA expression in PLS20.The CpGs was suggested to be responsible for AR expression in rat prostate cancer,and found to be located-9 and-1 nucleotides upstream of the transcriptional initiation site.All of the examined rat prostate and seminal vesicle cancers demonstrated hypermethylation at these CpG sites. Conclusions These data clearly demonstrate that aberrant hypermethylation in the AR promoter region may play a critical role in AR expression in rat prostate cancers.Loss of AR expression might result in development of hormone-resistant prostate carcinoma.

4.Clinical Observation of Acupuncture plus Hyperbaric Oxygen for Post-stroke Depression

Objective To observe the therapeutic efficacy of acupuncture plus hyperbaric oxygen in treating post-stroke depression. Method Sixty patients with post-stroke depression were randomized into a treatment group of 30 cases and a control group of 30 cases. The treatment group was intervened by acupuncture plus hyperbaric oxygen, while the control group was by Fluoxetine. Before and after intervention, the Hamilton Depression scale (HAMD) was adopted for evaluation, and the efficacies were compared. Result The total effective rate of the treatment group was significantly higher than that of the control (P<0.01);after intervention, the HAMD score of the treatment group was markedly lower than that of the control (P<0.01). Conclusion Acupuncture plus hyperbaric oxygen can obviously improve the depression symptom and promote the recovery of neurologic function in patients with post-stroke depression.

Objective]To appreciate Professor Lin's rigorous attitude of research,discuss and analyze his research findings in Traditional Chinese Pharmacy, academic thinking and achievements. [Methods]We put in order his academic papers,articles,as well as the materials science books of half a century, followed Professor Lin's visiting and studying across the country,summed up and gave comparative analysis as well as referring to detailed quotation and abstract.[Results]Professor Lin,about the spirit of learning,concentrated on the research thoroughly,made his academic achievement gradually and improved himself step by step.He had extensive interest and wide reading,committed to inheritance and improvement.His academic thinking reflected the value of history,respected tradition and followed the code and had comprehensive and innovative advantages.In his professional field,he also showed a wonderful career ethics such as caring for the people, simple prescription, attention to compatibility of Chinese Medicinal Herbs and thought highly of the classics. His academic achievements included textbooks and monographs, medical history and the history of medicine,herbal cuisine and tea treatment, as well as studies of both prescriptions and pharmacies since ancient times till now.All his achievements involved culture both at home and abroad. [Conclusion] Professor Lin Qianliang was born of poor parentage, but he concentrated on studies. He has excellent academic attitude and rich academic achievements,a wide rang of knowledge,which is worth learning and passing on to the younger generations.
6.Antibiotic resistance of Neisseria gonorrhoeae isolates from female patients

Objective To investigate antibiotic resistance of Neisseria gonorrhoeae strains isolated from female patients. Methods:Antimicrobial susceptibility of N.gonorrhoeae to 7 antibiotics was tested by disk diffusion method and penicillinase- producing N.gonorrhea(PPNG)was confirmed by nitrocefin disk.Results:Of the 95 strains collected,38(40%)were found to be PPNG.No resistance to spectinomycin was identified.The susceptibility rate of these isolates to penicillin,tetracycline, and ciprofloxacin were much lower,only from 0% to 2.6%.More than 90% of the isolates were susceptible to cefuroxime, ceftriaxone,and ceftizoxime.Conclusions:Our study suggests that spectinomycin,cefuroxime,ceftriaxone,ceftizoxime are the first choice for treatment of gonorrhea in female patients.
7.A diagnostic protocol for ipsilateral femoral neck and shaft fractures

Objective To evaluate a serf-designed diagnostic protoeol which can early detect a femoral neck fracture for patients with a femoral shaft fracture. Methods From September 2005 to June 2007, a self-developed protocol was used to detect an ipsilateral femoral neck fracture for all the patients with femoral shaft fracture who had sought treatment in our department. This protocol consisted of anteroposterior plain radiography of internal rotator, intraoperative fluoroscopy of the hip, a fine (2 mm) cut computed to-mographic scan through the femoral neck, postoperative anteroposterior and lateral plain radiography of the hip in the operating room prior to awakening the patient, at the time of follow-up anteroposterior and lateral plain radiography of the hip in the presence of hip pain. The diagnostic effects of the protocol were compared with those of conventional diagnosis used for all the patients with femoral shaft fracture who had sought treatment in our department from September 2003 to August 2005. A chi-square analysis comparing the protocol group (September 2005 to June 2007) and the non-protocol group (September 2003 to August 2005) was used to assess the early and delayed diagnosis rates for an associated ipsilateral femoral neck fracture. Results The earlydiagnosis rate of an associated femoral neck fracture by the protocol was 93.8%, markedly higher than that by conventional method (46.2%), with statistically significant difference (χ2 = 4.069, P =0.044). Conclu-sion In presence of a femoral shaft fracture. this protocol consisting of plain radiography of intemal rotator, intraoperative fluoroscopy of the hip, fine cut computed tomographic scan of the femoral neck, postoperative plain radiography of the hip, and follow-up plain radiography of the hip in the presence of hip pain, may sig-nificantly improve the diagnostic rate of an associated femoral neck fracture.
8.Perioperative nursing of patients with primary venous reflux disease of lower extremity

Objective To discuss the perioperative nursing experience of patients with primary venous reflux disease(PVRD)of lower extremity undergoing external valvuloplasty of superficial femoral vein.Methods Patients(146 cases)with PVRD of lower extremity were given psychological nursing,observation of postoperative complications and rehabilitation exercise instruction.Results Five limbs appeared short lymphatic seepage and other limbs recovered completely.No thrombus Was seen.Venous claudication,swelling and pain disappeared in 90.4% patients.Limbs symptom alleviated in 9.6% patients.All varicos evein disappeared.The function of deep vein valve recovered in 92% patients by color doppler ultra sonoscope re-examination.Conclusions General evaluation before operation,good perioperative psycho logical nursing and attention to the complication and rehabilitation exercise instruction are significant for reduction of postoperative complication and improvement of success rate of operation.
9.The physiological responses of healthy youth to doing chest compression as a feedback of the quality and intervention effects of CPR after their rapid ascent to high altitude

Objective To investigate the physiological responses of healthy youth to doing chest compression as a feedback of quality of CPR after their rapid ascent to high altitude and to evaluate the feedback in the intervention effects of CPR.Methods Prospective,single sample,before-after comparison method was used in this study.Fifteen young adults from plains natives were enrolled as trial subjects in this study.All of them received basic life support training course in advance.In Chongqing (259 m above sea level),subjects performed empiric chest compressions on the model body for 4 minutes followed by feedback compressions for 4 minutes after at least 30 minutes rest.Compression depth,rate and other compression quality parameters were measured and recorded at each turn of compressions with an AED PLUS device.Subjects performed empiric compressions based on their knowledge and experiences,and practiced feedback compressions according to the audiovisual guidance of AED PLUS device.Blood pressure,heart rate and SpO2 were taken before and after each turn of compressions.One week after arrival to Lhasa (3658 m above sea level) by flight,all subjects were asked to do the same procedure as did in Chongqing to see their physiological response to.Paired t tests or Wilcoxon matched pair rank test were used for comparisons of measurements before and after trials.Results Systolic pressures,diastolic pressures,heart rates at baseline in Lhasa were significant different from those in Chongqing,including systolic pressure (125.9 ±9.5) mmHg vs.(112.7 ±13.4) mmHg,diastolic pressure (75.3 ±7.7) mmHg vs.(64.2 ±7.3) mmHg,heart rate (86.3 ± 13.0) beat/min vs.(72.7 ± 11.6) beat/min,SpO2 (90.4 ± 1.7)% vs.(97.8 ±0.9)%,all P < 0.01.In Lhasa,empiric compressions only caused an increase in heart rate (91.1 ± 14.9) beat/min vs.(86.3 ± 13.0) beat/min,P < 0.01.However,feedback compressions resulted in a significant decrease in SpO2 [(88.3 ± 3.4) % vs.(90.6 ± 1.9) %,P < 0.01] as well as change of systolic pressure [(130.9 ± 11.7) mmHg vs.(120.1 ± 11.9) mmHg,P <0.05] and heart rate [(87.9 ± 17.5) beat/min vs.(80.9 ± 11.7) beat/min,P <0.05].In Lhasa,the compression quality during feedback compressions was closer to guideline recommendation than that during empiric compressions.The median (interquartile range) of composite qualification rate was 43.6% (55.9%) vs.0.6% (5.3%) during feedback compressions and empiric compressions respectively,P < 0.01.Conclusions Compression quality decreased significantly among youth after rapid ascent to high altitude.Feedback techniques for CPR could effectively guide the rescuer to improve their CPR quality,but it may be at the expense of more physical consumption.
10.Study on Cold-soak Extraction of Fat-soluble Components in Radix Salviae Miltiorrhizae

Objective To explore the optimal process conditions for cold-soaking extraction of fat-soluble components in Radix Salviae Miltiorrhizae.Methods Tanshione Ⅱwas determined by HPLC.The effect of different concentrations of ethanol and comminution on the extraction rate of tanshione Ⅱwas observed,and the process conditions were optimized by orthogonal test.Results The optimal process conditions for obtaining higher extraction rate of tanshione Ⅱwere as follows: the medicinal material was extracted by cold-soak extraction with 95% ethanol for two times(3 hours for each time),and the volume of ethanol was as much as 8 times of the medicinal material.Conclusion The extraction process is simple,stable and efficient for Radix Salviae Miltiorrhizae.
