3.Study of the Therapeutic Effects of Bushenshengsui I on Mice With Experimental Aplastic Anaemia

OBJECTIVE:To study the possible mechanism of Bushenshengsui I(BSSS I),a kind of compound Chinese herbs,in treatment of aplastic anaemiaMETHODS:The animal model of mice with aplastic anaemia was established by 60Co irradiationThe therapeutic effects of BSSS I on aplastic anaemia were observed with the changes of peripheral blood cell count,bone marrow GM-CFU-c and activities of blood IL-1,IL-2 and IL-6RESULTS:After treatment with BSSS I,the peripheral blood cell count,amount of bone marrow karyocyte and GM-CFU-c of bone marrow of the aplastic anaemia mice elevated markedly,and the blood IL-1 and IL-6 activities increased obviouslyThe levels of IL-6 increased to the level of normal and showed positive relationship with IL-1(r=0904)The levels of IL-2 of the treatment group showed a decrease but with no significant differencesCONCLUSION:The results showed that BSSS I may improve the function of hematopoiesis by elevating the peripheral blood cells and the amount of bone marrow karyocytes,accelerating GM-CFU-c of bone marrow,and through moderating the activities of IL-1,IL-2,IL-6

Bronchial asthma is the most common chronic diseases in children.Asthma can not be fully explained by imbalance of Thl/Th2.With the research progress of CD4+ CD25+ Treg cell,it has been found that CD4+ CD25+ Treg cell related factors such as forkhead/winged helix transcription factor,heine oxygenase-1,transforming growth factor-?,cytotoxic T lymphocyte-associated antigen-4 are closely linked to asthmatic mechanisms.
6.Effect of qingchang huashi recipe on IL-17 in the plasma and colonic mucosa of patients with ulcerative colitis.

OBJECTIVETo detect the expression level of interleukin 17 (IL-17) in the plasma and colonic mucosa of patients with ulcerative colitis (UC), and to explore the synergistic mechanism of qingchang huashi recipe (QHR) combined with Mesalazine.
METHODSRecruited were 24 mild or moderate UC patients of damp-heat inner accumulation syndrome (DHIAS). Their samples of intestinal tissues were histologically graded. They were assigned to the combination group and the Western medicine (WM) group, 12 in each group. Besides, another 12 healthy volunteers were recruited as the healthy control group. QHR combined Mesalazine were given to patients in the combination group, while those in the WM group took Mesalazine. The therapeutic course for all was 3 months. By the end of treatment the expression level of IL-17 in the plasma and colonic mucosa was detected using ELISA. The infiltration of IL-17 in the intestinal mucosal tissue was detected by immunohistochemical SP method.
RESULTSThe expression level of IL-17 in the plasma and colonic mucosa was significantly higher in UC patients than in healthy controls (P <0. 05). The higher the histological grading the higher the expression level. The expression level of IL-17 in plasma and colonic tissues decreased after treatment in the two treatment groups (P < 0.05). Besides, the expression level of IL-17 was lower in the combination group than in the WM group (P <0.05).
CONCLUSIONQHR combined Mesalazine could synergically enhance the effect and effectively inhibit intestinal inflammation through down-regulating the expression of IL-17.

7.Effect of Pyridoxamine and Telmisartan on the Indexes of Myocardial Remodeling in Spontaneous Hypertensive Rats

Objective: To observe the effect of pyridoxamine and telmisartan on the indexes of myocardial remodeling in spontaneous hypertensive rats (SHR). Methods: A total of 48 male SHR at 22 weeks of age were randomly divided into 4 groups:①Hypertension (H) control group, the rats received distilled water 2 ml/d,②telmisartan (T) group, the rats received telmisartan 6 mg/(kg.d),③pyridoxamine (P) group, the rats received pyridoxamine 200 mg/(kg.d),④Combination (TP) group, the rats received both telmisartan and pyridoxamine. All animals were treated for 16 weeks and the systolic blood pressure (SBP) was measured before and after treatment. After treatment, the serum levels of nitric oxide (NO) and super oxide dismutase (SOD) were examined by chemiluminescent method, advanced glycation end-products (AGEs) was detected by ELISA, left ventricular weight index and collagen volume fraction (CVF) in myocardial tissue were calculated, the mRNA expression of myocardial receptor of advanced glycation end products (RAGE) was evaluated by real time RT-PCR. Results: Compared with H group, SBP levels were decreased in T and TP groups,P<0.01, while SBP in P group stayed similar,P>0.05. Compared with H group, the serum levels of NO and SOD were increased in T, P and TP groups,P<0.01; while compared with T group and P group, the levels of NO and SOD were further increased in TP group,P<0.05. Compared with H group, left ventricular weight index and CVF were decreased in T, P and TP groups,P<0.01; while compared with T group and P group, left ventricular weight index and CVF were further decreased in TP group,P<0.05. Pyridoxamine and telmisartan had the coordinative effect on left ventricular weight index and CVF, P<0.05. The serum levels of AGEs were lower in T group (5.99 ± 0.51) mg/L, P group (5.57 ± 0.91) mg/L and TP group (5.24 ± 0.63) mg/L than that in H group, (6.71 ± 0.50) mg/L,P<0.01; while compared with T group and P group, AGEs was further decreased in TP group,P<0.05. The mRNA expressions of RAGE were reduced in T group (0.035 ± 0.010), P group (0.036 ± 0.005) and TP group (0.024 ± 0.007) than that in H group (0.053 ± 0.010),P <0.01; while compared with T group and P group, the mRNA of RAGE was further reduced in TP group, P<0.05. Conclusion: Pyridoxamine could improve the oxidative stress and the indexes of myocardial remodeling independently from decreasing blood pressure in SHR. Combined intervention of P and T may break the coordination systems between AGEs-RAGE and renin-angiotensin, therefore further improve the indexes of myocardial remodeling in SHR.
8.Application of robotic system in gastrointestinal surgery.

Robotic system helps surgeons in performing surgery. Currently Da Vinci system is the most popular. Da Vinci system has been used for the stomach and bowel diseases in 27 cases(18 cases of stomach and 9 cases of colon and rectum) in the Zhongshan Hospital, Fudan University. Accurate preoperative staging is crucial, and Da Vinci system is advantageous in lymph node dissection, preservation of nerve plexus, and complete resection of mesorectum. Adoption of gastrointestinal tract reconstruction technique should depend on the operation and experience in surgery. Though Da Vinci system has limitations and the cost is high, it is believed to be the future trend.

9.Application of Chemometrics in Quantitative Characterization of Traditional Chinese Medicine

With the deepening of modernization of traditional Chinese medicine (TCM), the method of quantification and standardization of TCM (i.e., quantitative characterization of TCM) has been more and more widely accepted by researchers. Chemometrics processes complicated data of TCM through applied mathematics, statistics and com-puter technology. And multivariable study was introduced into the quantitative characterization of TCM with great achievements. This article reviewed existed problems of quantitative characterization in TCM, the principles, char-acteristics, limitations, commonly used statistical methods and application conditions on quantitative characteriza-tion of TCM. With this review, a reference for further study of quantitative characterization of TCM was provided and a further research idea of combination with main methods of chemometrics was given.
10.Preparation and characterization of a new microporous bacterial cellulose material as a potential scaffold

Objective When bacterial cellulose (BC) is used as a scaffold material in tissue engineering,the nano-structure of BC may not provide enough space for animal cell growth and differentiation which would not achieve a perfect application in tissue engineering.In order to solve this problem,a novel green approach is developed in this research to produce bacterial nanocellulose materials with micropores ranging 50-800 μm.Methods Several ratios of hydrogen peroxide to sodium chlorite were used to react instantly to produce a large number of bubbles in BC hydrogels,which formed micropores with diameters ranging 50-800 μm.Optical microscopy and scanning electron microscope were used to evaluate microporous BC hydrogels and verify the existence of micropores.Results The size of pores could be regulated along with the changes in the amount of reactants used in the experiment.Fourier transform infrared spectroscopy verified that no cellulose was oxidized.Water content of the microporous BC hydrogels was similar to that of the original BC hydrogels.The Young's modulus of microporous BC hydrogels was 26.1 kPa,which was lower than that of the original BC hydrogels (69.9 kPa).Thiazoyl blue tetrazolium bromide (MTT) test displayed a higher viability on the microporous BC hydrogels compared to the growth on the unmodified BC substrates.Conclusions This study provides a convenient and promising way to prepare microporous materials,which may not be limited to only BC material,but could be used in other hydrogels.The proposed approach is suitable for extensive industrialization.
