1.Effect of Pidotimod on Expressions of IL-2 and IL-10 of Patients with Genital Herpes and Curative Effect Observation

Objective:To investigate the effect of pidotimod on the expressions of IL-2 and IL-10 of patients with genital herpes(GH) and obverse its curative effect and quality of life of the patients.Mehtods:45 cases of outpatients with GH were selected as experimental group wich were given pidotimod orally 0.8g per day for 3 months.Observe the curative effect and value the quality of life before and 3 months after and 6 months after the treatment respectively.Detect the change of the expressions of IL-2 and IL-10 of the blood plasma of the GH patients.Select 30 healthy people as control group.Results:The recurrence rate of the experimental group during 1-3 month,4-6 month,7-9 month after treatment were 60.0%、42.2%and 22.2%respectively.The scores of quality of life and the total evaluation of quality of life itself (G1) and health condition(G4) at 3 and 6 month after treatment rising significantly than before treatment,and it is more obviously at 6 months after treatment than 3 months after treatment.Before treatment,the level of IL-2 of experimental group is significantly lower than that of control group,while the level of IL-10 of the experimental group is significantly higher than that of control group.3 and 6 months after treatment,the level of IL-2 rising significantly,while the level of IL- 10 reducing significantly.The change is more obviously at 6 months after treatment than 3 month after treatment either in the IL-2 rising or the IL-10 reducing.Conclusion:There is cellular immune function disorder in the GH patients.Pidotimod has a certain curative effect on the GH patients.It can reduce the recurrence rate of GH and improve the quality of life of the patients.Maybe it can enhance the cellular immune function of the body through rising the level of IL-2 and reducing the level of IL-10.
2.Clinical study on 156 cases with hydrops fetalis

Objective To investigate the ultrasound characteristics,etiology and prognosis in hydrops fetalis.Methods From September 2002 to May 2010,156 hydrops fetalis presented in Shenzhen Maternity and Child Healthcare Hospital were studied retrospectively,including ultrasound characteristics,etiology,and prognosis.Results All of the 112 typical hydrops fetalis,20 cases with isolated ascites,8 cases with isolated pleural effusion,7 cases with isolated pericardial effusion,5 cases with isolated subcutaneous edema,4 cases with isolated placental thickening were observed by ultrasonography.The major etiology and associated diagnosis consisted of 35.9% (56/156) of non-immune anemia,9.6% (15/156) of cardiac abnormalities,7.1% (11/156) of intrauterine infection,6.4% (10/156) of twin problems,5.8% (9/156) of meconium peritonitis,5.1% (8/156) of thoracic-lung disease,4.5% (7/156) of chromosomal abnormalities,1.9% (3/156) of immune anemia.Alpha thalassemia was the most common non-immune anemia (96%,54/56).An etiology and associated diagnosis could be determined in 81.4% ( 127/156 ) of cases.Follow-up data showed that 7 cases were fetal death,110 women elected to terminate their pregnancies,3 cases lost follow-up,the other 36 cases preserve continuing pregnancy,including 28 liveborn infants and 8 fetal deaths.Etiology of twin-twin transfusion syndrome,meconium peritonitis,congenital chylothorax,intrauterine infection,cardiac abnormalities and so on had survived fetuscases.The survival rate of typical hydrops fetalis in the present series was 3.6% ( 4/112 ).Conclusions Ascites is the most common characteristics of sonogram in hydrops fetalis.The etiology of hydrops fetalis is extremelycomplex.The prognosis is associated with the etiology and hydrops subtype.
3.Validity and reliability of the Chinese version of Snaith-Hamilton Pleasure Scale (SHAPS)in assessment of patients with depression

Objective:To test the validity and reliability of the Chinese version of the Snaith-Hamilton Pleasure Scale (SHAPS).Methods:Totally 166 patients with major depression completed the Chinese version of SHAPS,Beck Depression Inventory (BDI-Ⅰ) and Hamilton Depression Scale (HAMD-17).The validity was tested by using correlation analysis and exploratory factor analysis.The reliability was tested by using internal consistency coefficient and Pearson correlation analysis.Results:The ICCs between three different Scale score were 0.90 and 0.91(Ps < 0.001).Exploratory factor analysis identified 2 common factors,which explained 62.8% of total variancel.The Cronbach coefficient of SHAPS was 0.94.The test-retest coefficient was 0.91 (P < 0.001).The intercorrelations of 14 items ranged from 0.30 to 0.93 and item-total correlations ranged from 0.61 to 0.79.Conclusion:The Chinese version of SHAPS has good validity and reliability,it could be used to assess anhedonia in Chinese patients with major depression.
4.Inhibition of lovastatin on proliferation and expression of proinflammatory cytokines in cultured human glomerular mesangial cells.

OBJECTIVETo study the effects and mechanism of lovastatin on cell proliferation and expression of proinflammatory cytokines in cultured human glomerular mesangial cells.
METHODSThe influence of lovastatin on HMC proliferation was evaluated with 3H-thymidine incorporation. mRNA expression of proinflammatory cytokines (IL-1 beta, IL-6, TNF-alpha, and MCP-1) and activation of NF-kappa B of HMC were measured using Reverse transcription-polymerase chain reaction (RT-PCR) and electrophoretic mobility shift assay (EMSA) respectively.
RESULTSLovastatin was found to have inhibitory effects on human mesangial cell (HMC) proliferation and lipopolysaccharide (LPS)-mediated human mesangine cell HMC mRNA expression of proinflammatory cytokines via activation of NF-kappa B. The effect of lovstatin on HMC could be prevented when the mevalonate and farnesol were added to the culture.
CONCLUSIONLovastatin may decrease HMC proliferation and production of proinflammatory cytokines through the inhibition of NF-kappa B activation. This provided experimental evidence for further evaluation of the renal protective effect of HRI, suggesting that it may be a potent agent for prevention of progressive renal diseases aside from its lipid-lowering effect.

OBJECTIVETo map the gene responsible for nonsyndromic hearing impairment in a consanguineous family.
METHODSFirstly, X chromosome scanning was used to exclude X chromosome. Secondly, candidate gene analyzing and genome scanning were performed by homozygosity mapping. Then, additional markers flanking the tightly linked marker were tested to confirm linkage and decide the candidate region.
RESULTSThe nonsyndromic hearing impairment of this family was autosomal recessive. Twenty-five known genes were excluded. Autosomal genome scanning indicated that D17S1293 was tightly linked with disease gene. And further study mapped the disease gene to a 5.07 cM interval bounded by D17S1850 and D17S1818.
CONCLUSIONThe disease gene of the family is mapped to a 5.07 cM interval between D17S1850 and D17S1818, which is a new locus of autosomal recessive nonsyndromic hearing impairment.

7.Efficacy and safety of metformin for Behcet's disease and its effect on Treg/Th17 balance: a single-blinded, before-after study.

OBJECTIVE:
Behcet's disease (BD) is an autoimmune disorder that causes most commonly mouth and genital ulcerations and erythema nodules of the skin and currently has limited options of therapeutic medicines. Metformin is recently reported to suppress immune reaction, and we hypothesized that metformin could be an option for treatment of BD.
METHODS:
Thirty patients with BD were enrolled in this perspective single-blinded, before-after study. We recorded the changes in the mucocutaneous activity index for BD (MAIBD), relapse frequency, C-reactive protein (CRP) level and erythrocyte sedimentation rate (ESR) after metformin treatment to assess the changes in the disease activity. We also analyzed the changes in the protein and mRNA expression levels of Foxp3, interleukin-35 (IL-35), transforming growth factor-β (TGF-β), Ror-γt, IL-17, and tumor necrosis factor- (TNF-) in these patients using ELISA and qRT-PCR.
RESULTS:
Of the 30 patients enrolled, 26 completed the trial. After the treatment, favorable responses were achieved in 88.46% (23/26) of the patients, and partial remission was obtained in 11.54% (4/26) of them. During the treatment, 8 patients complained of gastrointestinal side effects, for which 4 chose to withdraw from the study in the first week. Our results showed that metformin treatment decreased MAIBD and relapse frequency in the patients, and significantly lowered the clinical inflammatory indexes including CRP and ESR. The results of ELISA and qRT-PCR revealed that metformin treatment obviously increased Foxp3 and TGF-β expressions at both the protein and mRNA levels and significantly decreased the levels of ROR-γt, IL-17 and TNF- as well as IL-35 level in these patients.
CONCLUSIONS
Metformin treatment relieves the clinical symptoms, reduces the inflammatory reaction indexes and regulates the Treg/Th17 axis in patients with BD, suggesting the potential of metformin as a candidate medicine for treatment of BD.
