1.The expression of miR-451 and its clinical significance in gastric cancer

Objective To investigate the expression of miR-451 and its clinical significance in gastric cancer. Meth?ods The miR-451 expression was detected in 92 samples of gastric cancer and 47 sapmles of normal stomach mucosa. The correlations of miR-451 expression with clinicopathologic characteristics, overall survival and disease-free survival were an?alyzed in patients with gastric cancer. Results The overexpression rate of miR-451 was 38.04%in gastric cancer, which was significantly lower than 76.60%in adjacent normal tissues (χ2=18.503,P<0.001). The overexpression rate of miR-451 was significantly lower inⅢ-Ⅳstages of gastric cancer than that inⅠ-Ⅱstages (27.3%vs 54.1%, P=0.009). The expres?sion of miR-451 was also lower in patients with lymph node metastasis than that in patients without lymph node metastasis (27.1%vs 57.6%, P=0.004). However, there were no significant differences in miR-451 expressions between different age, gender, histological grade and T stage groups (P>0.05). The overall survival rate was significantly higher in patients with high expression of miR-451 than that in patients with low expression of miR-451(69.2%vs 39.1%, P=0.003). Conclusion miR-451 is associated with the malignant transformation, progression, and prognosis of gastric cancer, and which can be used as a potential target of therapy and a biomarker of diagnosis and prognosis of gastric cancer in the future.
2.Analysis on the results of liquid-based thinprep cytologic test in 9 012 patients

Objective To investigate the application value of liquid-based thinprep cytologic test ( TCT) in diagnosis of cervical cancer and precancerous lesion. Methods The results of liquid-based thinprep cytologic test and TBS cytology subdivision from 9 012 patients were retrospectively analyzed, including 12 cases of ASC-US, 10 cases of ASC-H, 35 cases of LSIL, 72 cases of HSIL, 2 cases of SCC, and 6 ca-ses of gland cell abnormal. The abnormal results and the results of biopsy pathological diagnosis underwent colposcopy were compared. Results The liquid-based thinprepcytologic test showed that there were 285 cases of atypical squamous cells of undetermined significance (ASC-US), the positive rate was 3. 2%. The accordance rate of liquid-based thinprep cytologic test and colposcope histopathological diagnosis was 91. 24%. Conclusion TCT and TBS system can accurately reflect cervical intraepithelial lesion. Early detection and treatment of precancer-ous lesions is the key point of preventing cervical cancer.
3.Proximal femoral intramedullary nail fixation for treating intertrochanteric fractures in young patients:joint function and complications in 3-month follow-up

BACKGROUND:Intramedulary nail with rigid biomechanics was firstly selected for proximal femoral fracture. Proximal femoral intramedulary nail can effectively control the shortening and rotation of the fracture, alow patients to do early exercise and weight bearing, and promote the improvement of joint function. OBJECTIVE:To investigate the effects of proximal femoral intramedulary nail for treating intertrochanteric fractures on the recovery of joint function in young patients. METHODS:80 young patients with intertrochanteric fractures were randomly divided into the treatment group and the control group, with 40 cases in each group. The control group received dynamic hip screw fixation, and the treatment group received proximal femoral nail fixation. Perioperative index, complications and functional recovery of knee joint after 3 months of treatment were compared between the two groups. RESULTS AND CONCLUSION:Compared with the control group, length of incision, intraoperative blood loss, operative time, fracture healing time and postoperative ambulation time were significantly reduced in the treatment group (P< 0.05). Complications such as postoperative nonunion, hip varus, limb shortening and deep vein thrombosis were significantly less in the treatment group than in the control group (P < 0.05). After folow-up at 3 months post treatment, the excelent and good rate of hip function was significantly higher in the treatment group than in the control group (93%, 80%;P < 0.05). These results suggest that proximal femoral nail for treating intertrochanteric fractures in young patients folows the principle of biomechanics, reflects the characteristics of minimal invasion, and has few complications, and promotes the recovery of hip function.
4.Mycophenolate acid inhibits hepatitis B virus replication in HepG2.2.15 cells

AIM: To investigate the effect of mycophenolate acid(MPA) on hepatitis B virus(HBV) replication in vitro.METHODS: In the presence or absence of guanosine,the HepG2.2.15 cells were treated with different concentrations of MPA(1-20 mg/L) for 4 days.Hepatitis B surface antigen(HBsAg) and hepatitis Be antigen(HBeAg) in supernatant were detected by ELISA.Intracellular HBV core mRNA and HBV DNA were analyzed by reverse transcription-polymerase chain reaction(RT-PCR) and slot blot hybridization,respectively.RESULTS: MPA suppressed the expression of HBsAg and HBeAg,and inhibited the replication of HBV DNA.The effect of MPA on HBV replication was reversed by addition of exogenous guanosine.CONCLUSION: MPA suppresses the expression of HBsAg,HBeAg and replication of HBV DNA in HepG2.2.15 cells.Reducing the synthesis of guanosine nucleotides may be involved in the mechanism of the inhibitory activity of MPA on HBV replication.
5.Application of dual-energy subtraction technique in diagnosis of occupational pneumoconiosis

Objective To explore the application value of chest dual-energy subtraction (DES) technology in the diagnosis of
occupational pneumoconiosis. Methods A total of 86 patients with suspected pneumoconiosis and 21 dust-exposure workers
were selected as the research subjects using random sampling method. The posterior and anterior chest radiographs were taken
by digital radiography (DR) and DES technology, and the difference of chest radiographs of DR, DES and combined groups were
compared. Results The positive rate of superior chest radiographs in DR group was higher than that in DES group (72.9% vs
56.1%, P<0.05). The determination of shadow shape and size, total density and stage of pneumoconiosis on chest radiographs in
DES group and combined group were consistent with those in DR group (all Kappa values >0.75, all P<0.01). However, the
judgment of small shadow intensity, small shadow aggregation and large shadow distribution in the two groups were not superior
to those in the DR group (Kappa value was 0.67, 0.74, both P<0.01). There was no significant difference between DES group and
DR group in the determination of small shadow intensity, small shadow aggregation and large shadow distribution (P>0.05).
However, there were statistically significant difference in this index between the combined group and DES group and DR group
(all P<0.01). There was no statistically significant difference in shadow shape and size, total density and stage of pneumoconiosis
in chest radiographs among these three groups (all P>0.05). Conclusion There was no difference between DES alone and DES
combined with DR for the diagnosis of pneumoconiosis in terms of shadow shape and size, total density and stage of
pneumoconiosis when compared with the gold standard DR. The value of DES in the diagnosis of pneumoconiosis needs further
study.
6.Expression and Significance of Heat Shock Protein 27 Druing Carcinogenesis of Esophageal Squamous Epithelium

Objective To investigate the expression of heat shock protein 27(HSP27)in esophageal squamous carcinoma,mucosa adjacent to carcinoma and normal esophageal mucosa,and its relationship with the carcinogenesis of esophageal carcinoma.Methods The expression of HSP27 was observed in 86 specimens from esophageal squamous carcinoma,86 from mucosa adjacent to carcinoma and 75 from normal esophageal mucosa by immunohistochemistry.Results The expression of HSP27 in esophageal squamous carcinomas was higher than those in mucosa adjacent to carcinoma(P < 0.05);the expression of HSP27 in mucosa adjacent to carcinoma was higher than those in normal esophageal mucosa(P <0.001).There was no significant difference in the expression of HSP27 in esophageal squamous carcinomas with different differentiation degree(P > 0.05).Conclusion Expression of HSP27 was associated with the carcinogenesis of esophageal squamous carcinoma.
7.Relationship between anti-cyclic Citrullinated petide antibody and cardiovascular complication associated with rheumatoid arthritis

Objective The aim of this study was to assess the relationship between the level of anti-CCP antibody and cardiovascular(CV) risk in rheumatoid arthritis(RA) patients.Methods A total of 78 RA patients were enrolled in the study.They were divided into two groups:40 cases with CV comphtations which were secondary from RA and 38 cases with no complications.The anti-CCP antibody was diagnosed by enzyme - linked immunoabsorbent assay(ELISA) and rheumatoid factor(RF) were assessed by immunoturbidimetry method.The relationship between the levels of the two sera auto-antibodies and CV disease associated with RA was analyzed.Results The results showed that the level of anti-CCP antibody in RA-CV disease group was significantly higher than that in simple RA group(74.6% VS 87.3% ).Compared with the simple RA group,the level of anti-CCP antibody in the RA-CV disease group was higher,and with significant difference between the two groups( x2 =5.07,9.04,6.48,all P < 0.05 ).RF positive rate was 95.8% of the patients with RA-CV disease while it was 76.5% in simple RA group,and the difference between the two groups was statistically significant( x2 =33.41,26.05,34.14,all P < 0.05 ).The morbidi ty between the different level of RF had remarkable difference.In RA patients with CV complations,we compared antiCCP antibody and RF by Spearman analysis,whish showed no relationship between them ( r =0.301,P =0.174 ).Conclusion The results indicated that CV disease secondary from RA was more common in old patients.In RA patients the high level of anti-CCP antibody and RF maybe play an important role in the appearance of CV disease.
8.Expression of miR-124 in gastric cancer and its clinical signiifcance

Background and purpose:miR-124 is considered to be a tumor suppressor in multiple tumors, including lung cancer, prostate cancer, bladder cancer, and breast cancer. However, its function is unclear in gastric cancer. This study aimed to explore the expression of miR-124 in human gastric mucosal epithelium cells and different gastric cancer cells, as well as gastric cancer tissues and matched para-cancerous tissues. The correlations of miR-124 expression with gender, age, histological grade, T stage, TNM stage, lymph node metastasis and prognosis of gastric cancer patients were analyzed.Methods:A real-time lfuorescent quantitative polymerase chain reaction (RTFQ-PCR) was employed for detecting the expression of miR-124 in human gastric mucosal epithelium cells and gastric cancer cells. The expression of miR-124 in gastric cancer tissues and matched para-cancerous tissues was detected by in situ hybridization.Results:The RTFQ-PCR results indicated that the expression of miR-124 was down-regulated in MKN-74, MKN-28, MKN-45, MGC-803, SGC-7901 and AGS cells compared to GES-1 cells.In situ hybridization showed that miR-124 was strongly expressed in normal gastric mucosa. However, low expression, focal positive expression or lack of miR-124 expression were observed in gastric cancer tissues. Statistical analysis showed that miR-124 was close-ly correlated to histological stage, TNM stage and node metastasis of gastric cancer patients, but not the age, gender and tumor size. The OS and DFS of the patients with low expression of miR-124 were shorter than that of the patients with high expression of miR-124. Multivariate analysis suggested that miR-124 down-regulation was an independent prog-nostic factor for survival in patients with gastric cancer.Conclusion:miR-124 is down-regulated in gastric cancer cells and tissues. The expression of miR-124 is correlated to histological stage, TNM stage, node metastasis and prognosis.
9.GSTM1 polymorphism,smoking,alcohol drinking and oral cancer susceptibility in Asians:A meta-analy-sis

Objective:To evaluate the association of glutathione S-transferase M1 (GSTM1 )polymorphism,smoking and alcohol drinking with oral cancer susceptibility in Asians by meta-analysis.Methods:A literature search of PubMed,Chinese BioMedical, Wanfang,VIP and CNKI databases from inception to July 30th,2013 was conducted.Crude odd ration (OR)with 95% confidence intervals (95%CI)was calculated.Results:27 case-control studies were assessed,and the results showed that the association be-tween GSTM1 null genotype and oral cancer susceptibility was significant in Asians (OR=1.31,95%CI:1.18-1.45,P<0.001), especially in South Asians (OR=1.35,95%CI:1.20 -1.52,P<0.001).Significant associations between oral cancer risk and smoking and alcohol drinking were found in East Asians (Smoking:OR=1.70,95%CI:1.36 -2.13,P<0.001;Alcohol drink-ing:OR=1.54,95%CI:1.24 -1.90,P<0.001).Conclusion:GSTM1 null genotype may be associated with increased oral cancer risk in Asians.Smoking and alcohol drinking confer significant susceptibility to oral cancer in East Asians.
10.Protective effects of low-concentration hydrogen sulfide on rheumatoid joint disease

BACKGROUND:Rheumatoid arthritis is a common chronic systemic autoimmune disease which is characterized by synovial lesions and bone erosion. The clinical treatment is stil a difficulty.
OBJECTIVE:To explore the effect of exogenous hydrogen sulfide on the articular lesions in rheumatoid arthritis rats.
METHODS:Sprague-Dawley rats were divided into four groups:control group, model group, high concentrations of hydrogen sulfide intervention group, low concentrations of hydrogen sulfide intervention group. At 2 and 4 weeks after intervention, the arthritis index, levels of serum inflammatory cytokines (tumor necrotic factor-αand interleukin-1β), pathological change of synovial tissue, average absorbance of knee joint specimens using col agen-II immunohistochemical staining were measured.
RESULTS AND CONCLUSION:The arthritis index, levels of serum inflammatory cytokines and inflammatory infiltration of synovial staining in the model group and two intervention groups were higher than that in control group, the high concentrations of hydrogen sulfide intervention group was significantly higher than the model group (P<0.05) while the low concentrations of hydrogen sulfide intervention group was lower than the model group (P<0.05). The average absorbance of knee cartilage specimens using col agen-II immunohistochemical staining was lower in the model group and two hydrogen sulfide intervention groups compared with the control group, the high concentrations of hydrogen sulfide intervention group was significantly lower than the model group (P<0.05) while low concentrations of hydrogen sulfide intervention group was higher than the model group (P<0.05). Hydrogen sulfide has a regulatory effect on rheumatoid arthritis, high concentrations of hydrogen sulfide aggravate arthritis and cartilage degradation, low concentrations of hydrogen sulfide reduce arthritis and protect cartilage.
