1.A study on the human oral squamous carcinoma cell apoptosis induced by arsenic trioxide in vitro

Objective To investigate the biological effects of As 2O 3 and its mechanism on human oral squamous carcinoma cell line in vitro . Methods The effects of As 2O 3 on Tca8113 cell line were investigated in vitro with morphological method, MTT, clone forming, DNA ladder, TUNEL assay and flow cytometry(FCM). Results As 2O 3 had marked suppressive effect on the growth of Tca8113 cell line. The growth suppression rate of the Tca8113 cells was 76.3% shown by MTT. DNA ladder, TUNEL and FCM showed As 2O 3 could induce the apoptosis of Tca8113 which might be related to cell cycle arrest shown by FCM. Conclusion As 2O 3 can inhibit the growth and induce the apoptosis of oral squamous carcinoma cell in vitro .

XIAP(X-linked inhibitor of apoptosis protein)is one of the most important members of IAPs family.It inhibits cell apoptosis mainly by inhibiting the caspases activation and interfering with other apoptotic pathway,which plays a critical role in tumor development.Recently,genetic therapy targeting on XIAP for malignant tumors has deeply developed.Research on XIAP as a malignant and prognositic indicator is ongoing.
4.Effect of microsomal triglyceride transfer protein(MTP) gene polymorphism on angiopathy in type 2 diabetes

Objective To investigate the relationship of MTP gene variation with type 2 diabetic angiopathy. Methods PCR and restriction-fragment-length polymorphism(PCR-RFLP) analysis and gene sequencing were used to investigate the influence of a common MTP gene polymorphism in promoter region on diabetic macroangiopathy. Results (1) A single nucleotide mutation of G→T was detected at 493 bp of upstream from the transcriptional start point.There was a significant difference in genotype frequency(χ2=9.162,P=0.01) and T allele frequency(χ2=10.322,P=0.001) between macroangiopathy group and non-macroangiopathy group.(2) MTP-493 TT group had the significantly higher serum levels of TC, TG, LDL-C, VLDL-C and lower HDL-C than other genotype groups.(3) Multivariate stepwise regression analysis showed that T allele, age, disease duration, diastolic blood pressure and total cholesterol were independent risk factors against type 2 diabetic macroangiopathy.The HDL-C level was negatively related with diabetic macroangiopathy. Conclusions MTP-493 TT genotype may be deeply involved in production of atherogenic dyslipidemia and associated with development of macroangiopathy in type 2 diabetic subjects.
5.Effects of drug-contained sera ofNaoluo Xintong versus Zuogui pill on the proliferation and differentiation ofin vitro cultured neural stem cells

BACKGROUND:Because of limited source and a relatively weak ability of differentiation and proliferation, how to take positive and effective measures to promote neural stem cel proliferation, differentiation has become the focus of research.
OBJECTIVE:To investigate the effects of drug-contained sera ofNaoluo Xintong versus Zuoguipil on the proliferation and differentiation ofin vitro cultured rat neural stem cels.
METHODS:Embryonic neural stem cels of Sprague-Dawley rats were isolated and culturedin vitro, and then were co-cultured with the serum medium containing 10%Naoluo Xintong and 10%Zuoguipil, respectively. Comparative observations were performed between two groups by inverted microscope and immunofluorescence staining.
RESULTS AND CONCLUSION:Under the inverted microscope, the cels began to grow in cluster and gather into a bal, but the diameter was relatively smal after 24-hour culture; the neurospheres expended further, with relatively regular shape, but no neurosphere differentiation appeared after 48 hours of culture. The average prominent length of the neurospheres in theZuoguipil group was significantly greater than that in theNaoluo Xintong group after 5 days of culture (P < 0.05). The rate of rat neural stem cels differentiating into MAP-2-positive cels in theZuoguipil group was significantly lower than that in the Naoluo Xintong group (P < 0.05), but the rate of differentiated cels positive for glial fibrilary acidic protein in theZuoguipil group was significantly higher than that in theNaoluo Xintong group (P < 0.05). After 48 hours of culture, neurospheres were cultured in the different drug-contained media, and 12 hours later, the neurospheres adhered to the wal, and a smal amount of cel migration occurred. Then, cel migration began to increase with time. Under the immunofluorescence staining: prominent neurons with long protrusions were increased in both two groups, but there were no significant differences in the proportion of neurons and astrocytes between two groups (P > 0.05). These findings suggest that drug-contained sera ofNaoluo Xintong andZuoguipil can both not only promote the proliferation and differentiation ofin vitro cultured rat neural stem cels, but also provide a suitable microenvironment for neural cel proliferation. Additionaly, there are significant differences between the two drugs. Consequently, it is feasible to induce neural cel proliferation and differentiation byNaoluo Xintong andZuoguipil.
6.Efficacy of pioglitazone combined with insulin in the trea tment of type 2 diabetes mellitus:a meta-analysis of randomized controlled trials

The efficacy and safety of pioglitazone in combination with insulin in type 2 diabetic patient were evaluated. CENTRAL, MEDLINE, EMBASE, CBM, CNKI(up to September 2014) were searched by computer. The quality of included randomized controlled trials(RCTs)was assessed according to the Cochrane Collaboration system review, and then meta-analysis was performed using Stata 12. 0. A total of 9 RCTs were included. The results of meta-analyses showed that HbA1C value was lower in the group taking pioglitazone plus insulin than in the group taking insulin without pioglitazone[standard mean difference -0. 48, 95% CI( -0. 55, -0. 40),I2 = 0. 0% ,P = 0. 531], but there were more patients with hypoglycaemic in the pioglitazone plus insulin groups than with insulin without pioglitazone[relative risk 1. 33, 95% CI(1. 21, 1. 46), I2 =75. 4% , P=0. 00].

8.Study of the Compliance of Hypertensive Medication

Objective:To investigate the influencing factors and medication compliance of hypertension.Method: 3 grade-Ⅲand 15 grade-Ⅱgeneral hospitals in Anhui Province were selected by random sampling.732 outpatients with a hypertension course of 6 months or above were interviewed during July and October of 2004 with the medication compliance schedule.All data were input into computers with the software Epi info 6.04,and analyzed with SPSS 11.5.Result:①The medication compliance schedule had high reliability and validity,which could be used in the surveys of medication compliance of hypertension.②The medication compliance rate of the 732 patients was only 46.4%.③Trust in medical pro- fessionals,more attention paid to the disease,more knowledge of non-compliance harm,satisfactory family income,and longer consulting time were the common advancing factors in improving the medication compliance,but a longer duration of the medication regimen,a larger number of preparations and drugs,and higher frequency of taking medicine were the bloc- king factors.Conclusion:The medication compliance of hypertensives was generally not good,and it was influenced by many factors.It is necessary that medical professionals,patients,families and society all take part in its improvement.

10.Frequency domain parameter of heart rate variability and its circadian rhythm in type 2 diabetes with lower extremity neuropathy

Heart rate variability(HRV)analysis and its circadian rhythm(CR)were determined in 58patients with type 2 diabetes mellitus with lower extremity neuropathy(diabetic neuropathy group), 59 patients with type 2 diabetes mellitus without lower extremity neuropathy(diabetes group), and 50 healthy controls according to 24-hour Holter recording. Frequency domain parameters of HRV were significantly decreased in both diabetes groups. Frequency domain parameter of HRV in healthy controls,and daytime/nighttime difference were statistically significant. CR of HRV was changed in diabetes group and disappeared in diabetic neuropathy group. Impaired and seriously impaired autonomic nervous function developed in type 2 diabetes mellitus without and with lower extremity neuropathy respectively.
