1.Differentiation of human umbilical cord-derived mesenchymal stem cells into male germ cells

ObjectiveTo study the possibility of differentiation of human umbilical cord mesenchymal stem cells(HUCMSCs) into human male germ cells,and to explore a new source of cells for the treatement of male infertility.MethodsHUCMSCs were transplanted into the seminiferous tubules of the testis of infertile mice by microinjection.The survival rate,migration and germ cell markers of HUCMSCs in the mice testis were detected via immunohistochemistry,immunofluorescence and confocal laser scanning. ResultsHUCMSCs can survive in the mice testis for at least 120 days,and they can migrate from the lumens to the basement membrane.Immunofluorescence showed that HUCMSCs can further differentiate in the mice testicular environment,and express the germ cell marker.ConclusionsHUCMSCs can survive,migrate and differentiate into early male germ cell-like cells in the infertility mice testis after transplantation.
2.THE ANTIOXIDANT ACTION OF MALTOL ON ERYTHROCYTE MEMBRANE

Maltol is a chemical derived from Chinese herb Panax Ginseng. The effect of maltol on hemolysis and hydroperoxidation of erythro-cytes induced by active oxygen radical is presented. Maltol could protect hemolysis induced by H2O2 in vitro. When erythrocyte membrane protein was exposed to butylhydroperoxide in vitro, membrane protein was polymerized, and membrane lipids were oxidized to hydroperoxide compound. These oxidative reactions could inhibited by maltol. The results showed that maltol might act as a scavenger of superoxide of hydroxy radical as demonstrated by biochemical method and ESR spin method。
3.Application of DWI and MRA to the diagnosis of acute cerebral infarction

Objective To explore the value of diffusion weighted imagine(DWI) and magnetic resonance angiography(MRA) for the clinical significance in acute cerebral infarction in the early stage.Method The results of DWI and MRA and traditional CT and routine MRI in 30 patients with acute cerebral infarction were analyzed.Result Ultra-acute and acute cerebral infarction could be shown on DWI,which may not be shown on CT and T 2.WI.The focus that were shown on T 2.WI were more clearly shown on DWI.Focus and degree of intracranial vascular lesion could be detected rapidly.Conclusion DWI and MRA are sensitive for diagnosis in the early stage of acute cerebral infarction.Cerebral constitution and cerebral vessels can be shown by DWI and MRA,thus offering reliable imaging data for thrombolytic therapy.
4.Application of radionuclide bone imaging in diagnosis and localization of osteoporotic vertebral compressive fracture

Objective To investigate the value of radionuclide bone imaging in the diagnosis and localization of osteoporotic vertebral compressive fracture.Methods Percutaneous vertebroplasty was applied to 40 patients suffering from osteoporotic vertebral compressive fracture.Responsible vertebral bodys located according to the radionuclide bone imaging was involved as bone scan group,and responsible vertebral bodys located according to the physical examination,X-ray,CT and MRI as standard group.Then the Kappa test was carried out.Result In bone scan group,56 vertebrae were located by radionuclide bone imaging,and in standard group,58 vertebrae were located by X-ray,54 vertebrae by CT and 52 vertebrae by MRI.The Kappa value was 0.76,indicating essentially excellent agreement and statistical significance.Conclusion Radionuclide bone imaging was another good choice for the diagnosis of osteoporotic vertebral compressive fracture.Especially for cases with contraindication of MRI,it offers a reliable choice.
5.Biocompatibility of electrospun carbon nanotubes/poly(L-lactic acid) nanofiber scaffolds with mouse neural stem cells

BACKGROUND:Poly(L-lactic acid) (PLLA) scaffold is a kind of widely used biomaterial in tissue engineering. However, low hydrophilicity and lack of surface cell recognition site of PLLA hinder its further application. OBJECTIVE:To study the biocompatibility of multi-walled carbon nanotubes/PLLA (MWCNTs/PLLA) nanofiber scaffolds with mouse neural stem cells in vitro. METHODS:Mouse neural stem cells were isolated. Then we used electrospinning to fabricate PLLA nanofibers and modified them with multi-walled carbon nanotubes. We assessed their biocompatibility with passage 3 mouse neural stem cells in vitro. RESULTS AND CONCLUSION:Scanning electron microscope showed that the neural stem cells could survive on both scaffolds. No cytotoxic effects were detected on both scaffolds by Cell Counting Kit-8 detection. The adhesion and proliferation abilities of neural stem cells on the MWCNTs/PLLA scaffold were significantly greater than those on the PLLA scaffold. Neural stem cells were found grow well and have normal morphology on both scaffolds under scanning electron microscope and by Hoechst 33342 staining. Besides, immunofluorescence staining showed MWCNTs/PLLA could promote neural stem cells to differentiate into mature neurons and neurites grew along with the nanofiber scaffold. In conclusion, the MWCNTs/PLLA nanofiber scaffold has better properties than the PLLA for transplanted cells and provides a good growth carrier for neural stem cells to be induced to differentiate into neurons, which is expected to have a great potential of applications in nerve tissue engineering.
6.Percutaneous vertebroplasty combined with granulated allogeneic bone grafting for thoracolumbar compressive fractures: the completeness and stability of fractured vertebrae

BACKGROUND:Percutaneous vertebroplasty is mainly used to treat senile osteoporotic compressive fractures,while for young patients,pedicle screw internal fixation is usually applied,but heavy trauma,nail broken,secondary surgery and other problems can be found occasionally.OBJECTIVE:To investigate the clinical efficacy of percutaneous vertebroplasty combined with granulated allogeneic bone grafting for thoracolumbar compressive fractures.METHODS:Twenty-one patients suffering thoracolumbar compressive fractures were recruited,and underwent percutaneous vertebroplasty combined with granulated allogeneic bone grafting.The Visual Analogue Scale scores,height ratio of fractured vertebrae and Cobb angle were detected before and after surgery.RESULTS AND CONCLUSION:(1) Compared with baseline,the Visual Analogue Scale scores were improved significantly at 1 day and 6 months postoperatively (P < 0.05).(2) The height ratio of fractured vertebrae at 1 day and 6 months postoperatively was significantly higher than that before treatment (P < 0.05).(3) The Cobb angle of fractured vertebrae at 1 day and 6 months postoperatively was significantly less than that before treatment (P < 0.05).(4) The mean operation time was (52±12.3) minutes,and the mean blood loss was (11.5±1.5) mL.(5) These results suggest that percutaneous vertebroplasty combined with granulated allogeneic bone grafting can effectively restore the anterior fractured vertebral height,eliminate the cavity in the fractured vertebrae,renew the completeness and stabilization of the fractured vertebrae,which keep the spinal range of motion;therefore,it is a minimal invasive method in the treatment of thoracolumbar fractures.
7.Investigation on Current Situation of Workplace Violence the Intern Experienced and Their Professional Identity

Objective:To understand the current situation of hospital violence medical students experienced during the internship period and their professional identity.Method:With the method of cluster sampling,a questionnaire survey was conducted among 500 medical students from a medical college in Ganzhou,to understand the characteristics of hospital violence and compare the difference between the medical students who experienced violence and those not.Results:The effective recovery rate of questionnaire was 90.8%,of which 61.4% of medical students had witnessed hospital violence,8.1% of students had experienced hospital violence;the hospital violence that medical students experienced were characterized by low frequency and emotional violence.The overall score of personal identity was (117.05 ± 19.65),was in a medium level.Among the comparison of personal identity between the medical students who had experienced violence and those not,there was a significant difference in occupational cognition dimension (P < 0.01),but there was no significant difference in the total score and other four dimensions (P > 0.05).Conclusion:We should pay attention to the current situation of hospital violence that students experienced and their professional attitude,carry out targeted training and psychological counseling to improve their ability to respond to hospital violence and enhance their professional identity.
8.The hypolipidemic and liver protective effect of asiatic acid on hyperlipidemic golden hamsters

Objective To observe the protective effect of asiatic acid on hyperlipidemia golden hamsters induced by high fat diet and explore its mechanism.Methods Hyperlipidemic golden hamsters fed with high-fat diet were administered orally with asiatic acid (8,16,32 mg/kg)for 4 weeks.Levels of serum lipid content,liver histology,hepatic GSH-PX and SOD levels,serum ALT and AST activities were evaluated in golden hamsters,fluorescence quantitative polymerase chain reaction(RT-PCR)is using to examine the liver lecithin cholesterol fatty acyl transferase(LCAT) and class B typeⅠscavenger receptor(SR-BⅠ)mRNA expression. Results Compared with model group,the levels of TC,TG,LDL-C,TC/HDL,ALT and AST in low and mediate asiatic acid groups were all significantly decreased but the levels of SOD and GSH-PX were significantly increased(P<0.01).HE staining results showed that fat deposition in the liver were improved by administration of asiatic acid,and the expression of LCAT and SR-BⅠmRNA were increased.Conclusion Asiatic acid can effectively reduce blood lipid levels,and alleviate liver damage of the hyperlipidemia golden hamsters by lipid regulation and antioxidative ability augmentation.The increased levels of LCAT and SR-BⅠmRNA expression maybe involved in the mechanism of hypolipidaemic effect of asiatic acid.
9.Shenmai Zhusheye vs vitamine C injection in adjuvant treatment of dilated cardiomyopathy

AIM: To observe the therupeatic effects of Shemai Zhusheye vs vitamine C injection in treating dilated cardiomyopathy (DCM). METHODS: Fifty-two patients with DCM were divided into 2 groups, 32 patients in treatment group (M 20, F 12; age 46 a± s 13 a), 20 patients in control group (M 11, F 9; age 47 a±14 a). Besides the patients in two groups were treated with conventional therapy, the patients in treatment group were given Shenmai Zhusheye 30 mL and 5% glucose injection 100 mL, iv, gtt, qd for 2 wk. The patients in control group were given vitamine C 5.0 and 5% glucose injection 100 mL, iv, gtt, qd for 2 wk. RESULTS: Treatment group total effective rate was 84%. Control group total effective rate was 65% (P>0.05). CONCLUSION: Shenmai zhusheye has better effect in treating DCM. Symptom improving rate of Shenmai Zhusheye has better than vitamin C.
10.The cardiac protection of non-mitogenic human fibroblast growth factor on cardiomyocytes treated with hydrogen peroxide in vitro

AIM: To study the protective effects of n on -mitogenic human fibroblast growth factor (nm-haFGF) on cardiomyocytes injured b y reactive oxygen free radicals. METHODS: The cardiomyocytes were isolated from neonatal SD mouse by trypsin digestion. The cardiomyocytes injury model was established by expos ing the cells to hydrogen peroxide (H 2O 2), and the injury status in cardiomy ocytes were evaluated by examining the cellular viability, measuring cell apopto sis and observing the change of cellular morphology. nm-haFGF was added to the c ulture medium, and the changes of cellular viability, superoxide dismutase (SOD) , malondialdehyde (MDA) and celluar apoptosis were observed. RESULTS: A dose-dependence relation between the concentration of H 2O 2 and the cardiomyocytes injury was observed. 10-80 ?g/L nm-haFGF dose -dependently increased cardiomyocyte viability and the general SOD activity, as well as decreased the content of MDA and the quantity of cardiomyocyte apoptosis . CONCLUSION: The higher the concentration of H 2O 2, the mor e serious the cardiomyocyte injury. nm-haFGF may have a good protective effects on cardiomyocytes treated with H 2O 2.
