2.Clinical selection of individualizd artificial liver support
Chinese Journal of Clinical Infectious Diseases 2017;10(2):113-118
Liver failure is characterized by hepatic encephalopathy, jaundice, hypo-coagulation and high mortality.Artificial liver support aims to temporarily replace hepatic function until liver function recover or bridge to liver transplantation.Individualized artificial liver support should be balanced and cover the following aspects: removing albumin bound toxins and/or water soluble toxins, eliminating inflammatory mediators and correcting blood coagulation disorders, in order to support metabolism and modulate immune function and to promote the regeneration of hepatocyte and reverse multiple organ failure that may lead to death of patients.Traditional non-biological artificial liver support can't meet individualized treatment.Recently developed hybrid blood purification model, such as albumin dialysis, fractionated plasma separation and adsorption (Prometheus system) and plasma diafiltration (PDF) have provided more and better choices for individualized artificial liver support.Based on the characteristics of pathophysiology and organ dysfunction of liver failure, the author has designed an alignment diagram of individualized artificial liver support for clinical selection.
3.Role of Proteasome Peptidase Activities in the Ethanol-Induced Cytotoxicity on PC12 Cells
Journal of Environment and Health 1993;0(01):-
Objective To clarify whether ethanol induced cytotoxicity of neurons was inhibited by proteasome peptidase activities. Methods A rat pheochromocytoma(PC12) was used in the present study. Cytotoxicity and proteasome peptidase activities were assessed in PC12 cells exposed to 50, 100, 150 mmol/L, 200 and 300 mmol/L ethanol. Results After 48 hours of treatment, ethanol inhibited proteasome peptidase activity in a dose-dependent manner at the concentration above 100 mmol/L. Moreover, the effect of ethanol on activities of proteasome was also in a time-dependent manner from 24 h. Cytotoxicity was still observed as ethanol concentrations increasing, started from 100 mmol/L, as assessed by MTT method. Conclusion Our results demonstrates ethanol may disturb the metabolism of protein by inhibiting proteasome peptidase activities, which is related to the toxic mechanism of ethanol on PC12 cells.

Supernumerary tooth is a rare case. This report described a case of nasal cavity supernumerary tooth association with maxillary sinusitis. A 28-year-old male patient reported with the chief complaint of nasal obstruction, headache and purulent secretion for the past three months. Clinic examination and CT examination showed that there was a supernumerary tooth in the right nasal bottom, and maxillary sinus was infected in the same side. This patient was performed supernumerary tooth removing and given antibiotics for 3 days. Ten days after the operation, there was no clinical symptoms, and nasal bottom mucosa was normal. After 3 months of follow-up, reexamination of coronal CT scan appeared normal.

Basic & Clinical Medicine 2006;0(05):-
Objective To investigate the association between a missense mutation (T/G, Ser 1369 Ala) in exon 33 of sulfonylurea receptor 1(SUR1) gene in type 2 diabetes patients and the glucose-lowering effect of Gliclazide. Methods One hundred and four type 2 diabetes patients were selected and orally administered with Gliclazide for 56 days. Venous fasting plasma glucose levels (FPG), plasma glucose levels half an hour after taking 75 g glucose (OGH) and two hours (OGT) after taking Gliclazide were measured. Ser1369Ala polymorphism genotypes(TT, TG, GG) of SUR1 gene were determined by Taqman method. The glucose-lowering effect of Gliclazide was compared among different groups. If FPG was reduced more than 20%, we define it as effective. People whose blood sugar level was effectively reduced and not significantly reduced were compared to find whether their genotype groups were different. Results The change of FPG,OGH and OGT on day 1 and day 57 among TT,TG and GG genotype groups had no statistically significant difference. There was no statistically significant difference of the efficacy of Gliclazide in recessive model (TT+TG vs GG). The combination of TG and GG was more effective in reducing blood sugar level than TT. Conclusion Patients carrying one or two copies of the G allele were more sensitive to Gliclazide than patients carrying T allele.

Basic & Clinical Medicine 2006;0(05):-
Objective To investigate the expression of angiotensin-converting enzyme 2(ACE2) in rat endocrine and exocrine tissue of pancreas and to compare its expression level with that in other visceral tissues. Methods The expression of ACE2 in rat visceral organs was detected by immunohistochemistry and reverse transcriptase polymerase chain reaction (RT-PCR). Results ACE2 was positively expressed in all the test tissues, including pancreas, heart, liver, kidney, lung, spleen, duodenum, ileum and bladder. The expression of ACE2 mRNA was detected in whole pancreas and ACE2 protein was found in both endocrine tissue and exocrine tissue of rat pancreas, while in the endocrine tissue much less ACE2 protein was expressed. The absorbance value of exocrine tissue of rat pancreas increased significantly as compared to that in pancreatic islet. Conclusion Our data demonstrated a tissue specific pattern of ACE2 expression in rat pancreas and this indicated ACE2 might be present in islet of pancreas of human, which might cause the elevation of blood glucose levels in SARS patients. Since ACE2 works as a negative regulator of the angiotensin system, the expression of ACE2 protein in endocrine tissue of pancreas may facilitate the treatment of diabetes.
