2.Study on Content Determination of Coumarin in Fraxini Cortex Extract

Objective To establish a UV spectrophotometry method and an HPLC method respectively for the determination of the total content of coumarin and contents of four main constituents of coumarin in Fraxini Cortex extract.Methods UV spectrophotometry was used for the determination of the content of total coumarin in Fraxini Cortex extract. The reference substance was Aesculin, and the maximum ultraviolet absorption wavelength was 334 nm. The HPLC method was used to determine the contents of Aesculin, Fraxin, Aesculetin and Fraxetin in Fraxini Cortex extract, using gradient elution with acetonitrile-phosphate solution (0.01%) as mobile phase on Agilent ZORBAX SB-C18 chromatographic column (4.6 mm × 250 mm, 5μm) at room temperature.Results For the UV method, the linear range of the mass concentration of Aesculin was 5.76-23.04μg/mL (r=0.999 9), and the average recovery was 100.6% (RSD=1.8%). For the HPLC method, the linear ranges of the mass of Aesculin, Fraxin, Aesculetin and Fraxetin were 0.055 0-3.850 0μg (r=0.9997), 0.053 9-3.773 0μg (r=0.999 8), 0.060 0-0.660 0μg (r=0.999 9), and 0.056 2-0.618 2μg (r=0.999 9), respectively, and the average recoveries were 96.97% (RSD=1.26%), 100.80% (RSD=2.22%), 99.04% (RSD=2.47%), and 98.77% (RSD=1.94%), respectively.Conclusion Both of the two methods are simple, accurate and reliable, and can be used for the quality control of total coumarin and the main constituents of coumarin in Fraxini Cortex extract.
3.Survey of knowledge, attitudes and practice of preventing birth defects in married reproduction women

Objective To evaluate the effect of health education on preventing birth defects in community in terms of knowledge, attitude and practice (KAP) for married women in reproductive age. Method A self-designed general information form and a questionnaire on KAP of married women′s preventing birth defects were used to investigate the KAP among 300 married nulliparous women. Result The average rate of cognitive knowledge of birth defects was 60.8%, the average rate of cognitive behaviors was 59.2%, and the average rate of cognitive attitude was 67.4%. Conclusion In order to reduce the rate of birth defects and improve the quality of births, we should implement three-level prevention education system, enrich the health education methods to enhance the knowledge, attitude and behavior of married women.
4.Expression of aurora-A gene in ovarian cancer tissue and its clinicopathologic significance

Objective To detect the genetic amplification and protein expression characteristics of aurora-A in ovarian cancer and to interpret the role of aurora-A gene in course of onset,progression and regression phases of ovarian cancer.Methods The amplification of aurora-A gene was detected by quantitative PCR in 6 normal ovarian tissues and 8 ovarian cancer samples,and its protein expression was examined by immunohistochemistry in 6 normal ovarian tissues and 40 ovarian cancer samples,furthermore,the relationships between over-expression of aurora-A protein in ovarian cancer tissue and its pathologic classification,tissue differentiation,clinical phase,tumor proliferation trait and prognosis were analyzed.Results Quantitive PCR showed that aurora-A mRNA was significantly higher in 8 ovarian cancer samples than that in normal ovarian tissues(P0.05).Conclusion There are abnormal amplification and protein over-expression of aurora-A in ovarian cancer tissue,aurora-A probably play an important role in the onset and progression of ovarian cancer,and the novel biological treatment concerning aurora-A gene and its protein is probably a useful route for curing tumor.
5.The design and application of CCS examination system based on ASP.NET technology

6.Determination of Sildenafil Citrate Illegally Added in Kidney-yang Reinforcing Chinese Medicinal Tonics by RP-HPLC

OBJECTIVE: To determine the content of Sildenafil citrate illegally added in kidney-yang reinforcing Chinese medicinal tonics. METHODS: The samples were separated on ODS C18(250 mm?4.6 mm,5 ?m) with mobile phase consisted of methanol-water-acetic acid glacial-triethylamine(60∶40∶0.8∶0.3) at a flow rate of 0.8 mL?min-1.The UV detective wavelength was 290 nm and the sample size was 10 ?L. RESULTS: At a concentration range of 10~100 ?g?mL-1, Sildenafil citrate showed a good linear relation with peak area score. The mean recovery rate was 100.52%(RSD=1.20%). CONCLU-SION:The method can be used to determine Sildenafil Citrate illegally added in kidney-yang tonics.
7.Effects of modified Siwutang aqueous extract on mobilization and plantation of bone marrow cells in mice after acute myocardial infarction

Objective To investigate the effects of Modified Siwutang aqueous extract (mSWT) on bone marrow cell mobilization and plantation in mice after acute myocardial infarction (AMI).Methods The AMI model was established by ligating the left anterior descending artery in Kunming mice.Bone marrow cells were marked by injection of DAPI into the cavitas medullaris of shin bones.Mice were administered with mSWT 60 mg/kg by intragastric administration once a day after surgery.After haematoxylin-eosin and immunofluorescent staining,the plantation of bone marrow cells in AMI area and microvessel density were detected by histopathological assay.The EPCs proportion of peripheral blood was measured by flow cytometry.Results Compared with model mice,infarct size obviously was decreased after mSWT 60 mg/kg treatment,with EPCs ratio increased significantly.Inflammatory cell infiltration of myocardium was much less.Myocardium cellular degeneration and necrosis,fibrosis were reduced,which survival cardiac muscle tissue was increased.DAPI positive cells of plantation and survive were found in AMI region of mSWT group.The number of positive cells were increased in mSWT group than that of model group.There were lot of microvessels of vWF positive expresseion in mSWT than model group.Conclusion:mSWT significantly improved histopathology of AMI mice.The mechanism may be involved in moved bone marrow cells,homed to AMI area and promote angiogenesis.
8.Total ginsenosides improve cardiac function via promoting angiogenesis in a rat model of acute myocardial infarction

Aim To investigate the effects of total gin-senosides ( TG) on microvascular regeneration and car-diac function in rat after acute myocardial infarction ( AMI ) . Methods The acute myocardial infarction model was created with left coronary artery ligated in male Sprague Dawley rats. The model rats were ran-domly divided into sham, model, TG low and high dose groups. TG groups were injected into abdominal cavity with TG(20 mg·kg-1·d-1, 40 mg·kg-1· d-1 ) . Sham and model groups were injected with e-qual-volume normal saline. On the 35th day post-opera-tion, heart function was examined by color doppler ul-trasoundination. HE, Masson and immunohistochemis-trical staining were used to observe the histopathologi-cal changes of myocardium and micro vessel density. The level of vascular endothelial growth factor( VEGF) and basic fibroblast growth factor( bFGF) mRNA were detected by real-time PCR. Results Compared with the model group, high and low dose TG obviously de-creased the left ventricular end diastolic dimension, the left ventricular end-systolic dimension, the left ventric-ular end-diastolic volume and the left ventricular end-systolic volume(P<0. 05 and P<0. 01), and signifi-cantly increased the ejection fraction and the fractional shortening ( P <0. 01 ) . The histopathological changes of myocardium on myocardial infarction and fibrosis were dramatically reduced by TG. But ventricular wall was thicker. Two dose TG remarkably increased the expressions of VEGF and bFGF mRNA and micro ves-sel density of compositive CD31 + cells in ischemic myocardial tissue and around of infarct area(P<0. 01, P <0 . 0 5 ) . Conclusions TG could improve the car-diac function of acute myocardial infarction rat. The mechanism may be related to the upregulation of VEGF and bFGF gene expression, the promotion angiogene-sis, then the improvement of blood supply in myocardi-al infarction area.
9.Analysis on Quality of Life and Its Influencing Factors in Patients with Chronic Gastritis

Objective To analyze the status of quality of life and influencing factors in patients with chronic gastritis. Methods 124 chronic gastritis patients were measured by the Chronic Gastritis scale of Quality of Life Instruments for Chronic Disease (QLICD-CG) before and after treatments. The scores were compared by paired t-test,and the influencing factors were analyzed by independent sample t-test and One-Way ANOVA. Results There were significant differences in all domains and total scores between the before and after treatments. The quality of life of chronic gastritis patients were influenced by treatments,gender,marital status,occupation and economic conditions ( <0.05) . Conclusion While reasonable treatment is very important to improve quality of life of the chronic gastritis patients, some influencing factors should be addressed, and mental health education and psychological service should be strengthened.
10.Construction of an antibody microarray based on direct labeling strategy for microalbuminuria measurement

Objective To build an antibody microarray based on direct labeling strategy for microalbnrninuria measurement, and evaluate it's technical potentiality for clinical application. Methods Urine samples of diabetic patients were collected. Antibody microarrays were constructed by preparation of array support, array fabrication, then protein assay and data analysis were performed. Procedure conditions for each step especially the labeling of samples were optimized. The set-ups were evaluated in terms of sensitivity, specificity and reproducibility. Urinary albumin excretion in the samples was detected by fabricated protein array, which was compared to that detected with immunoturbidimetry. Results The signal intensity was best when protein quality ratio of pure albumin or urine sample against NHS-biotin was 2:1. A calibration curve with a correlation coefficient of 0.9995 was established. The lower limit of detection was 0.0617 mg/L. Interehip and intrachip variation studies conducted on patient urine demonstrated CVs as 6.78%-9.22% and 3.35%-7.59%, respectively. Compared with the immunoturbidimetry, the antibody microarray was able to detect the extremely lower grade albumin in urine samples. The correlation coefficient of the results obtained by the two methods was 0.9199 (P <0.01). Conclusion An antibody microarray based on direct labeling strategy for microalbuminuria measurement is successfully established, which is comparable to immunoturbidimctry in its accuracy and will have great potential for clinical use with its high throughput, sensitivity, specifity and reproducibility.
