1.Functions and characteristics of calcium-sensing receptorsand effects of CaSR on characteristics of adult stem cells

Calcium-sensing receptor(CaSR) is a widely distributed G-protein coupled receptor.The activated CaSR plays an important role in many kinds of signaling pathway regulation, such as Ca2+ signaling pathway.It not only maintains the body calcium balance, but also is involved in the regulation of a variety of cell secretion, proliferation, apoptosis, and ion channel opening processes.CaSR expression is involved in stem cell migration, adhesion and homing in hematopoietic stem cells and mesenchymal stem cells.The activated CaSR also regulated the function of itself and characteristics in stem cells through a variety of cell signaling pathways.We introduce the functions and characteristics of CaSR, the relationship between CaSR and disease, and review the effects of the biological characteristics on hematopoietic stem cells and mesenchymal stem cells.
2.Performance verification for high performance liquid chromatography detection of glycated hemoglobin

Objective To verify the detection performance of glycated hemoglobin (HbA1c)by using the BIO-RAD D-10 high performance liquid chromatography method,and to evaluate whether the detection performance can satisfy the requirements of the laboratory and clinical diagnosis and treatment.Methods The method of CLSI EP1 5A2 was used to verify the accuracy and preci-sion.The linear correlation and reference range were verified respectively by linear regression analysis and the method of the WS/T 402 2012.Results The calibration value of HbA1c calibrator level 1 was 5.3% and the validation limit of measured results was 1. 63%-8.78%.The calibration value of HbA1c calibrator Level 2 was 10.0%,and the validation limit of measured results was 4.76%-13.64%.Both calibration values were included in the 95% confidence range of measured values,so the accuracy verifica-tion passed.Laboratory variation coefficient (CV)for HbA1c with concentration of 5.24% was 1.01% and for 9.84% was 0. 54%,both less than the precision that the manufacturer declared,so the precision verification passed.The equation of linear regres-sion analysis for HbA1c linearity calibrator measured value and target value wasY =1.008X +0.023,with correlation coefficient in-dex r 2 of 0.999,so the linearity was good.The measured results of 20 healthy adults specimens were in the reference interval(4.1%-6.2%)that the manufacturer provided.Conclusion Accuracy,precision,linear correlation and reference range of HbA1c meas-ured by BIO-RAD D-10 HPLC detection method were verified.It can provide the accurate and credible measurement results for clin-ical diagnosis and treatment,and can be used in the detection of clinical specimens.
3.Compatible Stability of Moxifloxacin Hydrochloride and Sodium Chloride Injection with Four Kinds of Antihemorrhagic Drugs

OBJECTIVE:To observe the compatibility of moxifloxacin hydrochloride and sodium chloride injection with four kinds of antihemorrhagic drugs(etamsylate injection,aminomethylbenzoic acid injection,vitamin K1 injection,and aminocaproic acid injection).METHODS:The concentration of moxifloxacin in the mixture of Moxifloxacin Hydrochloride and sodium chloride injection with four kinds of antihemorrhagic drugs at 25℃ and 37 ℃ were determined by UV-spectrophotometry and dual wavelength spectrophotometry within 8 h.The changes of the mixture in appearance and pH were also observed.RESULTS:The mixture of moxifloxacin hydrochloride and sodium chloride injection with four kinds of antihemorrhagic drugs experienced no obvious changes in both contents of moxifloxacin and pH and appearance.CONCLUSION:The mixture of moxifloxacin hydrochloride and sodium chloride injection with four kinds of antihemorrhagic drugs was stable in contents of moxifloxacin and property.However,further studies remain to be done to investigate whether there are changes in the contents of the other four injections.
4.Performance verification of chemiluminesent micropaticle immunoassay for 6-indicator sex hormones

Objective To verify and evaluate the methodological performance of chemiluminesent micropaticle immunoassay(CMIA) in the ARCHITECT i2000SR analyzer for detecting estradiol(E2),progesterone(Prog),luteinizing hormone(LH),follicle stimulating hormone(FSH),prolactin(PRL) and testosterone(Tes).Methods The precision,accuracy,linearity range,reference interval and low detection limit in the 6-indicator sex hormones detection were verified according to the CLSI related documents.The results were compared with the performance indicators or acceptable quality target claimed by the manufacturer.Results The within-run imprecision of 6-indicator sex hormones was ≤2.62%,the total imprecision was ≤3.17%.Accuracy was within ±9.41%.The slope(a) within linearity was 1.00 ±0.05,and the correlation coefficient(r2) was >0.995.The coincidence rate of reference interval was ≥90%.The low detection limits were less than the indicator claimed by the manufacturer.Conclusion The methodological performance of 6-indicator sex hormones detected by the ARCHITECT i2000SR analyzer is satisfactory,reliable in detection results and suitable for clinical needs.
5.Establishment of an animal model for coxsackievirus A16 infection related immunological evaluation

Objective To establish a simple and reliable experimental rodent model sensitive to coxsackievirus A16 ( CVA16) .Methods Mongolian gerbils with different age were selected and inoculated intraperitoneally with live CVA16, and the gerbils were observed daily until 14 days postinoculation to screen for the most optimal ages sensitive to the virus.The dose-dependent symptoms were evaluated and the 50% lethal dose (LD50) was determined.The virus titers were measured in blood and various tissues of CVA 16-infected Mongolian gerbils 3 days post-infecton.Finally, the gerbils were immunized twice with inactivated CVA 16 vaccine at day 1 and day 11, respectively, followed by challenge with the virus with a dose of LD50 at day 14.The gerbils were then observed for another 2 weeks to record their body weight , symptom and mortality rate .Their blood samples were collected from the eyes , and CVA16-specific neutralizing antibodytiters and total antibody titers was checked by microneutralization test and ELISA , respectively .Results Various clinical symptoms, such as inactivity, hind limb weakness, paralysis and even death occurred in gerbils following CAV 16 infection. 7-day-old and 14-day-old gerbils are susceptible to CVA 16 infection whereas 28-day-old gerbils are resistant .The most sensitive and appropriate age is 14-day-old.The 50%lethal dose was determined to be 1×104.5 CCID50.High titers of the virus were confirmed in blood and various tissues of Mongolian gerbils contracted CAV 163 days post-infecton.The survival rate is 87.5%for 14-day-old gerbils preimmunized with two doses of inactivated CVA 16 vaccine and challenged with the virus.The geometric mean titers ( GMTs) of neutralizing antibody was 28.14, and the seroprevalence was 87.5%.Conclusions Mongolian gerbils is sensitive to CVA16 and the virus reproduces actively in Vivo.Thus, it can be used as a reliable small animal model for studies of CVA 16 pathogenesis , vaccine development and drug evaluation .
6.Effects of Ginkgo biloba extract EGb761 on the expressions of XIAP and Smac following focal cerebral ischemia/reperfusion in rats

Objective To investigate the effects of Ginkgo biloba extract EGb761 on the expressions of XIAP and Smac following focal cerebral ischemia in rats. Methods A total of 40 male Wistar rats were randomized into 4 groups: sham-operation group, cerebral ischemi-a/reperfusion group, low-dose EGb761 group, and high-dose EGb761 group (n=10 in each group). A rat model of middle cerebral artery occlusion (MCAO) for 1.5 hours and reperfusion for 24 hours was built. EGb761 50 mg/kg and 100 mg/kg were injected intraperitoneally at one hour before the model building in the low-dose EGb761 and high-dose EGb761 groups. The expressions of XIAP and Smac in brain tissue were detected by immunohistochemistry method. Results The expressions of XIAP were 18. 33±4. 01 and 26. 7±3.27 respectively in the low-dose EGb761 and high-dose EGb761 groups, and they were significantly higher than 12. 13±3.44 in the cerebral ischemia/reperfusion group (all P<0.01), and the high-dose EGb761group was higher than the low-dose EGb761 group (P<0.01). 1he expressions of Smac in brain tissue were 21.33±3.15 and 11.33±2. 10 respectively in the low-dose EGb761 and high-dose EGb761 groups, and they were significantly lower than 28.93±4. 96 in the cerebral ischemia/reperfusion group (all P<0.05). The high-dose EGb761 group was significantly lower than the low-dose EGb761 group (P<0.05). Conclusions Cerebral ischemia/reperfusion could induce the expressions of XIAP and Smac. EGb761 intervention could inhibit the expressions of Smac while upregulating the expression of XIAP, and increase the XIAP/Smac ratio. 1his may be one of the protective mechanisms of EGb761 intervention.
7.Clinical Observation of Oral Use and Acupoint Application of Chinese Herbal Medicine for Strengthening Spleen and Regulating Stomach, Lowering Rebellious Qi to Stop Vomiting in Treating Spleen-stomach Deficiency Pregnant Pernicious Vomiting

Objective To observe the clinical effect of oral use and acupoint application of Chinese herbal medicine with the actions of strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting in treating pregnant pernicious vomiting. Methods One hundred and twenty-two spleen-stomach deficiency pregnant pernicious vomiting cases were randomly divided into observation group ( N=63) and control group (N=59). At the end of the trial, 2 cases of the control group dropped out, and the total case number was 57 in the control group. Both groups were treated with intravenous infusion, dietary care, daily life and emotional nursing, and oral use of Chinese herbal medicine with the actions of strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting. Additionally, the observation group was given acupoint application of herbs for strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting. The treatment was performed once a day, 4 days constituted one treatment course, and the treatment lasted two continuous courses. The improvement of vomiting, therapeutic effect and the safety were compared between the two groups after treatment. Results ( 1) The total effective rate was 96.8%in the observation group and was 71.9%in the control group, the difference being significant (P<0.01). (2) The time for stopping vomiting in the observation group was significantly less than that in control group (P<0.01). (3) During the treatment, no obvious adverse reaction was found in the two groups. Conclusion Acupoint application of herbs for strengthening spleen and regulating stomach, lowering rebellious Qi to stop vomiting has obvious synergistic effect on the clinical efficiency of spleen-stomach deficiency pregnant pernicious vomiting treated by oral use of herbal medicine, intravenous infusion, dietary care, and daily life and emotional nursing.
8.Prediction and identification of immunodominant B cell linear epitopes in capsid proteins VP1 to VP3 of Coxsackievirus A16

Objective To identify immunodominant B linear cell epitopes in capsid proteins VP1 to VP3 of Coxsackievirus A16 (CVA16) strain YY157.Methods The protean algorithms of bioinformatic software Lasergene were used to analyze antigenicity, hydrophilicity and surface probability of amino acid sequence of CVA16 capsid proteins VP1 to VP3.Multiple regions containing potential lineal B cell epitopes were predicted and their corresponding average indexes were calculated by BepiPred 1.0 Server.Corresponding peptides were synthesized and examined in peptide-enzyme-linked immunosorbent assay (ELISA) individually to check whether it reacted positively or negatively toward sera from children with confirmed CVA16 infection.Results Totally 21 possible B cell linear epitopes were predicted according to their relatively strong antigenicity, hydrophilicity and surface probability.The corresponding synthetic peptides reacted positively with sera of CVA16-infected children in a varying extent.Conclusion Immunodominant B cell linear epitopes of capsid proteins VP1 to VP3 of CVA16 strain YY157 are successfully predicted and confirmed.
9.Radiation protection effect and its mechanism of multiple nilestriol administrations on the mice with bone marrow type of acute radiation syndrome

Objective To understand the radiation protection effect of pre-irradiation administrations of nilestriol on the mice with bone marrow type of acute radiation syndrome after irradiation with 60Co γ-rays,along with its mechanisms for improvement of hematopoiesis.Methods The nilestriol administration protocols were prepared by analysis of peripheral blood cell counts and survival rate experiment on mice.The mechanisms by which the pre-irradiation twice administrations improved the post-irradiation recovery of bone marrow hematopoiesis were studied by the analysis of the surface marker of bone marrow hematopoietic stem/progenitor cells of mice and by the inspection of hematopoietic progenitor cell colony and by using histopathological assessment of bone marrow.Results Pre-irradiation administration of nilestriol at two-or three-day intervals had been shown to increase survival rates up to 100% in mice exposed to 9.0 Gy γ-rays,which was superior to a single administration (20%,x2 =21.66,21.66,P <0.05).The pre-irradiation administration both at one-day or two-day intervals were capable of improving the recovery of peripheral blood counts,including white blood cell (WBC),red blood cell (RBC),and platelet in mice exposed to 6.5 Gy (F =21.33,100.9,49.34,19.19,P < 0.05),showing the better effects than a single administration (F =17.11,63.38,21.89,14.37,P < 0.05).The two-day-interval administration of nilestriol could significantly increase the numbers of bone marrow hematopoietic stem/progenitor cell counts (t =8.58,2.80,P < 0.05) in mice on day 10 after 6.5 Gy irradiation.This also could be capable to significantly improve colony formation,with there being statistical difference compared with single administration(t =4.29,6.34,P < 0.05).Also the administration at two-day-interval were also usefull in reconstruction of hematopoietic cell hyperplasia of bone marrow of irradiated mice.Conclusions As compared with conventional single admination,the pre-irradiation multiple administrations of nilestriol showed significantly improved radiation protection effects.Considering a nuclear medical emergency rescue,it is recommended to follow the pre-irradiation administration of nilestriol at two-day interval,which could obtain the best protection effects at minimum administration frequency.
10.Reconstruction of laryngotracheal defect using the musculo-periosteum flapof the sternocleidomastoideus with clavicular periosteum

Objective:To investigate the effect of the musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum on the reconstruction of extensiveness laryngotracheal defect. Method: Retrospectively studied 49 patients, who admited in our department from 1996 to 2005 years for severe laryngotracheal defect. There were 28 males and 21 females aged from 15 to 69 years old (mean age was 47 years old). The causes of laryngotracheal defect were laryngotracheal stenosis(31 cases) and surgery for thyroid carcinoma invading trachea (18 cases). All of 49 cases were treated with the graft of musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum and placed a silicon T-tube stenting for 3 to 6 months. Result; All of 49 cases were successfully decannulated with recuperative normal airway patency and effective phonation. The follow up ranged from 2 to 10 years, and the effect of operation was steady. Conclusion:The musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum is an ideal graft for the reconstruction of cervical extensiveness tracheal defect.
