1.Analysis of GDNF(△N39)-R9 Fusion Protein Delivery Across The Cellular Membrane,Blood Brain Barrier and Biological Function

In order to study the application of glial cell line-derived neurotrophic factor(GDNF)in clinic,gene mutation,fusion protein expression in E.coli and purification methods have been used to obtain the fragments of GDNF,GDNF(△N39),GDNF(△N39)-R9.Using primary cultured dopaminergic neurons and PC12 cells with transfected with GFR?1 and Ret to observe their biological function and cytotoxicity.Using B-Endo3 cells and Transwell method to analyze their delivery across the cellular membrane and blood brain barrier.The results show that GDNF(△N39)-R9 has the same neurotrophic function with wild GDNF and nearly no cytotoxicity to dopaminergic neurons and PC12-GFR?1-Ret cells and can get through effectually the cellular membrane and simulacrum of blood brain barrier with matrigel and B-Endo3.
2.Differentiation of Cortex Fraxini and its three kinds of confusable species

Object To provide a scientific basis for identification and rational use of the herbal medicine by the pharmacognostic study on Cortex Fraxini. Methods Microscopic identification, TLC, HPLC were used. Results There were remarkable differences in the genuine product, confusing product and fakements of Cortex Fraxini in characteristics of microscopic identification, fluorescence, TLC and HPLC. Conclusion The scientific basis has been established and provided for differentiation of confusing product and fakements.
3.Determination of aesculin and aesculetin in periderm and leaves of Cortex Fraxini among different provenances

Objective: To determine the contents of aesculin and aesculetin in periderms and leaves of Cortex Fraxini among differnet provenances. Methods: HPLC was used with acetonitrile-water(15∶85) as the mobile phase and detected wavelength at 348nm. C 18 column was adopted. Results: The calibration curves were linear. The value of correlation coefficient were 0.9992 and 0.9994, respectively. The average recovery of aesculin was 98.2% and aesculetin was 99.2%. RSD were 2.24% and 2.15%, respectively. The quality differences of Cortex Fraxini among different provenances were remarked. The quality of Cortex Fraxini from Shanxi province was the best. Conclusion: The method is applied in determination and analytics of content of Cortex Fraxini and is rapid, simple and easy to carry out. The method is with the feature of accuracy, repetition and stability.
4.Molecular mechanisms for arecoline against atherosclerosis

AIM To investigate the anti-atherosclerosis mechanisms of arecoline. METHODS Fifty-eight male rats were assigned to control, model, arecoline 1 mg?kg -1 and arecoline 5 mg? [FQ(12。46,X-WZ]-kg -1 groups randomly. The rats in model and arecoline-supplied groups were fed with hypercholesterol diet. The following variables were measured: HE staining of rat aorta; serum level of TC, LDL-CHO, HDL-CHO, IL-8, ET-1 and NO; expression of eNOS protein on rat aorta; expression of MCP-1, ICAM-1, CXCR-2 and eNOS mRNA on rat aorta. RESULTS Arecoline increased plasma level of NO and the expression of eNOS protein and mRNA, decreased plasma level of IL-8 and the expression of ICAM-1, MCP-1 and CXCR-2 mRNA on rat aorta. CONCLUSION The anti-atherogenic effects of arecoline seems to be closely involved in increasing plasma level of NO and eNOS protein and mRNA expression, decrease in plasma IL-8 level and down-regulation of the expression of ICAM-1, MCP-1 and CXCR-2 genes.-
5.Molecular mechanisms for the novel compound PPVP against atherosclerosis in rat fed with hypercholesterol diet

AIMTo investigate the anti-atherosclerosis mechanisms of the novel compound PPVP. METHODSThirty-six male rats were a ssigned to control, model and PPVP 5 mg?kg -1 groups randomly. The rats in model and PPVP 5 mg?kg -1 groups were fed with hypercholesterol diet for inducing atherolcerosis model. The following variables were measured: HE stainin g of rat aorta; serum level of TC, LDL-CHO, HDL-CHO, IL-8, ET-1, PGI 2, TXA 2 and NO. RESULTSPPVP increased serum level of NO, decreased plasma level of TXA 2 and inhibited the over expression of IL-8. CONC LUSIONThe anti-atherogenic effect of PPVP seems to be closely involved in increasing serum NO level, decreasing plasma level of TXA 2, and inhibiting over expression of IL-8. The result also suggests that the anti-atherogenic ef fect of PPVP in high cholesterol-fed rat may not due to the regulation of plasm a lipid profile, plasma level of ET-1 and PGI 2.
6.Effects of bone marrow mesenchymal stem cells and benefiting-Qi nourishing-Yin and dissolving-congestion prescription on lower extremity arterial restenosis in model dogs with diabetes mellitus

BACKGROUND:At present, a great quantity of research has shown the effectiveness of traditional Chinese medicine and bone marrow mesenchymal stem cel s for vascular restenosis. However, studies concerning their combined application to restenosis after percutaneous transluminal angioplasty with diabetes mel itus are presently lacking.
OBJECTIVE:To observe the effects of combined application of bone marrow mesenchymal stem cel s and benefiting-Qi nourishing-Yin and dissolving-congestion prescription on restenosis after percutaneous transluminal angioplasty in dogs with diabetes mel itus.
METHODS:A dog model of vascular restenosis with diabetes mel itus was established by bal oon injury of femoral artery and intravenous injection of al oxan. After successful model induction, 22 dog models were randomly divided into three groups:model group (n=6), treatment with Chinese medicine (n=8), and combined treatment with bone marrow mesenchymal stem cel s and Chinese medicine (n=8). Serum vascular endothelial growth factor levels were measured using enzyme-linked immunosorbent assay preoperatively and at 1, 2, 4 and 8 weeks postoperation. Samples of vessels were taken to conduct pathomorphological observation and quantitative analysis of proliferation degree. Tissues, including heart, liver, kidney and pancreatic gland, were col ected to evaluate the safety of stem cel transplantation using hematoxylin-eosin staining at 8 weeks postoperation.
RESULTS AND CONCLUSION:Serum vascular endothelial growth factor levels began to increase at 1 week postoperation in the Chinese medicine group and combined treatment group, at 4 weeks postoperation in the model group compared with preoperation (P<0.05). At al time points, serum vascular endothelial growth factor levels were highest in the combined treatment group, but lowest in the model group (P<0.05). Quantitative analysis of vascular proliferation demonstrated that at 8 weeks postoperation, new intimal area, new intimal/medial areas and stenosis rate were highest in the model group, but lowest in the combined treatment group at 8 weeks postoperation (P<0.05). Safety assessment of stem cel transplantation showed morphological structures of the heart, liver, kidneys and pancreas were normal, no necrosis. In a word, the effects of the combined application of bone marrow mesenchymal stem cel s and benefiting-Qi nourishing-Yin and dissolving-congestion prescription were much pronounced in preventing restenosis after percutaneous transluminal angioplasty in dogs with diabetes mel itus rather than single therapy of Chinese medicine. It is a safe and effective treatment to prevent vascular restenosis after percutaneous transluminal angioplasty in dogs with diabetes mel itus.

8.Reconstruction of laryngotracheal defect using the musculo-periosteum flapof the sternocleidomastoideus with clavicular periosteum

Objective:To investigate the effect of the musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum on the reconstruction of extensiveness laryngotracheal defect. Method: Retrospectively studied 49 patients, who admited in our department from 1996 to 2005 years for severe laryngotracheal defect. There were 28 males and 21 females aged from 15 to 69 years old (mean age was 47 years old). The causes of laryngotracheal defect were laryngotracheal stenosis(31 cases) and surgery for thyroid carcinoma invading trachea (18 cases). All of 49 cases were treated with the graft of musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum and placed a silicon T-tube stenting for 3 to 6 months. Result; All of 49 cases were successfully decannulated with recuperative normal airway patency and effective phonation. The follow up ranged from 2 to 10 years, and the effect of operation was steady. Conclusion:The musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum is an ideal graft for the reconstruction of cervical extensiveness tracheal defect.
9.Use of silicon T-tube in laryngotracheal reconstruction.

OBJECTIVE:
To discuss the function and operative method of silicon T tube in laryngotracheal reconstruction.
METHOD:
Two hundred and ninety-seven patients of laryngotracheal stenosis were operated with laryngotracheal reconstruction in our department. All of patients were tracheostomy dependent before reconstruction and were placed a silicon T-tube stenting for 3 to 6 months after reconstruction.
RESULT:
Two hundred and eighty-nine patients (97.3%) were successfully decannulated with good airway patency and effective phonation. They were followed up from 1 to 10 years, and no recurrence was found.
CONCLUSION
Silicon T-tube is an effective and safe stent for laryngotracheal reconstruction. Paying attention to some application details may avoid the complication and obtain a satisfactory effect.
