1.SYNTHESIS AND ANTIFUNGAL ACTIVITY OF SOME IMIDAZOLYL OXIME ETHER DERIVATIVES

Oxiconazole, a new broad-spectrum antifungal agent, was synthesized accordi ng to the method reported by Mixich G et al and some improvement was made in the preparation of N- (2, 4-dichlorobenzoylmethyl) imidazole, an important intermediate.In an effort to search for more effective antifungal drugs by chemical modification, several imidazolyl oxime ether derivatives were synthesized.The structures of the synthetic products were proved by elemental analysis.In vitro fungistatic tests showed that compounds Ⅰ-Ⅷ were more effective than clotri-mazole against most of the fungal strains tested and that compound Ⅲ possessed antifungal action against Aspergillus while others didn't.The structure of compound I(oxiconazole)was confirmed by UV, IR, 1H-NMR and MS.Its X-ray powder diffraction data are also provided.In a clinical trial,94.10% of the patients with tinea cruris were cured after a two-week oxiconazole treatments cure rate significantly higher than that of econazole(84.24%, P
2.Experimental study on anti-proliferation of mitomycin C-chitosan microspheres sustained release microspheres during glaucoma filtering surgery in rabbit

Objective To study the effect of mitomycin C(MMC)‐chitosan (CS)sustained release microspheres on the scar‐ring prevention of the filtering passage during glaucoma filtering surgery .Methods Filtering surgery model was established on 96 eyes of 48 healthy adult New Zealand white rabbits which were divided into 4 groups ,24 eyes in each group .The right eyes implan‐ted MMC‐CS microspheres in 24 rabbits(group A) ,whereas that of the fellow eyes implanted empty CS microspheres(group B) , and in other 24 rabbits which right eyes only used 0 .20 mg/mL MMC cotton(group C) ,on the opposite sides with no adjunctive treatment(group D) .Intraocular pressure ,filtering bleb ,anterior chamber flare ,complications and the numbers of corneal endotheli‐al cells were observed after surgery .Rabbits were killed on the 7th ,14th and 21th day postoperatively in batch and histopathological examination was carried out .Results (1)the intraocular pressure:group A can maintain the low level of intraocular pressure for a long time ,followed by group C ,group B and group D ,and difference between groups was statistically significant (P< 0 .01) .(2) The filtering blebs :the filtering bleb survival time of group A was longer than other group(P<0 .01) .(3)Complication:there was not any complication in the 4 groups .(4)The number of corneal endothelial cells :the number of corneal endothelial cells of group A preoperative and postoperative had no statistical significance differences(P>0 .05) .(5)Histopathologic findings :group A had bet‐ter conjunctival epithelial integrity ,subconjunctival fibroblast less than other groups ,filtration had no obvious inflammation infiltra‐tion around the mouth area ,but also had fewer new collagen fibers .Conclusion MMC‐CS sustained release microspheres is a safe and effective treatment to inhibit inflammatory cells activity and fibroblast activity in surgery sites ,and can significantly improve outcome of filtration surgery .
3.ANTIINFLAMMATORY ACTIVITY OF BUFEXAMAC ON TOPICAL SKIN INFLAMMATION MODEL INDUCED BY FORMALDEHYDE

The paper reported the topical skin antiinflammatory activity of bufexamac on inflammation model induced by formaldehyde in mice. Comparing with the blank cream, the effect of 0.5% bufexamac cre-am on inflammation had no statistically difference, but all those of 1% bufexamac cream at 1 and 2 h after treatment, of 2 % and 5 % bufexamac cream at 1, 2 and 4h after treatment, showed significantly difference. The antiinflammatory activity was enhanced with the increase of bufexamac concentration in cream. The antiinflammatory activity of 2 % bufexamac cream was close to the effects of 1 % indomethacin cream and 0.05 % clobetasol Cream.Furthermore, our study showed that steroid antiinflammatory agents and nonsteroid antiinflammatory agents were all active against erythema and edema produced by formaldehyde in mice. Formaldehyde and mice are cheap and easy to obtain in our country. It is of some value to use the animal model for screening the topical skin antiinflammatory agents.
4.Preparation and Quality Control of Anti-Microbicide Contraceptive Gel

OBJECTIVE:To prepare anti-microbicide contraceptive gel and establish its quality control method. METHODS: The gel was prepared with octoxynol and policresulen as main ingredients and HPMC as matrix. The content of octoxynol in the gel was determined by HPLC,and the content of policresulen in the gel was determined by titration method. RESULTS:Prepared gel was well-proportioned and of good viscidity and its identification and test were up to the standard. The linear rang of octoxynol was 250~1 500 mg?L-1(r=0.999 6)with an average recovery of 100.84%(RSD=0.74%,n=9). The average labeled amount of policresulen was 106.64%(n=3). CONCLUSION:This preparation is feasible and stable in quality,and the quality control method is simple and accurate.
5.Change of G?q/11-mediated signal transduction pathway in aorta of spontaneously hypertensive rats

AIM: To investigate the changes of G?q/11 and phospholipase C(PLC) of the aorta and to evaluate the role of signal transduction pathway mediated by G?q/11 in the pathogenesis of spontaneous hypertension. METHODS: Blood pressure was measured by intubation of carotid, and the level of plasma angiotension Ⅱ was measured by radioimmunoassay. In addition, G?q/11 and PLC contents in aorta were determined by Western blot analysis. RESULTS: Arterial blood pressure was not changed in 4-week-old spontaneously hypertensive rats(SHR), but it was increased markedly in 12-week-old SHR. 6.Management of chronic acromioclavicular joint dislocation with hook plate and fascia lata graft

Objective To evaluate the therapeutic efficacy of hook plate and fascia lata graft in management of chronic acromioclavicular joint dislocation.Methods From May 2012 to March 2014,a total of 16 cases of chronic acromioclavicular joint dislocation (Rockwood type Ⅲ) were treated with hook plate and fascia lata graft.They were 10 men and 6 women,from 19 to 57 years of age (mean,34.7 years).Seven cases were at the left side and 9 at the right side.All injuries resulted from direct violence.The time from injury to surgery ranged from 8 to 54 weeks (mean,12.4 weeks).At the final follow-ups,the shoulder function was assessed using the Quick Version of the Disabilities of the Arm,Shoulder and Hand Score (DASH) and visual analogue scale (VAS),and the efficacy using Karlsson's evaluation system.Results All surgical wounds primarily healed.The dislocation of acromioclavicular joint was corrected,without any vascular or neural damage.The patients were followed up for an average of 12 months (from 6 to 18 months).No plate breakage,screw loosening or redislocation was observed.The DASH and VAS scores of the shoulder at the final follow-ups (25.42 ± 8.34 and 2.22 ± 1.70,respectively) were significantly improved,compared with the preoperative ones (43.72 ± 12.35 and 6.33 ± 1.37,respectively) (P < 0.05).According to the Karlsson's criteria,10 cases were excellent,4 good and 2 moderate.Conclusion In management of Rockwood type Ⅲ chronic acromioclavicular joint dislocation,hook plate and fascia lata graft can restore dynamic balance and stability of the acromioclavicular joint,resulting in solid fixation,early joint motion and limited complications.

AIM: To investigate the crosstalk between angiotensin Ⅱ (AngⅡ)-mediated and platelet-derived growth factor (PDGF)-mediated signal transduction in vascular smooth muscle proliferation.METHODS: A model of renal hypertension was made by two kidney/one-clip operation. Objective To detect the expression of stem-cell related factors Nanog and CD44 in spheroid body-form?ing cells of gastric cancer cell line MKN-45. Methods The gastric cancer cell line MKN-45 was used to culture spheroid bodies in non-adherent condition in a serum-free medium supplemented with epidermal growth factor (EGF) and basic fibro?blast growth factor (bFGF). Using Western blot analysis, immunofluorescence staining and quantitative real time polymerase chain reaction(qRT-PCR), the expression levels of stem cell-related genes Nanog and CD44 were studied. Results In this study, we observed that MKN45 cells formed spheroid bodies in non-adherent condition in a serum-free medium, and the levels of Nanog and CD44 mRNA expression in spheroid body-forming cells were 2.34 ± 0.22 and 1.18 ± 0.04,respectively, which were higher than those in parental cells (1.00±0.00 and 1.00±0.05). The levels of Nanog and CD44 protein expression in spheroid body-forming cells were 0.18±0.02 and 0.24±0.04, respectively, which were significantly higher than those in pa?rental cells (0.07±0.02 and 0.18±0.01, P<0.05). Nanog protein was positively stained within the perinuclear and cytoplasm of the spheroid body-forming cells, and CD44 was positively stained mainly in the membrane. Dual staining of Nanog/CD44 indicated that the embryonal protein Nanog was co-localized with CD44 in the spheroid body-forming cells. Conclusion Spheroid body-forming cells developed from human gastric cancer cell line MKN-45 in serum-free medium supplemented with EGF and bFGF show characteristics of cancer stem cell (CSC). The cells co-expressed of CD44 and Nanog maybe a phe?notype of gastric CSCs.
9.Factors influencing the reception of home care services of old patients with chronic diseases

Objective To explore the needs of the elderly patients for home care services,explore factors affecting their reception of home care services.Methods A self-designed questionnaire was used to investigate the needs of the elderly patients with chronic diseases for chronic home care services and the influential factors.Results 92.3%of the elderly patients with chronic diseases in the study needed home care services.Medical insurance,financial problems and caregiver’s quality were the key factors affecting their reception of home care,accounted for 95.5%,84.8%and 80.3%respectively.Conclusions Elderly patients with chronic diseases need home care.We should meet the needs of patients with chronic diseases by understanding their needs,carrying out multifaceted cooperation with governments and institutions,completing medical insurance system and expanding healthcare services.
10.Clinical study of paclitaxel liposomes and paclitaxel in the treatment of advanced esophageal cancer

Objective To compare the safety and clinical efficacy between paclitaxel liposomes and paclitaxel in patients with advanced esophageal cancer .Methods A total of 90 patients with advanced esophageal cancer were enrolled into this study and were randomly divided into paclitaxel liposomes treatment group(treatment group) and paclitaxel treatment group(control group) . The patients of each group were treated with paclitaxel liposomes or paclitaxel 80 mg/m2 intravenously on day 1 and 8 ,and nedapla‐tin 75 mg/m2 intravenously on day 1 ,respectively .After two cycles of chemotherapy ,three weeks per period ,we evaluated the short term efficacy and adverse reactions according to the WHO standard .Results The short term efficacy between control group and treatment group showed no significance statistically (P>0 .05) .However ,the incidence of allergic reactions in the treatment group was significantly lower than control group (P<0 .05) .Conclusion Paclitaxel liposomal or paclitaxel combine with nedaplatin was effective equivalently in the treatment of patients with advanced esophageal cancer .But the allergic reactions of paclitaxel liposomal were lower than paclitaxel .It is worthy of clinical promotion .
