1.Damage to the central noradrenergic system and changes of P3-like cognitive potential in rats

BACKGROUND: It is well known that central noradrenergic system takes part in learning and memory, but there are a few researches focused on the effects of impairing the central noradrenergic system on rat' s P3-like cognitive potential.OBJECTIVE: To study the effects of impairing the central noradrenergic system on rat's P3-like cognitive potential.DESIGN: Randomized controlled study based on the experimental animals.SETTING: Department of physiology in a medical college of a university.MATERIALS: Totally 30 male SD rats, clean degree, weighing from 500 to 600 g, were selected from the Experimental Animal Center of Sun Yat-sen Medical University(certificate: 26-99A018), drank freely and fed timely under the normal condition. After Y-maze test of all the rats, 24 rats with normal learning and memory function, which met the criteria specified in the description of Y-maze test, were randomly divided into three groups(8 rats in each group): control group, 6-oxidopamine (6-OHDA) injection group and saline control group.INTERVENTIONS: The models of central norndrenergic system impairment were made by injecting 6-hydroxysopamine(6-OHDA) into the bilateral dorsal noradrenergic bundle in dentate granule(DG). Then Y-type maze test and elicitation of P3-likewere carried out respectively before and after the models were built.MAIN OUTCOME MEASURES: Main outcome: P3-like latency, total reaction time(TRT) and error number(EN). Secondary outcome: Results of P3-like test recorded in the 12 days after the models were built.RESULTS: As compared with the controls, the model rats' P3-like latency and total reaction time(TRT) prolonged and error number(EN) increased statistically ( P < 0.05 ).CONCLUSION: Central NE takes part in the activities of learning and memory and plays a certain role in the production and integration of P3 -like.
2.Clinical effect of mifepristone combined with misoprostol for termination of pregnancy in high risk pregnant women

Objective To explore the clinical effect of mifepristone combined with misoprostol for termina-tion of pregnancy in high risk pregnant women and provide reference for clinical treatment.Methods 182 cases of high -risk maternal were divided into observation group and control group by the random number table method,each group 91 cases.The control group was treated with conventional abortion termination of pregnancy,the observation group was treated with the artificial abortion before oral administration of mifepristone tablets and misoprostol tablets. The operation time,the amount of blood loss,the degree of pain,the degree of cervical softening and the occurrence of complications of induced abortion were compared between the two groups.Results The operation time of the observa-tion group was (2.21 ±1.41)min,intraoperative bleeding was (15.29 ±4.06)mL,which were significantly less than those of the control group (t =7.122,8.256,all P <0.05);The pain degree grade I in the observation group was 58.24%,which was significantly higher than that in the control group,grade III accounted for 5.49%,which was sig-nificantly lower than the that in the control group (χ2 =44.887,44.121,all P <0.05),the observation group of cer-vical softening full was 59 cases,full softening rate was 64.84% and the control group in the cervical softening full was 41 cases,softening full rate was 45.05%.Fully softening rate in the observation group was significantly higher than that in the control group (χ2 =7.191,8.411,P <0.05),the control group,the incidence of uterine perforation in 2 cases,the occurrence of artificial abortion syndrome 7 cases,the incidence of complications for 9.89%,the obser-vation group had no uterine perforation occurred,1 case of artificial abortion syndrome and complications occurred rate was 1.10%.The incidence of complications in the observation group was significantly lower than that in the control group (χ2 =6.772,P <0.05).Conclusion Mifepristone combined with misoprostol for high -risk maternal termi-nation of pregnancy could significantly shorten the artificial abortion operation time,reduce intraoperative blood loss, reduce the patient pain,reduce the incidence of complications.

7.Construction of hepatitis B virus C gene retroviral vector and its expression in immortalized human peripheral blood B cell lines

Objective In order to study the biological significance of HBV/C gene variant in vitro.Methods Retroviral vector pXT1 was used to construct the HBV/C gene expression vector and the recombinant plasmid pXT1-HBV/C was used to transfect immortalized human peripheral blood B cell lines to express HBcAg steadily in the host cells.Results plasmid pXT1-HBV/C was detected positive by PCR as well as enzyme digestion with Bgl Ⅱ and Xho Ⅰ.Meanwhile.transfected cells was detected to contain HBV/C gene by PCR and HBcAg was expressed in 47.4%of the ceils by means of flow cytometry.Conclusion Retroviral expression vector with HBV/C gene can transfeet into eucaryotic cells effectively and express the goal gene steadily.The recombinant cells may be used in the systematic studies on the biological significance of HBV/C gene variant.

Tyrosine phosphorylation is a key post-translational mechanism that regulates cellular processes and maintains homeostasis.Aberrant changes in tyrosine phosphorylation are often associated with disease states such as metabolicdisorders,cancer and cardiovascular disease.Protein tyrosine phosphatases (PTPs) are the enzyme family that regulates protein phosphorylation level of tyrosine residues in the cellular processes and signaling ways.So far,scientists have discovered 112 kinds of human PTPs.Among them,PTP1B is widely and clearly studied.Recently,as an enzyme that play a role in oncogenesis,PTP1B has been wildey studied by scientists.Here,we highlight the relationship between protein tyrosine phosphatase 1B and hematologic neoplasms.
9.Differentiation of bone marrow and umbilical cord blood mesenchymal stem cells into osteoblasts in vitro:comparison of their osteogenic potentials

BACKGROUND:Mesenchymal stem cels isolated from cord blood and bone marrow have multi-directional differentiation ability under a certain condition of induction. OBJECTIVE:To compare the difference of differentiation of umbilical cord blood and bone marrow mesenchymal stem cels into osteoblasts. METHODS:Human umbilical cord blood and bone marrow mesenchymal stem cels were isolated and cultured by density gradient method. When reached 90% confluency, mesenchymal stem cels were digested by trypsin for subculture. At the third passage, umbilical cord blood mesenchymal stem cels and bone marrow mesenchymal stem cels at 8×104/wel were incubated. When reached 80% confluency, cels were treated with low-glucose DMEM supplemented with 10% fetal bovine serum, 0.1 μmol/L dexamethasone, 50 μmol/L vitamin C and 10 mmol/L β-sodium glycerophosphate. RESULTS AND CONCLUSION:There was no significant difference in morphology and biological properties of the two kinds of mesenchymal stem cels. Cels were highly expressed CD44, CD29, but did not express CD34. They had the ability to differentiate into osteoblasts, which had a positive staining for known markers: alkaline phospatase and calciumin vitro mineralization. There was no significant difference in the activity of osteoblasts of two kinds of cels. Results verify that umbilical cord blood and adult bone marrow mesenchymal stem cels can be induced into osteoblasts with a similar ability, and they can be used as seed cels for bone tissue engineering.
10.Comparison of clinical and coronary angiographic characteristics in acute myocardial infarction patients with new-onset atrial fibrillation in early or later stages

Objective To explore the clinical and coronary angiographic characteristics in acute myocardial infarction (AMI) patients with new-onset atrial ifbrillation in early or later stages. Methods From Jun. 2010 to Jun. 2013, 1358 cases of AMI were hospitalized in which 88 were proved to have AMI complicated with new-onset atrial ifbrillation. Eligible patients were divided into early onset group (group1, n=40 cases) and the later onset group (group2, n=48 cases) according to the occurrence of atrial ifbrillation within or after 24 hours of admission. The clinical characteristics and the pathological changes of coronary arteries of the two groups were compared respectively. Results The incidence of inferior wall AMI was signiifcantly higher in group 1. The incidence of anterior wall AMI was signiifcantly higher in group 2 (P<0.05). The incidence of congestive heart failure, the incidence of three-vessel lesions and the in-hospital mortality in group 2 were higher than which in group 1 (P < 0.05). Conclusions The new-onset atrial ifbrillation in different stages may be regarded as a useful indicator for evaluating the clinical characteristic and the infarct-related coronary artery lesions and prognosis of patients with AMI.
