1.Clinical observation of Bushen Tiaochong method in treatment of 90 cases early unexplained recurrent spontaneous abortion

Objective To observe the clinical curative effect of Bushen Tiaochong method on treating early unexplained recurrent spontaneous abortion (URSA). Methods 200 patients who conformed to the inclusion criteria were randomly and paralleled control study divided into two groups.The control group (routine tocolysis group,n =100 ),with 8 cases fell off. The experimental group (routine tocolysis group +traditional Chinese medicine group,n =100 ),with 10 cases fell off. Results After the treatment,the total efficiency of the control group was 70.65%,and the experimental group was 88.89%. The total efficiency of comparative difference was statistically significant (p<0.05 ). And compared the experimental group with the control group,the former group improved the level of human chorionic gonadotropin(HCG)in patients. Conclusion Bushen Tiaochong method with traditional Chinese medicine combined western medicine treatment can significantly improve the symptoms of URSA patients with threatened abortion. Compared with simple western medicine therapy,the former group has better curative effect on early unexplained recurrent spontaneous abortion.
2.The significance of automated breast volume scanner of breast carcinoma and the research of its imaging manifestations

Objective: To investigate the diagnostic value of automatic breast volume system (ABVS) for breast carcinoma in clinical practice so as to use these reliable data to provide imaging reference for further therapy. Methods: 300 patients with suspected breast cancer were selected and the postoperative pathological examinations were chosen as the golden standard to implement screening. At the same time, 2 radiologists were selected to carry out the grading diagnoses of breast imaging reporting and data system (BI-RADS) so as to observe the consistency between diagnoses of ABVS and pathological results. Results: 210 patients were diagnosed as breast tumor by the post-operative pathological examination, and these patients included 89 malignant tumor (29.67%, 89/300). In these malignant cases, there were 42 cases were infiltrative ductal carcinoma, and its rate was 47.19% (42/89). Besides, there were 121 cases were benign tumor (40.33%), and the rate of patients with fibrosum adenoma (78 cases) was the highest in the 121 cases (64.46%, 78/121). And the diagnostic consistency of them was preferably (K=0.724). The sensitivity and specificity of ABVS in diagnosing breast carcinoma were 92.86% and 85.56%, respectively. Its positive predictive value and negative predictive value were 93.75% and 83.70%, respectively. And the Youden index was 0.78. Conclusion: ABVS has relatively high sensitivity and specificity, and the screening test has good authenticity and reproducibility. Therefore, it can provide a reliable basis for clinical diagnosis and treatment.
3.Path analysis of resilience and anxiety, depression on quality of life among esophageal carcinoma patients undergoing radiotherapy and (or) chemotherapy

Objective To explore the impact of resilience and anxiety, depression on quality of life (QOL) among esophageal carcinoma patients undergoing radiotherapy and (or) chemotherapy. Methods A total of 125 patients were recruited into our study. Date were collected by European Organization for Research and Treatment of Cancer, Quality of Life Questionnaire-Core 30 (EORTC QLQ-C30) , Resilience Scale (RS) and Hospital Anxiety and Depression Scale (HADS). Analysis of Moment Structures (AMOS) version 7.0 software was used to conduct the path analysis. Results The total score of patients′ QOL, RS and HADS were(51.98±19.42)points,(56.32±17.09)points,(14.59±7.04)points. Pearson′s analysis showed that QOL was correlated with RS and HADS ( r =0.423-0.588,P<0.05). RS was negatively correlated with HADS (path coefficient was-0.76 and-0.37) and HADS negatively correlated with QOL (path coefficient was-0.56 and-0.79). RS as a protective factor indirectly influenced QOL via HADS and the paths′ total coefficients was 0.69. Conclusions The level of QOL and RS were at poor level for esophageal carcinoma patients, as well as participants experienced anxiety and depression. HADS as intermediary variable has a greater moderating effect on relationships between RS and QOL. RS is a indirect protective factor of QOL. Interventions developed on the basis of influencing factors of QOL should be applied to improve patients′integral health status.
4.MODULATION OF MDR1 mRNA EXPRESSION AND REVERSION OF MULTI-DRUG RESISTANCE BY PPMP IN KBv200 CELL LINE

The current study was designed to investigate the effects of PPMP (DL-threo-1-phenyl-2- palmitoylamino-3-morpholino-1-propanol), a kind of glycolipids synthase inhibitor, on the modulation of mdr1 mRNA expression and the reversing effect of multi-drug resistance by PPMP in human malignancy KBv200cell line. In vitro KBv200 cells were treated with PPMP in different concentration, the alterations of mRNA expression of drug-resistant gene mdr1 in KB (sensitive cell line) and KBv200 (before and after the treatment of PPMP) cells were analyzed by RT-PCR. Intracellular rhodamine(Rh123) concentration was measured by flow cytometry. PPMP was found to inhibit mdr1 gene expression of KBv200 at the mRNA level, and complete inhibition appeared at 25μmol/L PPMP treatment for 48h. PPMP could increase intracellular Rh123 accumulation in resistant cell lines. This modulation of gene expression and Rh123 accumulation was directly correlated with the concentration of PPMP. It suggested that PPMP, a chemical inhibitor of glycolipids synthase, could modulate mdr1 expression at the mRNA level in a content dependent manner, PPMP possesses MDR-reversing activity.
5.Construction of eukaryotic expressing vector of human melanin-concentrating hormone receptor 1 and establishment of stable transfectant CHO cell line

Objective To construct eukaryotic expressing vector of human melanin-concentrating hormone receptor 1(MCHR1), then to transfect CHO cells with the vector for establishment of stable CHO cell line. Methods The full-length MCHR1 cDNA fragment was amplified by PCR from the human fetal brain cDNA library and then inserted into eukaryotic expression vector pcDNA3.1(+).The recombinant was transfected into CHO cells by lipofectamine TM 2000 after identification of digestion and sequencing on the recombinant eukaryotic expression vector pcDNA3.1(+)/MCHR1. The stable transfected CHO cell line was then established by screening cultures with G418, and the transcription and expression of MCHR1 were identified by RT-PCR, Western blot and immunofluorescence. Results The eukaryotic expression vector pcDNA3.1(+)/MCHR1 was constructed successfully, stable transfected CHO cell line was established, the MCHR1 protein was expressed successfully. Conclusion The construction of eukaryotic expression vector pcDNA3.1(+)/MCHR1 and the establishment of stable transfected CHO cell line provided a solid experimental foundation for further studies on the function of MCHR1.
6.Clinical experience of UOTLA in treatment of complicated appendicitis in children

Objective To analyze the efficacy of umbilical one trocar laparoscopic appendectomy (UOTLA) in treatment of complicated appendicitis in children. Methods Clinical data of 78 cases of children patients with complicated appendicitis from January 2012 to October 2015 was summarized, including 44 cases as UOTLA group received umbilical one trocar laparoscopic appendectomy, 34 cases as OA group received open appendectomy. Then statistically analyzed all the patients' operation time, postoperative hospital stay, postoperative abdominal abscess, incision infection, early inflammatory intestinal obstruction and pain level. Results The laboratory test results of C reaction protein (CRP) had no significant difference between the two groups, while peripheral white blood cell count decreased more significantly in UOTLA group than that in OA group; the operation time of UOTLA group was shorter than that in OA group with no statistical difference [(66.59 ± 33.24) vs (72.86 ± 30.36) min, P > 0.05], but postoperative hospital stay was shorter [(8.21 ± 1.67) vs (9.21 ± 2.01) d, P < 0.05]. Abdominal abscess after operation: 3 cases in UOTLA group, while 1 case in OA group (P > 0.05); incision infection: 6 cases in UOTLA group, 9 cases in OA group (P > 0.05); early inflammatory intestinal obstruction: 1 cases in UOTLA group, 5 cases in OA group (P > 0.05); the pain level, postoperative recovery time was significantly shorter in UOTLA group compared with OA group (P < 0.05). The average expenses comparison of the two groups has no statistical difference [(10639.37 ± 2970.92) vs (10765.04 ± 2902.64) yuan, P > 0.05]. Conclusion UOTLA is safe and effective for complicated appendicitis in children due to minimally invasive, less pain and faster recovery without significant increase in the cost and postoperative complications. It can be applied in children with purulent, perforated appendicitis and gangrene, perforated appendicitis and other complicated appendicitis.

8.Expression of IL-1ra mRNA and dysfunction of phagocytosis and intracellular killing of peripheral blood neutrophils in HD patients

Objective: To investigate the function and IL 1ra gene expression of neutrophils in hemodialysis (HD) patients. Methods: Gene expression was studied by reverse transcribe polymerase chain reaction (RT PCR) from untreated neutrophils in healthy controls and HD patients. Phagocytosis and bactericidal assays decreased in the number of viable extracellular and intracellular bacteria by colony counts. Results: Freshly isolated neutrophils spontaneously expressed icIL 1ra, sIL 1ra mRNA in HD patients. Phagocytosis and intracellular killing of neutrophils in HD patients impaired obviously when exposed to staphylococcus aureus in comparison with healthy controls. Conclusion: Untreated neutrophils in HD patients spontaneously express icIL 1ra and sIL 1ra mRNA, which is consistent with the suppressed phagocytosis and intracellular killing capacity. [

Endotoxin,which is also called lipopolysaccharide (LPS),is a unique element of Gram-negative bacteria.Liver is the main site for removing bacteria and endotoxin.Meanwhile,it is also the most vulnerable organ.Liver injury initially occurs in the hepatic sinu-soids.This article mainly introduces the impact of LPS on Kupffer cells,hepatic sinusoidal endothelial cells,oxidant/antioxidant balance in the hepatic sinusoids,and liver blood flow.It is believed that the function of hepatic sinusoids in LPS-induced liver injury should not be ig-nored,and the hepatic sinusoids would play an important role in the prevention and treatment of LPS-induced liver injury.
