1.Application of lectin microarray technology for the diagnosis of common gynecological tumors

Objective To probe the clinical values of the human serum glycoprotein profiles for the diagnosis of common gynecological tumors. Methods A total of 123 clinical serum samples which included 31 breast cancer, 24 cervical cancer, 19 ovarian cancer and 49 healthy individuals were collected. A lectin microarray consisting of 15 lectins with different glycan binding specificities was used to determine the glycoprotein profiles of serum sam-ples. Stepwise discrimination analysis method was adopted to establish function model of clinical serum samples classification with SPSS 15. 0 software. Results Two grades of diagnostic discrimination function models were es-tablished. The first grade discrimination function could differentiate gynecological tumors from healthy individuals, the diagnostic accuracy rates of retrospective inspection were 85. 7% and 83. 8% respectively, and the total diag-nostic accuracy rate was 84.6%. The second grade discrimination function was used to differentiate breast tumor, cervical tumor and ovarian tumor, the diagnostic accuracy rates of retrospective inspection were 96.8%,75.0%and 78.9% respectively, and the total diagnostic accuracy rate was 85.1%. Conclusion The human serum gly-coprotein profiles are associated with gynecological tumors, and the established discrimination function models based on lectin microarray data have a helpful reference value for the clinical diagnosis of gynecological tumors.
2. Salvianolic acid b inhibits oxidative damage of bone marrow mesenchymal stem cells and promotes differentiation into cardiomyocytes

BACKGROUND: In recent years, bone marrow mesenchymal stem cells have been used in clinical treatment of heart injury caused by permanent cardiomyocytopenia. However, the best inducer to promote the proliferation and differentiation of bone marrow mesenchymal stem cells into cardiomyocytes is still in the research and selection. OBJECTIVE: To investigate the protective effect of salvianolic acid B on oxidative stress injury and cardiomyocyte differentiation of bone marrow mesenchymal stem cells. METHODS: Bone marrow mesenchymal stem cells were used as study objects to establish oxidative stress injury model by 300 μmol/L H2O2 for 24 hours. Simultaneously, bone marrow mesenchymal stem cells were treated with salvianolic acid B for 24 hours. The contents of lactate dehydrogenase and creatine kinase in the supernatant and malondialdehyde and superoxide dismutase in the cell lysate were determined in each group using the kit. Western blot assay was used to detect the expression of nuclear factor E2 related factor 2 (Nrf2), Keap1, Bcl-2 and Bax in bone marrow mesenchymal stem cells. RT-PCT was used to detect cardiomyocyte differentiation-related markers 14 days after intervention. RESULTS AND CONCLUSION: (1) Compared with the model group, the levels of lactate dehydrogenase and creatine kinase in the supernatant were significantly decreased in the salvianolic acid B group (P < 0.05). (2) Compared with the model group, the level of malondialdehyde in cell lysate was decreased, and the level of superoxide dismutase was increased in the salvianolic acid B group (P < 0.05). (3) Compared with the model group, expression of Nrf2 protein in cells was significantly increased, and the expression of Keap1 protein was significantly decreased in the salvianolic acid B group. (4) The expression of Bcl-2 in salvianolic acid B group was significantly higher than that in model group, and the expression of Bax in salvianolic acid B group was significantly lower than that in model group. (5) Compared with the model group, the expression of GATA4 and cTnT mRNA was significantly increased in the salvianolic acid B group. (6) Salvianolic acid B can induce the expression of proteins that activate Nrf2-ARE signaling pathway in bone marrow mesenchymal stem cells for anti-oxidation, and salvianolic acid B can inhibit apoptosis and improve the ability of bone marrow mesenchymal stem cells to differentiate into cardiomyocytes.
3.Diagnostic Value of Color Doppler Flow Imaging in Subclinical Atrial Septal Defects

Objective To evaluate the usefuluness of the color Doppler flow imaging(CDFI) in subclinical atrial septal defects(ASD).Methods We reviewed 122 cases of subclinical ASD in adult which were first detected with CDFI.Results We found most of the patients were women and elder people,and no more special clinical symptoms and signs can be found.Color Doppler flow imaging had characteristic abnomallities making the dosis of ASD possible and easily with a high degree of accuracy in patients in whom the clinical presentation was atypical different magnitudes of left-to-right shunt were revealed in all these patients.Conclusion CDFI is a good noninvasive technique in the diagnosis of subclinical atrial septal defects.
4.Influencing factors for the short-term prognosis in elderly patients with posterior circulation infarction after intravenous thrombolysis

Objective To investigate the influencing factors for the short-term prognosis in elderly patients with posterior circulation infarction after intravenous thrombolysis.Methods We retrospectively analyzed the clinical data of 67 elderly patients with posterior circulation infarction treated with thrombolysis from March 2011 to December 2012.Clinical data of age,sex,hypertension,hyperlipidemia,glucose abnormalities,and thrombolysis,National Institute of Health stroke scale (NIHSS) score on admission,hyperhomocystinemia,the infarction area,anticoagulation and antiplatelet treatment were collected.All patients were followed up after discharge.The prognosis was assessed by the Rankin scale (mRS) after 3 months of thrombolysis.Patients were divided into good (MRS score ≤2) prognosis group and poor (mRS>2) prognosis group.Results There were 43 cases (64.2%) in good prognosis group and 24 cases (35.8%) in poor prognosis group.Logistic regression analysis showed that NIHSS score>12 (95%CI:1.087-1.569,P=0.005),thrombolysis time>4.5 h (95%CI:1.362-2.258,P=0.004),high homocysteine levels (95%CI:0.612-0.956,P =0.000),platelet after 24 h (95% CI:0.785 1.133,P=0.001) were the independent risk factors for the poor prognosis of posterior circulation infarction in elderly patients after intravenous thrombolysis.Conclusions Early thrombolysis and timely anticoagulant treatments have certain significances in improving the short-term prognosis in patients with posterior circulation infarction.
5.Application of early rehabilitation nursing model in the course of clinical nursing in patients with cerebral infarction

Objective To clear the clinical efficacy of using early rehabilitation nursing model in the course of clinical nursing in patients with cerebral infarction.Methods Divided 60 patients with cerebral infarction into the observation group (30 cases) and the control group (30 cases) randomly,traditional nursing measures were used in the control group,the early rehabilitation nursing model was used in the observation group in addition.Compared the clinical efficacy,ADL scores,NIHSS scores and the satisfaction between the two groups.Results In the observation group,the satisfaction was 93.33% (28/30),the efficiency was 90.00%(27/30),which were significantly higher than those in the control group [76.67%(23/30),[66.67%(20/30)].The ADL and NIHSS scores in the observation group was (91.25±11.38) and (8.01±1.05) respectively,which were significantly different with those of in the control group,the ADL and NIHSS scores was (79.37±10.27) and (10.22±1.27) respectively in the control group.Conclusions Early rehabilitation nursing model can effectively improve the never function of patients with cerebral infarction,and then improve their quality of life.
6.The plasticity of adult stem cells and their application in myocardial regenerative medicine

Current therapies for myocardial infarction and congestive heart failure are limited in efficacy or in applicability. The plasticity of adult stem cells and cellular transplantation offer a novel therapeutic approach to improve cardiac function. This review describes the latest progress in research, summarizes recent studies of adult stem cells and their application in myocardial regenerative medicine in China and abroad, and discusses the future directions of cell transplantation as a new therapy to repair injured hearts. 7.Effects of substrate stiffness on the proliferation of primary human umbilical vein endothelial cells and the release of NO and ET-1 during dengue virus infection

Objective To investigate the effects of substrate stiffness on the proliferation of human umbilical vein endothelial cells ( HUVEC) during dengue virus infection.Methods Polyacrylamide gels were prepared for cell culture [(0±4) kPa].The proliferation of HUVEC cultured on substrates with differ-ent stiffness was determined by using 3-(4,5-diethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfo-phenyl)-2H-etrazolium,inner salt ( MTS) assay.The cycle and apoptosis of HUVEC were determined by flow cytometry analysis.Dengue virus serotype 2 (DENV-2) strains were propagated and identified by con-ventional assays.The HUVEC were infected with DENV-2 strains at a MOI of 10 and cultured on traditional plastic and hydrogel substrates, respectively.The levels of nitric oxide (NO) and endothelin-1 (ET-1) were detected by nitric acid reductase assay and double antibody sandwich ELISA.Results Young′s modulus E value of the hydrogels was (3030 ±0.44) Pa.The proliferation of HUVEC and the expression of NO and ET-1 were enhanced along the increased substrate stiffness.However, no significant differences with the cell cycle and apoptosis were observed between cells cultured on different substrates.Conclusion The stiffness of substrates affected not only the proliferation of HUVEC, but also the release of cytokines during DENV-2 infection.The development of dengue fever was associated with the decreased secretion of vascular active substances as a result of blood vessel injury.The establishment of hydrogel substrates as the model of vascu-lar basement membranes might provide a new way for the in vitro investigation of the pathogenesis of DENV infection.
8.Investigation on smoking status of hypertensive elderly in Waigang Town of Shanghai: a cross-sectional study

Objective · To investigate the smoking hypertensive elderly, find out the status of smoking in this population, and explore effective system intervention. Methods · The smoking hypertensive elderly in Waigang Town of Shanghai who received free antihypertensive drugs were recruited to investigate and descriptively analyze the smoking status of them with questionnaires. Results · There were 77.6% of the smoking hypertensive elderly who began to smoke in the age of 13 to 25, and nearly 70% smoking for more than 40 years. There were 59.5% of these people smoking 11 to 29 cigarettes daily in recent 3 years, and 41.6% spending 200-299 yuan on smoking monthly. Conclusion · Currently almost 70% of the smoking hypertensive elderly, who receives free antihypertensive drugs, have been smoking for more than 40 years, and the average number of cigarettes smoked daily is big. It is clear that the health education attached to the policy of providing free antihypertensive drugs has not changed the behavior. Therefore, medical personnel still needs to contact the illness and smoking hazards to help the elderly reduce smoking. Besides, it proves that tobacco control should be implemented in the young population to control epidemic at the early stage.
9.Passive smoking status and its influencing factors among pregnant women in Shanghai

Objective · To investigate the prevalence of passive smoking among pregnant women in Shanghai and to analyze its influencing factors and to provide the basis and suggestion for making related intervention measures to reduce the harm of secondhand smoke on the health of pregnant women. Methods · From July to October 2014, pregnant women and their spouses were recruited for a questionnaire investigation at the antenatal care clinics of 3 maternity hospitals and 3 community hospitals in Shanghai. The questionnaire included demographic information, family basic situation and cognitive status of passive smoking. Results · The passive smoking rate before pregnancy in the 2831 pregnant women, who were involved in the investigation, was 17.1%. The passive rate during pregnancy was 7.8%. The difference between these two rates was statistically significant (P<0.05). 71.0% of the pregnant women have passive smoking for 15~59 min each day. Relative to home and work place, the passive smoking rate of pregnant women in public place was the highest (P<0.05). The passive smoking rate of pregnant women was significantly associated with the educational level of smoking spouses and the attitude of pregnant women when they faced the smokers. Conclusion · The passive smoking rate of pregnant women is influenced by the educational level of their spouses and the pregnant women's feedback to the smokers face to face. Public places are the most important exposure places to secondhand smoke for pregnant women. In order to reduce the harm from passive smoking to pregnant women and their fetuses, the related measures of smoking control at home education and smoking ban in public place should be made.
10.Effects of DENV-2 infection on the expression of IL-29 in primary HUVECs cultured on hydrogel

Objective To analyze the effects of dengue virus 2 ( DENV-2 ) infection on the ex-pression of IL-29 in primary human umbilical vein endothelial cells ( HUVECs) cultured on hydrogel sub-strates .Methods Primary HUVECs were isolated and cultured on hydrogel substrates .DENV-2 stains were used to infect the primary HUVECs at a multiplicity of infection( MOI) of 10.Flow cytometry analysis was performed to detect the apoptosis and infection rate of HUVECs after 48 hours of culturing .The gene chip profiling was performed to analyze mRNA expression .The expression of IL-29 at mRNA and protein levels were measured by real-time fluorescent quantitative PCR analysis and double antibody sandwich ELISA as -say, respectively.Results Compared with 96.36%of baby hamster kidney (BHK) cells that were infected with DENV-2 stains, only 4.71%primary HUVECs cultured on hydrogel substrates were infected .The pri-mary HUVECs cultured on hydrogel substrates with or without DENV-2 infection showed no significant differ-ences with the rates of cell apoptosis and infection (P>0.05).A significant difference was observed with the expression of IL-29 at mRNA level between primary HUVECs cultured on hydrogel substrates and the cells cultured in plastic bottles (P<0.05).The results of the real-time quantitative PCR analysis and ELISA as-say showed that IL-29 was highly expressed in DENV-2 infected primary HUVECs cultured on hydrogel sub-strates as compared with those in control groups (P<0.05).Conclusion The expression of IL-29 was de-tected in DENV-2 infected primary HUVECs cultured on hydrogel substrates , which was significantly differ-ent from that in DENV-2 infected primary HUVECs cultured in plastic bottles .The successful establishment of hydrogel substrates as the model of vascular basement membranes might provide a new way for the investi -gation of the pathogenesis of DENV infection .
