1.High density lipoprotein:the double-edge sword on endothelial function
Tianjin Medical Journal 2015;(4):439-442
Chronic exposure to risk factors at the beginning stage of atherosclerosis development can damage endotheli?um and ultimately initiat endothelial dysfunction. Endothelial dysfunction is one of the major mechanisms in atherosclerosis development. High-density lipoprotein (HDL) is a lipoprotein with complicated function and is composed of a variety of pro?teins and lipids. Epidemiological studies showed an inverse correlation between HDL and atherosclerosis. The protective role of HDL on endothelium might related to its function in reversing cholesterol transport, reducing peripheral cholesterol accu?mulation, preventing foam cells formation, dilating vessels, antioxidant and anti-inflammation, all of which antagonize devel?opment of atherosclerosis. But HDL obtained from patients with cardiovascular disease which has known dysfunctional HDL may induce endothelial dysfunction. This review addresses the relationships between functional HDL or dysfunctional HDL with the endothelial function.

The discovery of microRNAs (miRNAs) has introduced a new paradigm into gene regulatory systems. Since inception, computational methods have been an invaluable tool complementing experimental approaches, and many discoveries have been obtained through combination of experimental and computational approaches. The knowledge that has been accumulated about the principles of miRNAs and target recognition were reviewed. The currently available computational methodologies and software for prediction of miRNA and their target genes also have been discussed.

Objective To compare the efficacy of amisulpride and other SGAs in treating the negative symptoms of Schizophrenia. Methods The randomized controlled trials (RCTs) about Schizophrenia treated with amisulpride and other SGAs from Jan 1995 to Mar 2013 were searched in The Pubmed,EMbase,Cochrane Library,WanFang Data,CNKI and VIP.Two reviewers independently screened the literatures, extracted the data, and evaluated the methodological quality.Than meta-analyses were conducted by using RevMan 5.1 and Stata 12.0 software. Results The totall3 RCTs were included.Among the 1814 patients involved. The results of meta-analyses showed that the score of PANSS-N was no significant differences between two groups (MD =-0.33,95% CI:(-0.87,0.21),Z =1.20,P =0.23) ; and the score of SANS was no significant differences between two groups (MD =-0.21,95% CI:(-1.51,1.50),Z =0.31,P =0.76).The side effects were more in other SGAs group than those in amisulpride group. Conclusion Amisulpride is as effective as other SGAs for the treatment of schizophrenia with predominantly negative symptoms, and it has more advantage than other SGAs in safety.

Objective:To study the bone-conductivity and absorbability of nano-hydroxyapatite/collagen(nHAC) composite implanted under the calvarial periosteum in rabbits.Methods:24 nHAC samples and 24 HA samples were prepaired in the shape of round disk with the diameter of 8 mm and thickness of 3 mm.nHAC samples were implanted under calvarial periosteum on the left side and HA samples on the right of 24 rabbits. The bone-conductivity and absorbability of the samples were examined by new bone height measuring and residual implant materia measuring 2,4,8 and 12 weeks after operation. Results:2,4,8 and 12 weeks after implantation new bone height(mm) in nHAC group was 0.54?0.09,0.72?0.12,1.83?0.14 and 2.63?0.07,that in HA group 0.13?0.11,0.31?0.12,1.23?0.05 and 1.75?0.14,respectively(P

Membrane type-1 matrix metalloproteinase (MT1-MMP or MMP14) plays the pivotal role in tumor development and metastasis, so it is a promising drug target in malignancy. To acquire MT1-MMP specific binding peptides, we first analyzed MMPs sequences to find the divergent and specific sequence of MT1-MMP by bioinformatics approach, then set the specific sequence as the sense peptide target and designed antisense peptide library. Finally, by means of molecular docking, molecular dynamics simulation and in vitro cell assays, we screened the antisense peptide library against MT1-MMP and further studied the obtained specific peptides. Here, we identified the divergent and specific sequence of AYIREGHE (Named MT1-loop) located in MT1-MMP loop by multiple sequence alignment and established the antisense peptides library with capacity of 1 536 sequences. After two rounds of virtual screening, we obtained five antisense peptides with Rerankscores in the top for further screening. They all interacted with MT1-MMP, and docked well at the active site composed of MT1-loop sequence. Analysis of the affinities of these five antisense peptides to other MMPs (MMP1-3, MMP7-13, MMP14 HPX, MMP16) revealed that the peptide FVTFPYIR was more specific to MT1-MMP. Molecular dynamics simulation showed that the peptide FVTFPYIR might affect the stability of MT1-MMP and thus have effects on its activities. Meanwhile, the peptide FVTFPYIR could specifically inhibit the growth of MG63 and MDA-MB-231 tumor cells both of which expressed MT1-MMP. The work provides a new insight and way for the development of antitumor lead peptides targeting MT1-MMP.

Objective To observe the association between adiponectin gene polymorphism, serum adiponectin lev-els with the incidence of coronary artery disease (CAD) and the severity of coronary artery stenosis in patients with essential hypertension (EH). Methods A total of 414 patients with EH (234 cases with CAD and 180 cases without CAD) and 185 control subjects were recruited in this study. Serum adiponectin levels were measured by enzyme-linked immunosorbent as-say (ELISA). Adiponectin single-nucleotide polymorphisms rs266729,rs7649121,rs1501299 and rs3774262 were geno-typed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results Serum adiponectin levels were significantly lower in EH with CAD group than those in control group and EH without CAD group. Adiponectin SNP rs7649121 AT genotype reduced the risk of CAD compared with AA genotype (adjusted OR=0.566,95%CI 0.346-0.925,P=0.023). Logistic regression analysis showed that age and LDL-C were risk factors of CAD, and adiponectin was the protective factor for CAD in EH patients. The severity of coronary artery stenosis was negatively related to the level of adipo-nectin. Adiponectin levels were not affected by the adiponectin gene polymorphism. Conclusion The decreased serum adi-ponectin level was the independent risk factor for CAD in EH patients, which was negatively related to the severity of coro-nary artery stenosis. Adiponectin SNP rs7649121 may contribute to the risk factors of CAD in EH patients.
