1.Rosiglitazone inhibition of A375 human malignant melanoma cell invasion

Objective To explore the effect ofrosiglitazone (RGZ), a ligand of peroxisome proliferatoractivated receptor γ (PPARγ), on the invasiveness of A375 cells in vitro and its mechanism of action.Methods A375 human melanoma cells were cultured in vitro and treated with different concentrations of RGZ. The proliferation of the cells, mRNA expression levels of matrix metalloproteinase (MMP) 2 and tissue inhibitor of metalloproteinase (TIMP) 2, protein expression of MMP2 in A375 cells were detected by MTT assay, semi-quantitative RT-PCR and Western-blot, respectively. The invasiveness of A375 cells was detected by Matrigel invasion assay. Results MTT assay showed that the proliferation of A375 cells was inhibited by (4.86±0.31 )% and (5.15±0.52)% under the 24-hour treatment with RGZ of 10 and 25 μmol/L, respectively, and no evident cytotoxity was observed for RGZ. Compared with untreated A375 ceils, a significant decrease was observed in the mRNA and protein expression of MMP2 in A375 ceils treated with RGZ of 10 and 25 μmol/L (all P < 0.05), along with an increase in the mRNA expression of TIMP2 (both P < 0.01 ).The count of A375 cells transmigrating through matrigel was 154.1±7.7 and 87.3±8.1 under the treatment with RGZ of 10 and 25 μmol/L, significantly lower than that of those without treatment (210.7±14.9,both P < 0.01 ). Conclusions RGZ could inhibit the transmigration of A375 ceils, likely by down-regulating the mRNA and protein expression of MMP2.
2.CT appearances of primary pulmonary histoplasmosis

Objective To study the CT appearances of primary pulmonary histoplasmosis (PPHP)and evaluate the diagnostic values of CT for PPHP.Methods The clinical materials and multi-slice spiral CT appearances in 8 patients with PPHP proved pathologically were reviewed retrospectively.Results Single or multiple pneumonia-like lesions were seen in 3 esses,single or multiple nodules or masses were in 4 cases,and both pneumonia-like lesions and nodules or mflsses in 1 case.Calcification and halo sign were common,and only slight enhancement was demonstrated after intravenous contrast administration on CT.Conclusion CT is helpful imaging method in the diagnosis and differential diagnosis of PPHP combining with clinical materials although the CT appearances are not specific.
3.The Short-term and Long-term Clinical Effect of Leep Knife Treatment of Chronic Cervicitis

Objective To explore the short-term and long-term clinical effect of leep knife treatment of chronic cervicitis. Methods A single-center, prospective, randomized, controlled study was used, 120 patients with chronic cervicitis were randomly divided into observation group and control group, each group of 60 patients. Patients in the observation group were treated with (LEEP) knife, parameters setting frequency was 3.8 MHz, power was 30 ~ 40W, patients in the control group were treated with microwave. The intraoperative and postoperative recovery and relapse were compared between two groups. Results The intraoperative blood loss, operative time, postoperative vaginal discharge time in the observation group were significantly less than control group, there was a significantly statistical difference (P<0.001) . After two weeks and three months , the cure rate in observation group were 70% and 98.33% respectively, the cure rate in control group were 16.67% and 51.67%, there was a significantly statistical difference (P <0.01) . After 2 weeks and 3 months, the infection rate and recurrence rate in the observation group were significantly lower than the control group, there was a significantly statistical difference (P <0.05) . Conclusion Leep knife for treatment of chronic cervicitis patients has lower postoperative recurrence, lower infection rate and higher cure rate in short-term and long-term, and less intraoperative trauma, shorter operation time, and has important clinical significance for the prognosis of patients.
4.Retrospection,stratagem,and practice on innovative drug research and development of Chinese materia medica

Taking the national key scientific and technological project in 11th Five-year and 10th Five-year Plan as its main clue,the innovative Chinese materia medica(CMM)research course has been reviewed in this paper.Based on the enlightenment from the first herbal medicine VeregenTM,which has been approved by FDA,together with the study and comprehension of many great achievements of new drug research,the strategy of innovative CMM research and development(R&D)and its practical route is going to be put forward in this article.With the innovative Compound Prescription of CMM research as the main innovative approach,innovation can be realized by the following three aspects:New substances,including new compound,new effective fraction/group or new components and their combinations;new indication of known compounds or extracts;the secondary development of high-quality Chinese patent medicine.Besides,in order to support the R & D of original drugs with Chinese characteristics,the international experiences in target-dependent screening and evaluation and optimization of leading compounds based on clinical application should be referenced,and the mechanism and targets of traditional Chinese medicine by genetic and molecular level be clarified.
5.Practice and Experience of Medication Consultation Wechat Platform in Our Hospital

OBJECTIVE:To provide reference for medication consultation Wechat platform in the hospital. METHODS:The medication consultation Wechat platform of our hospital was introduced,including main method,content and effect. The consulting pharmacist team was set up,and Wechat public number was applied and operated experimentally;information pharmacist was re-sponsible for its daily maintenance. We released pharmacy news,accumulated consumers and expanded influence through other me-dia at regular intervals. RESULTS & CONCLUSIONS:According to the medical practice,two modules“medication consultation”and“pharmacy news”were set up. From Dec. 2014 to Jul. 30th 2015,medication consultation system had accumulated 1 121 con-sumers,with an increase of 5 to 8 ones everyday;average number of consultation records was 10 everyday;we also had published 22 batch of medication news(79 messages),including 31 original messages and 3 video messages. Several messages gained much attention from network media and offline media. As new pharmaceutical care mode,Wechat platform contribute to realize a individ-ual,convenient and mobile medication consultation service,medication health science popularization,and improves medication compliance of patients and the awareness of medication safety in field of pharmaceutical administration Wechat platform,so as to improve pharmaceutical care effect of“taking patients as the center”and promote the propaganda of hospital brand. Thus it has a good prospect of application and popularization.
6.Bone morphogenetic protein receptor Ⅱ signaling pathway and pulmonary arterial hypertension:updates and expectations

The identification of the relationships between mutations of the bone morphogenetic protein type Ⅱ receptor (BMPR2) and pulmonary arterial hypertension (PAH) has been considered to be one of the most significant discoveries in this area in the 21st century. And BMPR2 mutation is responsible for the majority of hereditary PAH as well as some of idiopathic PAH. Furthermore, clinical and animal expreimental research over the past few years has revealed that BMPR2 signaling pathway plays a critical role in the initiation and progress of PAH, by participateing in the pathogenesis of PAH. In addition, the potential that BMPR2 signaling pathway is used as a therapeutic target is being evaluated. This review summarizes our current understanding of the role of BMPR2 mutations in PAH from the perspectives of genetics, epigenetics, inflammation as well as interactions with other significant pathways.
7.Effects of propofol on apoptosis signaling pathways in a rat model of renal ischemia/reperfusion injury

BACKGROUND:Renal ischemia/reperfusion during renal transplant surgery induces the process of apoptosis signaling pathways.Propofol possibly protects kidney from renal ischemia/reperfusion injury.So it is important to investigate propofol's mechanism underlying apoptosis.OBJECTIVE:To investigate the effects of propofol on apoptosis signaling pathways in a rat model of renal ischemia/reperfusion injury and the possible mechanism of action.DESIGN,TIME AND SETTING:An animal experiment,cell morphology observation was performed at the laboratory of the Department of Urinary Surgery,Beijing Friendship Hospital between 2004 and 2005.MATERIALS:A total of 99 male adult outbred Sprague Dawley rats were included in this study.Rabbit anti-rat Bcl-2,Bax,caspase 3,and cytochrome C were produced in Wuhan Boster Bioengineering Co.,Ltd.,China.METHODS:Rats were randomly divided into three groups:control(n = 26),ischemia/reperfusion(n = 35),and ischemia/reperfusion+propofol(n = 38).Rat model of renal ischemia/reperfusion injury was established in each group as follows.Following single intravenous transfusion of 5 mL/kg ringer lactate solution,the right kidney was excised through a median abdominal incision.The left renal pedicle was occluded for 45 minutes using an atraumatic vascular clamp,followed by reperfusion and full-layer suture.At 0,3,12,24,and 72 hours after reperfusion,the left kidney was excised,and simultaneously,rat was sacrificed through bloodletting.In the ischemia/reperfusion+propofol group,propofol(1 mg/kg per minute)was administered from 15 minutes prior to ischemia to 30 minutes after reperfusion,for a total of 75 minutes.In the control and ischemia/reperfusion groups,rats were administered the same amount of ringer lactate solution.MAIN OUTCOME MEASURES:Morphological changes of injured kidney and expression of apoptosis-related protein Bcl-2,Bax,caspase 3 and cytochrome C.RESULTS:Renal ischemia/reperfusion-caused kidney damages could be observed through an optical microscope,and proximal convoluted tubule was severest,followed by distal convoluted and collecting duct,and lastly renal glomerulus.Immunohistochemistry results demonstrated that compared with the control group,Bax,caspase 3,and cytochrome C expression was increased,but no obvious change in Bcl-2 expression was observed in the ischemia/reperfusion group;compared with the control group,Bax,caspase 3 and cytochrome C expression was significantly decreased,but Bcl-2 expression was significantly increased in the ischemia/reperfusion+propofol group(P<0.05).CONCLUSION:Propofol is likely to exhibit protective effects on cellular apoptosis caused by renal ischemia/reperfusion.Propofol inhibits pro-apoptotic protein Bax,caspase 3 and cytochrome C expression but does not produce effects on Bcl-2 expression.The underlying mechanism correlates with apoptosis signaling pathways in mitochondrion.
8.Fusion expression of antimicrobial peptides in Escherichia coli.

Due to their potential application as novel antibiotics, antimicrobial peptides are attracting much attention. Large quantities of highly purified peptides are crucial to basic and clinical studies. Natural resources of antimicrobial peptides are limited and hard to purify, chemical synthesis is of high-cost and unstable, so recombinant expression of antimicrobial peptides is a cost-effective way. Escherichia coli has been the most widely used as host to express antimicrobial peptides with fusion protein, which can not only avoid the lethal effect towards the host, but also protect the peptide from degradation by proteases. Combined with our research, the present article reviews the progress of fusion vector, cleavage methods and optimization options for antimicrobial peptides production with fusion protein in Escherichia coli.

9.Evaluation of the simultaneous amplification and testing for diagnosis of Mycobacterium tuberculosis

Objective To evaluate the clinical value of the isothermal RNA amplification assay (SAT) for detection of Mycobacterium tuberculosis in sputum samples.Methods Sputum specimens from 230 patients with diagnosed tuberculosis and 78 cases of other respiratory diseases during September to December 2011 were detected using SAT,BD960 culture,LowenStein-Jensen( L-J ) culture and concentrated smear simultaneously.The samples with different results between SAT and BD960 culture were tested by Mycobacterium tuberculosis PCR fluorescence diagnosis kits.Strains were identified by amplification and sequencing the BD960 culture-positive isolates and SAT amplification products.Positive detection rate of SAT and other three methods for patients with tuberculosis were compared by chi-square test.Results Using the results of BD960 culture as the golden standard (7 cases of pollution bacteria in BD960 culture was rejected ),the sensitivity,specificity,positive predictive value,and negative predictive value of SAT was 90.5% (95/105),84.2% (165/196),75.4% (95/126),94.3% (165/175),respectively.The agreement rate of SAT and BD960 culture was 86.4% (260/301).For 223 tuberculosis patients,the positive detection rate of SAT,BD960 culture,L-J culture and concentrated smear was 56.5% ( 126/223 ),45.7% ( 102/223 ),41.7% ( 93/223 ) and 37.2% ( 83/223 ) respectively.The positive detection rate of SAT is significantly higher than the other three methods (x2 =4.087,P < 0.05 ).Conclusion SAT,as a new technology for laboratory diagnosis of TB,has high specificity and sensitivity.The operation is fast and simple,and the pollution rate is low.It is a promising laboratory diagnosis method.
10.SPECT cerebral perfusion imaging and its quantitative analysis for the assessment of stent implantation for cerebral artery stenosis

Objective To evaluate the role of SPECT cerebral perfusion imaging in assessing the stent implantation for cerebral artery stenosis.Methods A total of 35 patients (31 males,4 females,average age (63.9±10.8)years) with cerebral artery stenosis confirmed by DSA for cerebral artery stent implantation were retrospectively analyzed.99Tcm-ECD cerebral perfusion imaging was performed for all patients before and after stent implantation.The images were realigned and normalized by SPM 2.0 and then analyzed by Brain Search software for quantitative analysis.The brain was automatically separated to 210 functional areas according to Talarich map.The normalized averaged counts (NAC) of each area were calculated and compared with the data of 28 health controls (8 males,20 females,average age (35.8± 9.4) years).Less than 1.96s was defined as low perfusion lesions.The NAC values before and after stent implantation were compared for classifying improved from non-improved group.The mean number of lesions and Essen stroke risk score (ESRS) were analyzed between the two groups.The mean number of lesions and postoperative improvement rate of the internal carotid artery (ICA) occlusion and stenosis were compared.Paired rank sum test,two-sample t test,two-sample rank sum test and x2 test were used for statistical analysis.Results In 35 patients with low perfusion areas,20 were significantly improved after stent implantation.The mean number of lesions in the improved group (34.05± 14.41)was significantly higher than that in the non-improved group (22.93±17.24; t=2.067,P<0.05).The mean ESRS of the improved patients (14.8)was significantly lower than that of the non-improved patients (22.3,Z=2.24,P<0.05).The improvement rate of 28 cases with ICA stent implantation was (60.7%,17/28)higher than that of 7 cases with middle cerebral artery (MCA) stent implantation (3/7; P>0.05).The mean number of the ICA occlusion lesions (34.36± 14.31)was higher than that of the ICA stenosis lesions(31.35± 16.37),but the difference was not statistically significant(t=0.498,P>0.05).The improvement rate of the ICA occlusion was higher than that of the ICA stenosis (7/11 vs 10/17),but the difference was not statistically significant (P>0.05).Conclusion SPECT cerebral perfusion imaging and its quantitative analysis can evaluate the low perfusion lesions before stent implantation and predict the perfusion improvement after stent implantation.
