1.Oxidative modification of rat β2-glycoprotein Ⅰ

Objective:To investigate oxidative modification of β2-glycoprotein Ⅰ in vit ro.Methods:Rat β2-glycoprotein Ⅰ was purified and characterized,then oxidize d by hypoxanthine plus xanthine oxidase as a supreroxide free radical generating system;carbonl groups of β2-glycoprotein Ⅰ were detected by the reaction w ith 2,4-dinitrophenylhudrazine.Results:There was a significant increase of carbonyl groups formation in β2- glycoprotein Ⅰ oxidized in comparison with native β2-glycoprotein Ⅰ (P <0.05). Conclusion:Carbonyl groups have been formed in vitro on rat β2-glycoprotein Ⅰ after oxidative modification using hypoxanthine plus xanthine oxidase system.
2.Administration of methylprednisolone suppresses the development of arteritis in a murine model of Kawasaki disease

Objective To investigate the inhibitory effect of methylprednisolone (MP) on the development of coronary arteritis in a murine model of Kawasaki disease (KD) induced with a candida albicaus watersoluble fraction (CAWS).Methods Forty-five C57BIL/6mice were evenly divided into three groups (the control group,the CAWS group and the MP group).Mice in the CAWS group were intraperitoneally injected phosphate buffer saline (PBS) for 5 days.MP or PBS was administered to the different group.The animals were sacrificed at day 3,day 10 and day 28,and the status of vasculitis in the coronary arteries and the aortic root was investigated histologically.One-way analysis of variance (ANOVA) was used to compare the differences among three groups,and t-test for two independent groups.Results The mice in CAWS group and MP group,which induced by CAWS,showed that the body weight and heart weight decreased significantly,and the spleen weight was increased at day 10 and day 28 (P<0.05).Vasculitis was induced in the mice of those two groups,and the severity score was the highest at day 10 (12.7±0.5).In addition,the severity of the inflammation of the aortic root and the coronary arteries were reduced in MP group (t=6.35,5.55,2.8,P<0.05).Elastic fiber staining showed that the layers of vascular walls were in disorder and elastic fibers were broken in the CAWS group.However,there was no disruption or breakage in the MP group.Conclusion MP can suppress the progression of coronary arteritis in this CAWS-induced murine vasculitis model,which indicates the efficacy of MP in KD patients with coronary artery lesions.
3.The role of promoter CpG islands methylation of leptin gene in osteoarthritis

Objective To investigate the effects of 5-Aza-CdR( methylation transferase inhibitor) on the expression levels of leptin gene in chondrocytes and methylation states of leptin promoter region between osteoarthritis (OA) group and control. Methods The chondrocytes in osteoarthritis group were treated with 5-Aza-CdR with different doses and time-points, and the expression level of leptin was detected by real-time polymerase chain reaction for picking up the optimum dose and time-point. Next, the chondrocytes in 5 osteoarthritis patients and 5 control patients (amputation due to severe trauma) were treated with 5-Aza-CdR. Lastly, leptin mRNA expression levels in the four groups osteoarthritis and control chondrocytes treated with/without 5-Aza-CdR were measured by real-time PCR and the methylation state of promoter region ( - 280- + 79) was detected by epityper quantitative DNA methylation analysis. Results ( 1 ) After treating the chondrocytes in OA groups with 10 μmol/L 5-Aza-CdR for 72 h, the mRNA expression levels of leptin were increased significantly. ( 2 ) The mRNA expression levels of leptin were significantly different among the four groups ( P < 0. 05 ), and the chondrocytes in osteoarthritis groups treated with 5-Aza-CdR showed a marked induction of leptin mRNA expression. (3) Analysis of quantitative methylation data using an unsupervised hierarchical clustering algorithm, showed that methylation patterns of leptin promoter was different between control and osteoarthritis chondrocyte treated with/without 5-Aza-CdR. Conclusion Demethylation of leptin promoter might up-regulate leptin gene expression level and it might contribute to osteoarthritis.
4.Influence of orthokeratology lens on vision, intraocular pressure and biometric measurement parameters of adolescent myopia

Objective To evaluate the influence of orthokeratology lenses on vision,intraocular pressure and biometric measurement parameters in adolescent myopia.Methods A total of 72 adolescent myopia patients (136 eyes) with orthokeratology lens,aged 8-16 years old,in Hebei Provincial Eye Hospital from Junuary 2013 to December 2015 were randomly selected.The vision,intraocular pressure,axial length,corneal topography,corneal thickness and anterior chamber depth were observed before wearing glasses,at 1 week,1,3,6 months after wearing glasses.Results After orthokeratology wearing,the uncorrected visual acuity was obviously improved (P<0.05),the average refraction diopter was declined (P>0.05) and the non-contact intraocular pressure was decreased (P<0.05).The axial length after orthokeratology wearing had little change (P>0.05).The curvature of the patient's cornea at one week after wearing glasses was deceased (P<0.05) and the posterior corneal curvature tended towards stability.The corneal thickness at 1 week after wearing glasses had no obvious change compared with before wearing glasses(P>0.05),which at 1 month after wearing glasses was decreased(P<0.05) and which at 3,6 months after wearing glasses trended to be stabilized.The anterior chamber depth after wearing glasses had no obvious change.Conclusion Orthkeratology lens can decrease the myopia degree,increases the uncorrected visual acuity and has obvious effect for controlling adolescent myopia.The intraocular pressure and biometric measurement parameters have corresponding change after wearing orthkeratology lens.
5.Assessment on health risk of drinking water in Hangzhou

Objective:
To assess the health risk associated with drinking water in Hangzhou from 2016 to 2017,and to provide evidence for the safety of drinking water .
Methods:
The monitoring data of 5 genetic toxic substances(arsenic,hexavalent chromium,cadmium,chloroform,tetrachloromethane)and 13 body toxic substances(lead,mercury,selenium,cyanide,fluoride,nitrate,iron,ammonia nitrogen,manganese,copper,zinc,aluminum,volatile phenol)from 36 source water samples,36 finished water samples and 288 tap water samples in the main urban areas of Hangzhou were collected from 2016 to 2017. The health risk of drinking water containing the chemical pollutants mentioned above were assessed based on the evaluation models recommended by United States Environmental Protection Agency .
Results:
The concentrations of 5 genetic toxic substances and 13 body toxic substances in source water,finished water and tap water were all within the reference limits issued by Standards for Drinking Water Quality(GB 5749—2006). The carcinogenic risk,non-carcinogenic risk and total health risk caused by the chemical pollutants in the source water were 2.18×10-5/a,7.75×10-9/a and 2.18×10-5/a. The carcinogenic risk,non-carcinogenic risk and the total health risk caused by the chemical pollutants in the finished water were 1.08×10-5/a,3.70×10-9/a and 1.08×10-5/a. The carcinogenic risk,non-carcinogenic risk and total health risk caused by the chemical pollutants in the tap water were 1.96×10-5/a,3.61×10-9/a and 1.96×10-5/a. The carcinogenic risk and total health risk caused by chemical pollutants ranged from high to low in the source water,tap water and finished water. The non-carcinogenic risks ranged from high to low in the source water,finished water and tap water .
Conclusion
The health risks of 18 chemical pollutants in drinking water in Hangzhou were at a low level,with the greater carcinogenic risk than the non-carcinogenic risk. Hexavalent chromium had the highest carcinogenic risk,while fluoride and aluminum had the highest non-carcinogenic risk.
6.One case of keratoconjunctival carcinoma in situ with microinvasion and literature review

Objective To recapitulate the clinical traits of conjunctiva-corneal carcinoma in situ with microinvasion,and explore the early diagnosis and precise therapeutic methods.Methods The clinical features,diagnosis,surgical methods and follow-up results of a hospitalized patient with keratoconjunctival in situ carcinoma with microinvasion were retrospectively analyzed and related literature was consulted.Results Surgical resection of the tumor and 2 mm marginal tissue outside the edge of the tumor was performed.Fresh corneal lamellar grafts were used to treat the disease.After 2 years of follow-up,the operation was effective and no recurrence was found.Conclusion Early diagnosis plays an important role in improving the prognosis of keratoconjunctival carcinoma in situ with microinvasion and preventing recurrence.For patients with keratoconjunctival carcinoma in situ acompanied with microinvasion,we have to expand the surgical resection of the tumor (≥2 mm at the edge of the tumor),and follow-up at regular intervals,if necessary,supplemented with chemotherapy or anti-metabolic treatment.
7.A possible receptor for beta 2 glycoprotein I on the membrane of hepatoma cell line smmc7721.

OBJECTIVESTo study the interaction of beta-2-glycoprotein I (beta 2GPI) with the membrane of hepatocytes and determine whether beta 2GPI participates in HBV infection.
METHODSLigand blotting, fluorescence microscopy, and fluorescence activated cell sorter (FACS) analysis were used to detect the specific interaction of beta 2GPI with the hepatoma cell line smmc7721, the gastric carcinoma cell line SGC7901, and the lymphoma cell line HL-60.
RESULTSA specific 40 kDa beta 2GPI band was observed by ligand blotting in the case of smmc7721 cells. No such band was observed in SGC7901 or HL-60 cells. Fluorescence microscopy also revealed specific binding of FITC-beta 2GPI to smmc7721 cells, but neither to SGC7901 nor HL-60 cells. FACS analysis demonstrated that the binding rate of FITC-beta 2GPI to smmc7721 cells was significantly higher than these in SGC7901 and HL-60 cells (P < 0.01). The binding rate to smmc7721 cells did not increase with increasing amounts of FITC-beta 2GPI.
CONCLUSIONSThere is a specific beta 2GPI-binding protein on the membrane of hepatoma cells in cell line smmc7721 which as the beta 2GPI receptor may participate in HBV infection of hepatocytes.

8.Effects of warm and tonify kidney-yang herbs on liver mitochondria proteome of kidney-yang deficiency rats.

OBJECTIVETo study the effects of warm and tonify kidney-yang herbs on the liver mitochondria proteome of the thyroidectomized kidney-yang deficiency rats.
METHODTwelve rats were divided into normal group, model group and treated group; each group had four rats. The rats of model group and treated group were excised the two side thyroid gland, and the rats of normal group were done the homologous operation, but didn't excise the thyroid gland. After seven days, the rats of model group and treated group appeared the symptoms of kidney-yang deficiency. From the eighth day, the rats of treated group were fed warm and tonify kidney-yang herbs 6.7 g x kg(-1) once daily, and the rats of other two groups were fed the equal normal. All rats of three groups were killed by decollation after six days of treatment, and liver mitochondria proteins were separated. Each liver mitochondria protein sample was labeled with Cy3 or Cy5 randomly, and one Cy3-labeled sample and one Cy5-labeled sample were mixed on the same 2-D gel along with a Cy2-labeled mixture of all samples as an internal standard and run on the same gel. The gels were scanned under different wavelength light after electrophoresis. All images were analyzed by DeCyder 6. 5 software, and the different proteins were identified by mass spectrum.
RESULTThe expression of HSP60, catalase, glutathione peroxidase, carbamoylphosphate synthetase I, ATP synthase, lactotransferrin, H(+)-transporting two-sector ATPase, alpha-ETF and calpain 12 were increased in the thyroidectomized kidney-yang deficiency rats, while the expression of oxoisovalerate dehydrogenase, Acyl-CoA dehydrogenase, ornithine aminotransferase, and GTP-binding regulatory protein were decreased. After the kidney-yang deficiency rats were treated with warm and tonify kidney-yang herbs, the expression of HSP60, catalase, glutathione peroxidase, oxoisovalerate dehydrogenase, Acyl-CoA dehydrogenase, ATP synthase, lacto-transferrin, H(+)-transporting two-sector ATPase, alpha-ETF and GTP-binding regulatory protein were increased, and the expression of carbamoyl-phosphate synthetase I and calpain 12 were decreased.
CONCLUSIONThe warm and tonify kidney-yang herbs perform its therapeutical effect by regulating the metabolism, protecting the stability of mitochondrial membrance and maintaining the signal conduction in the cells.

9.Relation between Beta-2-glycoprotein I and hepatitis B virus surface antigen.

OBJECTIVETo clarify the binding character between Beta-2-glycoprotein I (Beta-2-GPI) and HBsAg.
METHODSBeta-2-GPI was purified from human plasma and labelled with biotin. Solid phase enzyme linked absorbance assay was used to investigate its binding with HBsAg.
RESULTSBiotinylated Beta-2-GPI was found to bind HBsAg and the reaction could be inhibited by excess unlabelled Beta-2-GPI.
CONCLUSIONSBeta-2-GPI may play a role in hepatitis B virus infection.

10.Clinical study on Xiaosheng Powder ultrasonic nebulization combined with meibomian gland massage for the patients with meibomian gland dysfunction dry eye and pattern of liver depression and Yin deficiency

Objective:To observe the clinical efficacy of Xiaosheng Powder ultrasonic nebulization combined with meibomian gland massage in treating liver depression and Yin deficiency pattern of meibomian gland dysfunction dry eye.Methods:Randomized controlled trial. From March 2021 to September 2021, a total of 50 patients (100 eyes) diagnosed with meibomian gland dysfunction dry eye of the liver depression and Yin deficiency pattern at the Ophthalmology Hospital of China Academy of Chinese Medical Sciences were selected. They were randomly divided into two groups using a random number table, with 25 patients (50 eyes) in each group. The control group received 0.1% sodium hyaluronate eye drops combined with meibomian gland massage, while the treatment group received Xiaosheng Powder nebulization combined with meibomian gland massage. Both groups were treated for 4 weeks. Before and after treatment, the tear film break-up time (BUT), Schirmer I test (SIT), corneal fluorescein staining (FL) scores, TCM symptom scores, and meibomian gland function scores were observed and evaluated. Adverse reactions were recorded.Results:After treatment, the treatment group exhibited statistically significant improvements compared to the control group in the following aspects: BUT was significantly longer in the treatment group (t=8.76, P<0.01); SIT values were significantly higher in the treatment group ( t=6.18, P<0.01); FL scores, TCM symptom scores and meibomian gland function scores were significantly lower than in the treatment group ( t values were 2.19, 5.36 and 12.09, P<0.01). The total effective rate in the treatment group was 96.0% (24/25), while in the control group, it was 68.0% (17/25), and the difference between the two groups was statistically significant ( χ2=4.88, P=0.027). Conclusion:The combination of Xiaosheng Powder ultrasonic nebulization and meibomian gland massage is effective in improving the clinical symptoms of patients with meibomian gland dysfunction dry eye of the liver depression and Yin deficiency pattern. Furthermore, this approach is considered safe and efficient.
