Objective To establish the method for extracting the cytokeratin filament aggregating protein (filaggrin) from human epidermis,to detect the anti filaggrin antibody (AFA) in rheumatoid arthritis (RA) and to determine the diagnostic value of AFA in RA.Methods Filaggrin was extracted from human epidermis and partially purified.AFA in 103 patients with RA and 140 controls was detected by Western blotting.Results Filaggrin extracted from the epidermis could be used as the antigen in AFA test by Western blotting.AFA test resulted in diagnosis of 35 9% of 103 RA samples,with a specificity of 93 7%.The result was significantly higher compared with patients′ and normal controls ( P
4.Biomechanical research of calcium phosphate cement in fixating distal radius fractures

[Objective]To test the biomechanical effects of calcium phosphate cement in fixating distal tadius fractures,and to provid biomechanical foundation for clinical treating distal tadius fractures.[Method]A models of 18 human distal radius fracture of adult upper limbs specimen were made and randomly divided into three groups:fixed with Kirschner wire(Kirschner group),CPC(CPC group),and CPC combined with Kirschner wire(combination group).Wrist joint was vertically loaded with 98 N,and palmar flexion with dorsal extention motion was dimulated,which range of movement was from palmar flexion 5?to dorsal extention 30? and frequency was 2000 cycles.Displace data of distal radius were measured by sliding caliper at every 500 cycles,we had tested Young' s modulus of two samples of CPC.[Result]Multiple factor variance analysis was used,Kirschner group and others had significantly statistical difference(P0.05).[Conclusion]CPC can provide enough fixation strength for distal radius fracture with bone defect and Young' s modulus of CPC is about 30 MPa,which is between those of cancellous bone and compact bone.

6.The relationship of platelet derived growth factor-BB and carotid atherosclerosis in maintenance hemodialysis patients

Objective To measure the level of platelet derived growth factor-BB (PDGF-BB) in maintenance hemodialysis patients and investigate the significance of carotid atherosclerosis.Methods Thirty-six maintenance hemodialysis patients (test group) and 20 healthy controls (normal control group) were enrolled in this study.The level of PDGF-BB was measured by enzyme-linked immunosorbent assay in two groups.Intima-media thickness (IMT) and atherosclerotic plaques of the extracranial common carotid artery were measured by echocardiography.Results The level of IMT and the positive rate of carotid atherosclerosis in test group were significantly higher than those in normal control group [(1991.8 ± 228.6) μ mol/L vs.(71.2± 15.9) μmol/L,(36.78 ±3.40) g/Lvs.(41.96±2.07) g/L,(4.97 ±0.57) mmol/Lvs.(4.48 ±0.84) mmol/L][(1.01 ±0.23) mm vs.(0.72 ±0.15) mm,69.4%(25/36) vs.10.0%(2/20)](P< 0.01).The level of plasma creatinine,albumin and total cholesterol in test group were significantly higher than those in normal control group (P < 0.01 or < 0.05).The level of PDGF-BB in test group was significantly higher than that in normal control group[(93.27 ± 31.58) ng/L vs.(31.71 ± 15.78) ng/L,P < 0.01].In test group,age and the level of PDGF-BB and triacylglycerol in carotid atherosclerosis patients were significantly higher than those in non-carotid atherosclerosis patients [(48.04 ± 9.97) years vs.(34.54 ± 10.35) years,(102.60 ± 32.87) ng/L vs.(72.06 ± 13.67) ng/L,(1.51 ± 0.59) mmol/L vs.(1.01 ± 0.27) mmol/L] (P < 0.01).Regression analysis showed that age (β =0.346,P < 0.01) and PDGF-BB (β =0.594,P< 0.01) were the important impacting factors for IMT.Conclusion The level of PDGF-BB is increased in maintenance hemodialysis patients,and it may play an important role in carotid atherosclerosis.
7.Spectral marker construction for evaluating antipyretic effects of 4 herbs

AIM: To construct suitable quality standard of evaluating antipyretic effect of 4 herbs,Bupleurum,Flos Lonicerae Japonicae,Radix Isatidis and Fructus Forsythiae. METHODS: Extracts,essential oils and its mixtures of extract and oil of above herbs,plus Ibuprofen Suspension,were used for antipyretic experiment on rats.Principal Component Analysis and Canonical Correlation Analysis were adopted as evaluation tools to examine the correlation between pharmacological effect and UV spectra of herb's chemicals,from which spectral marker of potential reliability for evaluating antipyretic effects of samples could be constructed. RESULTS: Antipyretic effect sequencing of UV spectral canonical variables was consistent with that of the four herbs. CONCLUSION: The first canonical variable U1 expressed by principal component scores of UV spectra of herbs could be used as spectral marker for evaluating antipyretic effects of the four herbs.
8.Clinical Observation on Brucea Javanica Oil Combined with Gemcitabine and Cisplatin in the Treatment of Advanced NSCLC

OBJECTIVE:To observe clinical efficacy and safety of Brucea javanica oil combined with gemcitabine and cisplat-in in the treatment of advanced non small cell lung cancer(NSCLC). METHODS:Totally 131 advanced NSCLC patients selected from Huanggang Hospital of TCM during Feb. 2014 to Jan. 2016 were divided into observation group(71 cases)and control group (60 cases)according to random number table. Control group was given Gemcitabine for injection(1st and 8th day)+Cisplatin injec-tion (2nd day),every 21 days,twice as a treatment course. Observation group was additionally given Breucea javanica oil oral emulsion 20 mL,po,2-3 times/d,for consecutive 14 d (3 days before chemotherapy). Both groups received treatment for 87 d and followed up until Jul. 1,2016. Clinical efficacy,the levels of T cell subsets (CD3+,CD4+,CD8+),survival time were ob-served in 2 groups. Single factor and multiple factor analysis was conducted for survival time. The occurrence of ADR was record-ed. RESULTS:The total response rate of observation group (32.39%) was higher than that of control group (25.00%),without statistical significance(P>0.05). Before treatment,there was no statistical significance in the levels of CD3+,CD4+ and CD8+ be-tween 2 groups(P>0.05). After treatment,the levels of CD3+ and CD4+ in observation group were increased significantly,while CD8+ level was decreased significantly;there was statistical significance compared to control group at corresponding period (P<0.05). Four patients withdrew from the study in observation group and 3 patients in control group. The survival time of observation group [(14.02 ± 4.10) months] was longer than control group [(13.21 ± 5.22) months],without statistical significance (P>0.05). Single factor analysis showed that the survival time of patients aged <70 were significantly longer than those of patients aged ≥70;those of patients with pleural effusion were significantly shorter than those of patients without pleural effusion;there was statis-tical significance (P<0.05). Multiple factor analysis showed that with or without pleural effusion,age were influential fac-tors for survival time of patients with advanced NSCLC(OR=1.685 and 0.647,P=0.012 and 0.043). There was no statistical significance in the incidence of ADR between observation group (50.70%) and control group (53.33%)(P>0.05). CONCLU-SIONS:Brucea javanica oil combined with gemcitabine and cisplatin in the treatment of advanced NSCLC patients,although not significantly improve the therapeutic effect,but can significantly improve the cellular immune function. With or without pleural effu-sion and age are infuential facters for survival time of advanced NSCLC patients.
9.The Clinic Study of Therapy in the Chronic Myelocytic Leukemia-Chronic Phase by As_2O_3 Combined with Homoharringtoninum

Objective To study the effctiveness and safety of arsenic trioxide(As2O3)combined with Homoharringtoninum in the chronic myelocytic leukmia-chonic phase(CML-CP).Methods Seven patients were treated with As2O3 10mg per day for 2~3 week and with Homoharringtoninum 3~4mg per day for 1~2.Results All patients were clinic remission,of which there were 4 complete remission who were all initial therapy,and 3 partial remission,2 of who were initial therapy and 1 was resume threapy.4 patients of CR were treated with second strengthen therapy and continued hematologic CR.The main side effects were grade 3 hematologic toxicity and light liver damage,and no significant nausea,emesis,diarrhea or mucositis.Conclusion As2O3 combined with Homoharringtonium in the CML-CP is a safe and effctive regimen.
10.Method Evaluation of HPLC Fingerprints for Quality Control of Chinese Herbal Medicine

Objective To investigate the effects of different chromatogram fingerprints analysis methods on the quality evaluation of Chinese herbal medicine. Methods With the chromatogram fingerprints of a Chinese patent medicine in different batches as the analytic object, the quality evaluation of three methods such as peak area ratio,peak area quantification and similarity coefficient were investigated. Results The method of peak area ratio has the highest accuracy rate and the precise rate for quality evaluation, and similarity coefficient and peak area quantification came next. The result of peak area quantification was similar to that by current evaluation method. Conclusion Different chromatogram fingerprints analysis methods would result in a great difference in the quality evaluation of Chinese herbal medicine.
