1.Neuroprotective mechanisms of histone deacetylase inhibitors in ischemic stroke
International Journal of Cerebrovascular Diseases 2010;18(5):358-362
Histone deacetylases (HDACs) catalyze the deacetylation of histone and nonhistone proteins. They play important roles in regulating chromatin remodeling and gene transcription, and that aberrant chromatin remodeling and transcriptional dysregulation are associated with many neurodegenerative diseases. Preclinical studies have suggested that histone deacetylases inhibitors (HDACIs) can target diverse pathophysiologies of ischemic stroke, not only reduce neuronal damage and infarct volume, but also promote the neuronal plasticity and functional recovery after ischemia. This article mainly reviews the neuroprotective mechanisms of HDACIs in ischemic stroke.

Journal of Jilin University(Medicine Edition) 2014;(5):997-1001
Objective To investigate the protective effects of Xuebijing(XBJ)on the injury of vascular endothelial cells(VEC)induced by lipopolysaccharide (LPS),and to study the mechanisms of the production of nitric oxide (NO)and the expressions of inducible nitric oxide sytnhase (iNOS)and signal transduction under XBJ intervention condition. Methods The cultured VEC were divided into control group, LPS (1 mg · L-1 )group, LPS (1 mg·L-1)+XBJ(25 g·L-1)group,LPS(1 mg·L-1)+pyrrolidine dithiocarbamate(PDTC,20μmol·L-1) group;XBJ and PDTC were administrated 1 h before incubation of with LPS.Western blotting method was used to detect the expressions of iNOS and NF-κB p65 protein. The level of NO in the supernatant was measured by Griess reagent. Results Comparaed with control group, the NO level and the expression levels of iNOS protein and NF-κB p65 protein in VEC in LPS group were significantly increased(P<0.01).Compared with LPS group, the NO level and the expression levels of iNOS protein and NF-κB p65 protein in VEC in LPS+XBJ group were significantly decreased (P<0.05 or P<0.01);the NO level and the expression levels of NF-κB p65 protein and iNOS protein in VEC in LPS+PDTC group were significantly decreased(P<0.05 or P<0.01).There were no significant differences of the NO levels and the expression levels of iNOS protein between LPS+XBJ group and LPS +PDTC group (P>0.05),but the expression level of NF-κB p65 protein in LPS+PDTC group was lower than that in LPS+XBJ group(P<0.05).Conclusion XBJ can inhibit the production of NO and the expression of iNOS protein in VEC;its mechanism may be related to inhibiting the activation of NF-κB to control inflammation.

Cancer Research and Clinic 2007;19(z1):47-50
Objective To study the variety and significance of immune function in lung cancer patients. Methods 68 pretherapy lung cancer patients and 20 healthy volunteers with peripheral blood samples by flow cytometry. Results The proportion of CD+3T cells,CD+4T cells and the ratio of CD4/CD8 decreased in lung cancer patients, while the proportion of CD+8T cells and natural killer cells increased.Serum levels of TNF-α,IL-4,IL-6,IL-10 were significantly increased in patients compared with healthy controls, while the ratio of Th1/Th2 was decreased.(2)The natural killer cells level was negatively correlated with the CD3,CD4,CD8 levels respectively. The CD4 level was negatively correlated with the levels of IL-6,IL-10 respectively.(3)No statistically significant relationship was found between the peripheral blood lymphocyte,Th1-Th2 cytokines and pathological type, lymph node metastasis, distant metastasis. Conclusion Immune suppression was very common in lung cancer patients, particularly cell mediated immunity suppressed markedly. The proportion and function of CD+4T cells decreased significantly.which may resulted from the Th1,Th2 imbalance.Natural killer cells and CD+8T cells increased significantly, but could not prevent tumor development, which may resulted from these factors that suppress immune response in tumor microenvironment.

Objective To investigate the expression of heat shock protein (HSP) 60 and analyze the significance in cervical carcinoma. Methods The cervical tissue samples of 57 cases of cervical cancer and 57 normal cervical tissues were collected. The expression of HSP60 were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). The protein expressions of HSP60 were determined by Western blot analysis. To observe the relationship between HSP60 expression and the occurrence and development of cervical cancer. Results HSP60 mRNA expression in normal cervical tissue was significantly lesser than that in cervical cancer tissue (t=2.65, P0.05). The levels of HSP60 protein expression in normal cervical tissue was significantly lower than the expression in cervical cancer tissue (t=3.132, P

Objective To evaluate the impact of PET/CT on the clinical therapeutical strategy of patients with non-small cell lung cancer (NSCLC). Methods Sixty patients with histologically confirmed NSCLC supposed to accept radical radiotherapy or surgical operation were firsthy divided into group A (stage Ⅰ-Ⅱ) and group B (stage Ⅲ) according to CT findings, and the individualized treatment strategy was designed, then restage and compare with different stage and management with initial treatment strategy by PET/CT scanning. Results ①PET/CT scanning restaged the TNM stage in 53.33% (32/60) patients, including 3 of T stage, 23 of N stage and 9 of M stage. Changes of stage occurred in 65.85% (27/41) patients with adenocarcinoma and in 26.32% (5/19) with squamous carcinoma (P<0.05), while in 34.29 (12/35) with central and 57.14% (20/35) with peripheral NSCLCs (P<0.05). ②PET/CT scanning resulted in alteration of the managements of NSCLC patients. The management changed from radical into palliative strategy with PET/CT information in 9 (15.00%) patients. In 25 patients who were supposed to accept surgical operation, the treatment changed to radical radiotherapy in 8 (8/25, 32.00%) and to palliative radiotherapy in 5 (20.00%) patients. In 35 patients who were supposed to accept radiotherapy, surgical operation or palliative radiotherapy was finally chosen in 5.71% (2/35) and 11.43% (4/35) patients, respectively. Conclusion PET/CT images can make significant alteration to clinical stage and treatment plan in patients with NSCLC.

Chinese Journal of Pathophysiology 2001;17(8):792-793
AIM: To determine the evolutionary pattern of parenchymal cell apoptosis in multiple organs early after intestinal ischemia-reperfusion(I/R) and its induction mechanisms and the role of apoptosis in triggering SIRS/MODS. METHODS: An I/R model was reproduced by clipping and releasing the superior mesenteric artery in rats and mice. Flow cytometry, electron microscope, DNA agarose gel electrophoresis, TUNEL method, fluorescent and Gomori's silver-HE staining were used to detect apoptosis. Distribution features of apoptotic parenchymal cells in multiple organs were observed. Immunohistochemical staining of HSP 70 and Bcl-2 were performd to study the induction mechanisms of apoptosis.RESULTS and CONCLUSION: 1. Damage of the liver, lung, gut and kidney was appeared in early phase of I/R. The percentages of apoptosis in parenchyma organs increased progressively. The percentages of cell necrosis increased with the prolonged I/R duration. 2. Percentages of apoptosis were much higher near the central veins of liver lobules, in the outer medulla of the kidney, and the antimescenteric border of intestinal mucosal epithelium because of ischemia. 3. The expression of HSP 70 increased and Bcl-2 reduced in the areas mentioned above because of hypoperfusion. 4. Apoptosis of I/R hepatocytes, splenocytes and thymocytes was obviously increased after Kupffer cell blockage with GdCl3, showing the functional state of Kupffer cells may play an important role in SIRS/MODS.
