Objective To elucidate the role of insulin receptor and its ?-subunit autophosphorylation in evoking insulin resistance in non-obese type 2 diabetes. Methods 14 patients with non-obese type 2 diabetes and 12 healthy subjects were enrolled in this study. Insulin receptor numbers and its insulin affinity in erythrocytes were measured using radio-labelled ligand binding assay described by Comi et al. Insulin receptor autophosphorylation in erythrocytes membrane was initiated by adding insulin and ?- 32 P ATP. Then the membrane proteins were resolved on 7 5% SDS-PAGE and subjected to autoradiography. Results The numbers of high and low affinity receptors in erythrocytes of type 2 diabetes were 21?13 and 955?427 per cell respectively, and they showed no significant difference as compared with control. The high affinity constant(K 1) and low affinity constant(K 2) of insulin receptors in erythrocytes of type 2 diabetes were (1 70?0 93)?10 -9 LM -1 and (1 44?0 86)?10 -7 LM -1 respectively, also displaying no significant difference in comparison with the control. Insulin-induced receptor ? subunit autophosphorylation was 8 02?5 6△A/mg protein in type 2 diabetes, which was lower than that (28 38?15 24)△A/mg protein) in control(P

Objective To study the expression levels of SCCA1 and SCCA2 mRNA in tissues of cervical squamous cell carcinoma. To investigate the role of this gene in the clinical diagnosis, evaluation of treatment and observation of prognosis of cervical squamous cell carcinoma. Methods Quantitative real-time RT-PCR was used to detect the expression of SCCA1 and SCCA2 mRNA in tissues of 60 cases of cervical squamous cell carcinoma and those of 30 cases of normal cervical tissues. Results The expression level of SCCA2 mRNA in tissues of 30 cases of cervical squamous cell carcinoma was higher than in those of 15 cases of normal cervical tissues (4.405 ± 2.310, 9.088 ± 2.195) (t =-6.513, P <0.001), while the expression level of SCCA 1 mRNA did not significantly differ between normal and malignant tissues (P >0.05). The expression of SCCA2 mRNA was relevant to FIGO stages and there was a tendency for this gene to increase with the stage getting worse (F =8.313, P <0.05). Moreover, the overexpression of SCCA2 mRNA was significantly correlated with lymph node metastases (t =2.853, P <0.05). The expression of SCCA2 mRNA was not correlated with age and pathological grading (P >0.05). However, the expression of SCCA1 mRNA was not correlated with age,FIGO stages, lymph node metastases and histological grade (P >0.05). Conclusion The expression of SCCA2 mRNA may provide help for more accurate diagnosis on the clinical stages and lymph node metastases of cervical squamous cell carcinoma.

Objective To survey and understand the endemic situation of schistosomiasis among 80 national surveillance sites in China in 2008, so as to provide the evidence for decision-making of schistosomiasis control. Methods According to the National Surveillance Protocol for Schistosomiasis, the data of schistosomiasis in residents, domestic animals and Oncomelania snails and other relevant factors were collected and analyzed in the 80 national surveillance sites. Results Among the 80 national surveillance sites, the average positive rate of residents with sera examination and the average infection rate of the residents were 9.86% and 0.67% , respectively, and the infection rate of domestic animals was 1.62% , all remarkably decreased compared with the rates of 2007, but the proportion of domestic animals stable breeding was only 6.07% . The areas of infected snail habitats and the new snail habitats were 177.70 hm~2 and 7.64 hm~2, respectively. The mean density of living snails and infection rate of snails were 0.32/0.1 m~2 and 0. 15% , respectively. Conclusions The endemic situation of residents, domestic animals and snails decreased in 2008 compared with those in 2007, and domestic animals are still the main infection sources of schistosomiasis, the management of domestic animals need to be further strengthened, meanwhile the surveillance and control of snails should be focused on.

Objective To investigate the relative activity of multidrug resistance P-glycoprotein in the plasma of breast cancer patients, analyze the expression of P-glycoprotein before and after chemotherapy and evaluate its clinical value. Methods The plasma of patients and chemical reagents were mixed and distributed into the 96-well plate. The plate was read at 340 nm absorbaace before and after reaction. The variation of absorbance for each well was calculated, and then relative activity of each sample was compared to the referent compound activity. The relative activity of over 30 % was taken as positive expression of the P-gp for each sample. Results The frequency of P-glycoprotein expression was significantly increased from 16.2 % before chemotherapy to 43.2 % after chemotherapy (P<0.05). The positive expression of P-glycoprotein before and after chemotherapy were 5/6 and 9/16, respectively, in patients with positive lymph node metastasis, and 1/6 and 7/16, respectively, in patients with negative lymph node metastasis. Conclusion The results of this preliminary study suggest in P-glycoprotein expression in plasma of patients play an important role in the selection of drugs, which may be useful to evaluate the prognosis for treatment of breast cancer patients.

Cancer Research and Clinic 2009;21(5):308-310
Objective To investigate the expression of NKG2D peripheral blood of patients with esophageal carcinoma and expression of the soluble form of major histocompatibility complex class Ⅰ-related chain A (MICA). To analyze and discuss the function of NKG2D in anti-esophageal carcinoma mechanism and its relation with escape of immunosurveillance of cancer. Methods Flow cytometry analysis was used to detect NKG2D in the peripheral blood of 50 patients with esophageal carcinoma and 25 normal control individuals. Enzyme-linked immunoabsorbent assay was used to measure serum levels of soluble MICA. Results The expression of NKG2D were(87.25±3.06), (88.38±4.24), (92.46±1.46) respectively. Compared with the normal control individual, the expression of NKG2D in esophageal carcinoma group was significantly low; Among patients categorized according to most disease parameters tested (tumor size, grade of differentiation, regional lymph node status, disease stage), soluble MICA levels in sera did not statistically differ from those in normal control individuals. Patients with stage Ⅳ disease and/or regional lymph node metastasis, exhibited significantly higher serum levels of soluble MICA than control individuals. Conclusion The activity of NK cell and the anti-cancer cellular immunity level reduce in patients with esophageal carcinoma. The decrease of the receptor NKG2D is a reason for the descend of the the activity of NK cells. The escape of immunosurveiilance with esophagal carcinoma may correlate with down-regulate of NKG2D expression and up-regulate of sMICA.
