Abstract Objective:To explore antitumor effect of NDV HN. Methods: pIRHN nucleic acid vaccin was constructed and was transfectedto Hela cells. Westem blot was used to analyssize the expression of pIRHN nucleic acid vaccine in eukarytic cell. The mode of cell death was de-tected by fluorescence microscope, gel electrophoresis and TUNEL assay. The effect of pIRHN nucleic acid vaccine on the contents of siaha acidin the Hela cell was examined. Results: pIRHN nucleic acid vaccine could be expressed in the eukarytic cell. pIRHN could induced apoptosisafter HeLa cells were transfected. The effect of antitumor responses of pIRHN was correlated with the contents of sialic acid in tumor cells. Con-clusion: The reults of our experiment will provide the theory evidence for the anti tumor mechamism of NDV.

Objective:To investigate the effect of the BmK-9-(2) on the sodium current in guinea-pigventricular myocytes. Methods:The whole cell patch-clamp technique was used. Results:The BmK-9-(2)increased the magnitude of the peak inward sodium current (27 ％) ,peak sodium conductace (17 %) and theprolong time course of sodium channel inactivation (8％) and there was no effects on calcium channels.Conclusion:BmK-9-(2) can activate sodium channel and prolong the inactivated course of myocytes.

Objective To evaluate the effect of alprostadil on acute lung injury in septic rats. Methods Thirty adult male Sprague?Dawley rats, weighing 200-250 g, were randomly divided into 3 groups (n=10 each) using a random number table: sham operation group (group S), acute lung injury group ( group ALI) , and alprostadil group ( group Q) . The animals were anesthetized with intraperitoneal 1% pentobarbital sodium 5 ml∕100 g. Sepsis was induced by cecal ligation and puncture. In group Q, al?prostadil ( 10 μg∕2 ml) 2 ml∕kg was injected via the tail vein at 30 min before cecal ligation and puncture. The equal volume of normal saline was given in S and ALI groups. At 24 h after operation, blood samples were taken for determination of serum tumor necrosis factor?alpha ( TNF?α) and interleukin?6 ( IL?6) con?centrations by enzyme?linked immunosorbent assay. The animals were then sacrificed. The left lungs were immediately removed for microscopic examination, and the right lungs were immediately removed for deter?mination of wet∕dry lung weight ratio ( W∕D ratio ) , and expression of TNF?α mRNA and high mobility group box?1 ( HMGB1) using real?time reverse transcriptase?polymerase chain reaction. Results Com?pared with group S, the concentrations of serum TNF?α and IL?6 were significantly increased, and the ex?pression of TNF?α mRNA and HMGB1 mRNA was up?regulated, and W∕D ratio was increased in ALI and Q groups ( P<0?05) . Compared with group ALI, the concentrations of serum TNF?αand IL?6 were signifi? cantly decreased, and the expression of TNF?α mRNA and HMGB1 mRNA was down?regulated, and W∕D ratio was decreased in group Q ( P<0?05) . The pathological changes of left lungs were significantly attenua?ted in group Q as compared with group ALI. Conclusion Alprostadil can reduce acute lung injury in septic rats, and the mechanism may be related to down?regulation of HMGB1 expression and inhibition of inflam?matory responses.

Objective To monitor the protective qualities of personal protective appliances and to ensure the health and safety of radiological working personnel.Methods The lead-equivalent thickness of personal protective appliances and materials was measured by means of standard lead slices.The lead equivalent thickness represents in terms of mm Pb.Results 77 pieces of products and samples were measured altogether.The results indicate that the specific lead equivalents of lead-rubber plates were between 0.20-0.39 mm Pb/mm for 37 pieces of lead-rubber plates and the values of 6 pieces of samples were less than 0.25 mm Pb/mm,which did not accord with the requirement of the relational standard.27 pieces of personal protection appliances were measured altogether.They were 12 pieces of protective clothes,4 pieces of protective headgears,5 pieces of protective neckpieces,4 pieces of protective gloves and 2 pieces of protective masks.13 pairs of lead-glass spectacles among them were measured altogether.The measured results for personal protective appliances and lead-glass spectacles showed that actually measured lead-equivalent were higher than the nominal lead-equivalent.Conclusions The protective qualities are reliable for personal protection materials and appliances to be made in home and imported abroad.But the protective qualities of interventional protection gloves should be improved and made them better.

Objective To investigate the incidence of aspirin resistance (AR) in patients with Type Ⅱ Diabetes mellitus (DM) and the correlation between AR and advanced glycosylation end products (AGEs) as well as the homeostasis model assessment for insulin resistance index (HOMA-IR). Methods A total of 69 patients with Type Ⅱ DM and another 23 patients without DM as control group were enrolled between October 2009 and July 2010. Blood lipid, blood routine, fasting blood glucose, Glycated hemoglobin (GHb/Hb A1c) ,fasting insulin were determined at first. After aspirin treatment for at least 7 days, platelet aggregation stimulated by arachidonic acid(AA) and adenosine diphosphate (ADP) were measured. In addition, the level of serum AGEs was measured by using ELISA assay. The degree of insulin resistance was obtained by using HOMA-IR. Results The incidence of AR in patients with Type Ⅱ DM was higher than that in the controls(30.4% vs. 8.7%, P = 0.037 ); the levels of serum AGEs and HOMA-IR in patients with Type Ⅱ DM were higher than those in the controls [ (359.56 ± 120. 14) pg/mL vs. (275.45 ± 118.06)pg/mL, P=0. 004; (4.42 ±4.78) vs. ( 1.5 ±0.78), P＜0.01, respectively]; platelet aggregation stimulated by AA in the diabetic group was correlated with serum AGEs and HOMA-IR( R =0.463, P ＜0.01; R=0.290, P =0.016, respectively); and platelet aggregation stimulated by ADP was only positively correlated with HOMA-IR(R =0.242, P = 0.045). Conclusions The incidence of AR in patients with Type Ⅱ Diabetes mellitus is higher than that in the controls, and diabetics with higher serum AGEs and HOMA-IR are more likely to develop aspirin resistant.

Background It has been demonstrated that αvβ3/αvβ5 and α5β1 integrins are overexpressed in neovascular tissue.Consequently,peptide containing the RGD (Arg-Gly-Asp) sequence,which exists in ligands of integrins,is effective in targeting therapeutic reagents to neovascular endothelium.ObjectivePresent study aims to investigate the antiangiogenetive effects of liposome mediated plasmid encoding endostatin with RGD sequence on alkali burn-induced corneal neovascularization (CNV) in rabbits.MethodsCNV models induced by alkali burn were established in 72 eyes of 36 New Zealand white rabbits by putting the filter paper with 1 mol/L NaOH at the central cornea for 20 seconds.The animal models were divided into four groups randomly.0.2mL of liposome and plasmid encoding RGD-ES complex (liposome mediated pCI-RGD-ES injection group),liposome and plasmid encoding ES complex (pCI-ES injection group),liposome and carrier plasmid (pCI) complex (pCI-ES injection group),and PBS were subconjunctivally injected respectively in the models from four groups twice a week for two weeks.The growth status of CNV was observed on day 1,3,7,14 after alkali burn under the slim lamp microscope.Experimental animals were sacrificed on the 3rd,7th and 14th day and the expression of VEGF in CNV was detected by immunohistochemistry.The number of corneal microvessels was counted based on the number of CNV cross-section under the light microscope.ResultsCNV area was significantly smaller in liposome mediated pCI-RGD-ES injection group and pCI-ES injection group compared with PBS group at different time points (P＜0.01),but no significant difference was seen in CNV area between pCI-ES injection group and PBS group at different time points (P＞0.01).The changes of number of corneal microvessels was followed a similar fashion as the change of CNV area.Expression of VEGF in cornea was obviously stronger in pCI-ES injection group and PBS group than liposome mediated pCI-RGD-ES injection group and pCI-ES injection group.ConclusionEndostatin with RGD sequence could effectively inhibit corneal neovascularization,and liposome is proved to be a potent carrier in gene transfer.

Objective To investigate the predictive value of heart rate variability to,and its correlation with ventricular arrhythmias in patients with structural heart disease.Methods 24 hours ambulatory electrocardiogram monitoring was administered during March 2012 and October 2013,and standard deviation of the NN interval (SDNN) in the time-domain analysis and sympathetic/vagal ratio in the frequency domain analysis were collected and retrospectively analyzed in 300 patients.Results The incidence rate of ventricular arrhythmias was higher in patients with structural heart disease in SDNN ＜100 ms group than in control group [4.1％ (3/74) vs.39.5％ (45/114),P＜0.01].The sympathetic/vagal ratio was significantly higher in group with structural heart disease and ventricular arrhythmia than in control group(19.9 ±2.3 vs.10.1 ±1.7,P＜0.01)The time-domain index SDNN was negatively correlated with the frequency domain index (sympathetic/vagal ratio) (r=-0.819,P ＜0.01).Conclusions SDNN ＜100ms and the increased sympathetic/vagal ratio can be considered as the predictors of ventricular arrhythmias in patients with structural heart disease.

Objective To evaluate the clinical implementation value of bedside ultrasound monitoring of gastric residual volume in the guidance of enteral nutrition (EN) in severe patients with mechanical ventilation. Methods One hundred and forty-three patients undergoing nasal feeding EN and mechanical ventilation &ge; 72 hours in the Department of Intensive Care Unit (ICU) of Jinhua People's Hospital from May 2017 to August 2018 were enrolled, and they were randomly divided into two groups: a traditional control group (71 cases) and an ultrasound monitoring group (72 cases). The traditional control group applied 50 mL syringe to withdraw gastric juice from gastric tube, once every 4 hours, and then the EN protocol based on the gastric residual volume was adjusted; the ultrasound monitoring group used bedside ultrasound to monitor the gastric residual volume once every 4 hours, and then the EN protocol was adjusted accordingly. The changes of EN tolerance indexes e.g. abdominal distension, reflux vomiting, diarrhea, bowel sound hyperactivity or disappearance, intra-abdominal pressure, etc, the changes of incidence of ventilator-associated pneumonia (VAP), mechanical ventilation time and ICU hospitalization time in these two groups were observed. Results The incidences of abdominal distension, reflux vomiting, diarrhea, intestinal hyperactivity or disappearance of bowel sounds were significantly lower in the ultrasound monitoring group than those in the traditional control group [abdominal distension: 23.6% (17/72) vs. 28.2% (20/71), reflux vomiting: 18.1% (13/72) vs. 22.5% (16/71), diarrhea: 13.9% (10/72) vs. 14.1% (10/71), bowel sounds hyperactivity or disappearance: 9.7% (7/72) vs. 11.3% (8/71), all P < 0.05], the intra-abdominal pressure was obviously lower in the ultrasound monitoring group than that in the traditional control group [mmHg (1 mmHg = 0.133 kPa): 8.9±5.6 vs. 9.6±6.1, P < 0.05], and the incidence of VAP was significantly lower in the ultrasound monitoring group than that in the traditional control group [19.7% (14/72) vs. 23.9% (17/71), P < 0.05], the mechanical ventilation time and ICU hospitalization time were also significantly shorter in the ultrasound monitoring group than those in the traditional control group [mechanical ventilation time (days): 10.1±3.1 vs. 12.2±3.4, ICU hospitalization time (days): 16.8±4.2 vs. 20.6±4.8, all P < 0.05]. Conclusion The bedside ultrasound monitoring gastric residual volume can improve EN tolerance, reduce VAP incidence, and shorten the mechanical ventilation time and ICU hospitalization time in patients with mechanical ventilation.

Objective: To investigate the mode of cell death caused by NDV strain Changchun and NDV strain Siping. Methods: Plaque formation, cell suppression test, gel electrophoresis, and TUNEL assay were used after the cells were infected by NDV. Results: The apparently pathological changes were observed in chicken embryo fibroblasts, BHK, Hela, Hep 2, HCT and OS 732 tumor cells, but not in Wish cells. The higher suppressed effect on tumor cells was found in the NDV strain Changchun than that in the NDV strain Siping. There was no dose effect relationship between NDV and tumor cell suppression, only optimum dose NDV could cause maximal tumor cells inhibitory effect. Conclusion: The mode of cell death might be different after infection of NDV. The NDV strain Changchun killed tumor cells mainly through apoptosis, while the NDV strain Siping killed tumor cells mainly through necrosis. \[
