1.Studies on fingerprint of bidens bipinnata L.by HPLC

Objective The fingerprint method based on high-performance liquid chromatography for the quality control of bidens bipinnata L were firstly described and optimized. Methods HPLC system was used to obtain the fingerprint spectrum of bidens bipinnata L. The chromatography condition was ODS C18 column (4. 6mm×255mm,25μm),mobile phase Methanol-0. 2% HAc,and flow speed 1. 0ml ? min-1, detection wavelength 363nm.Results In this experiment,the standard fingerprint had been established and the similitude degree of 10 batches bidens bipinnata L. has also been obtained(0.941~0.999). Conclusion The method was accurate,reliable,good reproducibility, strong characteristic and could be used for the standard of general judgement and the quality estimation about the bidens bipinnata L.
2.Modified radical mastectomy-stage Ⅰ expansion of latissimus dorsi muscle flap breast reconstruction

Background and purpose:Physical and psychological trauma has been a challenge for both clinician and patients with breast cancer after modified radical mastectomy,and now more and more patients have begun to accept breast reconstruction,especially Ⅰ stage reconstruction after surgery.The aim of our study was to investigate a breast reconstruction procedure that can not only achieve radical tumor excision but also retain the aesthetic appearance of breast.Methods:15 patients from January 2006 to April 2007 were enrolled with early stage breast cancer have received surgical treatment with conservation of breast skin,breast tissue excision and axillary lymph nodes;latissimus dorsi muscle flap has been used for breast reconstruction.Results:The results of the 15 cases:excellent in 11 cases(83.3%),good in four cases(16.7%).Conclusions:Immediate breast reconstruction after radical mastectomy with conservation of skin has shown good results in terms of aesthetic appearance of breast,this procedure has been proved to be a safe and feasible method of treatment for the patients with early stage breast cancer.
3.Intravascular ultrasound-guided interventional treatment of coronary true bifurcation lesions:a clinical ;study

Objective To investigate the application of intravascular ultrasonography in guiding the performance of interventional management for coronary true bifurcation lesions. Methods A total of 62 patients with coronary true bifurcation lesions, who were admitted to authors’ hospital during the period from April 2010 to Jan. 2014 to receive double stenting treatment, were included in this study. The patients were randomly divided into intravascular ultrasonography group (IVUS group, n=32) and coronary angiography group(CA group, n=30). The minimal lumen diameter(MLD), the minimal lumen area(MLA), the reference lumen diameter (RLD), the length of the lesion and the diameter and length of the implanted stent were determined, and the results were compared between the two groups. During the follow-up period lasting for 12 months after percutaneous coronary intervention, the incidence of in - stent thrombus, target lesion revascularization (TLR) and major adverse cardiac events were recorded. Results The MLD values of the main artery and branches in IVUS group were higher than those in CA group, but the difference was not significant (P>0.05). The MLA, RLD, the diameter and length of the implanted stent in IVUS group were significantly larger than those in CA group (P<0.05). The incidence of in-stent thrombus, TLR and major adverse cardiac events in IVUS group were lower than those in CA group (P>0.05). Conclusion Intravascular ultrasound- guided double stent implantation for coronary true bifurcation lesions can help optimize the performance of stenting and improve the long-term outcome of percutaneous coronary intervention.

Aim TostudytheroleofASIC1aonthe matrix turnover and MAPK expression of the rat articu-lar chondrocytes with extracellular acidosis. Methods ArticularchondrocyteswereisolatedfromSprague-Dawley rats, and their phenotype was determined by toluidine blue and immunocytochemical staining. The GAG content of cell culture supernatant was deter-mined by dimethyl-methylene blue spectrophotometric assay, while Hyp content by chloramine T assay. ELISA assay was used to measure MMP-2 , TIMP-2 content. Furthermore, the ERK1/2, p38 MAPK phos-phorylation protein expression levels were tested by Westernblotassay.Results ASIC1acontributedto the effect of GAG, Hyp and TIMP-2 levels reduction induced by extracellular acidification, while the effect of MMP-2 was weaker. Moreover, ASIC1a could in-crease the ERK1/2 , p38 MAPK phosphorylation pro-teinexpressionlevels.Conclusion ASIC1acould regulate rat articular chondrocytes matrix turnover via ERK1/2 and p38 MAPK signaling pathway, and there-by inhibit the rat articular cartilage damage induced by acidosis.
5.Study on Simultaneous Determination Method of Zolpidem,Dexzopiclone and Zaleplon in Human Plasma by UPLC-MS/MS

OBJECTIVE:To develop a method for rapid,accurate and simultaneous determination of dexzopiclone,zolpidam and zaleplon in human plasma. METHODS:After the plasma sample was processed by liquid-liquid extraction,UPLC-MS/MS was established to detect plasma sample with carbamazepine as internal standard. The separation was performed on a Waters ACQUITY UPLC HSS T3 column with mobile phase composed of 0.1% ammonium solution-acetonitrile (gradient elution) at flow rate of 0.2 ml/min. The column temperature was set at 40 ℃,and sample size was 5 μl. The detection was performed by multiple reaction monitoring (MRM) via electrospray ionization (ESI) source in positive mode. The mass transition ion-pairs were as follows:m/z 308.2→263.1(zolpidem),m/z 389.3→245.0(dexzopiclone),m/z 306.2→236.1(zaleplon),m/z 237.3→151.2(internal standard). RE-SULTS:The linear range of zolpidem,dexzopiclone and zaleplon were 0.02-20.00,0.50-20.00,0.02-20.00 ng/ml,respectively (r=0.990 1,0.996 8,0.991 7). LLOQ of them were 0.02,0.50,0.02 ng/ml,and detection limit were 0.01,0.20,0.01 ng/ml. RS-Ds of intra-day and inter-day were all lower than 15% ;extraction recovery were 88.9% -106.5% ;matrix effect were 94.8%-106.3%. CONCLUSIONS:The method is simple,rapid,sensitive and specific,and it can be used for simultaneous deter-mination of zaleplon,zolpidem and dexzopiclone in human plasma.
6.Effects of targeted nursing interventions on patients with diabetic retinopathy after panretinal photocoagulation

Objective:To explore the effects of targeted nursing interventions on the quality of life in patients with diabetic retinopathy (DR) after panretinal photocoagulation (PRP) .Methods:Totally 92 DR patients who received PRP in the People's Hospital of Hechi from May 2016 to May 2018 were selected using convenient sampling and divided into control ( n=46) and observation ( n=46) groups. Patients in the control group received routine nursing care postoperatively, while patients in the observation group received targeted nursing interventions on this basis. Patients in both groups received follow-up visits for 3 months. The 25-item National Eye Institute Visual Functioning Questionnaire (NEI-VFQ-25) was used to evaluate the patients' quality of life postoperatively and at the last follow-up visit. Results:There was no statistically significant difference in NEI-VFQ-25 scores between the two groups before intervention ( P>0.05) . Single item scores and total NEI-VFQ-25 scores of the observation group were increased after intervention than before intervention, and the differences were statistically significant ( P<0.05) . There was no statistically significant difference in the score of driving between the observation and control groups after intervention ( P>0.05) . The scores of another 11 items and total NEI-VFQ-25 scores of the observation group were higher than those of the control group, and there were statistically significant differences between the two groups ( P<0.05) . Conclusions:Targeted nursing interventions have significant effects on the quality of life in DR patients after PRP, which can improve their quality of life.

The vagina contains at least a billion microbial cells,dominated by lactobacilli.Here we perform metagenomic shotgun sequencing on cervical and fecal samples from a cohort of 516 Chinese women of reproductive age,as well as cervical,fecal,and salivary samples from a second cohort of 632 women.Factors such as pregnancy history,delivery history,cesarean section,and breastfeeding were all more important than menstrual cycle in shaping the microbiome,and such information would be necessary before trying to interpret differences between vagino-cervical micro-biome data.Greater proportion of Bifidobacterium breve was seen with older age at sexual debut.The relative abundance of lactobacilli especially Lactobacillus crispatus was negatively associated with pregnancy history.Potential markers for lack of menstrual regularity,heavy flow,dysmenor-rhea,and contraceptives were also identified.Lactobacilli were rare during breastfeeding or post-menopause.Other features such as mood fluctuations and facial speckles could potentially be predicted from the vagino-cervical microbiome.Gut and salivary microbiomes,plasma vitamins,metals,amino acids,and hormones showed associations with the vagino-cervical microbiome.Our results offer an unprecedented glimpse into the microbiota of the female reproductive tract and call for international collaborations to better understand its long-term health impact other than in the settings of infection or pre-term birth.
