2.Problem and development of the hospital medicine preparation in Beijing

Objective To realize the actuality and analyze the problem of the hospital medicine preparation in Beijing,then to discuss its developing methods.Methods By summarizing questionnaire of the hospital medicine preparation and combining with the problem showed in daily supervision,we analyzed the main existing problems and reasons for hospital medicine preparation and put forward the developing methods.Results The main existing problems of hospital preparation are the decreasing variety of medicine preparation,insufficient infrastructure and software.Conclusion Therefore we should enhance the input on the infrastructure,set up the software,improve the quality of the staff,and consummate the regulation means and system.
3.Infection of respiratory syncytial virus in BALB/c mice and nude mice

Objective To compare respiratory syncytial virus(RSV)infection and inflammatory responses between immunocompetent BALB/c mice and immanodeficient nude mice.Methods At various time points after BSV infection of BALB/c mice and nude mice,pulmonary viral titers were assayed.Leukocvtes in bronchoalveolar lavage fluid(BALF)and pulmonary histology were identified.F4/80+cells and CD49b+cells in lung tissue were examined by immunohistochemistry,and the cytokines of TNF-α,IL-12,IFN-r and IL-10 in BALF were assayed by ELISA.Results RSV titers in infected BALB/c mice and nude mice peaked on the 3rd day postinoculation,and nude mice had higher-level and more durative viral replication than BALB/c mice.RSV infection induced more severe pulmonary histopathology and larger number of leukocytes in airway in nude mice than in BALB/c mice.RSV infection enhanced more pulmonary F4/80+macrophages,CD49b+ NK cells in both mice.Furthermore infected nude mice had larger amount of pulmonary macrophages and NK cells than infected BALB/c mice.RSV infected BALB/c mice secreted more TNF-α,IL-12,IFN-r and IL-10 as compared with control BALB/c mice,and infected nude mice had hisher level of TNF-α.IL-12 and IL-10 than infected BALB/c mice.Conclusion Nude mice are good model for severe and pemistent RSV infection in immunocomprised hosts.The inflammation induced by RSV infection is not parallel with the immune response of T cells,and macrophages and NK cells are potent immunocytes and inflammatory cells in RSV infection especially when T lymphocytes are absent.
4.Relation Between Anticardiolipid Antibodies and Brain Infarction

We measured IgG and IgM ACA levels by a modified enzyme一linked immunosorbent as-say(ELISA) in 122 patients with brain infarction and 86 healthy controls. Significant difference in positiveIgG and IgM ACAs was found between the patients with infarction(50.8%)and the control group(11.6%).The relation between positive ACAs and brain infarction was studied. The pathogenetic mecha-nism remains unclear.
5.Tissue distribution and excretion of Hylotelephin

Objective To investigate tissue distribution of Hylotelephin in Beagle dogs and excretion of Hylotelephin in rats. Methods A high performance liquid chromatography (HPLC) with UV detection was developed to study the concentrations of Hylotelephin in biological samples, taking anthracene as an internal standard, Benzoyl chloride as the pre-column derivatization of Hylotelephin and methanol-water as the mobile phase. Results After a single intravenous dose of 10.6 mg/kg Hylotelephin in beagle dogs, parent drug was widely distributed to virtually all tissues in 5 min and the concentration of Hylotelephin in most tissues at 90 min were lower than those at 5 min obviously. Hylotelephin was mainly distributed in kidney, liver and spleen, secondly in heart, lung and intestine. The parent drug concentration in kidney, liver and spleen was similar to that in blood at the same time point. After a single intravenous dose of 36 mg/kg Hylotelephin in rats, the excretion of the parent drug in urine, feces and bile amounted to 88.45%, 0.61% and 1.08% of the dose, respectively. The parent drug excretion amounted to 67% of the dose in 3 h. Conclusion Hylotelephin was distributed and eliminated in Beagle dogs rapidly. It was mostly distributed in kidney, liver, spleen and plasma. The parent drug excretion was 88% via urine.
6.Inhibitory effect of poly (lactic acid)electrospun membranes loaded with cisplatin and chloroquine on proliferation of oral squamous cell carcinoma CAL-27 cells

Objective:To investigate the effect of poly (lactic acid)(PLA)electrospun membranes loaded with cisplatin and chloroquine on the oral squamous cell carcinoma CAL-27 cells,and to explore the method to prevent the recurrence of oral cancer.Methods: The DDP/PLA membranes, CQ/DDP/CQ/PLA membranes and CQ/DDP/PLA membranes were prepared by electrospinning.Then the micro morphology of three kinds of membranes were observed by scanning electron microscope (SEM);the degradation rate of PLA membrane was measuredby UV spectrophotometric.The LC3-Ⅱ expression level in CAL-27 cells was detected by laser scanning confocal microscope.The survival rate of CAL-27 cells was detected by MTT method.Results:The SEM results showed that the nanofibers of DDP/PLA,CQ/DDP/PLA and CQ/DDP/CQ/PLA membranes were continuous and smooth with uniform diameters.The degrated time of membranes was about 21 d.The MTT result showed that compared with control group,at first,the effects of cell killing of DDP/PLA membranes,CQ/DDP/CQ/PLA membranes and CQ/DDP/PLA membranes were not obvious;as the extension of time,the survival rates of CAL-27 cells in DDP/PLA membranes group,CQ/DDP/CQ/PLA membranes group and CQ/DDP/PLA membranes group were decreased (P <0.05).The immunofluorescence results showed that the fluorescence intensity of LC3-Ⅱ in CQ/DDP/CQ/PLA membranes group and CQ/DDP/PLA membranes group were lower than that in DDP/PLA membranes group.Conclusion:CQ/DDP/PLA membranes with sustained-release effect can increase the sensitivity of CAL-27 cells to DDP and enchance the killer effect of DDP on the CAL-27 cells.
7.Analysis of contact allergens in 1065 patients with dermatitis or eczema in Wuxi city

Objective To determine contact allergens and their clinical significance in patients with dermatitis or eczema in Wuxi city.MethodsPatch test was conducted in 1065 patients with a clinical diagnosis of dermatitis or eczema.A statistical analysis was carried out.Results Positive patch test reactions were observed in 83.19% of the patients,with no statistical differences in the frequency of positive patch test reactions between female and male patients or between patients of different age groups.The most frequent 6 allergens were potassium dichromate,cobalt chloride,nickle sulfate,formaldehyde,carba mix and fragrance mix in these patients.Female patients showed a higher frequency of positive patch test reaction to nickle sulfate and formaldehyde,but a lower frequency to potassium dichromate and carba mix compared with male patients (all P< 0.01 ); young,middle-aged,and old patients exhibited an increased frequency of positive reaction to cobaly chloride compared with child patients,while young and middle-aged patients showed a decreased frequency to nickel sulfate compared with child patients (all P < 0.05).Conclusion Potassium dichromate,cobalt chloride,nickle sulfate,formaldehyde,carba mix and fragrance mix are the main allergens causing dermatitis and eczema in Wuxi city.
8.The role of Huang-qi in balance of THl/TH2 in asthma on dendritic cells level

Objective To investigate the role of Huang-qi in balance of TH1/TH2 in asthma on dendritic cells level. Methods DCs from human peripheral blood mononuclear cells(PBMC) were induced by rhGM-CSF and rhIL-4, and then were identified. The level of IL-12 and IL-10 produced by DCs were de-tected by ELISA assay. After autoreactive T cells, mRNA of T-bet and GATA-3 was measured by RT-PCR. Simultaneously, IL-4 and IFN-γ were determined by flow cytometer. Results After 7 days culture, IL-12 was significantly decreased in asthma group compared to control group (P < 0.01), whereas IL-10 on the opposite. At the 7th day of co-culture with T cells derived from floating cells, the IFN-γ/and T-bet mRNA level in asthma group were significantly decreased than that in control group, whereas IL-4, GATA-3 mRNA level, the ratio of IL-4/IFN-γ and GATA-3/T-bet were apparently increased in asthma group than that in control group(P<0.01). After Huang-qi treatment, the IFN-γ/and T-bet mRNA level were significantly in-creased, whereas the ratio of IL-4/IFN-γ and GATA-3/T-bet, and the IL-10 level were apparently de-creased, but the level of IL-12, IL-4 and GATA-3 mRNA were not changed significantly. Conclusion DCs in asthma regulated the balance of TH1/TH2 by means of secreting decreased IL-12 and increased IL-10, that made TH2 playing a dominance role which is the key factor in initiating asthma. Huang-qi regulated DCs through decreasing the level of IL-10, and thus decreased the ability of inhibiting the differentiation of TH1 from TH0, that is also inhibiting the differentiation of TH2 from TH0 directly.
9.Down-regulation of monoamine neurotransmitter synthetase expression involved in CUS-induced depression of rats

Aim To investigate whether chronically un-predictable stress (CUS)-induced depression-like be-haviors of rats is associated with the variant expression of tryptophan hydroxylase (TPH)and tyrosine hydrox-ylase (TH).Methods 30 male SD rats were ran-domly divided into depression model group(MG)and control group (CG),the former was established using CUS plus solitary condition for 28 d,whereas the latter was fed normally as five rats per cage without CUS. The open field test(OFT)and the sucrose preference test were used to evaluate depressive behaviors.Both mRNA and protein expressions of TPH and TH in hip-pocampus and forebrain cortex were determined by re-al-time fluorescent quantitative PCR and western blot (WB),respectively.Results MG rats showed obvi-ous depressive behaviors with much lower locomotive activity and sucrose preference than CG.Meanwhile, the mRNA and protein expressions of TPH and TH also significantly decreased in MG rats,compared with CG rats.Conclusion The depression behaviors of rats in-duced by CUS may be associated with down-regulation of TPH and TH expression.
10.Biological effects of lipopolysaccharide, transforming growth factor-β1 on murine bone marrow-derived dendritic cells

AIM: To explore method of stimulating murine bone marrow-derived dendritic cells by lipopolysaccharide(LPS), transforming growth factor-β1 (TGF-β1)and to study their biological character. METHODS: Murine bone marrow-derived dendritic cells were cultivated with cytokine GM-CSF and IL-4 for 6 days, BMDC was stimulated by control, LPS, TGF-β1, LPS +TGF-β1 for 48 hours respectively. Morphological characters of BMDC were observed by a inversed microscope, surface molecules such as CD_(11C), CD_(80), CD_(86)and MHC Ⅱ were detected by flowcytometry, Interleukin-6 and interleukin-12 p70 in co-culture medium was quantified by ELISA. RESULTS: In LPS group it presented the most typical DC morphology with the highest expression of CD_(80), CD_(86) and MHC Ⅱ, the strongest ability in mixed lymphocyte reaction, higher level of IL-6 and IL-12 p70 compared with control, TGF-β1, LPS + TGF-β1 ( P < 0. 05). While in TGF-β1 group it presented the less typical DC morphology with the lower expression of CD_(80), CD_(86), MHC Ⅱ, weaker ability in mixed lymphocyte reaction, and lower levels of IL-6 and IL-12 p70 compared with control and LPS (P < 0.05). CONCLUSION: LPS can stimulate maturation of BMDC in its late differentiation which makes it presents a more significant biological characteristics. TGF-β1 can inhibit maturation but not differentiation of BMDC thereby can prevent its biological characteristic presentation.
