Objective To study the relationship between tumor angiogenesis and expression of ras p21 protein as well as the prognosis in serous ovarian neoplasms.Methods Microvessel density (MCD) and expression of ras p21 protein in issues of 10 adenomas and 50 serous ovarian cancers were observed by immunohistochemical staining methods with monoclonal antibody CD 34 to mark vascular endotheliocytes.Results Microvessel density in specimens from over expression of ras p21 protein and from grade Ⅱ and Ⅲ was significantly higher than that in specimens from low expression or negative expression of ras p21 protein as well as from grade I (P
2.Evaluation of transvaginal color Doppler sonography and magnetic resonance imaging in diagnosis for myometirial invasion of endometrial cancer

Objective To investigate the value of transvaginal color Doppler sonography (TVCDS) and magnetic resonance imaging (MRI) in diagnosis for myometrial invasion of eudometrial cancer before surgical operation, in comparison with pathological examinations. Methods TVCDS and MRI were undergone before surgical operation in 34 patients with endometrial cancer confirmed by biopsy to evaluate depth of its myometrial invasion, as compared to those with postoperative pathological examinations. Results Predictive accuracy for myometrial invasion by TVCDS was 85%, with sensitivity, specificity, positive predictive value and negative predictive value of 80%, 9/9, 100% and 9/14, respectively, and predictive accuracy by MRI was 68%, with sensitivity, specificity, positive predictive value and negative predictive value of 60%, 8/9, 15/16 and 8/18, respectively. Resistance index (RI) averaged 0.51 for flow signal to detect superfical myometrial invasion by color Doppler flow imaging (CDFI) and 0.43 for deep myometrial invasion. Conclusions TVCDS and MRI can be used before surgical operation to judge the depth of myometrial invasion for patients with endometrial cancer, particularly more suitable for its screening, which will be helpful to judge the depth of myometrial invasion incorporated with resistance index by color Doppler flow signal.
3.Valproic acid exerts differential effects on cytokine synthesis in human peripheral lymphocytes

AIM: Valproic acid (VPA) is a histone deacetylase inhibitor and is believed to have anti-tumor activity. The present study aims to investigate the effect of VPA on the, apoptosis and cytokine synthesis of human peripheral lymphocytes. METHODS: The activation and cytokine synthesis in lymphocytes in whole blood stimulated with phorbol dibutyrate (PDB) and ionomycin were evaluated with flow cytometry after fluorescent staining. The mitochondrial membrane potential was examined using 3, 3-dihexyloxacarbocyanine iodide [DiOC6(3)]staining. RESULTS: VPA at low doses (1 and 5 mmol/L) promoted CD69 expression in activated lymphocytes, whereas it turned to inhibit the expression of CD69 at a high dose (25 mmol/L). Meanwhile, VPA at low doses increased the mitochondrial membrane potential, while a high dose of VPA decreased it in activated lymphocytes. Furthermore, interleukin-2 (IL-2) synthesis was enhanced by low doses of VPA but inhibited by a high dose. However, interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) synthesis were dose-dependently enhanced by VPA as compared with those of PDB plus ionomycin-treated cells. CONCLUSION: VPA exerts biphasic effect on the further activation and apoptosis of human peripheral lymphocytes stimulated with mitogens and exhibits differential activity on the synthesis of several important cytokines in human lymphocytes.
4.Effect of Silencing Ran Gene by siRNA on Apoptosis and Expressions of Caspase-3 and PARP in Colon Cancer Cell Lines

Colon cancer is one of the common malignant tumors of digestive system.In our previous study, it has been demonstrated that colon cancer specific antigen thioredoxin like-2b (Txl-2b) can interact with Ras-related nuclear protein (Ran).However, there are few reports about the role and mechanism of Ran in tumorigenesis of colon cancer.Aims: To investigate the effect of Ran-targeting RNA interference on apoptosis and expressions of caspase-3 and poly(ADP-ribose) polymerase (PARP) in colon cancer cell lines.Methods: 20, 40 and 60 nmol/L siRNA-1 (si-1 group), siRNA-2 (si-2 group), siRNA-3 (si-3 group) targeting to Ran gene and normal control siRNA (NC group) were transfected into colon cancer cell line HCT116 and DLD-1, respectively.The interference efficiency of siRNAs and expressions of caspase-3 and PARP were detected by Western blotting.Cell apoptosis was determined by flow cytometry.Results: 20 nmol/L siRNA-1 and siRNA-2 had the best effect on inhibiting expression of Ran.Early apoptosis rates of HCT116 and DLD-1 cells in si-1 group were significantly higher than those in NC group (19.37%±7.57% vs.4.83%±1.72%;16.53%±3.38% vs.6.27%±3.13%;P all <0.05).Late apoptosis rates of HCT116 and DLD-1 cells in si-1 and si-2 groups were significantly higher than those in NC group (15.97%±3.31%, 16.33%±5.40% vs.6.40%±1.05%;22.93%±1.57%, 11.50%±0.70% vs.6.20%±0.98%;P all <0.05).Compared with NC group, expressions of cleaved caspase-3 (active form) and cleaved PARP (inactive form) were significantly increased in DLD-1 cells of si-1 and si-2 groups.Conclusions: Silencing Ran gene can significantly promote the apoptosis of colon cancer cells, and its mechanism is related to the regulation of caspase-3 and PARP expression.
5.Clinical value of transvaginal ultrasound, MRI and hysteroscopy in the assessment of endometrial cancer lesion size

Objective To investigate the clinical significance of transvaginal ultrasound, hysteroscopy and MRI in the assessment of endometrial cancer lesions size. Methods Data from 56 patients who successively underwent transvaginal ultrasound, MRI and hysteroscopy inspection preoperative endometrial carcinoma were retrospectively analyzed to assess the accuracy of lesions size. Results The pathologic lesions size measured mean maximum diameter of 56 cases was (3.05 ± 0.23) cm, while the mean maximum diameter measured by vaginal ultrasound, MRI and hysteroscopy were respectivelly (2.46±0.31) cm, (3.12± 0.08) cm, and (3.18 ± 0.21) cm. Compared with the pathologic measured values, the compliance rates of transvaginal ultrasound, hysteroscopy and MRI were respectively 54%(30/56), 71%(40/56)and 75%(42/56), which vaginal ultrasound measurement value was significantly different than that by pathologic measured ( P=0.031), while there were significant difference between the hysteroscopy measured lesion size and pathologic measured, or between MRI measured values and pathologic measured (all P>0.05). Conclusion Preoperative assess the endometrial cancer lesions size, significance of vaginal ultrasound examination is limited, and MRI and hysteroscopy examination is accurate, but easy to over-estimated lesion size.
6.Application of endometrial sampling device during the follow-up visit for the conservative treatment of endometrial cancer

Objective To study the feasibility of endometrial sampling device as a sampling tool during the follow-up visit for endometrial cancer patients undergone conservative treatment.Methods Before the hysteroscopy examination,endometrial sampling device was used to take the endometrium specimens 43times in 19 patients who had been diagnosed as endometrial cancer or atypical hyperplasia,and were undergone conservative treatment during May 2012 to Mar.2013.All cases accepted vaginal ultrasound screening before every sampling by endometrial sampling device.The histological results were compared with those done by hysteroscopy.Results The average age of those patients was (30 ± 6) years old.The mean thickness of the endometrium during the treatment was (0.81 ± 0.65) cm.The qualified rate for the sampling was 95% (41/43).Compared with the specimens undergone by hysteroscopy direct sampling,32samples got by the endometrial sampling device with thicker endometrium (0.93 ± 0.70) cm had the same histological results,while the other 9 patients with thinner endometrium (0.40 ± 0.14) cm were not (P =0.031).Conclusion The endometrial sampling device could be used during the follow-up visit for the conservative treatment patients with endometrial cancer or atypical hyperplasia,the vaginal ultrasound screening should be used together to figure out those with thinner endometrium.
7.Protection Effects of a New Combined Anti-G Measure

Objective To observe the protection effects of a new combined anti-G measure, which is composed of new bladder anti-G system, unassisted positive pressure breathing for G (PBG) and PHP maneuver. The problem of fatigue and pain when using this measure is also discussed. Method Each of the six fully qualified centrifuge subjects experienced 5 groups of centrifuge runs:1) relaxed +Gz tolerance without anti-G equipments and PHP maneuver;2) +Gz tolerance with FLH-x+KT-x;3) +Gz tolerance when performing PBG with FLH-x, KT-x, YM-x and TK-x;4) +6.5 Gz for 45 s using the same anti-G equipments as the 3rd group;5) +9.0 Gz for 15 s using the same anti-G equipments as the 3rd group and performing PHP maneuver. Result There were no incidents of G-induced loss of consciousness in this study. The protective effects of FLH-x+KT-x, PBG and PHP maneuver were 2.5 G, 1.67 G and 1.23 G respectively. All the subjects had accomplished the 6.5 G 45 s and 9.0 G 15 s runs with the new combined anti-G measure. The pain occured on neck, waist, arm and hands. Conclusion The new combined anti-G measure can provide enough anti-G protection for modern high performance aircraft. How to prevent the occurrence of neck injury and alleviate the pain induced by G when using this measure needs further investigation.
8.The value of Revolution CT in identifying the tumor-lung interface between central lung cancer and accompanying atelectasis

Objective To study the application value of multi-parameter imaging of Revolution energy spectrum CT in identifying the tumor-lung interface between central lung cancer and accompanying atelectasis.Methods The spectral CT imaging data of 73 patients with central lung cancer and accompanying atelectasis confirmed by pathology were collected.The polychromatic image,the best monochromatic image,the best monochromatic image combined with iodine concentration map and the effective atomic number(Eff-Z)of the tumor-lung interface in the arterial,venous and delayed phases were compared,and the CT value,iodine concentration(IC)value,Eff-Z and the slope of energy spectrum curve(λHU)of tumors and atelectasis in the venous phase were also compared.Results(1)On tri-phase CT,the subjective score of the tumor-lung interface in the venous phase was the highest.(2)There were differences among the subjective scores of tumor-lung interface on polychromatic image,best monochromatic image,best monochromatic image com-bined with iodine concentration map and Eff-Z on tri-phasic enhanced CT.The subjective score of the best monochromatic image combined with iodine concentration map was the highest,followed by the best monochromatic image.(3)There were significant differences in CT values,IC values,Eff-Z and λHU between tumors and atelectasis in venous phase,with the highest difference of IC value.Conclusion Revolution energy spectrum CT can identify the tumor-lung interface between central lung cancer and accompanying ate-lectasis through multi-parameter imaging,which may provide helpful information for the diagnosis and therapeutic evaluation of the disease.
9.Efficacy evaluation of different anti-G physical training programs for pilots

Objective To establish a scientific training program that takes into account both anaerobic and aerobic training for pilots,and to explore the appropriate ratio of aerobic and anaerobic training.Methods According to the physical examination standards for pilots,a total of 16 healthy subjects aged 18-24 were selected from two batches.The two batches of subjects were trained with different aerobic and anaerobic ratios.Training period was 3 months.The changes in cardiopulmonary function of the subjects before and after training were evaluated using the cardiopulmonary function exercise testing system(CPET),and the changes in anaerobic capacity were evaluated using changes in strength as an indicator.Results After training,the weight load of the subjects in the two training programs,including barbell squats,leg flexion and hard pull,and barbell under 10RM and 3RM,was significantly increased(P<0.001),and there was no statistically significant difference in anaerobic strength growth between the two groups.The results of CPET showed that the maximum load,maximum heart rate,and respiratory quotient in the two groups were significantly increased after than before the training(P<0.01).The maximum load(Experiment group 1:29.12±19.69,Experiment group 2:72.00±46.24)and respiratory quotient(Experiment grouop 1:0.11±0.09,Experiment group 2:0.28±0.16)of the subjects in experiment group 2 before and after training were greater than those in experiment group 1.The difference was statistically significant(P<0.05).Conclusion The anaerobic and aerobic capacities of the subjects in the experiment group 2 are effectively improved,indicating that ratio of aerobic and anaerobic of the training scheme is better.
