1.Determination and Pollution Control of Microcystins

As a most common algal toxin in eutrophic freshwater body, microcystins can be produced by the bloom-forming Cyanophyta microcystis, it has become a potential hazardous substance in aquatic environments for its toxic, distribution and stability. With the increasing recognition of microcystins, China had added microcystin-LR into the related water quality standards. It is very important to detect and control the microcystins in the aquatic environment. This paper systematically introduced the current situation of researches about the determination and pollution control technique of microcystins in drinking water at home and abroad in resent years and then some issues worth of paying more attention to and doing further study in pollution control in future were presented.
2.Endoscopic Diagnosis and Treatment of Endogenous Adenoma of the Duodenal Papilla:Report of 8 Cases

Objective To explore the diagnosis and treatment of endogenous adenoma of the duodenal papilla by endoscopy.Methods Between June 2005 and October 2008,eight patients who had dilated common and intrahepatic bile ducts with strictured opening of the papilla and lower segments shown by B-ultrasonography or MRCP,were enrolled in this study. The patients were assigned to receive ERCP and IDUS for diagnosis and treatment. Results In this series,three patients received pancreatico-duodenectomy because of tubulovillous adenoma (two cases) or tubular adenoma (one case) complicated with moderate atypical epithelial hyperplasia; one of the two cases with tubulovillous adenoma was confirmed as having local cancer by postoperative pathological examination,and then received interventional therapy and embolization because of liver metastasis in 3 months; the patient who had tubular adenoma was diagnosed with local signet ring cell cancer after the operation,and was followed up at 6 and 14 months afterwards by physical examination,which showed no abnormalities. Endoscopic snare papillectomy followed by bile duct stenting for drainage was performed on two patients,one of them was diagnosed with tubulo-papillary adenoma after the surgery and then showed recurrent tumor at 6 months,resulting death at 10 months; the other one was confirmed as having tubulovillous adenoma complicated with moderate atypical epithelial hyperplasia,this patient showed no abnormalities during follow-up at 6 and 12 months. Plastic bile duct stent was employed in one patient with severe atypical epithelial hyperplasia,but was replaced by metal stent because of repeated infection of the bile duct. One patient with papillary adenoma was lost to follow-up after bile duct stenting for intradrainage. One patient refused any treatment for the tubular adenoma; follow-up by phone showed no abnormality in this case in 12 months.Conclusions Endogenous adenoma of the duodenal papilla is rare detected. The tumor is very likely to turn to malignant. Multi-point biopsy is necessary for the patients with hyperplasia of the mucosal and muscle layers.
3.Analysis of the new mutations of the gene in Chinese patients with congenital retinoschisis

Objective To investigate the mutations of the gene in Chinese patients with X linked juvenile retinoschisis (XLRS), and to provide the genetic diagnosis and consultation of heredity for the patients and their families. Methods Genomic DNA was isolated from leukocytes of 29 male patients with XLRS, 38 female carriers and 100 normal controls (the patients and the carriers were from 12 families). All 6 exons of XLRS1 gene were amplified by polymerase chain reaction (PCR) and analyzed by single strand conformation polymorphism (SSCP) assay. The positions and types of XLRS1 gene mutations were determined by direct sequencing. Results Eleven different XLRS1 mutations were identified in these 12 families, including one frameshift mutation due to base loss of the first exon: c.22delT(L9CfsX20), one nonsense mutation due to base loss of the first exon (Trp163X), one splice donor site mutation(c.52+2 T→C; IVS1+2T to C), and eight missense mutation due to base replacement(Ser73Pro, Arg102Gln, Asp145His, Arg156Gly, Arg200Cys, Arg209His, Arg213Gln, and Cys223Arg). No gene mutation was detected in the control group. Four new mutations included frmaeshift mutation(L9CfsX20) and mutations of Asp145His, Arg156Gly, and Trp163X at the fifth exon. A newly discovered non-disease-related polymorphism (NSP) was the c.576C to T (Pro192Pro) change at the sixth exon. Conclusion Eleven different XLRS1 mutations were detected, which is the cause of XLRS in Chinese people. The detection of gene mutations may provide the guidance of genetic diagnosis and the consultation of family heredity for the patients and their families.

Long non-coding RNA (lncRNA) widely exists in living organisms.It,whose length is more than 200 bp,can regulate the genic expression in the epigenetic,transcriptional and post-transcriptional level.Therefore,it can influence cell proliferation,apoptosis,vitality,immune response and oxidative stress.In recent years,the study of lncRNA is developing rapidly in the ophthalmic research,and this article reviews the latest research progress on mechanism of lncRNA in the ophthalmology disease.
5.Influence of Continuous Nursing Intervention on Psychological Quality and Medication Compliance in Ulcerative Colitis Patients after Discharge

Objective To explore the effect of follow-up continuous nursing intervention on the psychological quality and medication compliance in patients with ulcerative colitis after discharge. Methods Ninety-seven patients with ulcerative colitis after discharge from October 2014 to October 2015 were recruited,and 48 cases were selected in the study group and 49 cases in the control group randomly. The control group received routine postdischarge care. The study group received postdischarge follow-up nursing intervention. After 3 months of postdischarge intervention, the psychological quality of patients in two groups was assessed by the emotion application measure scale separately. The Morisky Medication Adherence Scale 8-item version (MMAS 8-item version) was used for the assessment of the medication compliance of the two groups of patients. Results After the intervention, the patients in the study group showed better psychological quality and medication compliance than the patients in the control groups.There were significant differences between two groups. Conclusion Post-discharge follow-up nursing intervention on patients with ulcerative colitis is effective for improving the psychological quality and the medication compliance and may be helpful to improve the life quality of the patients.
6.Extraction of polysaccharides from Asterias amurensis and preliminary study on their antibiotic activities in vitro

Objective To explore the extraction technology of polysaccharides from Asterias amurensis and study their antibacterial activities in vitro. Methods Determined by spectroscopic methodology with phenol -sulfuric acid,the polysaccharide content was taken as inspecting index to select the extraction technology such as enzymatic hydrolysis,alkali,and alkali-enzymatic combined. Filter paper method was used to test the polysaccharides antibiotic activities in vitro. Results The optimum extraction technology was adding 2 times amount of 1 mol?L-1 sodium hydroxide,heating for 2h,under these conditions the greatest yield of starfish polysaccharides would be obtained which showed inhilitory effect on Staphylococcus aureus. Conclusion The alkal hydrolysis extraction technology is superior to the other methods; polysaccharides from Asterias amurensis showed antibiotic activity to some kinds of bacteria.
7.Mitochondrial DNA deletion of the brain tissue of aged rats with learning and memory deficit

AIM and METHODS: The ratio of mitochondrial DNA (mtDNA) deletion was measured to find the relationship between mtDNA deletion and aged learning and memory deficit. The aged rats were divided into two groups, aged learning and memory deficit group and aged learning and memory normal group. The ratio of mtDNA deletion was measured by dilution polymerase chain reaction. RESULTS: There are deleted mtDNA (about 4834 bp) in the cerebral cortex, hippocampus and cerebellum of both young and aged rats. The ratios of deleted mtDNA were similar in the cerebral cortex,hippocampus and cerebellum of young rats (about 0.00018%). The ratio mtDNA of aged learning and memory normal rats had increased by five-fold in the cerebral cortex and hippocampus, or one-fold in the cerebellum over young rats. The ratio of aged learning and memory dificit rats had increased by one-fold in the cerebral cortex or 0.8-fold in the hippocampus or two-fold in the cerebellum over aged learning and memory normal rats.CONCLUSIONS: There was really the increase of mtDNA in aging rat brain. And this increase was double in amount in aged learning and memory deficit rats compared to the normal learning and memory aged rats. It is suggested that the mtDNA deletions in the brain regions associated with learning and memory may be contributed to the cellular and molecular mechanism of learning and memory deicit with aged rats.
8.Pathogenic risk factors correlation study of developmental dysplasia hip in Shigatse of Tibet

Objective To perform the epidemiological analysis of the developmental dysplasia hip(DDH) in Shigatse of Tibet in order to explore the correlation between the onset of DDH with some risk factors such as altitude and take targeted intervention measures to reduce the disability rate of DDH.Methods A total of 5900 children aged 0 to 14 year-old from different altitudes in Shigatse of Tibet were surveyed randomly from June 2013 to June 2014.SPSS 18.0 statistical software was used to analyze the correlation of the onset of DDH with residence altitude,production and living ways,binding method of the lower limbs,delivery situation and genetic factor.Results One hundred and ninety-one DDH cases were screened out,and the average incidence of DDH in Shigatse of Tibet was about 32.4‰,and unilateral was higher than bilateral,fight side(33 cases)∶left side(68 cases) =1.00∶ 2.06.The result of Pearson correlation analysis of the onset of DDH with residence altitude showed that there was significant correlation (r=0.820,P=0.046).The result of Spearman correlation analysis of the onset of DDH with production and living ways showed that there was significant correlation(r=0.530,P =0.048).The result of x2 test analysis of the onset of DDH with binding method of the double lower limbs and breech delivery showed that there was statistical significance (P=0.0017,0.0082).Conclusions The DDH incidence of children living in high altitude areas in Tibet has certain regional characteristics indicating that it has clear correlation with some environmental risk factors such as altitude,methods of the lower limbs binding,production and living ways and breech delivery,especially the bundled package of lower limbs is the main environmental risk factor of DDH.
9.Study on Fap1 glycosylation and maturation regulating by ORF3 coded by fap1-orf4 gene locus of Streptococcus parasanguis

Objective To study whether ORF3 coded by fap1-orf4 gene locus of Streptococcus pa-rasanguis is involved in the regulation of Fap1 glycosylation and maturation and to investigate whether ORF3 influences Streptococcus parasanguis adhesion. Methods A gene replacement strategy was adapted to con-struct orf3 alleic replace mutant of Streptococcus parasanguis. Complementation assay and Western blot were used to test Fap1 expression levels. Whole saliva-coated hydroxyapatite (SHA) adhesion assay was adapted to determine Streptococcus parasanguis adhension. Results (1) Non-polar was found in strain VT1774, the orf3 alleic replace mutant of Streptococcus parasanguis. (2) Western blot showed that mature Fapl (Mr about 220 × 103) disappeared and were substituted with high molecular weight Fapl (Mr about 470 × 103) in strain VT1774, furthermore, complementation assay showed VT1775, the complementation strain of VT1774, re-stored mature Fapl expression. (3) The binding ability reduced significantly in strain VT1774. Conclusion ORF3 coded byfapl-orf4 gone locus was required for Fap1 glycosylation and maturation in Streptococcus pa-rasanguis, orf3 alleic replacment resulted in Fap1 glycosylation and mature disorder and decreasing of adhen-sion ability of Streptococcus parasanguis.
10.Effects of stromal derived factor-1 and CXC chemokine receptor-4 in rats with white matter damage treated with human umbilical cord mesenchymal stem cells

Objective To investigate the effects and the migration mechanisms of stromal derived factor-1 (SDF-1) and CXC chemokine receptor-4 (CXCR-4) in rats with white matter damage treated with human umbilical cord mesenchymal stem cells (hUC-MSCs).Methods A total of 108 three-day old Sprague-Dawley rats were randomly divided into the experimental group,control group and sham group.The left common carotid artery was ligated and then exposed to hypoxia of 6% O2 and 94% N2 in rats in the experimental and control groups.Rats in sham group were neither ligation nor hypoxia.After 24 hours,rats in the experimental group were administered 0.5 ml hUC-MSCs (1 × 106/ml) intraperitoneally,and rats in control and sham groups were administered 0.5 ml saline by the same way.Immunohistochemistry and reverse transcription-polymerase chain reaction were used to determine the expression of SDF-1 and CXCR-4 protein and mRNA in 5-,7-and 14-day-old rats.Analysis of variance and the LSD test were used for statistical analysis of the data.Results HE staining showed that,in 14 day-old rats of the experimental group,bilateral cerebral ventricles were similar with no cellular edema or necrocytosis.In the sham group,bilateral cerebral ventricles were also normal.However,in the control group,ventriculomegaly,cellular degeneration and necrocytosis were observed on the left side.On the 5th,7th and 14th day,SDF-1 protein levels were 0.15±0.06,0.24±0.01 and 0.12±0.01,and CXCR-4 protein levels were 0.35±0.16,0.60±0.21 and 0.72±0.25,respectively,in the experimental group; SDF-1 protein levels were 0.13 ± 0.01,0.16± 0.01 and 0.08± 0.01,and CXCR-4 protein levels were 0.18 ± 0.04,0.17 ± 0.09 and 0.25 ± 0.06,respectively,in the control group,and all were higher than those in the sham group (SDF-1 protein levels were 0.03 ± 0.01,0.04± 0.01 and 0.02±0.01; and CXCR-4 protein levels were 0.04±0.02,0.05±0.03 and 0.05±0.03,respectively) (LSD test,all P<0.05).SDF-1 protein increased to a peak on the 7th day and decreased on the 14th day in the experimental group,however,these values were both higher than those in the control group (LSD test,both P<0.05).CXCR-4 protein increased on the 5th day and continued to increase up to the 14th day in the experimental group,and these values were higher than those in the control group at the three time points (LSD test,all P<0.05).In 5-,7-and 14-day-old rats,SDF-1 mRNA levels were 3.52 ± 0.33,4.18± 0.28 and 2.60± 0.21,respectively,in the experimental group,which were higher than those in the control group (2.07± 0.34,3.73 ± 0.28 and 2.08± 0.15,respectively),and were even higher than those in the sham group (0.99±0.17,1.00±0.16 and 1.31 ±0.32,respectively) (LSD test,all P<0.05).In the experimental group,SDF-1 mRNA levels reached a peak on the 7th day,and on the 14th day,it decreased to the level lower than that on the 5th day (LSD test,all P<0.05).In the control group,SDF-1 mRNA levels also reached a peak in 7-day-old rats,but not in 14-day-old rats,which was similar to 5-day old rats (LSD test,9>0.05).In 5-,7 and 14-day-old rats of the experimental group,CXCR-4 mRNA levels were 1.32±0.29,1.75±0.36 and 2.33±0.49,respectively,higher than those in the sham group (1.00±0.16,0.94±0.16 and 0.81±0.14,respectively) and the control group (0.97±0.14,0.97±0.15 and 1.07±0.25,respectively) (LSD test,all P<0.05).In the experimental group,CXCR-4 mRNA levels were higher in 14-day-old rats than that in 5-and 7-day-old rats (LSD test,both P<0.05).Conclusions SDF-1/CXCR-4 may play a vital role in the migration of hUC-MSCs homing to damaged brain.
