1.Progress in enhancing radiosensitivity by inhibition of DNA double strand break repair
Journal of International Oncology 2010;37(5):358-361
DNA double strand breaks are potentially lethal DNA lesions induced by ionizing radiation, which can be repaired by homologous recombination and non- homologous end joining. Inhibition of the expression and function of genes in double strand breaks repair pathways using RNA interference and anti- sense techniques could improve the radiosensitivity of cancer cells.

Journal of International Oncology 2014;41(5):327-331
Hypoxia is an inherent feature of the majority of solid tumors, which can increase the resistance of tumor cells to radiotherapy and chemotherapy, promote tumor angiogenesis and lead to poor prognosis. Therefore, targeting the hypoxic tumor cells has become a spot in cancer therapy.Bioreduction drugs can be activated by a specific reduction to become cytotoxic metabolites, thus killing hypoxic tumor cells, while small molecular targeting inhibitors can selectively act on the key point of hypoxia pathway. They have paved a new way for hypoxia targeted therapy.

Objective To investigate the effects of stress intensity and the expected duration of stress on the inhibition ability of individual responses to stress. Methods A total of 60 cases of hospitalized patients in respiratory department were selected in the study, including 31 male cases and 29 female cases. Incorporated patients were divided into the high-stress group and the low-stress group ( 30 cases in each group) according to whether the patient accepted a invasive examination or not. Then, within each group,pa-tients were further randomly sub-divided into the acute expectation group and the chronic expectation group ( 15 cases in each group) in the form of a lottery. Detection risk disclosure was conducted at 2 hours and at 24 hours before the examination. Visual analogue scale ( VAS) and stop-signal task were used to detect the level of psychological fear and the inhibition ability of individual responses to stress of each group following informing of the detection risk, and the comparative analysis was conducted afterwards. Results ( 1) The score of psychological fear in the high-stress group was significantly increased when compared to the low-stress group ((3.90±2.71) vs (0.80±1.24)),showing statistical difference (F(1,58)=30.16, P<0.01);addi-tionally,there was no statistical difference in the score of psychological fear in subjects between the acute and chronic expectation group ((2.60±2.90) vs (2.10±2.41);F(1,58)=0.785, P>0.05);meanwhile,no statisti-cal difference of the interaction between stress intensity and the expected duration of stress on the level of psychological fear (F(1,58)=0.031, P>0.05). (2) As for stop-signal task, the signal execution error rate of the high-stress group was significantly increased than that in the low-stress group ((9.40±5.80)%vs (8.30± 12.60)%),and the statistical difference was significant (P<0.01).Signal execution responses duration was obviously prolonged in the acute expectation group than that in the chronic expectation group ((677.25±201. 26)ms vs (588.24±127.10)ms),and the difference was statistically significant (P<0.05); meanwhile,stop-signal error rate at 400 ms was significantly decreased ((57.00±26.00)% vs (70.00±23.80)%),and the difference was statistically significant (P<0.05). There was no statistical significance in the interaction be-tween stress intensity and the expected duration of stress (P>0.05) . Conclusion There is no interaction be-tween the effect of the stress intensity and the expected duration of stress on the inhibition ability of individu-al responses to stress. The stress intensity is more important than the expected duration of stress to exert more important influence in the inhibition ability of individual responses to stress.

Objective To prospectively investigate the characteristics and correlative influential factors of pulmonary hypertension (PHT) in patients on long-term maintenance hemodialysis (MHD). Methods Pulmonary artery systolic pressure (PASP) was assessed by echocardiography according to the guideline from the American Society of Echocardiography in 2010 and PASP more than 35 mm Hg was diagnosed as PHT.Echocardiography and pulse wall velocity (baPWV) was performed in the next day after hemodialysis.Arteriovenous fistula (AVF) flow was evaluated by the ultrasound dilution method.Hemodialysis-related informations and laboratorial parameters were detected in the same period. Results One hundred and eleven MHD patients [male 45,female 66,mean age (57.32±12.49) years old] in our hemodialysis center were included in the study. All of the patients received MHD treatment for more than 6 months with AVF as the vascular access. The patients with any possible diseases causing PHT were excluded. The mean MHD period was (70.51±44.98) months.Twenty-eight patients (25.32%) were diagnosed as PHT with mean PASP (45.68±10.83) mm Hg.Left ventricular diastolic dysfunction was severer in patients with PHT than that in patients without PHT.The prevalence of moderate to severe diastolic dysfunction was statistically higher in PHT group compared to non-PHT group (53.60% vs 6.02%,P<0.01).Ejection fraction (EF),fractional shortening of left ventricular diameter in PHT group were also significantly lower than those in non-PHT patients (62.06%±14.90% vs 69.72%±8.60%,36.46%±10.04% vs 40.20%±7.86%,P<0.01).The patients with EF less than 50% were 21.43%and 3.61% in PHT and non-PHT group respectively. However, there were no significant differences in age, sex,MHD periods, body mass index (BMI),interval dialysis weight growth, blood pressure before dialysis,hemoglobin,albumin,pre-albumin,serum calcium and phosphorus,iPTH,nPCR,Kt/V,baPWV and AVF flow between the two groups. Conclusions PHT is a common complication of patients on long-term MHD.There is close relationship between PHT and left ventricular insufficiency.PHT is not significantly relevant to mineral metabolic disturbance,AVF flow, hemoglobin, dialysis adequacy and nutrition status.
