1.Multi-Teaching in Traditional Rehabilitation Approaches in Higher Vocational Education

Objective To explore the effects of multi-teaching mode on traditional rehabilitation approaches in higher vocational education.Methods The students from 2007 rehabilitation class as the control group (n=35) were taught with routine mode and the students from 2008 rehabilitation class as experimental group (n=60) were taught with multi-teaching mode. The result of the examination and the questionnaire was compared after one semester. Results The experimental group got higher scores in examination of theory and practice than the control group (P<0.05). The result of questionnaire showed that the experimental group acquired more increase in some aspects such as the interest of study and self-study ability than the control group. Conclusion The multi-teaching mode may be easily accepted by students and can help them learn more knowledge effectively than before.
2.Study on the Effects of Disassembled Formulation of Xiaosuanzao Decoction on Spontaneous Motion and Sleep Time in Mice

OBJECTIVE:To investigate the effects of Xiaosuanzao decoction on spontaneous motion and sleep time in mice, and to study its disassembled formulation. METHODS:Seven ingredients of Xiaosuanzao decoction were selected as the influential factors, including Ziziphus jujuba, Poria cocos, Ligusticum chuanxiong, Anemarrhena asphodeloides, Glycyrrhiza uralensis, Zingiber officinale and Cortex cinnamoni. Two levels of with or without were designed for each factor,and the experiment was ar-ranged according to L8(27) orthogonal test table. The effects of Xiaosuanzao decoction and each ingredient on spontaneous motion within 2 min and sleep time induced by superthreshold dosage of pentobarbital sodium were compared by visual analysis method and analysis of variance. RESULTS:Z. jujube and A. asphodeloides were the main active ingredients of Xiaosuanzao decoction on spontaneous motion;Z. jujube was the main active ingredient of Xiaosuanzao decoction on sleep time induced by superthreshold dosage of pentobarbital sodium. CONCLUSIONS:Xiaosuanzao decoction can inhibit spontaneous motion and prolong sleep time in-duced by superthreshold dosage of pentobarbital sodium,and Z. jujube is the main ingredient of Xiaosuanzao decoction.
3.Differential expression of miR-143 in colon cancer tissue and its effect on apoptosis and migration of colon cancer cell

Objective · To study the expression of microRNA-143 (miR-143) in human colon cancer and adjacent normal tissues, and to explore its effect on the function of colon cancer cells and downstream target genes. Methods · Lentiviral expression vector of miR-143 was designed and used to stably transfect colon cancer cell lines HCT116 and RKO. Real-time PCR was used to detect the expression of miR-143 in colon cancer, adjacent normal tissues, HCT116 cells and RKO cells infected with lentivirus. The apoptosis ratio of HCT116 cells and RKO cells was detected by Annexin V-APC single staining and flow cytometry. The migration ability of HCT116 cells and RKO cells was detected by Transwell method. The expression of FOSL2 protein was detected by Western blotting assay. Results · The expression of miR-143 in colon cancer tissue was significantly lower than that in adjacent mucosal tissue (0.339±0.454 vs 1.003±0.003) (U=16.000, Z=-4.231, P=0.000). By up-regulation of miR-143, apoptosis rates were significantly higher in HCT116 and RKO cells than those in the negative controls (P=0.000). The count of migrating HCT116 cells in up-regulated group was significantly lower than that in the negative control group (P=0.000). The count of migrating RKO cells in down-regulated group was significantly higher than that in the negative control (P=0.003). By down-regulation of miR-143, the expression of FOSL2 protein was increased. Conclusion · The expression of miR-143 was significantly decreased in colon cancer tissues. Up-regulation of miR-143 can promote the apoptosis rate in both cell lines, and inhibit the migration ability of HCT116 cells. Down-regulation of miR-143 could promote the migration ability of RKO cells. By inhibiting miR-143, the expression of FOSL2 protein in colonic cancer cells would be increased.

6.Clinical Research of Electric-impulse Stimulation to Jiaji Points in the Rehabilitation of Spinal Injury

Objective To observe the effect of electric-impulse stimulation to Jiaji points (EX-B2) on spinal cord injury (SCI). Methods Totally 126 eligible SCI patients were randomized into a treatment group and a control group, 63 cases in each group. The control group was intervened by conventional rehabilitation training plus acupuncture, while based on which the treatment group additionally received electric-impulse stimulation to Jiaji points (EX-B2). The spasticity, function, and activities of daily living (ADL) were evaluated 90 d later. Results The spasticity score was significantly changed after intervention in the treatment group(P<0.01). Tendon reflex and spasticity score were significantly changed in the control group (P<0.01). After intervention, there was a significant difference between the two groups in comparing spasticity score (P<0.01). The changes of motor function and sensory function scores in the treatment group were significantly different from that in the control group (P<0.01, P<0.05). The Barthel Index (BI) scores were significantly changed in both groups after intervention (P<0.05). There was a significant difference in comparing the BI score between the two groups after intervention (P<0.05). Conclusion Electric-impulse stimulation to Jiaji (EX-B2) plus conventional rehabilitation training can significantly improve spasticity, and the motor and sensory functions, and promote the recovery of activities of daily living.
7.Genetic Toxicity of Magnesium Sulfate to Root Tip Cells of Vicia faba

Objective To study the genetic toxicity of magnesium sulfate to the root tip cells of Vicia faba. Methods The root tip cells of Vicia faba with 1.5-2 cm root tip were taken as the subjects and were exposed to magnesium sulfate of different concentrations(0.50%-2.00%) for 6 h. In the other test,the cells were treated with 1.5% magnesium sulfate for 2,4,6,8 and 10 hours respectively. The distilled water and potassium dichromate (0.05%) were used as the negative and positive control respectively. The micronucleus and the chromosomal aberration were calculated after 24 hours of culture. Results Compared with the negative control group,0.1%,0.15%,0.175% and 0.2% of magnesium sulfate increased the micronucleus and 0.05%,0.1%,0.15%,0.175%,0.2% of magnesium sulfate increased the chromosomal aberration in Vicia faba root tip cells. 9.The effect of diabetic education on the patients using insulin treatment

Objective To study the effect of diabetic education on the patients using insulin treatment.Methods 41 cases of diabetes were trained in three different ways:group education,individual education and self education in the hospital for three to four weeks by a team of nurses.Results The results showed that the total average test score was increased by 4.44 after education.The average test scores was 9.37±0.45 after education and higher than before education (4.93±0.59) ( P <0.001).12.4% of the patients got full scores after education,34.1% of the patients got the excellent scores (≥8) before education.82.9% of patients got excellent scores after education.41.5% of the patients got low scores (<3) before education,and it was only 3.1% after education.For diabetic knowledge accumulating,the effects were significant ( P <0.001).The self care skill and knowledge such as self monitor of blood glucose and urine glucose,foot care were improved ( P <0.001).By education,diet control,exercise,oral hypoglycemic durgs and insulin treatment,FBS and PBS of patients were efficiently controled ( P <0.001).Conclusion Education is an important way for the patients to learn diabetic knowledge,improve the ability of self management
10.Construction of chimeric CblN/Grb2 and its expression in prokaryon

Objective To construct the pGEX-4T-2-CblN/Grb2, express and purify the corresponding chimeric protein; To investigate whether the chimeric CblN/Grb2 possesses ubiquitin ligase activity. Methods Total RNA of SK-BR-3 cells were isolated and reversely transcribed into cDNA, which were used as templates to amplify Grb2 SH2 by PCR. The gene fragment encoding the N-terminal part of Cbl protein (named as CblN) was amplified by PCR using pEFHACbl plasmid encoding human Cbl as templates. BamH I and EcoR V restriction enzyme digestion sites were introduced into both flanks of SH2 by overlapping extension PCR and the modified gene were cloned into pcDNA3.1(+). The pcDNA3.1(+)-CblN/Grb2 was obtained by replacing SH2 of CblN with Grb2SH2 and then used as templates to amplify CblN/Grb2 by PCR. The pGEX-4T-2-CblN/Grb2 was constructed by subcloning CblN/Grb2 into the prokaryotic expressing vector pGEX-4T-2. The GST-CblN/Grb2 fusion protein was expressed in E. coli of DH5? under IPTG induction and further purified with Glutathione Sepharose 4B. In vitro ubiquitination assay was performed to investigate whether the GST-CblN/Grb2 fusion protein is able to mediate auto-ubiquitinating reaction, namely whether it possesses ubiquitin ligase activity. Results The fusion expressing vector of pGEX-4T-2-CblN/Grb2 was successfully constructed; The GST-CblN/Grb2 fusion protein was correctly expressed and purified; In vitro ubiquitination assay indicated that GST-CblN/Grb2 fusion protein is able to mediate auto-ubiquitinating reaction and therefore possesses ubiquitin ligase activity. Conclusion Expression, purification of GST-CblN/Grb2 and identification of its′ activity have laid the foundation for further study of chimeric ubiquitin ligase′ effects on the growth of HER2 positive tumor cells.
