Objective To investigate the difference of gene expression in two human gastric adenocarcinoma cell lines, 5-fluorouracil resistant cell line SGC7901/R and its parental cell line SGC7901, and to screen related genes of JAK/STAT signaling pathway by gene chip. Methods The mRNAs of two cell lines were extracted and purified. The two cDNA probes were made from these two mRNAs which were labelled by (biotin-)16-dUTP, hybridized with human JAK/STAT signaling pathway gene array and scanned for intensity, respectively. The acquired images were analyzed by software. Different expression genes were then screened out. The mRNA expressions of Stat3, NF-?B1 and bcl-x were analyzed by semi-quantitative RT-PCR, (respectively.) Results A parallel comparison between the gene profiles of JAK/STAT signalling pathway in two cell lines showed that a total of 70 genes were screened out whose expressive level was more than 2 folds in 5-flurouracil resistant cell line SGC7901/R and its parental cell line SGC7901. Among these genes, 40 were upregulated and the other 30 were down-regulated. The results of RT-PCR were consistent with those of microarray scanning. Conclusions The knowledge of gene expression profile of JAK/STAT signaling pathway, which was changed in 5-fluorouracil resistant cell line of human gastric adenocarcinoma, has proven to be useful for illustrating the multi-drug resistance mechanisms of gastric cancer.
3.Examination and analysis of recurrent spontaneous abortion couples karyotype

Objective To investigate the correlation between recurrent spontaneous abortion and karyotype correlation,and provide reference for clinical diagnosis and treatment of spontaneous abortion.Methods 51 couples peripheral blood lymphocyte were collected and cultured from March 2010 to March 2013 in the region.Karyotype analysis were performed to observe the distribution of the rate of chromosomal abnormalities situation.Meanwhile the correlation between the number of abortions and chromosomes were compared.Results Among the 102 subjects,32 cases(31.37%)of them were found chromosomal abnormalities in RSA.The ratio of polymorphic mutation accounted for 56.25 % of the chromosome abnormality.accounting for 17.65 % the total.Which was significantly higher than other chromosomal abnormalities(P<0.05).Among the RSA and chromosomal karyotype cases,females accounted for 68.75 %,which was significantly higher than males (31.25 %)(P<0.05).46XY(Y≥18)has the highest incidence in chromosome polymorphism,accounting for 33.33 %,while abortions accounting for 30.77 %.RSA number and incidence of chromosomal abnormalities to miscarriages three times the highest rate of abnormal,accounting for 43.48%.Conclusion Chromosomal abnormalities is an important factor in recurrent spontaneous abortion.Recurrent abortion is not only related to the structure of chromosomes,but also has a certain relationship with the variation of chromosome polymorphism.
4.The primary prevention effect of losartan potassium on patients with hypertension and stroke

ObjectiveTo study the effect of angiotensin Ⅱ type 1 receptor antagonist (ARB) losartan on reducing the incidence of stroke in patients which suffer from hypertension and atrial fibrillation(AF).MethodsProspective randomized analysis was used to divide one hundred and eighty hypertemion patients with atrial fibrillation into two groups.ARB treatment group was treated with losartan (n =90) and beta-blockers treatment group was treated with metoprolol ( n =90),all patients were treated for three years and followed up.Blood pressure,pulse pressure,incidence of stroke and myocardial infarction and mortality of cardiovascular events were evaluated.ResultsAfter antihypertensive treatment,blood pressure was reduced in two groups,the pulse pressure in losartan group was reduced obviously( all P <0.01 ).The incidence of stroke and myocardial infarction and mortality of cardiovascular events in losartan group were 22.2%,10.0% and 13.3%,respectively,lower than that in metoprolol group 70.0%,40.0% and 44.4% ( all P <0.01 ).ConclusionLosartan reduced the incidence of stroke in the hypertension patients with AF.

Pharmacological studies have shown that Bruceae Fructus has unique antitumous effects, which has broad market prospects. In recent years, the demand of Bruceae Fructus has increased year by year. Production and marketing thrive, and prices continue to rise. Therefore, it has become one of the Chinese medicinal herbs with antitumour effects that obtained people’s attention. This author summarized the dynamic condition of production and marketing of Bruceae Fructus during the last 10 years, and analyzed the major reasons for price changes and forecast development tendency so as to provide references for the reasonable layout of production and marketing of Bruceae Fructus.

Objective To investigate the drug-resistance of Staphylococcus aureus induced by erythromycin in vitro and its changes in growth and susceptibility of antibiotics.Methods Erythromycin in vitro induction was conducted with the S.au-reus reference strain ATCC25923 on the serial of erythromycin agar plates.The growth of S.aureus,and the susceptibility a-gainst other antibiotics was compared after induced to the parent strain.Results Resistance to erythromycin was successful-ly,and themaximum MIC was over 256 mg/L.The Erythromycin-resistant S.aureus grew much slower than the susceptible parent,and the strains didn’t have cross-resistance to other antibiotics.Conclusion S.aureus could be induced resistance in vitro by erythromycin,and this resistance inherited stably.Some phenotype and biochemical characteristic features of the strain were changed after induced.
7.Effects of Prophylactic Antibiotics on Infections after Coronary Stent Implantation

Objective To analyze whether routine prophyrlactic antibiotic administration is necessary for the patients undergoing coronary stent implantation. Methods The clinical data of 156 patients from January 2010 to December 2010 (prophylactic antibiotic therapy),and 466 patients from January 2014 to December 2014(no-prophylactic antibiotic therapy), who underwent coronary stent implantation, were retrospectively analyzed. The prophylactic antibiotics and the infection rates in two groups were compared. Results The rate of infections related to coronary stent implantation in no-prophylactic antibiotic therapy group and prophylactic antibiotic therapy group, such as surgical site infection (0.2% vs 1.3%,P>0.05) and catheter-related infection(0.6% vs 1.9%,P>0.05), was not significant different(P>0.05). Similarly, the unrelated to coronary stent implantation was not significant different, too ( P > 0. 05). Conclusion Routine prophylactic antibiotic administration is unnecessary for the patients undergoing coronary stent implantation.
8.Investigation of working situation of community nurses and care needs of the residents in a prefecture-level city

Objective To survey the working situation of community nurses and care needs of the residents in a prefecture-level city.Method One hundred and thirty-five community nurses and 338 residents were selected and investigated by self-designed questionnaire.Results Community nurses still focused on the area of nursing and treatment rather than prevention,rehabilitation, health care,health education and family planning,which need to be paid more attention to.The needs of residents on community nursing service shift from disease treatment to disease prevention and health promotion.The current community nursing service cannot satisfy residents’care needs.Conclusion Community nurses should provide demand-oriented community care to the residents, strengthen the concept of disease prevention and health promotion,and set up community health centers to be a blend of prevention, health care,rehabilitation,health education and family planning as soon as possible.
9.The roles of abnormal expressions of Caspase-8 and protein kinase C-β of cardiomyocytes in the development of the apoptosis of cardiomyocyte in diabetic rat

Objective To study the roles of abnormal expressions of Caspase-8 and protein kinase C(PKC)-β of cardiomyocytes in the development of the apoptosis of cardiomyocyte in diabetic rat. Methods Rats were divided into 4 groups:(1)normal control (NC, n=37),(2)rats given STZ injection and normal diet(STZ,n= 42), (3) rats fed with high fat and high sngar ( HFS, n= 37), (4)rats given STZ injection and high fat and high sugar diet (type 2 DM, n=64). Plasma glucose, insulin and lipids were detected. At the end of experiment, the animals were sacrificed, and their hearts were examined. Pathological changes were observed and the expressions of Caspase-8, PKC-β mRNA were determined by real time-PCR method; apoptosis was analyzed by flow cytometry. Results (1)The body weight was higher in HFS group than in other three groups, and progressively decreased in type 2 diabetes group. The glucose level was highest in diabetic group, and was similar between groups of HFS and NC. (2)The apoptosis showed tendency to ascend during course of disease in diabetes model group. (3)The expressions of Caspase-8 and PKC-β mRNA were significantly enhanced in diabetes model group than in normal control group, and had a tendency to ascend during the course of disease.(4)The myocardial cells of the diabetic rats were rarified and swelling, fibrosis was observed. (5)At the 16th week, the level of plasma glucose was correlated positively with the expressions of Caspase-8 and PKC-β mRNA. Conclusions The enhancement of expressions of Caspase-8 amd PKC-β may play iportat rols in the pathogenesis of diabetic cardiomyopathy,in which apoptosis of the cardiomyocytes increased.
10.Study on cardiomyopathy complicated with type 2 diabetes mellitus in rats

Objective To study the development of cardiomyopathy complicated with type 2 diabetes mellitus in the rats. Methods The 120 health male Sprague-Dawley rats, weighing 180-220 g, were divided into 4 groups: (1)STZ-modeled diabetes, fed with high-carbohydrate plus high-fat diet (n=40);(2)fed with regular diet (n=30);(3)and (4)SD rats with citrate buffer instead of STZ injection, fed with high-carbohydrate and high fat diet (n= 25);and fed with regular diet (n= 25); At the 4th, 8 th, 12 th and 16 th week after the intra-peritoneal injection of STZ solution or citrate buffer solution, rats from each group were scarified and examined. Results There were no significant differences in body mass and blood glucose among those groups after one week of feeding (P>0. 05).After 4 weeks of feeding before injection, the body mass, fasting insulin (FINS) and insulin sensitive index (ISI) were obviously increased in diabetes group and high-carbohydrate plus high-fat control group as compared with STZ control group and normal control group (P< 0. 05). There were no significant differences between diabetes group and high-carbohydrate plus high-fat control group,between STZ control group and normal control group (P>0. 05). After injection, the blood glucose,body mass, ventricular mass, TG and TC were higher in diabetes group and high-carbohydrate plus high-lipid control group than in STZ control group and normal control group (P<0.05). The above parameters were much higher in diabetes group than in high-carbohydrate plus high-lipid control group, but there was no difference between STZ control group and normal control group (P<0. 05).Pathological examination showed that the weight of the heart was significantly increased, the myocardial cells were hypertrophied accompanying degenerative changes and apoptosis, the interstitial collagen fibers were hyperplasia in the diabetic rats. The ultrastructures also presented severe damage.These changes indicated that cardiomyopathy was induced in the diabetes rats. Although similar changes were found in the rats fed with high-carbohydrate plus high-fat diet, they were much less significant than those in the diabetic rats. Conclusions Cardiomyopathy developes frequently in the rats with type 2 diabetes mellitus induced by feeding high-carbohydrate plus high-lipid diet and single intra-peritonial injection of 30 mg/kg STZ solution.
