1.Effect of Liraglutide on Toll-like Receptor 4 and Na+/K+-ATPase Activity in Proximal Tubular Epithelial Cells of Rats Induced by High Glucose

Objective To explore the protective effect of liraglutide ( Li) on the proximal tubular cells ( PTECs) of rats induced by model control. Methods The PTECs of rats were obtained by primary culture and divided into normal control group ( NC) , model control group ( HG) , HG+low-dose Li group, HG+medium-dose Li group, and HG+high-dose Li group ( n-6 in each group) . After 72 h of incubation, the activity of Na+/K+-ATPase in PTECs was measured by the liquid scintillation counter. Protein expression of Toll-like receptor 4 ( TLR4 ) was measured by Western blotting. Interleukin-6 ( IL-6 ) , tumor necrosis factor-α (TNF-α) and plasminogen activator inhibitor-1 (PAI-1) in culture supernatants were measured by the enzyme linked immunosorbent assay (ELISA). Results Compared with NC, activity of Na+/K+-ATPase [(2 737. 88±317. 22)μmol·g-1 ·h-1 ] and the protein expression of TLR4 in PTECs, IL-6, TNF-α and PAI-1 in culture supernatants were significantly increased in HG (P<0. 05, or P<0. 01). Compared with HG, activity of Na+/ K+-ATPase and the protein expression of TLR4 in cells, IL-6, TNF-αand PAI-1 in culture supernatants reduced significantly in HG+medium-or high dose Li groups (P<0. 05, or P<0. 01). Conclusion Liraglutide can inhibit the expression of inflammatory cytokines and stabilize the Na+/ K+-ATPase’ s activity in PTECs in high glucose environment. Therefore, it may play a role in kidney protection.
2.A comparative study of the effects of group psychological quality training between field soldiers and military students cohorts

Objective To explore the effects of group psychological training on mental health of military students and field soldiers.Methods A total of 60 students and 48 soldiers received group psychological quality training and studied a textbook Mental Quality Training for armymen for 3 months.Mental Quality Questionnaire for Amymen (MQQA),Symptom CheckList 90 (SCL-90),Self-rating Anxiety Scale (SAS),Self-rating Depression Scale (SDS) and State-Trait Anxiety Inventory(STAI) were employed to evaluate mental health and psychological quality of subject before and after the training.All the data were analyzed by independent-samples t test.Results (1) The difference in MQQA score of field soldiers before and after training was significantly larger than that of military students in loyalty and general score,and lower in willpower((-16.58 ± 7.75) vs.(-1.75 ± 8.68),(-27.74 ± 28.74) vs.(-12.57 ± 30.96),P < 0.05).(2) The SCL-90 difference of field soldiers between before and after training was significantly larger than that of military students in hostility and phobic anxiety((0.26 ±0.47) vs.(0.07 ± 0.24),(0.13 ± 0.40) vs.(0.02 ± 0.13),P < 0.05).(3) The difference in emotion score of field soldiers between before and after training was significantly larger than that of military students in SAS,SDS,SAI,TAI and STAI (P < 0.01).Conclusion The effects of group psychological quality training on field soldier group are better than that of military students,which is helpful to improve mental quality and mental health,as well as to relax anxiety and depression of soldiers.
3.Effect of adaptive group psychological training on mental quality and emotion of soldiers

ObjectiveTo study the effect of adaptive group psychological training on mental quality and emotion of soldiers.MethodsA total of 48 soldiers ( experiment group) received group psychological training and read a textbook,Mental Quality Training for soldiers,for 3 months,and the other 45 soldiers served as control group.Armyman mental quality inventory (AMQI),self-rating depression scale (SDS) and state-trait anxiety inventory(STAI) were employed to evaluate the mental quality and emotion of soldiers before and after training.All data were analyzed by paired-sample t test.Results ①As compared with pre-training,the AMQI scores of training group increased significantly in bravery ( 71.50 ± 9.59 ),self-confidence ( 68.24 ± 8.36 ),aptitude ( 74.54 ± 10.81 ),loyalty(75.29 ± 8.08 ) and general score ( 359.07 ± 20.30 ) (P < 0.05,P < 0.01 ) ; and no significant difference were found in those of control group(P > 0.05 ).②When compared with pre-training,the scores of SDS (39.00 ±8.38),SAI(40.14 ±9.92),TAI(41.16 ±7.47) and STAI(81.30 ± 12.52) of training group decreased significantly (P < 0.05,P< 0.01 ) ; and no significant difference was found in those of control group(P >0.05 ).ConclusionGroup psychological training can improve mental quality of soldiers,which is helpful to relax anxiety and depression of soldiers.
4.Design, synthesis and biological evaluation of novel 1,3 dioxolo 4,5-fisoindolone derivatives.

A series of [1,3]dioxolo[4,5-f]isoindolone derivatives were designed, synthesized and evaluated as inhibitors of acetylcholinesterases (AChE). Furthermore, their effects on memory impairment of mice induced by scopolamine were investigated with step-through test. The results suggested that most of the target compounds exhibited potential inhibition on AChE with IC50 values at micromolar range. Compounds I1 (IC50 value of 0.086 μmol · L(-1)) and I2 (IC50 value of 0.080 μmol · L(-1)) showed the strongest AChE inhibitory activity, which are equipotent to donepezil (IC50 value of 0.094 μmol · L(-1)). Moreover, compounds I1-I4 could improve the memory impairment induced by scopolamine in mice.
5.An investigation on freshmen's psychological trouble and needs for mental health services in in military medical university

ObjectiveTo investigate psychological trouble of freshmen and their needs for mental health services in military medical university. MethodsAn half-open questionnaire was employed to evaluate 854 freshmen by group-style in military medical university.Results①Freshmen were most concemed about their training stress,soldier role adaptation and emotional trouble,etc.As compared with the male,the rate of emotional trouble and interpersonal conflict of female were increased obviously ( P<0.05 ).②The most important factor resulting in psychological problems was poor self-control,learning and training stress,military management,and so on.③The most attracting style of mental health services was communication with psychological personnel,psychological manual and movie,psychological lectures,and so on.As compared with the male,the rate of female were increased in individual counseling obviously ( P<0.01 ).④Freshmen most focused on education of general psychological knowledge,emotion and stress management,soldier role adaptation,and so on.As compared with the male,the rate of female were increased in emotion and stress management,decreased in service role adaptation obviously ( P<0.05 ).⑤Freshmen were most interested in emotion management,self-awareness,soldier role adaptation,and so on.As compared with the male,the rate of female were increased in emotion management training and profession programming,decreased in soldier role adaptation obviously ( P<0.05 ).Conclusion Freshmen's psychological problems and needs for mental health services showed obvious gender difference.Military mental health services should be focused on development objectives,specification of ser-vices ,andimprovement of the service availability in military medical university.
6.Study on Nasal Mucosa Absorption of Paeonol of Xingbi Microemulsion-based Thermo- sensitive Gel in Rats

Objective To study the nasal mucosa absorption of paeonol ofXingbi microemulsion-based thermo-sensitive gel in rats; To evaluate the nasal mucosa absorption of medicine and the rationality of preparation form.Methods According to the nasal administration method, the blood medicine concentration of paeonol was determined at different time points.Results The nasal administration bioavailability of paeonol in Xingbi microemulsion-based thermo-sensitive gel was 78.68%, the plasma concentration reached the maximum (0.3404 μg/mL) after 3 min of administration, and the average residence time was 9.23 h. After reaching Cmax, The plasma concentration changed similar to intravenous administration of paeonol. The concentration data of paeonol in plasma was consistent with a two-compartment model, with a weight of 1.Conclusion Xingbi microemulsion-based thermo- sensitive gel has a higher bioavailability, and the preparation form was reasonable.
7.Application of OTSU-based self-attenuation correction PET reconstruction technology in 18F-AV45 imaging

Objective:To explore the application of OTSU-based self-attenuation correction PET (sacPET) reconstruction technology in 18F-florbetapir (AV45) imaging. Methods:From November 2018 to December 2019, 7 confirmed Alzheimer′s disease (AD) patients (4 males, 3 females, age (69.6±4.5)years) and 3 healthy controls (HC; 1 male, 2 females, age (68.0±4.6) years) were recruited prospectively for 18F-AV45 PET imaging in the First Affiliated Hospital of Sun Yat-Sen University. Original data collected by PET acquisition was processed with sacPET reconstruction and then compared with standard PET images by visual analysis and semi-quantitative analysis. Fisher exact test, Kappa test and Pearson correlation analysis were used to analyze data. Results:In HC group and AD group, the radioactive distribution showed by sacPET images and that by standard PET images were similar, and the contrast of gray-white matter in sacPET images was weaker than that in standard PET images. Moreover, the positive uptake area of the cortex in the AD group was smaller than that in standard PET images. Visual analysis showed 19 positive regions in sacPET images and 22 in standard PET images, with no statistical difference of positive rates of the sub-regions in the cortex between the two PET images (all P>0.05), and the overall consistency of 88.00% (44/50; Kappa=0.75 (95% CI: 0.57-0.94), P<0.05). Semi-quantitative analysis showed that the standardized uptake value ratio (SUVR) of frontal lobe and cingulate gyrus measured by sacPET was lower than that measured by standard PET (0.93±0.06 vs 0.96±0.06 and 0.99±0.04 vs 1.01±0.04; t values: 5.30 and 5.10, both P<0.01), while SUVR of parietal lobe, temporal lobe and occipital lobe measured by sacPET was higher than that measured by standard PET (0.78±0.08 vs 0.68±0.07, 0.97±0.07 vs 0.91±0.08 and 0.94±0.11 vs 0.71±0.12; t values: 6.27, 7.36 and 16.90, all P<0.01). The overall SUVR of sacPET images was significantly correlated with the standard PET images ( r=0.75, P<0.001). Conclusion:For 18F-AV45 imaging, sacPET reconstruction technology can obtain reliable and effective PET images without CT data, but its accuracy and precision still need to be improved.
8.Combination therapy of FK228 with rapamycin synergistically promotes human breast cancer cell apoptosis by DNA damage and cell cycle arrest

[ ABSTRACT] AIM:To investigate the depressant effect of FK228 combined with rapamycin on the human breast cancer cell line MCF-7 and MDA-MB-435.METHODS:FK228, a new histone deacetylase inhibitor, and rapamycin, the specific inhibitor of the mammalian target of rapamycin ( mTOR) protein, were used in the study.MCF-7 cells and MDA-MB-435 cells were exposed to different concentrations of FK228 and rapamycin.The inhibitory rate of cell growth was de-termined by SRB assay.Combination index ( CI) was used to evaluate the interaction between FK228 and rapamycin.The expression of the apoptotic proteins, cycle proteins and nucleic acid proteins were detected by Western blotting.The cell cycle was analyzed by flow cytometry.RESULTS: Both FK228 and rapamycin showed growth inhibitory effects on the breast cancer cell lines in a time-and dose-dependent manner.CI of the 2 drugs was less than 1 when the inhibitory rate of the cell growth was 50%effective dose (ED50)~ED70, indicating a synergistic effect.The combination therapy of FK228 with rapamycin increased the apoptotic proteins, and induced the down-regulation of phosphorylated Akt and over-expres-sion of caspase-3 compared with a single use of the drugs.The combination therapy of FK228 with rapamycin reduced the cycle proteins, and the cell cycle was arrested in G2/M.The levels of phosphorylated H2AX and acetylated H3 were ob-viously increased after combination therapy.CONCLUSION:The combination therapy of FK228 with rapamycin inhibits the cell proliferation and increases apoptosis with a synergistic effect, which may become a new trend for treating endometri-al cancer.
9.Thoughts on breaking away from the " paper-centric" orientation and establishing a new evaluation model of scientific and technological innovation in academic universities

Objective:To analyze the disadvantages of " paper-centric" in science and technology evaluation, explore how to establish a new evaluation model of scientific and technological innovation in academic universities in China on the premise of breaking the " paper-centric" orientation.Methods:Analyze problems and disadvantages of the " paper-centric" orientation in the evaluation of science and technology at academic universities in China, take account into the in-depth interpretation of key policies of breaking away from " paper-centric" in recent years, and finally make proposal for the establishment of evaluation system and mode of scientific and technological innovation in the future.Results:There are many pitfalls in the " paper-centric" orientation in the evaluation of science and technology in academic universities in China, thus, it is urgent to establish a new evaluation mode of scientific and technological innovation.Conclusions:Based on the current domestic and international context, academic universities in China should deploy the strategy of scientific and technological innovation in advance, break the " paper-centric" orientation, and establish a new evaluation system and mode of science and technology that proactively match the national strategy and the development requirement.
10.Application of self fixing mesh with " Y-shaped" placement in laparoscopic transabdominal preperitoneal repair of inguinal hernia of female inguinal hernia

Objective:To investigate the effect of self fixing mesh with " Y-shaped" placement in laparoscopic transabdominal preperitoneal repair of female inguinal hernia.Methods:From February 2019 to September 2020, a total of 40 female patients who underwent TAPP (Keyhole method) in Shaanxi Provincial People′s Hospital were researched.Random number table method was used.According to different patch placement methods, the patients were divided into the self fixing mesh with " Y-shaped" placement group (21 cases) and with normal placement group (19 cases). The operative time, placement time, length of stay, Visual Analogue Scale (VAS), postoperative complication were compared between two groups.Results:All 40 patients completed TAPP, and all patients were followed up successfully, with a median follow-up time of 12.5 months.The operation time and patch placement time were (54.9±9.0) min and (10.8±2.5) min in the Y-shaped placement method group and (62.8±11.2) min and (20.3±3.5) min in the conventional placement method group, respectively; the differences between the two groups were statistically significant ( t=2.490, P=0.017; t=.925, P<0.001). There was no statistically significant difference between the two groups in length of stay, VAS score 1 day after surgery and postoperative complications(all P>0.05). Conclusion:In laparoscopic female inguinal hernia repair (keyhole method), the " Y" shape placement method can effectively shorten the time of self fixing patch placement, thus shorten the operation time, and does not increase the risk of complications.
