2.Effect of intravenous immunoglobulins G on rat subjected to chronic unpredictable stress

Objective To investigate the effect of intravenous immunoglobulins G on rat subjected to chronic un-predictable stress. Method Rats were randomly divided into 2 groups (IVIG+stress group and stress-only group). Sucrose preference test, force swim test and neurogenesis analyze were used to analyze the effect of IVIG on rats in stress group. Results Sucrose consumption in IVIG group was significantly more than CUS group(P<0.01), while non floating time of force swim was significantly lower than CUS group(P<0.01). The difference of BrdU positive cell number in Hippocampus between two groups was significant(P<0.01). Conclusion IVIG show an antidepressant like effect, the mechanism may be involved in eliminate inflammatory cytokine of peripheral circulation.
3.Infection Rate of HBV among High School Graduates in Jingjiang City of Jiangsu Province

Objective To investigate the positive rates of HBsAg and abnormal rates of Alanine aminotransferase among high school graduates in Jingjiang city.Methods HBsAg were detected by ELISA and ALT was detected by rate assay among 20 988 high school graduates during 2004—2007 in Jingjiang city.Results The HBsAg positive students were 1754.The total positive rate of HBsAg was 8.36%.The positive rate of HBsAg of male(9.46%)was higher than that of female(6.95%).The HBsAg positive rate of rural students(9.44%)was higher than that of urban students(7.28%).HBsAg positive rates of urban students showed a decreasing trend during 2004—2007,the rates were 9.31%,8.08%,6.74% and 5.24%,respectively(P0.05).There were 155 cases(8.4%)with increased ALT among 1754 HBsAg positive students.Conclusion The total positive rate of HBsAg among high school graduates in Jingjiang city from 2004—2007 was 8.36%.The HBsAg positive rate of male students was higher than that of female.The HBsAg positive rate of rural students was higher than that of urban students.8.4% students were found ALT abnormal.We should intensify Hepatitis B vaccine in time to prevent the spreading of HBV.
4.Transport of melanocytes by chitosan membrane: an animal experiment

Objective To study the feasibility of using chitosan membrane to carry and transport melanocytes, in order to refine the technique for melanocyte transplantation with chitosan membrane. Methods Melanocytes were inoculated onto chitosan membrane and cultured for a period of time, then, electron microscopy,MTT assay and NaOH assay were carried out to estimate the adherence, growth and melanogenesis of the melanocytes. Skin wound surface was prepared in 12 nude mice, which were equally divided into 3 groups, test group inoculated with melanocytes on chitosan membrane, negative control group I treated with chitosan membrane without melanocytes, and negative control group II directly dressed immediately after the preparation of wound surface. On day 10 and 20 after the transplantation, confocal laser microscopy and immunohistochemistry were performed to observe the migration of melanocytes into the skin wound surface. Results Scanning electron microscopy and inverted microscopy showed that melanocytes were evenly distributed on and adhered well to the underlying chitosan membrane. As the growth curve of melanocytes demonstrated, chitosan membrane could support the normal growth of melanocytes, and no significant difference was observed in the synthesized melanin content between melanocytes cultured on the chitosan membrane and those in culture disks (0.087 ± 0.027 vs. 0.101 ± 0.036, t = 0.79, P > 0.05). Melanocytes were seen at the transplantation sites by confocal laser microscopy, and biopsy specimens from the transplantation sites stained positive for antimelan-A monoclonal antibody. Conclusions Melanocytes can adhere to and grow on the chitosan membrane,which can facilitate the migration of melanocytes to the transplantation sites in animals with the maintenance of biological activity of melanocytes.
5.A qualitative research on gestational diabetic patients' primary use of real-time blood glucose monitoring

Objective To explore the inner feelings of gestational diabetes at their primary use of the real-time blood glucose monitoring. Method Using the qualitative method, 10 gestational diabetic patients monitored their blood glucose using the real-time blood glucose monitoring for the first time and then were interviewed, the acquired results treated with phenomenological analysis. Result The inner feelings of the 10 patients were summarized including 5 themes:worry about the safety of the fetus, fear on alarming at monitoring, different views on four times input of finger blood glucose values, worry about inaccuracy in blood glucose monitoring data, changes in mental conditions and diets. Conclusion Health care workers should provide the gestational diabetic patients with targeted care strategies based on presence of psychological status and knowledge needs so as to improve their compliance, reduce the psychological burden of patients and control their blood glucose level.
6.Factors influence elderly patients with hip fracture sitting early away from bed after operation and countermeasures

Objective To explore the factors influencing the sitting early away from bed after operation in elderly patients having hip fractures.Method Forty-six elderly patients with hip fractures who were taking postoperative functional exercises of sitting away from bed and 32 nurses were investigated on the influential factors by a questionnaire.Results More than 52.1%of the patients believed the factors influencing their sitting early away from bed including lack of family support,worry about insufficient physical strength and fear of pains.65.6%of the nurses thought the factor was their inadequate assessment.Conclusion Such measures as enhancing family support,nutritional support,relief of pains from sitting from bed and improving nurse’s ability in rehabilitative training instruction are important for the patients to sit early away from bed after operation on hip fractures.
7.Optimization of cell seeding density in the treatment of vitiligo by transplantation of cultured autologous melanocytes

Objective To evaluate the relationship between cell seeding density and clinical efficacy of autologous cultured melanocyte transplantation in the treatment of vitiligo.Methods A total of 632 patients with vitiligo were enrolled in this study,and randomly classified into 4 groups to be treated with transplantation of autologous cultured melanocytes at 4 different seeding densities respectively,i.e.,(3.0-4.9)× 104/cm2 (n =201),(5.0-7.9) × 104/cm2 (n =303),(8.0-9.9) × 104/m2 (n =82),(10.0-12.0) × 104/cm2 (n =46).Epidermal sheets were obtained by suction blister biopsy from the normal skin of the vitiligo patients,and subjected to the isolation and culture of melanocytes.After 2 to 5 passages,the cultured autologous melanocytes were transplanted at different seeding densities to vitiligous lesions,which were abraded previously by ultra-pulsed CO2 laser,of these patients.All the patients were followed for 6-12 months.Results At 6 months after the transplantation,52.85%of these patients achieved more than 90% repigmentation,and 82.28% more than 50% repigmentation,with no differences in the cure rate and response rate between the 4 groups (both P < 0.05).The percentage of patients obtaining excellent color matching was significantly higher in the group treated with transplantation of melanocytes at a seeding density of (5.0-7.9) × 104/cm2 than in the other 3 groups at 6,12 and 24 months after treatment (all P < 0.05),and higher in all the 4 groups at 12-and 24-month points compared with the 6-month point (all P < 0.05),but no statistical difference was observed between the 12-and 24-month point in any of these groups (all P > 0.05).Conclusions The transplantation of autologous cultured pure melanocytes is effective for the treatment of stable vitiligo with the optimal cell seeding density of melanocytes being (5.0-7.9) × 104/cm2,and the color matching appears to improve with time.
8.Detection of Schistosoma japonicum cercaria DNA by loop-mediated isothermal amplification

Objective To establish a method for detecting cercaria of Schistosoma japonicum.Methods The loop-mediated isothermal amplification(LAMP)was used.The cercaria DNA of Schistosoma japonicum was extracted by using GeneReleaser.Four primers which recognized 6 distinct regions on the calcium-binding protein gene of cercaria of Schistosoma japonicum were designed and used for LAMP assay and Clonorchis sinensis was used as the negative control for evaluating the specificity and 20,10,5,1 cercariae of Schistosoma japonicum were amplified by LAMP for evaluating the sensitivity.The LAMP results were judged with the naked eyes and electrophoretic analysis.Results After LAMP reaction,the positive signal was observed with cercariae of Schistosoma japonicum.By contrast,no positive signal was observed for Clonorchis sinensis.The detection limit of LAMP assay was 1 cercaria of Schistosoma japonicum per reaction.Conclusion LAMP assay has usefulness for rapid detection of cercariae of Schistosoma japonicum.
9.Therapeutic Observation of Electroacupuncture plus Moxibustion for Infantile Cerebral Palsy

Objective To observe the clinical efficacy of electroacupuncture plus moxibustion in treating infantile cerebral palsy. Method Eighty eligible patients with infantile cerebral palsy were randomized into a treatment group of 42 cases and a control group of 38 cases. The control group was intervened by physiotherapy (PT), while the treatment group was given electroacupuncture at scalp acupoints, body acupuncture, and moxibustion along the Governor Vessel in addition to the treatment given to the control group. The two groups were treated once a day, 6 months in total. The comprehensive ability and the Gross Motor Function Measure (GMFM) were evaluated before and after the interventions in the two groups, and the clinical efficacies were compared.Result After 6-month treatment, the total effective rate was 92.9% in the treatment group versus 78.9% in the control group, and the total effective rate of the treatment group was superior to that of the control group (P<0.05). The motor function and activities of daily living were significantly improved in both groups after the interventions (P<0.05,P<0.01); after the treatment, there were significant differences in comparing the motor function and activities of daily living between the two groups (P<0.05). The GMFM scores of each domain were significantly changed after the treatment in both groups (P<0.05); after the treatment, there were significant differences in comparing the GMFM scores of each domain between the two groups (P<0.05).Conclusion Compared with being used alone, conventional PT can more significantly benefit the improvement of motor function of cerebral palsy patients when electroacupuncture and moxibustion were added.
10.Influence of evidence-based nursing on the compliance of sitting at early stage on post-operative elders with hip fracture

Objective To investigate the compliance of evidence-based nursing on post-operative elders with hip fracture sitting at early stage. Methods A total of 108 post-operative elders with hip fracture were divided into the observation group and the control group with 54 cases in each group with random digit table. The patients in the control group sit from bed based on their state of vital signs and mental status on the 2nd or 3rd postoperative day. And those in the observation group employ evidence-based nursing on the basis of the control group. The degree of anxiety, postoperative fatigue and compliance of patients were compared. Results Two weeks after the operation, the degree of anxiety scored 40.96±5.13, postoperative fatigue scored 1.89±0.65 in the observation group, which were lower than those of the control group, 56.40±4.28, 4.55±0.71, the difference was significant (t=288.41, 412.35, both P<0.01). The rate of compliance in the observation group was 94.44% (51/54), and the rate of the control group was 77.77%(42/54). The rate of compliance in the observation group was higher than that of the control group(χ2=8.57, P<0.05). Conclusions Using evidence-based nursing on post-operative elders with hip fracture sitting at early stage is able to improve the compliance of patients. It is good for reducing the postoperative complications, improving activities of daily life and mental state, promoting rehabilitation in patients as well.
