1.Analysis on the Detection of PTA Blood Current of the PIH Pregnant Women by Means of the Ultrasonic Wave

ObjectiveTo provide a new method for the predictive diagnosis of PIH through the detection of the PTA blood flowing indexes of the pregnant women. MethodTo adopt the color Doppler ultrasonic wave the conduct the contrast normal pregnant women and PIH women of post-pregnancy 30 cases respectively. Results①The peak value flow rate of blood at diastolic period (short for D), the average blood flow rate (short for MN), the ratio of the peak value blood flow rate (S/D) for PIH group are higher than those for the other two groups respectively (P<0.01); ②There is no statistic difference in the changes of the PTA artery internal diameter between the three groups. ConclusionThe changes in the PTA blood flow indexes of the PIH patients provides a new method for the diagnosis of this disease, which is of some reference value. Such indexes as S, MN and S/D of the PTA are better targets for observation.
2.Quality Control for Chanshukang Granule

Objective To study the quality control method for Chanshukang granule. Methods TLC and HPLC method were used to identify the Chinese herbal medicines. The determination was carried on HPLC, using Kromasil C18 column (200 mm?4.6 mm, 5 ?m), ethanol-2% glacial acetic acid (40∶60) as mobile phase and detected at the wavelength of 324 nm. Results The Chinese herbal medicines, Radix Angelicae Sinensis, Rhizoma Chuanxiong, Placenta Hominis and Radix Glycyrrhizae Preparata, in granules were identified by TLC. The calibration curve was linear in the range of 0.625~3.750 ?g for ferulic acid. The contents in three batches were 0.054%, 0.037%, 0.042% respectively. The average recovery was 96.2% and RSD=2.37%. Conclusion The TLC method can be used for the quality control of four herbal medicines in the granule. The HPLC method is simple, sensitive, accurate and provides a certain evidence for controlling the quality of Chanshukang Granule.
3.Determination of isopimaric acid in leaves of Platycladus orientalis and its preparations Cebai No.V Capsula and Isopimaric Acid Capsula by HPLC

Objective To establish an HPLC method for the determination of isopimaric acid in the leaves of Platycladus orientalis and its preparations Cebai No.V Capsula and Isopimaric Acid Capsula to evaluate the quality of them. Methods HPLC Method was developed. Kromasil C18(200 mm?4.6 mm, 5 ?m) column was used and the mobile phase: methyl alcohol-water-acetic acid (60∶37∶3), float rate: 1 mL/min, temperature: 25 ℃, wavelength: 231 nm, injected volume: 10 ?L. Content of isopimaric acid in samples was calculated by external standard. Results The linearity relationship of isopimaric acid was good in the range of 1.25-10.00 ?g. The equation of standard curve: Y=66 214 X+2 487.9, r=1.000. The average recovery was 95.8%, RSD=2.7% (n=5). The determination results of isopimaric acid in three batches of P. orientalis were 3.09, 3.20, and 3.56 mg/g; three batches of Cebai No.V Capsula were 10.39, 9.67, and 10.99 mg/g; three batches of Isopimaric Acid Capsula were 553.89, 541.50, and 528.83 mg/g. Conclusion The method is reliable, simple, and precise, which could be used for the quality control of P. orientalis and its capsules.
4.The Sensitivity of ERIC-PCR in the Mixed Bacteria System

Genomic DNA of the two bacterial strains including Escherichia coli DH5? and Enterobacter cloacae E 26R were amplified by PCR with specific primers of ERIC sequence Each strain showed the stable and unique DNA fingerprint when PCR products were analyzed in agarose gel electrophoresis The stability of the genomic DNA fingerprint wasn't influenced by the templet from 10ng to 100ng In the fingerprints there were characteristic bands The unique band was influenced difficultly due to the change of the environment and condition of experiment DH5? and E 26R were mixed proportionately, and examined by ERIC PCR The result showed that the DNA fingerprints of the mixture were the superposition of each pure bacterium's Analysis of the main bands showed that DH5? can be examined by ERIC PCR when its concentration gets to 0 5% of the total mixed bacteria The study provides the reference for the research of soil microbe and environmental microbe, and especially can be used for quick identification and examination of the sundry bacterial contamination in the fermentation industry
5.Identification and characterization of peptide mimics of blood group A antigen.

In order to investigate peptide mimics of carbohydrate blood group A antigen, a phage display 12-mer peptide library was screened with a monoclonal antibody against blood group A antigen, NaM87-1F6. The antibody-binding properties of the selected phage peptides were evaluated by phage ELISA and phage capture assay. The peptides were co-expressed as glutathione S-transferase (GST) fusion proteins. RBC agglutination inhibition assay was performed to assess the natural blood group A antigen-mimicking ability of the fusion proteins. The results showed that seven phage clones selected bound to NaM87-1F6 specifically, among which, 6 clones bore the same peptide sequence, EYWYCGMNRTGC and another harbored a different one QIWYERTLPFTF. The two peptides were successfully expressed at the N terminal of GST protein. Both of the fusion proteins inhibited the RBC agglutination mediated by anti-A serum in a concentration-dependent manner. These results suggested that the fusion proteins based on the selected peptides could mimic the blood group A antigen and might be used as anti-A antibody-adsorbing materials when immunoabsorption was applied in ABO incompatible transplantation.

6.Clinical manifestation and MRI feature on three patients of toxic encephalopathy after inhaling caffeine and sodium benzoate

Objective To study the clinical manifestation and MRI feature of toxic encephalopathy after inhaling caffeine and sodium benzoate. Methods Three patients with toxic encephalopathy induced by inhaling caffeine and sodium benzoate were observed clinically and with MRI. Results The 3 patients were male. They all had definite addiction histories (the time of addiction for 3 years to 7 years ). There were special language and retropulsive dystonia besides the common characteristics of toxic encephalopathy:reaction retardation and disturbance of intelligence. MRI showed the change of generalized demyelination in bilateral symmetrical cerebral hemisphere, posterior limbs of internal capsule, corpus callosum, medial lemniscus, and cerebellum hemisphere dentate nucleus. The clinical symptoms of 3 patients were significantly improved 40 days after the adrenocortical steroid and neurotrophic treatment. The mild mood disorder remained in case 1. Conclusion Clinical manifestation and MRI of toxic encephalopathy after inhaling caffeine and sodium benzoate are known, so we could diagnose the patients of the disease in time.
7.Clinical value of serum CA19-9,CA125 and CP2 in mucinous ovarian tumor:a retrospective study of 273 patients

Objective To evaluate the diagnostic and prognostic value of serum CA19-9,CA125 and CP2 in mucinous ovarian tumors.Methods In this retrospective study,the serum CA19-9,CA125 and CP2 levels of 273 hospitalized patients with ovarian tumors of either mucinous or non-mucinous type were analyzed.Results(1)CA19-9 had the biggest area under chive(AUC)in mucinous tumors followed with CA125 while CA125 and CP2 had bigger AUC in non-mucinous tumor.(2)For the diagnosis of mucinous tumors,CA19-9 and CA125 combination showed a greatly increased sensitivity compared with CA19-9 or CA125 alone(93.8%versus 75.0%and 66.7%,P<0.05)with no significant improvement of the specificity(P>0.05).For the diagnosis of non-mucinous tumors,CA125 and CP2 combination showed an increased sensitivity compared with CA125 or CP2 alone(85.0%versus 80.7%,P>0.05,85.0%versus 70.6%,P<0.05)with no significant improvement of the specificity(P>0.05).(3)Seventy percent of tumor marker-positive patients could undergo cytoreductive surgery.Compared with those who could not undergo cytoreductive surgery,they were more likely to have normal tumor marker two months after surgery (P<0.05)and longer interval to re-elevation of tumor markers(P>0.05),with lower reeurrence and death rate (P<0.05).All of the 20 tumor marker-negative patients could have eytoreduetive surgery with only 10%recurrence.(4)CA19-9 inereased mainly in recurrent mucinous tumor,while CA125 increased dominantly in recurrent non-mueinous tumor.(5)The survival rate of CA125 and CP2 positive patients was much lower than CA125 and CP2 negative patients(P<0.05),while the survival rate was similar between CA19-9 positive and CA19-9 negative patients.Conclusions CA19-9 is a sensitive index for diagnosis of mucinous ovarian tumors.Combination of CA19-9 with CA125 can improve the sensitivity of diagnosis and postoperative monitoring of mucinous ovarian tumors.Combination of CA125 with CP2 is more valuable in the diagnosis of non-mucinous ovarian tumors.
8.Origin and Phylogenetic Characteristics of Dengue Virus Isolated from the Large Outbreak of Dengue in Guangdong Province in 2014

[Objects]To isolate and identify the pathogen of the large outbreak of dengue in Guangdong province in 2014. To understand the origin and the phylogenetic characteristics of the isolates ,and provide scientific foundation for the surveillance and prevention of dengue fever.[Methods]Collected the patient serum samples over all the Guangdong province during the 2014 outbreakperiod,isolated and identified the virus from these samples. Amplified complete E gene and complete genome with certain primers and sequenced all the products. Then the Phylogenetic ,Bayesian phylogeography and mutations analysis were carried.[Results]40 DENV-1 strains were isolated and identified. 40 complete E gene sequences and 6 complete genome sequences of DENV-1 were obtained. Phylogenetic analysis with E gene sequences revealed that the 40 isolates were classified into two genotypes including 16 genotypeⅠ(Asia)and 24 genotypeⅤ(America/Africa). 14 genotypeⅠisolates were clustered closest with isolates from Guangdong province(2013)and Sigapore(2013)which share the nucletide identities of 99.6% ~ 99.9%,other two genotypeⅠisolates were clustered with strains from Malaysia (2013) and both share the nucletide identities of 99.7%;24 genotypeⅤisolates were all classified in one clade with striains from Bangladesh(2009),China(2009)and Bhutan(2013)which share nucletide identities of 99.0%-99.9%. Further analysis with six complete genome sequences showed that five isolates were clustered closest with strains isolated from Guangdong province(2013)share the nucletide identities of 99.6%-99.8% while the sixth stains closest with strains isolated from Myanmar(2002)share the nucletide identities of 98.8%. The isolates have five amino acid mutations compared with strains epidemic in Guangdong province in 2013,three mutations(S88V,E203G,T275R)are in the EⅡdomain and one mutation (S305P)is in the EⅢdomain which associated with virulence.[Conclusions]During the outbreak in Guangdong province in 2014, DENV-1 is the predominant causative serotype,and there are at least two different kinds of genotypes of DENV-1 largely epidemiced in the whole province. Evolution analysis reveals the multiple origins of the isolates which may origin from Guangdong province , Sigapore,Malaysia,Myanmar so that we should enhance the study and surveillance of autochthonous and vectors in order to understand the epidemic way of dengue in Guangdong province. The isolates have had four mutations in the domain associated with virulence which remain further study to know their biological effects.

Purpose To assess the clinical and pathological features of small gastrointestinal stromal tumours (sGIST).Methods To reevaluated the clinical,histological and immunohistochemical parameters of 21 sGISTs.The standard immunohistochemical panel antibodies were studied on the tumor sections.All data were compared with clinical sGIST.Results There were a total of 7 females and 14 males of sGISTs.The median age was 63 years old.The tumors were predominantly located in the stomach showing a spindle cell morphology and the tumor sizes ranged from 0.5 cm to 1.5 cm.9 sGISTs combined with malignant tumors,which were gastric cancer have been incidentally detected during surgery.As the lesions were small in size,with infrequent bleeding,necrosis,mucosal invasion,ulceration and less mitotic index,sGISTs reoccurred less compared with clinical sGIST.p53,Ki-67 labeling index and microvascular density (MVD) in sGIST were significantly lower than clinical sGIST (P < 0.05).Conclusion sGIST may occure with digestive tract cancer synchronously.p53,Ki-67 labeling index and MVD were lower than clinical GIST,which means better prognosis.
10.Study of the expression of the cyclin D1 in chronic myelogenous leukemia

Objective To examine the expression levels of cyclin Dl in the patients with chronic myelogenous leukemia (CML), and evaluate the pathogenesis and clinical significance of cyclin Dl in CML Methods The real-time quantitative polymerase chain reaction (RQ-PCR) was performed to detect the expression levels of cyclin Dl in the bone marrow samples of 18 patients with CML, and 16 samples of benign hemopoietic patients. The relationship between the expression levels of cyclin Dl and the progression and prognosis of patients with CML were analyzed. Results The level of cyclin Dl was higher expressed in 18 patients with CML than the control group (P <0.001). The levels of cyclin Dl was apparently higher expressed in accelerated phase /blast crisis phase than in chronic phase (P <0.05). And the RQ-PCR method showed the tendency that a significant increase was observed in the levels of cyclin Dl from 0.1980 in control group to 1.4002 in chronic phase and 5.4540 in accelerated phase /blast crisis phase. Conclusion The cyclin Dl overexpressed in CML, the roles of cyclin Dl in CML might be an oncogene expressed. The expression level is correlated with the progression and prognosis of patients with CML.
