Lentiviral vectors can transduce a wide range of different cell types.It can transduce not only dividing cells but also nondividing cells.It can be the immunotherapy tool for cancer therapy.Lentiviral yeators have several advantages,amongst which long-term expression,low toxicity,high capacity and low antivector immunity,they will play an important role and will attract more and more interest as an immunotherapeutic.Here,we discussed the clinical application and safety of the lentiviruses encoding tumor-associated antigen(TAA).
2.Safty observation in children with acute lymphoblastic leukemia by different doses of Methotrexate

Objective To investigate the toxicities of methotrexate in the treatment of the children with acute lymphoblastic leukemia Method 28 children with acute lymphoblastic leukemia were randomly divided into two groups(A and B), they were treated with methotrexate 3.0 g/m 2 (n=16) and 5.0 g/m 2(n=12). The toxicities of two doses of methotrexate were observed. Results The main toxicities of methotrexate were marrow suppression,gastrointestinal reaction and liver damage. They were mostly mild.No statistically significant difference was found between group A and group B .Conclusion The toxicities of methotrexate are common and can be tolerated.
3.Influence of glypican-3 gene on proliferation,adhesion and invasion of hepatoma carcinoma cancer cell line SK-Hep-1

Objective:To study the influence of GPC3 gene on the proliferation,adhesion and invasion of hepatoma cell line SK-Hep-1.Methods:SK-Hep-1 cells were transfected with pEGFP-N2-GPC3 using Lipofectamine2000.RT-PCR was used to examine the GPC3mRNA expression in GPC3-SK-Hep-1 cells.MTT assay was used to examine the proliferation and calculate the adhesion rate of SK-Hep-1 cells.Transwell system was used to assess the migration and invasion of the cells.Results:SK-Hep-1 cells were successfully transfected with pEGFP-N2-GPC palsmid.GPC3 mRNA was detected in SK-Hep-1 cells.Transfection with CPC3 significantly suppressed the growth of SK-Hep-1 cells(P
4.Diabetic autonomic neuropathy is a risk factor of cerebrovascular disease

Objective To study the relationship between the diabetic autonomic neuropathy (DAN) and the cerebrovascular disease. Methods The heart rate variability (HRV) was measured in 77cases of type 2 diabetes 〔49 men, 28 women, age (63.1?11.4)years〕. Forty-one cases suffered from DAN (group A), 36 cases without DAN (group B) according to the results of HRV. All the cases received the examination with transcranial Doppler ultrasonography (TCD). Results Compared with group B, the abnormality detected with TCD was higher in group A, cerebrovascular compliance was commonly decreased and cerebrovascular stenosis was also extensively observed. Conclusion DAN seems to be one of the risk fcators in inducing cerebrovascular disease.
5.Study on the relationship of change in hemodynamics of portal system and liver reserve in patients with hepatocirrhosis

Objective To investigate the relationship of change in hemodynamics of portal system and liver reserve of Chile-Pugh classification in patients with hepatocirrhosis.Methods Color Doppler ultrasound were applied to detect the inner diameter,average velocity of blood flow,quantity of blood flow of portal vein and splenic vein for 173 patients with post-hepatitis hepatocirrhosis(61 of Child grade A,53 of grade B,59 of grade C),which were sub- sequently compared with the healthy controls.Results In patients with hepatocirrhosis,the inner diameter of portal vein and splenic vein were widened,the average velocity of blood flow of portal vein and splenic vein were slowed down and the quantity of blood flow of splenic vein was increased,and the worse the damage of liver function was, the more obvious the change became.Conclusion The change in hemodynamics of portal system varies with the damage in liver function in patients with hepatocirrhosis.Measuring the hemodynamics of portal system is significant in determining the severity of the disease and the prognosis.
6.Evaluation of application effect of risk management in nursing of department of nephrology

Objective To analyze the related factors of nursing risk in department of nephrology and to explore strategies of risk management.Methods Risk management was implemented to 39 nurses in department of nephrology of our hospital.The KAP scores,the incidence of nursing risk events,nursing complaints and patients' satisfaction degree before and after the implementation of nursing risk management were compared.Results After the implementation of nursing risk management,nurses' KAP score was significantly higher; the incidence of nursing risk events and complaints significantly reduced; patients' satisfaction degree to nursing were significantly higher compared with those before the implementation.Conclusions The implementation of nursing risk management can effectively improve the quality of nursing and reduce the incidence of nursing risk events.
7.Clinical value of three-dimensional CT angiography in diagnosing blood vessel diseases

ObjectiveTo valuate the three-dimensional CT angiography(3DCTA) in diagnosing blood vessel diseases.MethodsCTA findings of 284 patients confirmed with blood vessel diseases were retrospectively analyzed.They were divided into two groups:141 cases of head and neck and 143 cases of trunk and extremities.Scan was performed with the beam collimation of 0.625 ~ 3.0mm,increment of 0.3 ~ 1.4mm,pitch of 1.0 ~ 1.5,with the gross contrast medium of 1.5 ~ 2.0ml/kg.3D imaging methods included volume rendering(VR),multi-planar reformatting (MPR),curved planar reformatting(CPR),surface shade display(SSD),maximum intensity project(MIP) and virtual endoscopy(VE).The diagnostic results of CTA were compared with those of DSA and/or surgery.ResultsIn this study,3D images of vasculature met the diagnostic criterion.The diagnostic sensitivity,specificity and accuracy of CTA was 97.28%,82.35% and 96.48%,respectively.The size of artery aneurysm shown with CTA and DSA had no significant difference( P >0.05 ),both smaller than that in surgery( P < 0.05 ).Conclusion3DCTA could be used in diagnosing the cardiovascular diseases and compensate the limitation of conventional CT.It had the characteristics of non-invasion,high accuracy and could partly substitute DSA.
8.Percutaneous transluminal angioplasty combined with autologous peripheral blood stem cells for lower extremity ischemia

Objective To observe the clinical effect of percutaneous transluminal angioplasty (PTA)combined with autologous peripheral blood stem cells(PBSC)transplantation in the treatment of lower extremity ischemic disorders.Methods Fourty-two cases of lower extremity ischemic disorders in the treatment group were treated with PTA and autologous peripheral blood stem cells injection and 40 cases in control group were treated with PTA exclusively.Results All the procedures were successful.In treatment group,ABI improved from 0.32 ±0.11 to(at the 3rd month)and 0.49 ±0.13(at the 6th month)(t=-6.765,-6.040,P<0.05)while TcPO_2 improved from(26.1 ± 2.3)mm Hg to(32.7 ±4.2)mm Hg(at the 3rd month)and(34.5 ±2.7)mm Hg(at the 6th month)(t=-8.901,-14.250,P<0.05).In control group,ABI improved from 0.30 ±0.12 to 0.47 ±0.15 and 0.47 ±0.130=-5.631,-5.873,P<0.05)while TcPO_2 increased from(25.9 ±2.4)mm Hg to(28.9 ±2.9)mm Hg(at the 3rd month)and(28.9 ± 2.1)mm Hg(at the 6th month)(t=-5.090,-5.389,P<0.05).There was significant difference in TcPO_2 on follow-up between the two groups after the treatment(P<0.05).Conclusion Autologous PBSC transplantation in combination of PTA was effective for the treatment of lower extremity ischemic disorders.PBSC injection helps to increase TcPO_2.
9.Observation of Curative Effect of Gastric Bypass Operation on Non-Obese Type 2 Diabetes and Change of Glucagon-Like Peptide-1

Objective To observe the curative effect on non-obese type 2 diabetes and the effect on change of glucagon-like peptide-1 (GLP-1) of gastric bypass operation. Methods Thirty-two cases of gastric ulcer with non-obese type 2 diabetes were suffered gastric bypass operation. Plasma glucose concentrations, insulin and GLP-1 were measured respectively in fasting and postprandial conditions before operation and in week 1, 2, 3 and month 1, 3, 6 after gastric bypass operation, and the body mass index (BMI), homeostasis model assessment ? cell function index (HBCI) and glycosylated hemoglobin (HbA1c, the index was detected only before operation and in month 3, 6 after operation) were also measured. The turnover of the diabetes condition in the 6th month after surgery was observed. Results Compared with the levels before operation, the fasting and postprandial plasma glucose levels were descending (P0.05). 10.The Effect of New Photosensitizer Combined with Adriamycin on Cell Proliferation and Cell Cycle

Objective: To study the effect of new photosensitizer Chlorophyl-derivative (CPD4) combined with Adriamycin on cell cycle and cell proliferation of MCF-7 breast cancer cell line and to investigate the mechanism of combination therapy. Methods: A new type of photosensitizer and traditional chemotherapy drug Adriamycin (ADM) were used to treat breast cancer cell line MCF-7. Flow cytometry was employed to detect apoptosis rate and cell cycle distribution in ADM group, group with photodynamic effect and the combined group. The influence of ADM on the mean fluorescence intensity and the changes in the mean fluorescence intensity after CPO4 (1.5μg/mL) treatment were analyzed. Results: The apoptosis rate of the combination group was higher than that in the other two groups, with statistical significance (P<0.01). Photodynamic effect caused G_0/G_1 phase arrest in MCF-7 cells. Low concentration of ADM increased the number of G_2/M phase cells. The percentage of G_2/M phase cells was increased in the combination group. No significant difference was found in the mean fluorescence intensity between ADM pretreated MCF-7 cells for 24 hours and 48 hours and the control group (P>0.05). Pretreatment of MCF-7 cells with ADM increased the volume of photosensitizer CPD4 into the cells. The mean fluorescence intensity at 2 hours after CPD4 incubation was the highest. Conclusion: ADM can increase the amount of CPD4 into the MCF-7 cells. Photodynamic therapy com-bined with Adriamycin has a synergistic effect on MCF-7 cells.
