1.Vascular endothelial growth factor and remedial angiogenesis
International Journal of Surgery 2008;35(10):675-678
Vascular endothelial growth factor (VEGF) is the endothelial cell-specific mitogen, facili-tates endothelial cell proliferation, increases vascular permeability and accelerates the formation of new blood vessels role. Angiogenesis is an important physiological and pathological significance of the process. In the human wound healing, inflammation, organ regeneration and tumor growth and metastasis, vascular prolifer-ative diseases, angiogenesis is an important role. Therapeutic angiogenesis is the use of inducible factor or vascular endothelial progenitor ceils, simulates in vivo angiogenesis mechanism, promotes angiogenesis and improves the collateral circulation. In this paper, VEGF and therapeutic ansiogenesis research progress were reviewed.

Aim To study on biological properties and hemopoiesis-supporting function of human bone marrow stromal cells cultured for long-term in vitro. Methods ① Bone marrow stromal cells from fetus, chidren and adults were cultured for long-time in vitro by static adherent wall assay. ② The phenotypes of the stromal cells were analyzed by immunocytochemical staining or FACA.③ Hemopoietic stem cells in umbilical cord blood were co-cultured and expanded with the stromal cells at different development stages. Results ① The fibroblast myoid cell lines were established and could be propagated to 10 generations and kept for 6 months, meantime, endothelial cells and macrophoges were obtained. ② The child stromal myoid cells were positive for viementin and negative for VIII factor by immunocytochemical staining and their phenotypes were CD33-,CD34-,CD38-and CDW90+ , while the phenotypes of adult stromal cells were CD33-, CD34-, CD38+ and CDW90+ . ③ The stromal cellls could support long-time survival or expansion of LTC-ICs. Survival time of LTC-ICs in the stromal cell-supporting system supplemented with SCF, IL-3 and IL-6 was even longer than that in system without stromal cells(P∨ 0.01). The productivity rate of LTC-ICs cultured in the stromal cell-supporting system was 2 to 4 times higher than that cultured in system without stromal cells. Conclusion The human bone marrow stromal cells could be cultured for long-time in vitro and have hemopoiesis-supp ortingfunction for the hemopoietic stem cells in cord blood.

This paper is to report the determination of the skeletal muscle cellular membrane potential (SMCMP) in vivo with the technique of "Semi-floating"glass microelectrode. The changes of SMCMP and the distribution of extra- and intracellular electrolytes and water in different phases of irreversible hemo-rrhagic shock in rabbits were studied systematically. It was found that SMCMP decreased significantly and immediately as soon as hypotension occurred, which means that the functions of the cellular membrane begin to deteriorate in the early stage of shock; SMCMP continued to decrease when the shock was progressing, which reflects that besides the failure of the cellular membrane function, there is also the degradation of the functions of the cell proper; the leakage of the intracellular potassium across the cellular membrane and the retention of the intracellular sodium and water occurred three hours after the onset of the shock, which indicates that the inability of cells to regulate their volume.

According to medicine high special educational reform’s mentality,we have extracted the knowledge points closely related to basic medical curriculum and the nursing spe-cialty,integrated the basic medical curriculum as "the basis of medical morphology"and"basic medical functions"and have it implemented in the nursing specialty in the three-year program.By doing so,we have gained some experience and provide practical basis for further integration of basic medical courses.
9. Clinical effect of low-dose cyclophosphamide and prednisone in treatment of multiple myeloma and its influence on serum VEGF, PDGF-BB levels
Academic Journal of Second Military Medical University 2012;33(4):413-416
Objective To investigate the clinical effect of low-dose cyclophosphamide plus prednisone (CP regimen) in treatment of multiple myeloma (MM) and its influence on serum levels of vascular endothelial growth factor (VEGF) and platelet-derived growth factor BB (PDGF-BB). Methods Totally 54 patients with refractory or relapse MM were continuously treated with CP regimes oral cyclophosphamide (CTX, 50 mg/d) and prednisone (Pred, 15 mg/d). The peripheral blood samples were collected from each group 0, 2, 4, and 6 months after treatment. Serum VEGF and PDGF-BB levels were analyzed by ELISA. Results The overall effective rate was up to 69% (36/52) in our study. The treatment was effective in 30 patients, including 2 complete response (CR) cases, 4 very good partial response (VGPR) cases and 24 partial response (PR) cases, and their serum samples were examined for 4 times. The serum levels of VEGF and PDGF-BB were significantly decreased in the 30 patients at 2, 4, and 6 months after treatment (P<0. 01); while their levels were not significantly change in the 7 invalid patients 2 months after treatment compared with before treatment. Conclusion Low-dose cyclophosphamide plus prednisone has noticeable effect in treatment of MM patients; it can also greatly down-regulate serum VEGF and PDGF-BB levels. The mechanisms of CP regimen in MM patients may be associated with the inhibition of new blood vessel generation.
