OBJECTIVE:To establish a method for simultaneous determination of gallic acid,protocatechuic acid,catechuic acid,corilagin and brevifolincarboxylic acid in Geranium carolinianum. METHODS:HPLC was performed on the column of Won-daSil C18 WR with mobile phase of acetonitrile-0.1% Phosphoric acid(gradient elution)at a flow rate of 1.0 ml/min,the detection wavelength was 280 nm,the column temperature was 40 ℃,and the volume injection was 20 μl. RESULTS:The linear range was 0.02-20.02 for gallic acid(r=0.999 9),0.01-20.10 for protocatechuic acid(r=0.999 7),0.02-19.78 for catechuic acid(r=0.999 6), 0.02-20.02 for corilagin(r=0.999 9)and 0.02-20.10 for brevifolincarboxylic acid(r=0.999 5);RSDs of precision,stability and re-producibility tests were lower than <3.0%;recoveries were 95.1%-100.6%(RSD=2.20%,n=6),95.8%-100.6%(RSD=1.74%,n=6),95.1%-101.9%(RSD=2.71%,n=6),97.7%-103.1%(RSD=2.04%,n=6)and 95.3%-99.0%(RSD=1.46%,n=6). CONCLUSIONS:The method is simple,accurate and reproducible,and can be used for the contents determination of gallic ac-id,protocatechuic acid,catechuic acid,corilagin and brevifolincarboxylic acid in G. carolinianum.
3.Study of complications simulated experience education improved self-management behavior in young patients with hypertension

Objective To explore the influence of complications simulated experience education on self-management behavior(medication adherence and blood pressure control and treatment attitudes and beliefs)in young patients with hypertension.Thus,tocontrol the risk factors,improve the quality of life and prevent the complication of the heart, brain and kidneys. Methods A total of 200 young patients aged 18 to 44 old with hypertension were selected according to the diagnostic criteria of Chinese guidelines for the management of hypertension from June 2016 to May 2017.Participants were randomly divided into intervention group and control group (100 cases in each group).In the control group conventional education was used one-to-one education mode.And the complication simulated experience model was used in the intervention group one-to-one education mode. The differences in medication adherences, blood pressure, treatment attitude and beliefs between the two groups of patients were compared before and one month after intervention. Results The were no differences of general situation, blood pressure and medication adherence and treatment attitudes and the score of beliefs between two groups before intervention (P>0.05). After intervention, the systolic and diastolic blood pressure were (132.67 ± 12.95) mmHg (1mmHg=0.133 kPa) and (83.21 ± 5.97) mmHg in the intervention group, which were lower than(137.87±13.97)mmHg and(89.68 ±6.12)mmHg in the control group(t=2.730,7.568,P<0.05).Medication adherence score was 7.15±0.75 in the intervention group,which was higher than 5.10± 1.89 in the control group (t=10.081, P<0.05). In the intervention group, after the intervention treatment, attitudes and beliefs score was 82.15±13.13,which was higher than 71.11±15.89 in the control group.(t=5.337, P<0.05). Conclusions The complications simulated experience education is superior to conventional health education, which can improve medication compliance, blood pressure control, and treatment attitude and belief level in young patients with hypertension.
4.Expression of tumor necrosis factor-α induced protein 8 like-1 in cisplatin-induced acute kidney injury models

Objective:To investigate the expression and role of the tumor necrosis factor-α (TNF-α) induced protein 8 like-1 (TIPE1) in acute kidney injury (AKI) induced by cisplatin in animal model and cells.Methods:Twelve male C57BL/6 mice aged 6-8 weeks were randomly divided into the control group and the model group. Mice in the model group received a single intraperitoneal injection of 20 mg/kg of cisplatin (20 mg/kg saline in the control group). All mice were euthanized after 5 days. Meanwhile, serum and kidney samples were collected. The levels of serum creatinine (Scr) and blood urea nitrogen (BUN) were detected by biochemical kits. Renal histopathological changes in mice were observed by HE staining. The expression of TIPE1 in kidney was examined using immunohistochemistry. qRT-PCR was used for testing the relative expression of TIPE1 mRNA in mice kidney. Western blotting was used for testing TIPE1 and NGAL protein relative expression in mice kidney. Human kidney proximal tubular cells (HK-2) were stimulated with 20 μmol/L cisplatin for 0, 6, 12 and 24 h to establish cisplatin-induced AKI cell model. The expressions of TIPE1 mRNA and protein were detected by qRT-PCR and Western blotting in HK-2 cells. The expression of TIPE1 gene in HK-2 cells was silenced by lentivirus containing TIPE1 siRNA sequence. Then, TIPE1 stable knockout HK-2 cell strains were treated with 20 μmol/L of cisplatin for 24 hours. The protein expression of tubular damage marker neutrophil gelatinase-associated lipocalin (NGAL), microtubule-associated protein 1 light chain 3 (LC3) and Beclin1 in HK-2 cells were detected by Western blotting. Results:Compared with the control group, the expressions of TIPE1 mRNA and protein were up-regulated and NGAL protein expression was increased significantly in renal tissue of the model group (all P<0.05). The expressions of TIPE1 mRNA and protein were remarkably increased with the prolongation of cisplatin treatment in HK-2 cells (both P<0.05). Compared with the scramble siRNA group, the protein expressions of NGAL, LC3-Ⅱ and Beclin1 were increased significantly in the TIPE1 siRNA group after lentivirus interfered with the expression of TIPE1 gene in HK-2 cells (all P<0.05). Conclusions:The mRNA and protein expressions of TIPE1 are increased in acute kidney injury models. Gene silencing of TIPE1 can promote the expressions of early renal tubular damage marker and autophagy-related proteins, which indicates the excessive autophagy aggravates renal tubular injury. It is suggested that TIPE1 may be involved in the pathogenesis of acute kidney injury.
5.Correlation study of connexin 43 expression in placenta tissue and gesta-tional diabetes

Objective To observe the expression of connexin 43(Cx43) in the placenta of patients with gestational dia-betes mellitus (GDM) and explore the role of Cx43 and Cx43-constituted gap junctional intercellular communication (GJIC) in the GDM placenta tissue. Methods The immunohistochemical method and Western blotting method were used to detect the Cx43 location and expression in the placenta tissue of GDM non-treatment group (18 cases), GDM treat-ment group(22 cases) and normal gestational group(12 cases). Results Immunohistochemical staining showed that Cx43 expressed in the cytoplasm and cell membrane of trophocytes. In the GDM non-treatment group,Cx43 positive particles mainly existed in cytoplasm and had reduced obviously. Statistical analysis showed that GDM non-treatment group had significantly lower Cx43 protein level than the other two groups (P<0.05). Conclusion Low Cx43 expression in the trophocytes of GDM non-treatment group indicates abnormal GJIC between the trophocytes, which leads to abnormal fusion and differentiation of GDM placenta trophocytes. This may be related to the adverse pregnancy of patients with GDM.
