The confirmation of maxillomandibular relation is the key step to manufacture complete denture. Certain kind of transformation and shift will lead to the inaccuracy in the progress of the confirmation of maxillomandibular relation, while these problems could be avoided by applying remodified vacuum-formed occlusal pad. In this way, accuracy of the confirmation of maxillomandibular relation and the clinical effect can be enhanced.
2.Effect of NG-nitro-L-arginine methyl ester on invasion and metastasis of human colorectal cancer cell line

BACKGROUND:Nitrogen monoxide (NO) plays dual effects in the occurrence and development of tumor. High concentration of NO exerts anticancer effect through cellular cytotoxicity and inducing the apoptosis of tumor; under the changed microenvironment in tumor tissue, its effect on tumor tissue is mainly promoting the growth of tumor.OBJECTIVE: To study the effect of the nitric oxide synthase(NOS)inhibitor NG-nitro-L-arginine methyl ester(L-NAME) on invasion and metastasis of human colorectal cancer cell line LS174T.DESIGN: Single sample observationSETTING: Department of Surgery, Tumor Hospital affiliated to Harbin Medical University MATERIALS: The study was conducted in Cancer Research Institute of Harbin Medical University between January 2004 and October 2004. The human colorectal cancer cell line LS174T was purchased from Cancer Research Institute of Harbin Medical University. L-NAME was purchased from Sigma Company.METHODS: LS174T cells were treated with L-NAME of 0.2,0.4,0.8 and 1.0 mmol/L. Effect of drugs on invasion and migration of LS174T cells was observed in reconstructed basement membrane invading experiment.MAIN OUTCOME MEASRES: Inhibitory rate of L-NAME on LS174T line invading reconstructed basement membrane and the inhibitory rate of cellular migration.RESULTS: ①0.2, 0.4, 0.8, 1.0 mmoL/L L-NAME were used to treate LS174T cells for 72 hours, and the inhibitory rate in cell invading reconstructed basement membrane was 10.29%, 19.62%, 34.08%,42.23% respectively (P < 0.05 or 0.01 ). ② 0.2, 0.4, 0.8, 1.0 mmol/L L-NAME were used to treate LS174T cell for 72 hours , its inhibitory rate in inhibiting cells migration was 20.76%,24.95%,39.43%,46.85% respectively(P < 0.01 ).CONCLUSION: L-NAME has anti-invasive and anti-metastasic effects on LS174T cells.
3.Changes of bone mineral density of the proximal femur in patients with revision total hip arthroplasty

[Objective]To understand the changes of periprosthetic bone mineral density(BMD) of the proximal femur in patients with failed hip arthroplasty by measuring with dual energy X-ray absorptiometry(DEXA). [Method]Periprosthetic BMD was determined in 9 patients with revision total hip arthroplasty by dual energy X-ray absorptiometry(DEXA),defining regions of interest according to Gruen(1~7),and comparing the operation side with the nonoperation side.Clinic results of the cases were evaluated according to Harris Hip Score(HHS).[Result]The mean HHS was 61 points in 9 patients at this follow-up examination.There was a significant decrease(8.9%~27.6 %) in BMD in every Gruen zone as compared with the nonoperation side.[Conclusion]The bone loss in the proximal femur in patients with failed hip arthroplasty is significant,and the pattern of bone loss around failed hip arthroplasty may be different from the typical remodeling seen after successful hip arthroplasty.
4.Inhibitory Effect of N~G-Nitro-L-Arginine Methyl Ester on Growth of Colorectal Cancer Xenografts in Nude Mice

Objective To investigate the effect of N~G-nitro-L-arginine methyl ester (L-NAME), an inhibitor of nitric oxide synthase, on the growth of colorectal cancer xenografts in vivo and on tumor-associated neovascularization. Methods Twenty BALB/c nude mice were randomly divided into control group and study group equally. Human colorectal cancer cell line SL174T was inoculated subcutaneously into nude mice to form transplantation tumors. Saline 0.2 ml was intragastric-administrated to mice in control group and L-NAME (4 mg toties guoties) was administrated orally to mice in study group three times per week for four weeks. The changes of tumors in both groups were recorded and the microvessel density (MVD) was also measured by immunohistochemistry assay. Results L-NAME significantly inhibited the growth of colorectal cancer xenografts in nude mice. The weight of transplantation tumor reduced with the inhibitory rate of 41.36%, and the inhibitory rate of tumor volume was 43.48 % in study group. Objective To study the change of chromosome aberration rate and micronucleus rate in patients with esophageal carcinoma before and after radiotherapy.Methods Twenty one patients with esophageal carcinoma who were fit for conditions were divided into two groups randomly.GroupⅠ(n=10),patients received conventional radiotherapy(radiation field was regular rectangular shape);groupⅡ(n=11),patients received conformal radiotherapy(radiation field was same to the shape of target volume),1.8-2.0 Gy,one time a day,5 times a week,the total dose was 60-70 Gy.The peripheral blood was obtained before and after radiotherapy,chromosome aberration rate and micronucleus rate were detected.Results The chromosome aberration rate and micronucleus rate were increased after conventional radiotherapy than before (P0.05).Conclusion Compared with the conventional radiotherapy,the conformal radiotherapy can decrease cytogenetics damage,the detection of chromosome aberration rate and micronucleus rate can be regarded as a simple method to evaluate cytogenetics damage in esophageal carcinoma patients induced by radiotherapy.
6.The effects of L-NAME on invasion and metastasis of human colorectal carcinoma cell line

Objective To study the effects of L-NAME on invasion and metastasis of human colorectal (carcinoma) cell line(LS-174T) and explore its possible mechanism.Methods The LS-174T cells were co-incubated with L-NAME at various concertrations.Griess techniqe was used to examine the effect of(L-NAME) on excretion of nitric oxide(NO) of the cells.The effects of L-NAME on invasion and migration of LS-174T were evaluated using Transwell chambers attached with polycarbonate filters and reconstituted(basement) membrane.Expression of MMP2 mRNA and TIMP2mRNA in LS-174T cells was measured by(RT-PCR).Results(1)L-NAME decreased the excretion of NO of the cells in a dose-dependent manner.(2)The ability of the L-NAME treated LS-174T cells to invade the reconstituted basement membrane(increased) significantly with the concentration and time,at the concentration of 0.2 mmol/L,0.4 mmol/L,0.8mmol/L and 1.0mmol/L,respectively,after 72 hour,the inhibition rates were 10.29%,19.62%,34.08% and 42.23%,respetively.(P

Objective: To study the effects of the alcohol extract of Loulu (AEL) on the ability of learning and memory in mice and on the activity of cholinesterase in brain and whole blood.Methods: In order to establish the effect of the alcohol ectract of Loulu on the ability of learning and membory in mice, the methods of one trial passive avoidance response(step down test) were used. Results: The AEL could antagonize the acquisition impairment of memory induced by scopolamine (SCPL) and the consolidation disruption of memory elicited by cycloheximide (CYC), inhibit the activity of cholinesterase in brain and whole blood. Conclusion: The improving effect of the AEL on the ability of learning and memory might be related with its effect of improving function of the central cholinesterase system.
8.Different fixation methods for artificial femoral head replacement: A biomechanical comparison of joint stability

BACKGROUND: Artificial humeral head replacement is an effective method for the treatment of complex proximal humeral fractures, which has received good results in relieving pain. However, the final functional recovery is unpredictable. OBJECTIVE: To compare biomeshanical stability between anatomical and overlapping reconstruction of the greater tuberosity in cadaveric humeral head replacement models.METHODS: Eight pairs of fresh-frozen shoulder cadavers (16 shoulder joints) were match-paired into two groups. Standardized humeral head replacement procedure was performed in all specimens, and anatomical and overlapping reconstruction of thegreater tuberosity was adopted in each group respectively. For overlapping group, the greater tuberosity was reattached to the proximal humeral shaft in an overlapping style, which was achieved by an additional 5 mm bone osteotomized from the medial cortex of the humeral diaphysis. Custom mounting apparatus and fixation jigs were designed for designated shoulder motion.RESULTS AND CONCLUSION: When the shoulder was external rotated to neutral position, the mean displacement of greater tuberosity in the anatomical reconstruction group was smaller than that of the overlapping reconstruction group (P < 0.05). When the gleno-humeral joint was elevated to 30~ and 60~ forward flexion (accounting for 45° and 90° shoulder forward flexion), there was no significant difference of greater tuberosity displacement between the anatomical group and overlapping group. The findings demonstrated that, although overlapping reconstruction can increase the bone healing area between the greater tuberosity and the humeral diaphysis, there may be some loss in mechanical stability as the trade-off. Even though we strictly follow the standardized postoperative rehabilitation protocol after humeral head replacement, prominent displacement between the greater tuberosity relative to the humeral diaphysis was detected. Accordingly, postponing of the postoperative rehabilitation program after humeral head replacement for a decent period may improve tuberosity healing.

Objective To study the effect of single short (SS) veno-venous hemofiltration and intermittent short (IS) veno-venous hemofiltration on imbalance of pro- and anti-inflammatory cytokines in severe acute pancreatitis (SAP) patients. Background Our previous study demonstrated that hyperglycemia aggravate the choroidal neovascularization (CNV) by promoting the chemotaxis process of bone marrow-derived cells (BMCs).Bioluminescence imaging (BLI) can dynamically monitor CNV in vivo.However,how diabetes mellitus (DM)participate in CNV is still in research.Objective This study was to dynamically observe the influence of BMCs to CNV under hyperglycaemia by using BLI combined with histopathology.Methods BMCs from luciferase-green fluorescent protein (Fluc-GFP) double transgenic mice were injected to adult wild type C57BL/6J mice (nine mice per group) via caudal vein to create the chimera models with a chimerism degree higher than 85%,and the chimeric mice were randomized into the control group and DM group based on randomized number table.Streptozotocin [60 mg/(kg · d)] was intraperitoneally injected daily for 5 days to establish the DM models in the chimeric mice of the DM group.CNV was induced in the chimeric mice of both control group and DM group with 532 nm laser photocoagulation.BLI signal of BMCsFluc+GFP+ was in vivo examined by IVIS Kinetics system 1,3,5,7,14,21 and 28 days after CNV modeling.At the seventh day after laser,part of mice were sacrificed,and choroidal and retinal sections were prepared for histopathological examination.The length and thickness of CNV were compared between the control group and DM group.The use and care of experimental followed Statement of ARVO.Results The chimerism degree of the chimeric mice was (88.85 ± 2.46) % 28 days after BMCs transplantation,and the blood glucose concentration in the DM group was (17.88±0.86)mmol/L.Histopathological examination revealed that CNV broke through the Bruch membrane toward subretinas.The length of the CNV was (338.67±33.17) μm in the DM group and (180.33±24.68)μm in the control group,showing a significant difference between the two groups (t =8.943,P<0.05).However,no significant difference was seen in the CNV thickness between the two groups (t =1.790,P>0.05).Light signals appeared 1 day and reach strongest 7 days after CNV modeling in both groups.The Light signals were stronger in the DM group than those in the control group on 5,7,14 and 21 days after CNV modeling (t =3.411,5.594,5.067,2.663,all at P<0.05).Conclusions Hyperglycemia can promote more BMCs to participate in the pathogenesis and aggravation of CNV.The behavior of BMCs in CNV can be evaluated using BLI in vivo.
