1.The transdifferentiation of Sca-1~+ cells from murine fetal liver

AIM: To explore transdifferentiation potential of Sca-1 + cells from murine fetal liver. METHODS: 2?10 3 of Sca-1 + cells from male murine fetal liver were transfused into female mouse irradiated lethally with ? ray from 60 Co source (10 Gy) via tail vein. Two months later, FISH and immunohistochemistry were used to detect the situation for transdifferentiating of the donor cells (male cells) in tissues of female recipient mouse. RESULTS: The renal tubular epitheliocyte-like and neurocyte-like cells with Y chromosome were found on the sections of renal and brain tissues from female recipient mice. These cells have phenotype characteristics of RCA+/CD - 45 F - 4/80 and NueN +/CD -45 F - 4/80, respectively. CONCLUSION: The evidence is provided for Sca-1 + cells from murine fetal liver to transdifferentiate into both renal and brain tissue cells.
2.Differentiation of Sca-1~+ cells from murine fetal liver into renal cells in mice with acute renal failure

AIM: To study the effect of acute renal failure (ARF) on the differentiated frequency of Sca-1+ cells from murine fetal liver in irradiated mice. METHODS: The Sca-1+ cells from murine fetal liver were isolated with magnetic cell sorting (MACS) technique, the sex of which was identified by PCR. The 2?104 Sca-1+ cells were transplanted into a lethally irradiated ([ 60Co], 8 Gy) inbred female mouse. After 8 weeks, these recipient mice were divided to A, B, and C groups at random (A group: irradiated; B group: ARF; C group: ARF and Sca-1+). The mice in B and C groups were induced to ARF with 50% (V/V) glycerin (11.6 mL/kg). 72 hours later, the mice in C group were injected with the fresh prepared Sca-1+ cells again. 8 weeks later, mice were sacrificed, and their kidneys were taken out, fixed and slices were prepared. Fluorescence in situ hybridization (FISH) of renal slices was performed and the pictures of them were taken and analyzed. RESULTS: The cells containing Y chromosome were found in renal slices from the mice in A, B and C groups, which located in epithelial cells of renal tubules, interstitium, glomeruli, and glomerular margin and increased gradually. The double and encircle zone of Y chromosome cells were found in the slices from the mice in B and C groups separately, which was consist of new renal tubules. The differentiation frequency of Sca-1+ cells in kidney in A, B and C groups were (1.65?0.18)%, (8.58?1.34)% and (18.13?1.91)%, respectively, which showed significant difference between former group and later group (P
3.Hybrid therapy in patients with complex arteriosclerosis obliterans

Objective To evaluate hybrid therapy in patients with complex arteriosclerosis obliterans.Methods From Sep 2006 to Oct 2011,66 patients with complex arteriosclerosis obliterans were treated by hybrid therapy.A retrospective review was performed in all cases.Primary patency rate analyses were performed using Kaplan-Meier life tables.Results The technical success rate was 100%,and no patients died in perioperative period.12.1% patients improved 3 levels,22.7% patients improved 2 levels,and 65.2% patients improved 1 level in Fontaine grade after procedures.The average ABI before and after procedures were 0.38 and 0.67 (P < 0.05).2 patients died and other 64 patients completed 24 months follow-up.The primary patency rates and second patency rates were 75.0% and 92.2% at 24 months respectively.Primary patency rates of Fontaine Ⅱ patients were better than Fontaine Ⅲ and Ⅳ patients (P < 0.05),and primary patency rates of Fontaine Ⅲ patients were better than Fontaine Ⅳ patients (P < 0.05).Conclusions Hybrid therapy procedures provide an effective treatment in complex arteriosclerosis obliterans patients.
4.Differentiation of murine fetal liver derived Sca-1~+ cells into cardiomyocytes in vivo

Objective:To explore potential differentiation from antigen-1 positive stem cells (Sca-1 +cells) in murine fetal liver tissue into cardiomyocytes in vivo. Methods: Sca-1 + cells from the murine fetal liver of 14.5 d were separated by Magnetic cell sorting (MACS).The sequence of the sex determination region of the Y chromosome was determined by PCR.Sca-1 + cells (2?10 3) of male mice were transfused to female mice receiving 8-12 weeks irradiation at a lethal dose of ?-ray(10 Gy) from a 60Co source.Two months later,the mice were killed and the hearts were removed to make slices. Fluorescence in situ hybridisation (FISH) tests with a Y chromosome probe were used to detect sex of the cells.Immunohistochemistry with antibodies of desmin,Flt-1,white blood cell common antigen CD45 and macrophage F 4/80 were used to detect tissue characteristics of cardiomyocytes. Results: Cells with Y chromosome were found in the cardiomyocytes of female mice transplanted with Sca-1 + cells,and showed phenotypic characteristics of Desmin +/Flt-1-/CD45-/F- 4/80 at the same time.Conclusion: Sca-1 + cells from the murine fetal liver are mostly hematopoietic stem cells,having the potential of differentiating into cardiomyocytes.
5.Tissue irradiation injury and past-transplantation distributing diversity of Sca-1 positive cells from murine fetal liver

liver.Combined informational analysis showed that the presenting frequency of the cells containing Y chromosome was consistent with the irradiative sensitivity of the organ.CONCLUSION: These findings suggest that the capability of differentiation of Sca-1 positive cells from murine fetal liver was potentially connected to the extent of damage in these organs when transferred in vivo.
6.Investigation of Nutritional KAP and Its Effect on Serum Ferritin Level of Elite Female Small Ball Athletes

Objective To investigate the status quo of nutrition knowledge,attitude and practice(KAP) among elite female small ball athletes of Guangdong province and its influencing factors.Their serum ferritin was also observed to explore its correlation with their KAP,so as to provide evidences for formulating and implementing scientific and reasonable nutrition education schemes.Methods Fifty-four female small ball athletes,including 20 handball,20 hockey and 14 softball players,were selected to complete a self-designed diet questionnaire based on KAP education mode and their serum ferritin indicators were analyzed at the same time.Results (1)The average scores(all out of 100)of nutrition knowledge (K),attitude (A) and practice (P) were 43.64 ± 11.66 (with a failure rate of 90.7%),78.37 ± 10.21 (with the rate of excellence of 59.3%)and 63.10 ± 10.17(with a passing rate of 37.0%),and the average KAP score was 185.10 ± 23.01,with a passing rate of 44.4%.(2)The one-way ANOVA found that there were significant differences in the average A score and the total KAP score (P<0.05) among the athletes of different small ball teams.The correlation analysis results showed that the average A score was of significant positive correlation to the average P score.Moreover,the average KAP total score was of significant correlation with sports events (P<0.05) and the education level (P<0.05),but not with the age,training period and exercise level(P>0.05).The multiple stepwise regression analysis results showed that training period(P<0.05) and sports events(P<0.05) was the influencing factor of nutrition knowledge and attitude respectively,and that education level (P<0.01) was the influencing factor of both nutrition practice and KAP.(3)Significant differences were found in the serum ferritin among athletes of different sports events(P<0.05)the serum ferritin was of significant positive correlation with the nutrition knowledge(P<0.05).Conclusion (1)The lack of sports nutrition knowledge and the unbalance of nutrition behavior are common in elite female small ball athletes,but they had a positive attitude.It is necessary to carry out a series of effective nutrition education programs to improve their sports nutrition knowledge,cultivate their good nutrition attitudes and finally formulate rational and effective nutritional intervention measures to promote the formation of healthy nutrition behavior of athletes.(2) The sport event is the key influencing factor of the nutrition attitude and dietary behaviors,which suggests that more more attention should be paid to athletes' dietary management and focus should be put on their diary behavior on no-training days and on their energy ratios control of each meal,especially breakfast and extra meals,and rational intake of snacks.(3)The incidence of iron deficiency is consistent with KAP questionnaire survey results,which indicates that it may be an effective measure to prevent iron deficiency anemia through strengthening athletes' dietary nutrition education.
7.Prevalence of dyslipidemia among adult professional athletesin Guangdong Province

Objective:
To investigate the prevalence of dyslipidemia among adult professional athletes in Guangdong Province, so as to provide insights into dyslipidemia screening and health management among professional athletes.
Methods:
In 2019, active athletes at ages of 18 to 30 years were recruited from 12 provincial sports teams in Guangdong Province using a cluster sampling method. A self-designed questionnaire survey was conducted to collect gender, age and sport items, and the levels of triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) were detected. The non-HDL-C level was calculated, and the gender- and sport item-specific prevalence of dyslipidemia was estimated.
Results:
Totally 460 athletes were investigated, including 245 males (53.26%) and 215 females (46.74%), with a mean age of (21.91±2.77) years. The overall detection of dyslipidemia was 25.87%, and the detection rates of high TG, high TC, high LDL-C, low-HDL-C and high non-HDL-C were 20.22%, 5.87%, 13.04%, 3.26% and 9.57%, respectively. The detection rates of high TG (9.39% vs. 1.86%; χ2=11.743, P<0.05), low HDL-C (5.31% vs. 0.93%; χ2=6.951, P<0.05) and high non-HDL-C (12.24% vs. 6.51%; χ2=4.351, P<0.05) were significantly greater in men than in women, and the detection of high TC was lower in men than in women (17.96% vs. 22.79%; χ2=8.627, P<0.05). There was a significant difference in the detection of dyslipidemia among athletes engaging in different sport items (χ2=47.552, P<0.05), and a high detection rate of dyslipidemia was seen in baseball athletes (34.29%), softball athletes (37.04%), shooting/archery athletes (52.73%).
Conclusion
The prevalence of dyslipidemia was 25.87% among adult professional athletes in Guangdong Province, and high TC in combination with high LDL-C were the predominant type of dyslipidemia. The management of blood lipids should be given a high priority to male athletes and baseball, softball and shooting/archery athletes.
8.Cytokine production in mice with experimental cardiomyopathy treated with anti-L3T4 monoclonal antibody at different stages

AIM:To clarify the mechanism of treating autoimmune cardiomyopathy at different stages with anti-L3T4 monoclonal antibody.METHODS:Mice immunized with human mitochondria ADP/ATP peptides were used as the cardiomyopathy(DCM) group,and the sham-immunized mice were regarded as the controls.Mice receiving early treatment were immunized with the same peptides,followed by the injection of 400 ?g of anti-L3T4 on day 0,1 and 2 post-immunization.Mice in the late treatment group were immunized as of the early treatment group but anti-L3T4 was administered 3 months post-immunization.The cytokine expression was measured with three-color flow cytometry to quantitate the splenic Th1/Th2 cell subsets in the different groups of mice.In addition,serum and myocardial cytokines were measured by enzyme-linked immunosorbent assay and real-time PCR.RESULTS:Th1 and Th2 subsets in the early treatment group were similar to those in control group,but were drastically lower than those in DCM group.Mice in the late treatment group showed an increased level of Th1-related cytokines,while the Th2 level was between the DCM and early treatment group.IFN-? and IL-6 levels in early treatment group were similar to those in control group.In the early treatment group,IL-4 level was higher than that in control and lower than that in DCM group,whereas IL-2 and TNF-? contents were lower than those in control and DCM group.In the late treatment group,IFN-? and IL-2 levels were higher than those in DCM group and lower than those in the early treatment group,while IL-6 and IL-4 levels were lower than those in DCM group.CONCLUSION:These results suggest that the cytokine production in cardiomyopathic mice may be repressed by treatment with anti-L3T4 at different stages.Early treatment with anti-L3T4 has better inhibitory function than treatment in late stage of autoimmune cardiomyopathy.

AIM:We examined the efficacy ofanti-L3T4 McAb in the T cell signaling pathway in treating ex-Derimental antoimmune cardiomyopathy in BALB/c mice,as a model of the autoimmune mechanism involved in human di-latedardiomyopathy(DCM).METHODS:ADP/ATP carrier peptides were used to induce autoimmune cardiomyopathy in BALB/c mice.Afier 3 months.anti-L3T4 McAb WaS administered to deplete CD4+ T ceHs in the mice.Real-time PCR were used to detect the expression of intracellular signaling molecules(p561ck,p59fyn and Zap-70)and cytokine production(IFN-r,IL-2 and IL-4)in T cells.The expression of CIM5 was determined by immunohistochemistry a-nalysis.RESULTS:Reduced expression of p561ck,p59fyn and Zap-70 and the reduced cytokine production of IFN-r, IL-2 and IL-4 in T cells of anti-IJ3T4-treated DCM mice were found.Also,the expression of CD45 in spleen T cells was significantiv decreased in the anti-L3T4-treated group.In contrast,immunization with irrelevant Ab did not protect the mice.the expression of T cell signaling molecules,CD45,and cytokine were not inhibited.CONCLUSION:Thesestudies provide direct evidenee that anti-L3T4 McAb can be all effective immunomodulator to T cell signal molecules and 8ubsequent cytokine production events in ADP/ATP carrier-induced DCM in BALB/c mice.
