Objective To investigate the molecular mechanisms that high glucose stimulate collagen Ⅰ synthesis in renal tubular epi-thelial cells. Methods Normal rat pwximal tubular epithelial (NRK52E) cells were cultured grown in RPMI-1640 medium and were divid-ed four groups: mannitol group, high glucose group, high glucose + neutralizing TGF-β1 antibody group, high glucose + IgG1 group. TGF-β1 in the supematant of cultured cells were measured by enzyme-linked immunosorbent assay (ELISA). Nuclear expression of p-Smad2/3 were examined by immunocytochemistry. Expression of collagen Ⅰ mRNA was detected by RT-PCR. Expression of collagen Ⅰ pro-tein was detected by Western blot. Results High glucose up-regulated the expression of collagen Ⅰ mRNA by time-dependent manor. Com-pared with mannitol group, high glucose markedly increased the level of TGF-β1 in supernatant of cultured cell after 24h and 48h and upreg-ulated p-Smad2/3 nuclear expression(t =4. 2, t = 3.25, P <0.01). Neutralizing TGF-β1 antibody inhibited high glucose- induced p-Smad2/3 nuclear expression, downregulated high glucose-induced collagen Ⅰ mRNA and protein expression(t = 3.12, t =3.02, P < 0.01). Conclusion High glucose stimulated collagen Ⅰ synthesis in renal tubular epithelial cells via activated TGF-β/Smad signaling path-way.

Objective To investigate the effect of smad7 on AGEs-induced NRK52E cell tranedifferentiation. Methods NRK52E cells were transferred with Smad7 gene and the cell lines of doxcycllne (Dox) -regulated Smad7 expression were selected for the study. The NRK52E cells were random divided into two groups, Dox - group, the NRK52E cells were cultured with medium only containing AGEs, Dox +group, the NRK52E cells were cultured with medium containing AGEs and Dox. Nuclear translation of p-Smad2/3 were examined with immunocytochemistry. The expression of SmadT, α-SMA, E-cadherin protein were detected by western blot. Results Expression of Smad7 protein in the Dox + group was increased higher than that in Dos- group( t = 3.05, P<0.01). High expression of Smad7 caused a marked inhibition of p-Smad2/3 transnuclear location at 30min and 24h(t=4. 5,t= 3.2, P<0.01). Overexpression of Smad7 dramatically de- creased protein expression of α-SMA, while evidently increased protein of E-cadherin in the NRK52E cells stimulated by AGEs(t=3.47, t=3.25, P<0.01). Conclusion Overexpression of Smad7 can inhibit AGEs - induced transdifferentiation in NRK52E cells.

Objective:To investigate expression of cytokines and apoptosis factors in islets of female NOD mice at different weeks of age.Methods:54 female NOD mice were randomly divided into three groups(C1,C2,C3)and each group consisted of 18 mice.The mice were killed respectively at 4th,15th,30th week (of onset of diabetes),and their panreases were removed.The levels of expression of cytokines and apoptosis factors mRNA of the pancreas were detected by RT-PCR.The expressions of cytokines and apoptosis factors protein were measured by using immunohistochemical method.Insulitis scores were observed by HE staining.Results:Expressions of Fas and IFN-? mRNA in pancreas were stronger in C3 vs C1 or C2,in C2 vs C1(P0.05).Expressions of Fas and CPP32 were stronger in islet cells in C2 and C3 vs C1(P

Objective To analyze the status quo and influencing factors of patient safety culture among medical staffs in the hospitals of Shenzhen City to provided reference for its improvement and conduct the patient safety culture research by other medical institutions.Methods The Patient Safety Culture Questionnaire of Medical Institutions(PSCHO) was adopted to investigate on 1000 medical persons from February to May 2016.Results The total score of patient safety culture in medical staffs was (3.53±0.40) and,the difference among different categories of medical persons had no statistical significance (P＞ 0.05).The differences among the medical staffs with different working ages,communication frequency with patients,training times,concern frequency had statistical difference(P＜0.05).Conclusion The patient safety culture level in Shenzhen City is higher.However,the sense of blame and shame is one of the important factors impeding the promotion of the patient safety culture in hospitnl.Hospital should conduct the targeted improvement to the related factors affecting the patient safety culture from the aspects of organism,department and individual level.

OBJECTIVETo evaluate the clinical outcomes and complications of Triple-Endobutton plates in treating Tossy type III acromioclavicular joint dislocation.

METHODSFrom January 2011 to January 2013,45 patients with Tossy type III acromioclavicular joint dislocation were treated with Triple-Endobutton plates. There were 35 males and 10 females with an average age of 30.5 (ranged from 19 to 60) years old. At the final follow-up, VAS, DASH, Constant-Murley criterion were used to evaluate shoulder function.

RESULTSAll patients were followed up from 15 to 36 months. No neurovascular injury, wound infection and stress fractures were found,but 3 patients had a re-dislocation. At the final follow-up,the mean VAS score was decreased from (5.7±1.6) preoperatively to postoperative (0.2±0.1); DASH score was significantly decreased from (19.6±4.3) preoperatively to (0.3±0.1) postoperatively; Constant-Murley score was improved from (34.4±4.3) before operation to (94.8± 3.5) after operation.

CONCLUSIONClinical outcomes of treating Tossy type III acromioclavicular joint dislocation with Triple-Endobutton plates is satisfactory. However, re-dislocation is still the most common complication. Careful perioperative management is an important factor in preventing re-dislocation.

The in vitro passage number and proliferation of non-immortalized cells are limited, which restrictions cell therapy or in vitro studies. Cells transfected with temperature sensitive simian virus 40 large T antigen (ts-SV40LT) gene could show the greatest proliferation. The cells can be amplified with compensating the lack of limited number of cells under the permissive temperature. Non-permissive temperature can be used in studying the cell therapy or its other physiological characteristics. This research field involves peritoneal stromal cells, satellite cells of urinary tract, oral epithelial cells, adre?nal medullary cells, bone marrow-derived endothelial cells, retinal progenitor cells, mesenchymal stem cells, hematopoietic stem cells, mast cells, podocytes and Kupffer cells. In this study, the current research on Ts-SV40-mediated temperature-sensitive cells was reviewed.

Objective:To study the relationship between serum level matrix metalloproteinase-9 (MMP-9) and atrial fibrillation (AF) and to assess its predictive value of AF recurrence in persistent AF patients after cardio-version.
Methods: Our research included in 4 groups: Control group, n=30 healthy subjects, Paroxysmal AF group, n=30, Persistent AF group, n=60 and Permanent AF group, n=30. Serum levels of MMP-9 were examined and compared among different groups. The patients in Persistent AF group received drug therapy or drug+electric cardio-version;with successful cardio-version, antiarrhythmia medication was used to maintain sinus rhythm and the patients were followed-up for prognosis, MMP-9 levels were further compared between the patients with or without AF recurrence to assess the predictive value of MMP-9 for AF recurrence.
Results: Serum levels of MMP-9 were as follows: Control group (151.43±88.14) ng/ml, Paroxysmal AF group (182.66±96.39) ng/ml, Persistent AF group (278.93±89.22) ng/ml and Permanent AF group (336.82±112.35) ng/ml respectively, which indicated that MMP-9 level in Persistent AF group was higher than Paroxysmal AF group and Control group;MMP-9 in Persistent AF group was higher than the other 3 groups, P<0.01. The patients in Permanent AF group were followed-up for (15.4±4.3) months after cardio-version, retrospective analysis found that the patients with AF recurrence had the higher MMP-9 level than those without AF recurrence (315.63±94.52) ng/ml vs (261.92±87.86) ng/ml, P=0.036. Multivariate regression analysis was conducted on age, gender, AF history, left atrial diameter and MMP-9, the consequence showed that serum MMP-9 level was the independent predictor for AF recurrence in persistent AF patients after cardio-version (P=0.035, OR=1.135, 95%CI 1.018-1.321).
Conclusion: Serum MMP-9 level was elevated in persistent and permanent AF patients;MMP-9 was the independent predictor for AF recurrence in persistent AF patients after cardio-version.

ObjectiveTo investigate the effects of different doses of obestatin on amylase secretion of pancreatic acinar and lobules of rats in vitro.MethodsPancreatic acinar cells of rats were separated in vitro and incubated with different doses of obestatin (0,0.1,1,10,30 nmol/L) for 1h,another group of pancreatic acinar ceils was incubated with obestatin for 30 min,then was incubated with 100 pmol/L CCK-8 for another 30 min.Pancreatic lobules,which containing intrapancreatic nerve terminals and islets,were prepared and were incubated with different concentrations of obestatin for 30 min at 37℃ with or without 75 mmol/L KCl.Amylase levels in the supernatants,acinar cells and pancreatic lobules were calculated as a percentage of total amylase content.ResultsObestatin (0,0.1,1,10,30 nmol/L) produced no significant change in basal amylase release of acinar cells [ (3.48 ± 1.44) ％,(3.70 ±- 1.39) ％,(3.36 ± 1.24) ％,(3.86 ± 1.41 ) ％,(4.54 ± 2.01 ) ％ ].CCK-8 significantly increased amylase release of acinar cells [ ( 13.84 ± 2.63 ) ％ vs (3.48 ± 1.44)％,P ＜0.05],but obestatin (0.1,1,10 nmol/L) has no effect on the amylase release inducedby CCK-8 [(14.55 ± 1.7)％,(13.79 ± 1.81)％,(14.39 ± 1.12)％].Obestatin (0.1,1,10,30 nmol/L) did not affect the amylase release of pancreatic lobuole.KCl significantly increased anylase release,which was ( 1.84 ± 0.29 ) folds higher than that of control group ( P ＜ 0.05 ),but obestatin has no effect on the amylase release induced by KCl,which were (2.01 ± 0.30 ),( 1.89 ± 0.41 ),( 1.74 ± 0.14 ),( 1.88± 0.33) folds higher than those of control group.ConclusionsExogenous obestatin has no effects on pancreatic exocrine of acinar cells and lobules of rats in vitro and cannot block or assist the increased amylase release induced by CCK-8 or KCl.

Objective This study was designed to study the distribution and developmental changes of 11?\|hydroxysteroid dehydrogenase 1(11?\|HSD1) in the neonatal rat brain. Methods Immunohistochemistry and Western blot were used to observe the distribution and changes of 11?\|HSD1 protein levels in the neonatal rat brain. Results 11?\|HSD1 protein was highly expressed in all layers of the cerebral cortex as well as all sub\|regions of the hippocampus by immunohistochemistry.Western blot analysis showed that the expression of 11?\|HSD1 protein in the neonatal rat cortex,hippocampus and hypothalamus was higher during the first two weeks of life,but started to fall from 15th day after birth.Conclusion\ The expression pattern of 11?\|HSD1 protein in different brain areas in the neonatal rat suggests that 11?\|HSD1 protein may play an important role in the development and maturation of the brain.\;[

Objective: To investigate the proliferation of three subtypes of odontogenic keratocysts(OKC), namely simple(not recurrent),recurrent and basal cell naevus syndrome(BCNS) associated lesions. Methods: Ki 67 expression was studied in 34 odontogenic keratocysts(simple, n =10;recurrent, n =12;syndrome, n =12)by biotin streptavidin method using Ki 67 monoclonal antibody after microwave treatment. Ki 67 positive cells were counted manually and related to the area of epithelial lining as determined by computer image analyzer. Results: Ki 67 positive cells per mm 2 in simple odontogenic keratocysts ①, recurrent odontogenic keratocysts ② and BCNS ③ were 1 812.29?606.47, 2 393.88 ?997.08 and 3 983.17?858.92 respectively(① or ② vs ③ P 0.05). Conclusion: The basal cell naevus syndrome associated OKC has a higher rate of epithelial proliferation than non syndrome OKC, the increased epithelial proliferation is correlated with increased recurrence potential. Ki 67 may be an alternative method to differentiate syndrome and non syndrome OKC.
