1.Effects of electroacupuncture on hippocampal epileptic burst discharges induced by intracerebroventricular injection of kainic acid in the rat

Epilepsy induced by intracerebroventricular injection of kainic acid in the rat could be attenuated by electroacupuncture(EA)at the "Du Mo point" since EA resulted in a reduction of the frequency and amplitude of the hippocampal EEG especially in the 1st min after EA.No unsynchronization of the hippocampal EEG occurred before or after EA.The hippocampal unit discharges after intracerebroventricular injection of kainic acid were classified into positive units,negative units and indifferent units,The positive units were inhibited by EA while the negative units,on the contrary,were activated.The positive units(21%)and the indifferent units(33%)were converted into negative units by EA.At the same time,EA reduced the NSEB value and IBSF value of hippocampal epileptic burst discharges.On the basis of these findings,it is believed that the inhibition of the amplitude of hippocampal epileptic EEG by EA plays an important role of the antiepileptic action of EA,hippocampal negative units are one of the factors of antiepileptic action,and EA can regulate the intrinsic characteristics of epileptic neurones.
2.Transfection of siRNA into rabbit cervical cells in transformation zone by adopting solid phase in vivo inhibits HPV-DNA replication in SCID mouse with cervical carcinoma

AIM: To investigate the possibility of transfecting siRNA into rabbit cervical cells in transformation zone by the method of solid phase in vivo and to verify the effectivity of siRNA transfection by modifying the permeability of the cervical epithelium. METHODS: A sense strand small-interference RNA (siRNA) for human papillomavirus type 16 (21 bp) was designed and labeled with Cy3. siRNA-Lipo2000-carbomer gum was prepared. Twelve rabbits were included in the study and divided into experimental group and control group. In order to modify the permeability of cervical epithelium, hypertonic saline solution at concentration of 200 mmol/L was used to infuse the cervix in the experimental rabbits for 10 min, and normal saline was used for the control animals. The siRNA-Lipo2000-carbomer gum was applied to the surface of the rabbit cervix. Twenty-four hours later, the rabbits were sacrificed, and the cervix was isolated, cut into 2 parts, one part was for rapid frozen sectioning and the efficiency of transfection was observed under fluorescence microscope, another part was prepared by paraffin embedding and sectioning, and the form of cervical histiocytes was observed. Twelve SCID mice with SiHa cell cervical tumor, divided into experimental group and control group, were also used in the study. The mice in experimental group were treated with siRNA-Lipo2000-carbomer gum for 7 d. The control mice were treated with Lipo2000-carbomer gum only. Five days later, the mice were sacrificed and the tumor was collected, and the HPV16-DNA was measured by PCR. RESULTS: (1) Red fluorescence (Cy3) in cervical epithelium was observed in all rabbits. However, no different effect of siRNA transfection was found between the ways of modifying the cervical epithelium permeability. (2) No abnormal change such as flare, swelling and ulcer at all cervical tissue was observed, the cervical cell form was normal. (3) The titer of HPV16-DNA was decreased significantly after siRNA transfection (P<0.05). CONCLUSION: Transfection of siRNA into rabbit cervical epithelium in vivo is successful by using the method of solid phase and inhibits the processes of HPV-DNA, indicating that using RNAi is a practical way to treat HPV infection in human cercix and to decrease the incidence of cercical carcinoma.
3.Effect and safety of two treatment methods in patients with stable phase of chronic obstructive pulmonary disease

Objective To investigate the effect and safety of Compound codeine phosphate syrup with Ambroxol treatment in the stable phase of chronic obstructive pulmonary disease. Methods 100 patients with stable COPD were divided into Compound codeine phosphate syrup with Ambroxolt treatment group (Group A) 50 cases and Compound codeine phosphate syrup treatment group( Group B) 50 cases,drug were given for 3 ~ 7d. The symptom score and efficacy of two groups before treatment and after treatment were compared. Results The symptom score of cough,sputum volume,sputum viscosity ,wheeze,sleep after treatment 8th day were improved significantly ( χ2 = 3. 891 ,χ2 =3. 992, χ2 = 4. 198, χ2 = 3.981, χ2 = 3. 879, allP < 0. 05; Total effective rate of cough, sputum volume, sputum viscosity,wheeze in group A(92. 0% ,94. 0% ,90. 0% ,90. 0% ) higher than that in group B(80.0% ,86.0% ,80. 0%,78.0% ) ( χ2 = 3.991, χ2 = 4. 012, χ2 = 3. 998, χ2 = 3.971, allP < 0. 05); The adverse reactions of A、 B group were 12.0% and 14.0%(χ2 =1.971,P>0.05). Conclusion Compound codeine phosphate syrup with Ambroxol is an effective and safe antitussive in the stable phase of chronic obstructive pulmonary disease.
4.Determination of Unsymmetrical Dimethylhydrazine in Soil by Gas Chromatography-Mass Spectrometry with the Pretreatment of Alkaline Distillation and Ultrasonic Derivatization

An analytical method was developed for the determination of unsymmetrical dimethylhydrazine ( UDMH ) in soil samples by gas chromatography-mass spectrometry with the pretreatment of alkaline distillation and ultrasonic derivatization. After pretreated by the alkaline distillation, the quantitative analysis of UDMH was conducted in selected ion monitoring ( SIM ) mode by using salicylaldehyde as derivatization reagent and ultrasonic to accelerate the derivative reaction. The characteristic ion of the derivative product named salicylaldehyde dimethylhydrazone was m/z 164 . Impact factors including types of the alkaline distillation and the derivative conditions with ultrasonic were investigated. The conditions of derivatization were optimized. Under the optimal conditions, linear range of the method was 0. 4-30 mg/L. The limit of detection (LOD, S/N=3) was 0. 0078 mg/kg. The method was used to detect total UDMH in real soil samples with known concentration. The recoveries ranged from 76% to 108% with relative standard deviations ( RSD) of 12%-19% at different spiked concentration levels from 10 mg/kg to 100 mg/kg. In comparison with the methods of spectrophotometry and Soxhlet extraction-ultrasonic derivatization, this method had lower detection limit, and could be used to detect total UDMH in soil samples.
5.Glucocorticoid combined with mycophenolate mofetil versus cyclophosphamide in the treatment for adult refractory nephrotic syndrome:a Meta-analysis

Objective To systematically review the efficacy and safety of mycophenolate mofetil versus cyclophosphamide in adults with refractory nephrotic syndrome. Methods The randomized controlled trials of mycophenolate mofetil and cyclophosphamide treatment for refractory nephrotic syndrome were searched from Cochrane library, PubMed, Embase, Wanfang, VIP and CNKI till March 2014. The relevant studies were screened according to inclusion criteria and exclusion criteria. The quality of the included studies was evaluated. Meta-analyses were performed by using RevMan 5.2 software. The indexes were analyzed including the complete remission rate, efficiency, serum albumin, and adverse reaction after completing the treatment for adults with refractory nephrotic syndrome. Results There were 9 RCTs, a total of 467 patients were enrolled. The result of the meta-analysis showed that mycophenolate mofetil could significantly increase complete remission rate (RR=1.45, 95%CI=1.17~1.81, P=0.000 7) and efficiency rate (RR=1.23, 95%CI=1.11~1.36, P<0.000 1). It can also enhance the level of serum albumin (WMD=2.73, 95%CI=1.42~4.04, P<0.000 1) and decrease 24-hour urinary protein (SMD=-0.63, 95%CI=-1.16~-0.10, P=0.02) compared with cyclophosphamide in the treatment of refractory nephrotic syndrome. There was no significant difference in the serum level of cholesterol between mycophenolate mofetil group and cyclophosphamide group (SMD=0.31, 95%CI=-0.23~0.84, P=0.26 ). The incidence rates of liver dysfunction (RR=0.13,95%CI=0.06~0.28, P<0.000 01), leukopenia (RR=0.10, 95%CI=0.04~0.23, P<0.000 01), gastrointestinal reaction (RR=0.21, 95%CI=0.11~0.39, P<0.000 01) and alopecia (RR=0.08, 95%CI=0.02~0.29, P<0.000 01) were significantly lower in mycophenolate mofetil group than those of cyclophosphamide group. There were no significant differences in respiratory tract infection rate (RR=0.68, 95%CI=0.41~1.14, P=0.14) and lung infection rate (RR=0.58, 95%CI=0.31~1.08, P=0.09) between the two groups. Conclusion The safety and efficacy are better in the treatment of refractory nephrotic syndrome using mycophenolate mofetil than that of cyclophosphamide.
6.Expression of Notch1, Jagged1 and NICD in epithelial ovarian carcinomas and a preliminary study on the activity of gamma-secretase in epithelial ovarian carcinoma cell lines

Objective To study the expression of Notch 1,Jagged1 and Notch intracellular domain (NICD) in epithelial ovarian carcinoma tissues and analyze the clinical significance.To explore the activity of γ-secretase in epithelial ovarian carcinoma cell line SKOV3 and the effect of N-[N-(3,5-dil uorophenacetyl)-L-alanyl]-S-phenylglycine t-butyl ester (DAPT),a γ-secretase inhibitor on the activity of γ-secretase in SKOV3.Methods Immunohistochemistry staining method was performed in 43 patients with epithelial ovarian carcinoma and 11 patients with benign epithelial ovarian tumor to detect the expression of Notch1,Jagged1 and NICD.The differences of expressionof Notch1,Jagged1 and NICD between malignant and benign ovarian tumors was compared and alsoanalyzed the correlation with clinicopathological parameters of ovarian carcinoma.Human serous ovarian cancer cell line SKOV3 and immortalized nontumorigenic ovarian epithelial cell line T29 were incubated in vitro.The activities of γ-secretase in SKOV3 and T29 with dimethyl sulfoxide (DMSO) and DAPT were detected respectively by Gal4VP16/UAS and dual luciferase reporter assay system.Results (1) The immunohistochemical composite scores (ICS) of Notch1 in epithelial ovarian carcinoma (6.7±2.2) were not significantly different with those in benign epithelial ovarian tumor (5.4± 2.7,P=0.153),while the ICS of Jagged 1 and NICD in epithelial ovarian carcinoma (5.3± 2.4,5.3± 2.3) were higher than those in benign epithelial ovarian tumor (1.6± 1.4,3.1± 1.7; all P<0.01).The expression of Notch 1,Jagged 1 and N ICD had no correlation with patients' aged,history of carcinoma,ascites,the level of serum CA125,maximum length of ovarian tumor,Federation International of Gynecology and Obstetrics (FIGO) stage,grade and pathology subtypes (all P>0.05).The hazard ratio between the high expression of Notch1,Jagged1,or NICD and the moderate to low expression of Notch1,Jagged1,or NICD,and Jagged1 were 0.771,1.648 and 1.316,respectively (all P>0.05).The 5-year survival rate and median survival time between the high expression of Notch,Jagged 1 or NICD in subgroup and moderate to low expression in subgroup were of no difference (all P>0.05).The activity of γ-secretase in SKOV3 was significantly higher than that in T29 [(12.2± 1.4)%,P=0.019].(2)After DAPT treated,the relative activity of γ-secretase in SKOV3 (50 μmol/L) was declined from (100.0±5.3)% to (6.6±0.8)% (P=0.001).Conclusions Jagged1 and NICD in Notch1 pathway may play a key role in the occurrence of ovarian carcinoma.The activity of γ-secretase in epithelial ovarian carcinoma was higher than that in ovarian epithelial cell which suggest that DAPT,γ-secretase inhibitor,may become the target of ovarian carcinoma treatment.
7.An experiment study on myocardial regeneration with implantation of bone marrow cells into infarct myocardium

Objective: To study the effect of bone marrow cells on myocardial regeneration. Methods: Bone marrow cells were implanted into the acute myocardial infarct via topical injection. Specimens were harvested at 1st, 2nd, 4th, 8th weeks after bone marrow cells implantation for morphologic examination. Results: Bone marrow cells with fluorescence had been observed in the implanted site. A portion of the implanted cells had differentiated into myogenic cells. The regenerating myocardial cells observed by electron micrography had been connected with the host myocardium through intercalated discs. The infarct size was significantly smaller in implant group. Conclusion: This study suggested that bone marrow cells are capable to differentiate and regenerate into myocardial cells in the acute myocardial infarct sites and reduce size of infarct.

Breast cancer is one of the most common malignant tumours in women. It threatens the health and life of women. The imaging technology of breast electrical impedance scanning based on the dielectric properties of breast tissues develops a new aspect of early detection and auxiliary diagnosis of the breast cancer. The dielectric properties of human breast tissues reflect the pathological and physiological state of breast tissues and the measurement of these properties serves as the basis for electrical impedance scanning(EIS) technology. In this paper, the measurements and results of dielectric properties of human breast tissues are discussed, and the problems of measurement and the application perspective of the EIS are also explored.
