1.Expression and function of cyclooxygenase-2 in endometrial carcinoma

Objective: To evaluate the cyclooxygenase-2 ( COX-2) expression at mRNA and protein levels , as well as its clinical significance, and to study the correlation of COX-2 with angiogenesis, apoptosis and estrogen or progestron receptors. Methods: Forty-one samples of normal endoraetrium tissues and 52 ones of endometrial carcinomas( EC) were collected,together with their corresponding clinical information. RT-PCR was adopted to determine the expression of COX-2 mRNA. Immunohistochemical staining was engaged to evaluate the protein expression of COX-2, VEGF, Bcl-2 and the MVD marked by CD105. And TUNEL was used to count the apoptosis cells. Results: (1) Significantly higher mRNA and protein levels of COX-2 were found in endometrial carcinomas than in normal endometrial tissues ( P<0.001) . (2) At protein level ,COX-2 expression was related to the grade(P =0.020) , but had nothing to do with the stage, pathological type, depth of myometrium invasion and lymph node metastasis. Neither did the level of COX-2 mRNA with all the factors above. (3)The level of COX-2 mRAN had nothing to do with that of protein(P =0. 125, r =0. 222). (4) The higher the expression of COX-2 protein was, the higher that of VEGF,MVD or Bcl-2 was,and the lower the count of apoptosis cells was P<0.05. (6)The upregulation of COX-2 protein had no correlation with ER-α and ER-β . But it had positive correlation with PRA and PRB,P was 0.031 and 0.007 .respectively. Conclusion: (1)The expression of COX-2 is significantly higher in EC than in normal endometrium tissue, not only at the mRNA level, but also at protein level. ( 2 ) At protein level, COX-2 expression is related to the grade, but has nothing to do with the stage, pathological type, depth of myometrium invasion and lymph node metastasis. Neither does the level of COX-2 mRNA with all the factors above. (3)The upregulation of COX-2 protein in EC correlates with the increase of VEGF protein and MVD. (4) The upregulation of COX-2 protein in EC correlates with the increase of Bcl-2 protein and the decrease of apoptosis cells. (5)The upregulation of COX-2 protein in EC has no significant correlation with ER expression, but has something to do with the increase of PRA and PRB.

4.Analysis of myelogram and clinical pathogenesis of 196 cases of patients with pancytopenia syndrom

Objective To explore the etiological spectrum of pancytopenia ,in order to improve the diagnostic accuracy . Methods Retrospectively analysed myelogram and clinical data of 196 cases of patients with pancytopenia syndrom .Results The dominant cause of pancytopenia syndrom in 196 cases of patients was hematological diseases (accounted for 68 .9% ) ,including acute leukemia (14 .8% ) ,myelodysplastic syndrome (12 .2% ) ,aplastic anemia(11 .2% ) and immune‐related pancytopenia(10 .2% );while non‐hematologic diseases accounted for 31 .1% ,including connective tissue diseases (10 .7% ) ,chronic liver disease(7 .2% ) and in‐fection(6 .2% ) .Conclusion Etiology of pancytopenia syndrom is complex ,which should be comprehensively analysed with closely contacting clinics ,in order to clarify the cause ,reduce misdiagnosis and missed diagnosis and improve the diagnostic accuracy .

6.Synovial mesenchymal stem cells for knee cartilage injury

BACKGROUND:Knee cartilage injury is difficult to heal, which is an urgent clinical problem to solve. OBJECTIVE:To summarize the advantages of synovial mesenchymal stem cel s in the treatment of knee cartilage injury. METHODS:A computer-based search of PubMed was performed for articles related to synovial mesenchymal stem cel s for knee cartilage injury using the keywords of“knee joint, cartilage, synovial membrane, tissues, injuries and repair”. A total of 48 articles were retrieved initial y, and 35 articles were included in result analysis. RESULTS AND CONCLUSION:Synovial mesenchymal stem cel s for treatment of knee cartilage injury can achieve more ideal outcomes. Mesenchymal stem cel s inherent in the damaged tissue are the optimal seed cel s for tissue repair.
7.Progress in research on relationship between microRNA-7 and lung cancer

MicroRNA-7(miR-7)was firstly found in Drosophila melanogaster,which participated in the formation of Drosophila wings,eggs and so on.In human being miR-7 is not only involved in cell prolifera-tion and differentiation,but also plays an important role in the development of tumor,especially in lung cancer. Most studies report that miR-7 is low expression in lung cancer with poor prognosis.Improving the expression of miR-7 can inhibit tumor growth.MiR-7 can regulate the expression of BCL-2,EGFR negatively,meanwhile it can regulate the sensitivity of tumor cells to radiotherapy chemotherapy and targeted therapy.Therefore,as a tumor suppressor,miR-7 is expected to become a new target for treatment of lung cancer.
8.THE CLINICAL SIGNIFICANCE OF ANTI-CCP ANTIBODY IN PATIENTS WITH RHEUMA-TOID ARTHRITIS

Objective To investigate the clinical application value of Anti -CCP antibody ( CCP) levels in patients with in rheumatoid arthritis ( RA) .Methods 132 patients with rheumatoid arthritis , 42 patients of other categories of connective tissue dis-ease, 40 health cases were collected as the research object.The anti-CCP antibody titers were detected in sera of three patients, and the anti CCP antibody in rheumatoid arthritis in the diagnostic process was calculated .Anti -CCP antibody titers were detected in three group patients, and the specific of anti CCP antibody in diagnosis of rheumatoid arthritis was counted .Results Anti-CCP an-tibody has high specificity for the diagnosis of rheumatoid arthritis , and was correlated with clinical symptoms in patients with rheuma-toid arthritis.Combination detection of anti -CCP antibody and C reaction protein can improve the diagnostic specificity , and the difference was statistically significant (p<0.05).Conclusion The specificity of anti-CCP antibody in diagnosis of rheumatoid ar-thritis is higher and its specificity is higher than that of RF .The combined detection of anti -CCP antibody and RF both can improve the diagnostic specificity of rheumatoid arthritis , and anti-CCP antibodies are related with the degree of clinical symptoms .
9.Exploration on value of CD64 and procalcitonin in early diagnosis of pediatric bacterial diarrhea

Objective To detect peripheral blood neutrophil CD64 and procalcitonin(PCT)in children with diarrhea ,and to ex‐plore significance for differential diagnosis of diarrhea in children .Methods A total of 180 cases of children with diarrhea treated in this hospital from July 2012 to May 2014 were selected as subjects ,and were classified into three groups :bacterial diarrhea group (n=60) ,viral diarrhea group(n=60)and non infectious diarrhea group(n=60) .The levels of peripheral blood neutrophil CD64 and PCT were determined in all three groups ,and the diagnostic values of CD64 index and PCT were evaluated .Results The peripheral blood neutrophil CD64 index and level of PCT in the bacterial diarrhea group were significantly higher than those in the viral diar‐rhea group and non infectious diarrhea group ,and the positive rates of CD64 index and PCT in the bacterial diarrhea group were sig‐nificantly higher than those in the viral diarrhea group and non infectious diarrhea group ,all had statistically significant differences (P<0 .05) .The specificity and positive predictive value of CD 64 index in diagnosing bacterial diarrhea were significantly higher than those of PCT ,both had statistically significant differences (P<0 .05) .The relative analysis showed that the CD64 index was positively correlated with level of PCT (r=0 .865 ,P<0 .05) .Conclusion The CD64 index and PCT level of children with bacterial diarrhea increase significantly ,which indicates that the detection of CD64 index and PCT have significant clinical value in early diag‐nosis of bacterial diarrhea .
10.Regulation of endometrial stromal cells through the Wnt/beta-catenin signaling pathway

BACKGROUND:Increasing number of genes and signaling pathways are involved in regulating stem cels periodicaly, and wherein Wnt/β-catenin signaling pathway is an important pathway of stem cels.
OBJECTIVE:To investigate the effects of Wnt/β-catenin signal pathway on mouse endometrial stromal cels.
METHODS:After injection of Wnt/β-catenin pathway inhibitor or activator, endometrial tissues from Balb/c mice were obtained, some of which were used for detection of invasion of endometrial stromal cels and western blot detection, and the rest of which were used for preparing animal models of endometriosis folowed by immunohistochemical detection.
RESULTS AND CONCLUSION:The expression of β-catenin and GSK3β proteins was significantly higher in the activator group than the inhibitor group (P < 0.05). The number of transmembrane cels was significantly higher in the activator group than the inhibitor and control groups (P < 0.05). Immunohistochemical findings showed positive expression of E-cadherin in ectopic endometrial tissues of the inhibitory group, and strongly positive expression of vascular endothelial growth factor in ectopic endometrial tissues of the activator group. These findings indicate that Wnt/β-catenin signaling pathway may cause endometriosis by strengthening ectopic endometrial implantation, invasion and metastasis.
