Objective To discuss the clinical technique and outcomes of the treatment with arthroscopically assisted percutaneous retrograde interlocking nailing for supracondylar femoral fractures. Methods From July 2001 to June 2003, there were 22 supracondylar femoral fractures which were 19 fresh fractures and 3 old fractures. There were 10 males and 12 females, and the average age of which was 39.5 years ranging from 22 to 65 years. 7 patients sustained falls, and the other 15 patients were caused by traffic accidents. According to the AO/ASIF classification system, 7 fractures were evaluated as type A1, 8 type A2 and 7 type A3. All the patients were treated by arthroscopically assisted percutaneous retrograde interlocking nailing. Some additional lesions were also treated under arthroscopy simultaneously, including 3 releases for the stiff knee, 2 removals of the loose bodies, 7 debridements and lavages for the osteoarthritis, and 3 bone grafts for the fracture union. Results The average operative time was 130 minutes ranging from 90 to 210 minutes, and the average blood loss was 100 ml. The reduction and fixation were proved to be satisfactory by the postoperative roentgenogram. 20 patients were available for follow-up, which ranged from 6 to 24 months with an average of 14 months. All the fractures were united on an average of 3.3 months postoperatively except one ranging from 2 to 6 months. The one with fracture union were reoperated with bone grafting and compressive plate fixation, and the fracture healed 3 months later. The range of motion was beyond 90? in all patients, no breakage of internal fixation instruments or wound infection occurred. The functional results of knee joint were assessed as excellent in 14, good in 5 and fair in 1, and the rate of excellent and good was 95%. Conclusion The percutaneous retrograde interlocking nailing for the supracondylar femoral fractures is proved to have many advantages, such as less trauma and blood loss, and accurate location, which are beneficial to both of the fracture union and functional recovery.

Objective To verify the ability of distal ends of internal mammary artery and vein used as recipient vessels on breast and chest wall reconstruction. Methods Four bilateral (left 2 and right 2) scent pig buttock island flaps pedicled circle deep iliac artery and vein were used by comparing the different hemodynamic parameters (pressure; flux volume; perfusion unions) between the original pedicl artery (deep iliac artery), proximal and distal ends of internal mammary artery. The free flaps were transplantated by anastomosed end-end to the distal-ends of internal mammary artery and vein after the proximal ends were ligated. Results The pressure of distal end of internal mammary artery was 61%-65% of the pressure with original pedicl artery (deep iliac artery), the immediate volume of anastomasis stoma was a little lower than that of original pedicl artery. The flap, 15 cm?30 cm?2 cm in size, completely survived more than 14 days after operation. Conclusion The slightly decreased distal arterial pressure does not compromise flap survival. Bilateral scent pig buttock island flap pedicled circle deep iliac artery and vein is an ideal animal flap model. This hymodynamic model can be widely used .

ObjectiveTo observe the difference in analgesia effects of ropivacaine with fentanyl used intraarticularly after the single knee arthroscopy procedure.Methods40 patients performed knee arthroscopy under combined spinal-epidural anaesthesia( CSEA),were randomly divided into 4 groups (n =10),at the end of operation 10mlof different drug at the group F,R,FR and N(fentanyl 10μg,0.5％ ropivacaine,fentanyl 10μg +0.5％ ropivacaine,normal saline)were injected intra-articularly.The antalgic effects of four groups based on standard of VAS were observed at the 2,4,8,12 and 24h after operation.ResultsThe 2h postoperative VAS scores were lower in four groups,the differences in four groups were not significant.The 4,8,12,24h postoperative VAS scores of F,R and FR group were much lower than that of N group ( all P ＜ 0.05 ).Moreover,VAS scores of FR group were much lower than that of F and R group( all P ＜ 0.05 ).No other adverse effects were observed.ConclusionIntra-articular administration of ropivacaine with fentanyl could provide superior postoperative analgesia without side effects.It was an excellent regimen for analgesia after knee arthroscopy.

This study assessed the effects of leukemia-related protein 16 (LRP16) on the regulation of pancreatic functions in mouse insulinoma (MIN6) cells. Cells with down-regulated expression of LRP16 were obtained by a shRNA interference strategy. Insulin content and glucose-stimulated insulin secretion (GSIS) were examined by radioimmunoassay. Western blotting was applied to detect protein expression. Glucose-stimulated sub-cellular localization of PDX-1 was immunocytochemically determined. Cell proliferation and apoptosis were detected by flow cytometry. Our results showed that LRP16 regulated insulin content in MIN6 cells by controlling expression of insulin and insulin transcription factors. LRP16 gene silence in MIN6 cells led to reduced cell proliferation and increased apoptosis. The observation of phosphorylation of serine-473 Akt and the localization of PDX-1 to the nucleus under glucose-stimulation exhibited that LRP16 was a component mediating Akt signaling in MIN6 cells. These results suggest that LRP16 plays a key role in maintaining pancreatic β-cell functions and may help us to understand the protective effects of estrogen on the functions of pancreatic β-cells.

BACKGROUNDTo identify the relationship of cervical cancer with pathogen infectious, cytokine and Se.

METHODSOn the one hand regarded tissues of the carcinoma of the uterine cervix with 195 cases as the experimental group and ordinary cervical tissues with 75 cases as the control group. Polymerase chain reaction (PCR) to detect them. On the other hand applied ELISA to detect cytokine IL-2R, TNF and fluorescent luminosity technique to detect element Se in the serums.

RESULTSIn the experimental group those infected pathogen were 166 cases (85.1%), In all of pathogen HPV 16,18,35 type were 60 cases (30.8%), HSV 2 were 60 cases (30.8%), While those ordinary tissues infected pathogen were 15 cases (20 0%),in the contrast group. HPV 16,18,35 type and HSV 2 mere 4.0% and 6.7% respectively,(P<0.001). In the serums of effective 62 experimental objects IL-2R (x=356.44 U/ml) and TNF (x=373.48 pg/ml) were much high than them in the serums of effective 36 contrast objects (P<0.001). But Se (x=0.058 mg/ml) was lower than it in the serums of contrast objects (P<0.05).

CONCLUSIONSThe occurrence of carcinoma of the uterine cervix is closely connected with infection of HPV 16,18,35 and HSV2, high level of cellular factors IL-2R, TNF and low level of element Se.

In order to identify rat ovarian germ cells, we expressed and purified rat RVLG protein in Escherichia coli cells and prepared a mouse anti-rat RVLG polyclonal antibody. The rat RVLG cDNA was obtained from rat testicle tissue by RT-PCR and was cloned into the vector pMD19-T. Sequence analysis proves that the cloned RVLG cDNA fragment was 60 bp longer than that released in the GenBank (NM_001077647), resulting from an alternative splicing of the RVLG pre-mRNA. The RVLG cDNA was double digested with the restriction endonucleases BamH I and EcoR I, and then was extracted from gel and inserted into the prokaryotic expression vector pGEX-4T-1. The recombinant expression plasmid pGEX-RVLG was verified for successful construction and then was transformed into Escherichia coli BL21(DE3) for induction to express the GST-RVLG fusion protein by IPTG. The GST-RVLG fusion protein was expressed in Escherichia coli BL21 (DE3) at a high level which accounts for more than 10% of the total bacterial cellular protein. The purified RVLG protein was used as an antigen to immunize KM mouse for the production of polyclonal antibody in ascetic fluid followed by celiacly injecting the mouse with S180 cells. The mouse anti-rat RVLG antibody was analyzed by ELISA, Western blotting and immunohistochemistry for its specificity and titer. The antibody could recognize RVLG protein specifically and its titer was about 1:20 000. These results confirm that the mouse anti-rat RVLG polyclonal antibody with high affinity and specificity has been prepared successfully, and lay a foundation for our ongoing research on the specific expression of RVLG in rat ovary.

Objective:To analyze the clinical characteristics of monkeypox patients.Methods:The clinical data and laboratory findings of 4 patients with monkeypox patients diagnosed at Yiwu Central Hospital in July 2023 were analyzed. Herpes fluid and skin tissue samples were collected, the viruses were isolation and cultured in African green monkey kidney cells (Vero) and identified with whole gene sequencing.Results:All four patients were male, aged 24-35 years. All patients had male-to-male behavior within 21 days before onset of the disease. Among them, one patient has AIDS and one patient has syphilis. Four patients presented with perineal skin lesions with itching, and 3 patients were found to have enlarged lymph nodes upon admission. Laboratory testing: lymphocyte abnormality (4.57×10 9/L) in 1 case; increased procalcitonin (0.25 ng/mL) in 1 case; elevated IL-10 levels ( 7.11 ng/L and 9.42 ng/L) in 2 cases; increased IL-6 (66 ng/L) and IL-4 (3.24 ng/L) in 1 case, respectively. One case had abnormal myocardial zymogram with a elevated lactate dehydrogenase level of 313 U/L. The monkeypox virus was isolated from lesion tissue and herpes fluid, and the whole gene sequencing identified it as the B. 1.3 subtype of the IIb evolutionary branch, exhibiting typical pathological effects on Vero cells. Conclusion:The clinical manifestations of the 4 monkeypox patients confirmed in Zhejiang province are mild, patients had a definitive history of male-to-male sexual behavior and the virus strains belong to the B. 1.3 lineage of the IIb evolutionary branch.

A high throughput measurement method of human red blood cells (RBCs) deformability combined with optical tweezers technology and the microfluidic chip was proposed to accurately characterize the deformability of RBCs statistically. Firstly, the effective stretching deformation of RBCs was realized by the interaction of photo-trapping force and fluid viscous resistance. In the current study, we aim to investigate whether post-traumatic stress disorder (PTSD) is associated with structural alterations in specific subfields of hippocampus comparing with trauma-exposed control (TC) in a relatively large sample. We included 67 PTSD patients who were diagnosed under Diagnostic and Statistical Manual of Mental Disorders (4th Edition) (DSM-Ⅳ) criteria and 78 age- and sex-matched non-PTSD adult survivors who experienced similar stressors. High resolution T1 weighted images were obtained via a GE 3.0 T scanner. The structural data was automatically segmented using FreeSurfer software, and volume of whole hippocampus and subfield including CA1, CA2-3, CA4-DG, fimbria, presubiculum, subiculum and fissure were extracted. Volume differences between the two groups were statistically compared with age, years of education, duration from the events and intracranial volume (ICV) as covariates. Hemisphere, sex and diagnosis were entered as fixed factors. Relationship between morphometric measurements with Clinician-Administered PTSD Scale (CAPS) score and illness duration were performed using Pearson's correlation with SPSS. Comparing to TC, PTSD patients showed no statistically significant alteration in volumes of the whole hippocampus and all the subfields ( > 0.05). In male patients, there were significant correlations between CAPS score and volume of right CA2-3 ( = 0.197, = 0.034), right subiculum ( = 0.245, = 0.016), and duration statistically correlated with right fissure ( = 0.247, = 0.016). In female patients, CAPS scores significant correlated with volume of left presubiculum ( = 0.095, = 0.042), left subiculum ( = 0.090, = 0.048), and left CA4-DG ( = 0.099, = 0.037). The main findings of the current study suggest that stress event causes non-selective damage to hippocampus in both PTSD patients and TC, and gender-specific lateralization may underlie PTSD pathology.
