1.Application of the systematic education platform for patient with permanent colostomy in improving self-care ability

Objective To evaluate the effect of the systematic education platform for patient with colostomy in improving self‐care ability .Methods A total of 198 colostomy patients from a Third‐Grade Class A Hospitals were randomly assigned to ei‐ther learn colostomy nursing knowledge by the systematic education platform or routine health education condition .The patients were investigated by a questionnaire about Exercise of Self‐Care Agency Scale ,ESCA before discharge ,1 month and 3 months after discharge .Results After the systematic education platform for 3 month ,the total score(120 .87 ± 10 .11) and each factor scores of ESCA scale[self‐protection skills(30 .51 ± 2 .57) ,self nursing responsibility(23 .95 ± 2 .64) ,self concept(26 .23 ± 1 .69) ,health knowledge level (39 .78 ± 3 .12 )] were all significantly improved after the systematic education platform ( P < 0 .05 ) . Conclusion The systematic education platform can improve ostomy patients′self‐care ability and quality of life .
2.Feasibility of regional platform for wound care technical training

Objective To study the feasibility of regional network platform for wound care technical training.Methods Micro-letter pubic number and internet technologies were used to develop a regional network platform of wound care which has one center with multiple branches,training nurses how to take care of wound by the educational column.Results There were 383 nurses had browsed the web site and took part in the learning since regional network platform was working 3 months,931 nurses paid close at-tention to Micro-letter pubic number.Performance examination was improved obviously after training,the percent of pass was 17.8% before training and 80.7% after training,the difference was with statistical significance (χ2=303.629,P<0.01).Conclu-sion The regional network platform of wound care based on micro-letter and internet technologies has a promising application.

Objective To detect the serum antibody to HPV6 in patients with condylomata acuminata (CA) by use of human papilloma virus(HPV)6bL 1 virus-like particles (VLPS), and to assess the significance of detecting HPV infection. Methods HPV6bL 1 VLPS were produced by a HPV6bL 1 recombinant baculovirus in insect cells,purified by CsCl density gradients centrifugation,and used for ELISA to screen 136 human serum samples in three groups. Results The prevalence of antibody to HPV6bL 1 VLPs was much higher in patients with codylomata acuminata (75%Ab+;mean abs 0.111?0.094) than that in patients with cervical cancer (14.3%Ab+;mean Abs 0.029?0.022)and that in control groups (2.9%Ab+;mean Abs 0.012?0.024). OBJECTIVE:To prepare Levofloxacin and triamcinolone acetonide double-loaded ophthalmic gel. METHODS:Us-ing levofloxacin hydrochloride and triamcinolone acetonide as main components,carbopol-940P as base material,HPMC K4M as tackifier,Levofloxacin and triamcinolone acetonide double-loaded ophthalmic gel was prepared. Using dissolution time as index, the contents of carbopol-940P and HPMC K4M were determined by single factor test,and dissolution time,viscosity and the con-tents of 2 main components were also determined. RESULTS:The concentrations of carbopol-940P and HPMC K4M were 0.4%and 1.2%,separately. The dissolution time was more than 24 h and viscosity was 1 142.67 Pa·s. The content of levofloxacin hydro-chloride was 97.3% of labelled amount (RSD=0.84%,n=3),and that of triamcinolone acetonide was 92.97% of labelled amount(RSD=1.32%,n=3). CONCLUSIONS:Levofloxacin and triamcinolone acetonide double-loaded ophthalmic gel has been prepared successfully.
6.Up-regulation effect of cyclosporine A on level of kidney inj ury molecule-1 in culture supernatant of human kidney cells and its mechanism

Objective To investigate the mechanism of increasing of the level of kidney injury molecule-1(KIM-1)in culture supernatant of human kidney cells(HKC)induced by cyclosporine A(CsA),and to clarify the relationships between the expression levels of KIM-1 and p38 MAPK pathway and ERK1/2MAPK pathway in HKC. Methods The HKC at logarithmic growth phase were randomly divided into control group, CsA control group, CsA + p38 kinase inhibitor group, p38 kinase inhibitor group, CsA + ERK1/2 inhibitor group and ERK1/2 kinase inhibitor group.The inhibitory rates of proliferation of HKC in various groups were detected by MTT assay, and the expression levels of KIM-1 in HKC supernatant in various groups were detected by ELISA;the survival rates,apopototic rates and necrotic rates of the HKC in various groups were detected by flow cytometry. Results Compared with control group,the expression level of KIM-1 protein in the supernatant of HKC in CsA control group was significantly increased (P<0.05),and the survival rate was significantly decreased (P<0.05), while the apoptotic rate and the necrotic rate were significantly increased (P<0.05 ). Compared with control group,the survival rates, the apoptotic rates and the necrosis rates of cells in p38 kinase inhibitor group and ERK1/2 kinase inhibitor group had no significant differences(P>0.05).Compared with CsA control group,the expression levels of KIM-1 protein in CsA+ p38 kinase inhibitor group and CsA+ ERK1/2 kinase inhibitor group were significantly decreased (P<0.05),and the survival rate was significantly increased (P<0.05),while the apoptotic rate and the necrotic rate were significantly decreased (P<0.05).Conclusion p38 MAPK pathway and ERK1/2MAPK pathway are involved in the process of up-regulation of the KIM-1 level in HKC culture supernatant induced by CsA,and the expression of KIM-1 may become the biochemical marker of clinical monitoring of CsA nephrotoxicity.
7.Function and mechanism of TRIM22 targeting eIF4E in the process of NB4 cells differentiation

Objective To investigate the function of tripartite motif protein 22 (TRIM22) and the interaction with eukaryotic translation initiation factor-4E (eIF4E) in the differentiation of NB4 cells, one kind of acute promyelocytic leukemia cells, which elucidates the mechanism of TRIM22 targeting to regulate eIF4E.Methods The model of NB4 cells inducing differentiation was established in vitro.The expression changes of gene and protein of TRIM22 and eIF4E were detected by using quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting.In addition, the effect on cell function and protein expression level of eIF4E after adopting electroporation technology to depress or over-express TRIM22 was detected by CCK-8 and flow cytometry.Finally, the interaction of TRIM22 and eIF4E was verified by using co-immunoprecipitation (CO-IP).Results The mRNA relative expression level of TRIM22 was gradually increasing from 1.01±0.15 to 30.98±2.79 (F=280.700, P=0.000), and the protein relative expression level was gradually increasing from 0.22±0.03 to 0.51±0.05 (F=51.430, P=0.000) after the all-trans-retinoic acid (ATRA) induction for NB4 cell.However, the mRNA relative expression level of eIF4E was gradually decreasing from 1.01±0.09 to 0.47±0.06 (F=20.520, P=0.000), with the same trend, the protein relative expression level was gradually decreasing from 0.97±0.02 to 0.64±0.09 (F=14.700, P=0.001).The expression level of PE-CD11b in the TRIM22 over-expression group with ATRA detected by flow cytometry [(78.80±2.00)%] was higher than that in the transfection group of empty vetor with ATRA [(58.70±2.70)%] (t=9.535, P=0.000) and the cotransfection group with ATRA [(61.60±3.80)%] (t=8.187, P=0.000).Meanwhile, the protein level of eIF4E changed reversely after over-expressing the gene level of TRIM22 (t=4.985, P=0.007).The CO-IP experiment was used to verify the interaction of TRIM22 and eIF4E.ConclusionTRIM22 is able to promote the cell differentiation during the process of NB4 cells differentiation.Furthermore, eIF4E is a target of TRIM22 for binding with, which plays an important role in depressing the expression of eIF4E.
8.Effects of Qigong Pills Formula on Uterine Artery Blood Flow and Endometrial Receptivity of Patients with Polycystic Ovary Syndrome with Infertility

OBJECTIVE:To investigate the effects of Qigong pills formula on uterine artery blood flow and endometrial recep-tivity of patients with polycystic ovary syndrome(PCOS)with infertility. METHODS:A total of 86 PCOS patients with infertility in our hospital during Jan.-Dec. 2016 were divided into control group and observation group according to random number table, with 43 cases in each group. Control group was given western medicine pituitary down-regulation therapy. Observation group was additionally given adjuvant therapy of Qigong pills formula,a dose per day,decocted with water to 400 mL,morning and night un-til artificial fertilization stopped,on the basis of control group. The uterus ultrasonic indexes and endometrial receptivity were com-pared between 2 groups before treatment and 3 months after treatment. The occurrence of ovulation,pregnancy and ADR was re-corded. RESULTS:Before treatment,there was no statistical significance in uterus ultrasonic indexes and endometrial receptivity in-dexes between 2 groups (P>0.05). Three months after treatment,endometrial thickness of 2 groups were increased significantly, while endometrial spiral arterial pulsatility indexes and resistance indexes were decreased significantly;the levels ofαvβ3 and GLUT4 were increased significantly;observation group was significantly better than control group,with statistical significance (P<0.05). The ovulation rate of observation group was 83.72%,which was significantly higher than 62.79% of control group,with statistical significance(P<0.05). There was no statistical significance in pregnancy rate(27.91% vs. 18.60%)between 2 groups(P>0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:The adjuvant therapy of Qigong pills formula help to regulate the pa-rameters of uterine artery blood flow in PCOS patients with infertility,and strengthen endometrial receptivity so as to increase ovu-lation rate.
9. Rapid construction of stable cell lines which inducible express HA proteins from various influenza virus strains

Objective:
An innovative technique was established to rapidly construct various cell lines that could be induced to express multiple influenza A virus (IAV) proteins.
Method:
The HA protein genes of multiple IAVs were cloned into the Cumate-induced expression system which was positioned between two PiggyBac transposon sites. These HA plasmids were transfected into the HEK293A cell line with the PiggyBac transposase plasmids. The transfected cells were screened with puromycin, and after that the corresponding virus proteins were induced with Cumate.
Results:
The results of flow cytometry and Western blotting showed that the virus proteins were expressed in most of the cells in corresponding lines after the induction of Cumate.
Conclusion
Cell lines which were inducible to express IVA HA protein can be efficiently constructed by using the PiggyBac transposon system.
10.Wristband pedometer-measured activity and its determinants among middle school students

Objective:
To improve physical activity by assessing pedometer-based physical activity during specific intervals over a week, among junior high school students,and to provide a reference for improving the level of students’ physical activity.
Methods:
Students (n=675) of grades 6-8 from six public junior high schools in Shanghai were recruited and instructed to wear a pedometer for a week, step counts and contents were recorded over a specific period. This period mainly included steps taken on the way to and from school, at school, at home in the evening on weekdays, and in the morning, afternoon, and evening on weekends.
Results:
The daily step counts recorded were 8 332 steps, with those on weekdays showing significantly higher values than step counts on weekends (9 065 steps vs. 6 392 steps)(t=22.9, P<0.01). Proportionately, the physical activity level at school contributed more to daily step counts (61.3%), followed by those on the commute to and from school(25.5%). Boys were more active than girls. For all intervals on weekdays, the step counts of students in rural districts, with overweight or obese, in grades 6 and 7, were higher than those in urban districts, with normal status, in grade 8 respectively (P<0.05). Low-activity students with physical education were more active than one without physical education(t boy=1.99,t girl=2.45,P<0.05).
Conclusion
These findings facilitate the implementation of effective, feasible interventions to enhance physical activity over a series of intervals during the day.
