1.Clinical effect of ethinyl estradiol cyproterone tablets combined with ovulation induction in the treatment of polycystic ovary syndrome infertility

Objective To explore the clinical effect of ethinyl estradiol cyproterone tablets ( Diane -35 ) combined with ovulation induction in the treatment of polycystic ovary syndrome infertility .Methods 70 patients with polycystic ovary syndrome were enrolled , and they were randomly divided into two groups according to the random number table of the computer ,35 cases in each group .The control group was treated with Diane -35,the observation group was given ovulation induction .The clinical parameters ,vaginal ultrasound results ,cycle ovulation rate ,pregnancy rate,adverse reaction rate and quality of life were compared between the two groups .Results There were no statistically significant differences in uterine volume , intima thickness , bilateral ovarian volume and bilateral follicle number between the two groups after treatment (t=0.055,0.308,0.991,0.435,0.473,0.248,all P>0.05).The cycle ovulation rate of the observation group was significantly higher than that of the control group (80.88% vs. 54.05%,χ2 =8.451,P<0.01).The shape,color,thought and energy score of the observation group were significantly higher than those of the control group (t=4.819,3.519,4.735,6.804,all P<0.01).The pregnancy rate of the observation group was significantly higher than that of the control group (25.71%vs.5.71%,χ2 =5.285,P<0.05). Conclusion Diane-35 combined with ovulation induction in the treatment of polycystic ovary syndrome infertility has significant clinical efficacy .
2.Comparative research on pharmacognostic characteristics and microscopic characteristics of Radix Paeoniae Rubra from different areas.

OBJECTIVETo compare pharmacognostic characteristics and microscopic characteristics of Radix Paeoniae Rubra (chishao) from different areas.
METHODPharmacognostic characteristics and microscopic characteristics of Radix Paeoniae Rubra were compared by microscope count methods.
RESULTChishao in duolun was more straighter and longer, cortex with a set of closely spaced rill, peel off easily, pink section, etc. The wild chishao were different from the cultivated chishao on pharmacognostic characteristics and microscopic characteristics, such as appearance shape, smell, vessel arrangement, and number of crystal and starch in unit area.
CONCLUSIONChishao in duolun were different form others, appearance shape, wood fiber, difference of appearance shape, vessel arrangement, and number of crystal and starch in unit area can be used as identificatin feature of the wild chishao and the cultivated chishao.

3.Optimization of preparation of rat kidney decellularized scaffold by combining freeze-thawing with perfusion.

We explored the improved method to prepare decellularized kidney scaffold and provide experimental basis for kidney tissue engineering and renal pathology and toxicology in vitro research. We perfused rat kidneys with PBS (group control) and prepared the decellularized kidney scaffolds with sodium dodecyl sulfate (SDS) (group S), Triton X-100 combined with SDS (group TS), and Triton X-100 combined with SDS after repeated freezing and thawing (group FTS) in different flow velocity. Meanwhile we measured their fluid distributions and vascular resistance. We examined the degree of decellularization of acellular scaffolds by HE, DAPI staining and DNA quantification. We examined the retention of main composition and structural integrity of decellularized scaffolds by Masson, PAS and immunohistochemical staining. We also detected the ultrastructure, cytotoxicity and the level of growth factor of the scaffolds by scanning electron microscope, MTT and ELISA, respectively. The results showed that the time of decellularization in group FTS was less than that in group S and TS. The vascular resistance of scaffolds decellularized at 10 mL/min flow velocity was lower. The fluid distribution in groups S, TS and FTS was different from that in control group. No residual cell was detected by HE and DAPI staining. DNA content was less than 50 ng/mg. Masson, PAS and immunohistochemical staining results showed that there was extracellular collagen, polysaccharide, type I collagen, type IV collagen, fibronectin and laminin in the decellularized scaffolds, and the scanning electron microscope result showed the scaffolds had the honeycomb structure. The cytotoxicity level of decellularized scaffolds was between grade 0 to 1. The level of VEGF, EGF, IGF-1 and PDGF-BB in group FTS were significantly higher than those in group S and TS. In concluding, combining freeze-thawing with perfusion can produce more ideal and effective whole organ decellularized scaffold of rat kidney, and make a foundation for the study of kidney tissue engineering and in vitro pathology and toxicology of kidney.
