1.The subjective well-being of type 2 diabetes patients and its influential factors

Objective To investigate the situation of subjective well-being feeling of type 2 diabetes patients and its influential factors.Methods One hundred and twenty-seven patients with type 2 diabetes were recruited as our subjects and all of them were performed the questionnaire investigation about well-being feeling.Influential factors were analyzed through several statistic methods.Results The index of subject wellbeing ranged from 3.70 to 13.70 and the average was 10.67 ± 2.10 of all type 2 diabetes patients.Of which The index of total affectation was 5.11 ± 0.92 and the range was from 2.63 to 6.63.The score of life satisfaction was 5.58 ± 1.22,and the range was from 1.10 to 7.70.Index of subjective well-being significantly associated with education,family income,therapy approach,HbA1 c level,complication,medicine burden.However there was no correlation with gender,age,diabetes duration.Conclusion The main types of subjective well-being feeling of patients with type 2 diabetes were positive effect and positive experience.Many factors could be the influence factor to well-being.Measures should be taken to improve Subjective well-being of type 2 diabetes patients.
2.Therapeutic Effect of the Mixture of Luteolin and Rutin in MPTP Induced Mouse Model of Parkinson′s Disease

Objective To study the therapeutic effect of the complex mixture of luteolin and rutin ( MLR) on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridin ( MPTP) induced Parkinson’ s disease ( PD) mouse model. Methods Seventy-two C57BL/6 mice were divided into six groups randomly ( n=12 in each group): the normal control , model control , madopar (50 mg·kg-1) group, MLR at low (140 mg·kg-1), middle (280 mg·kg-1) and high (560 mg·kg-1) dose groups. PD mouse models were established by intraperitoneal injection of MPTP ( 30 mg · kg-1 ) . Pole test and traction performance were recorded to access the body coordinate capability and strength. The tyrosine hydroxylase (TH), dopamine transport protein ( DAT) , and glial fibrillary acidic protein ( GFAP ) positive cells were detected by immunohistochemical method. Dopamine ( DA ) , dihydroxyphenylacetic acid ( DOPAC ) , homovanilic acid ( HVA ) , 5-hydroxytryptamine ( 5-HT ) and 5-hydroxyindoleacetic acid (5-HIAA) in striatum were quantified by HPLC-ECD. Results MLR significantly ameliorated mouse motor coordination ability (P<0. 01 or P<0. 05). MLR at 280 and 560 mg·kg-1 could increase TH-positive neurons by 69. 00%and 77. 95% compared with the normal control group (P<0. 01) and DAT-positive neurons by 68. 53% and 70. 40% compared with the normal control group(P<0. 05), and decrease GFAP-postive astrocyte reactivity. The treatment with MLR at three doses attenuated the monoamine neurotransmitter disorder. Conclusion MLR markedly improves MPTP caused movement coordinate ability injury in mice and exerts therapeutic action on PD by regulating neurotransmitters in brain, inhibiting the inflammatory reaction and alleviating the neuron injury.
3.Effect of baicalein on brain injury induced by 6-hydroxydopamine at different sites in rats

Aim To explore the therapeutical effect and mechanism of baicalein on two 6-hydroxydopamine (6-OHDA ) induced Parkinson′s disease (PD ) rat models,which received unilateral lesions of the left medial forebrain bundle (MFB ) or caudate putamen (CPu ) made by stereotaxic injection of 6-OHDA (MFB-M,CPu-M).Methods PD rat models were established by microinjection of 6-OHDA into MFB or CPu.The anti-tremor effect of baicalein on PD rat models was examined.Spontaneous activity was recor-ded. Dopamine (DA ), dihydroxyphenylacetic acid (DOPAC)and homovanilic acid (HVA)in striatum were quantified by HPLC-ECD.The tyrosine hydroxy-lase (TH)and OX-42 positive cells were detected by immunohistochemical method.The morphological vari-ation of the neurons was confirmed by analysis at an ul-trastructural level.Results Baicalein significantly in-creased the spontaneous activity in CPu-M.The elec-tromyography (EMG ) recordings revealed that com-pared with 6-OHDA group,the tremor frequency in ba-icalein group was decreased by 55% in MFB-M,and by 60% in CPu-M.6-OHDA treatment decreased DA levels in the striatum,while treatment with baicalein attenuated the DA decreases in CPu-M.Moreover,ba-icalein treatment could increase TH-positive neurons and decrease OX-42-postive microglia compared with 6-OHDA group in both MFB-M and CPu-M.Conclu-sions In the present study,it is illustrated that ①microinjection of 6-OHDA into the MFB and the CPu could cause different pathological changes of PD, which is important for efficacy evaluation;②baicalein showed the ability to alleviate the behavior symptoms in PD-rats at different stages by improving motor function and attenuating muscle tremor;③therapeutic effect of baicalein was produced by inhibiting the inflammatory medium production and release,anti-apoptosis,chan-ging dopamine catabolism, and inhibiting dopamine turnover.
4.The anti-gastric ulcer effects of combined administration of Bletilla striata and Radix aconiti preparata in mice on the basis of uniform design

Objective To study the influence of the combined administration of Bletilla striata and Radix aconiti preparata with different doses or ratios on anhydrous ethanol-induced gastric mucosal injury in mice with gastric ulcer. Methods The uniform design method was adopted on the basis of two factors and seven levels, which aims to investigate the influence of combined oral administration of Bletilla striata and Radix aconiti preparata with different doses or ratios on gastric mucosal injury in the mice with gastric ulcer. The indices like gastric ulcer index and stomach mucosa damage index were used to help selecting the right dose or ratio of decoctions for further study. Results The combined administration of Bletilla striata and Radix aconiti preparata at a certain proportion could reduce gastric ulcer index and stomach mucosa damage index. According to the regression analysis, the gastric ulcer index and stomach mucosa damage index were obviously inhibited by Bletilla striata, while this function decreased with the combined administration of Bletilla striata and Radix aconiti preparata. However, an interactive effect between each other has not been found. The influence of decoctions on the gastric ulcer index and stomach mucosa damage index became less when the compatibility ratio of Radix aconiti preparata increased, which referred to the total dose ranged between 3.64 g/kg and 29.32 g/kg. Conclusions The effect of the combined administration of Bletilla striata and Radix aconiti preparata showed relationship among the doses and ratios on anhydrous ethanol-induced gastric mucosal injury in mice with gastric ulcer.
5.Correlation between central venous catheter-related staphylococcus epidermidis icaA, icaD, transforming growth factor beta 1 and biofilms in lung cancer patients

BACKGROUND: It is confirmed that pathogenicity of biomaterials-centered infection is positive correlated to bacterial biofilm formation of Staphylococcus epidermidis on the surface of catheter-related materials. OBJECTIVE: To analyze the relations between expressions of central venous catheter-related Staphylococcus epidermidis icaA, icaD, transforming growth factor β1 (TGF-β1) and formation of bacterial biofilm. METHODS: The type of Staphylococcus epidermidis in lung cancer cases with catheter-related bloodstream infections (CRBSI) was indentified, followed by bacterial genomic DNA extraction. The expression of biofilm formation-related genes icaA, icaD mRNA and phenotype of biofilm were detected by PCR. The serum TGF-β1 levels in cases with or without CRBSI were measured by enzyme-linked immunosorbent assay.RESULTS AND CONCLUSION: The expression of Staphylococcus epidermidis operon icaA and icaD gene was positive correlated to biofilm formation in lung cancer cases with CRBSI (P < 0.01); particularly, the TGF-β1 levels in CRBSI cases were greater than that of non-CRBSI cases (P < 0.05). The results demonstrated that, central venous catheter infection causes positive Staphylococcus epidermidis icaA and icaD gene expressions in lung cancer cases and is prone to form biofilm, high level of peripheral TGF-β1 may play a positive role in bacterial biofilm formation.
6.Establishment and application of a molecular typing system of Staphylococcus aureus based on resolution melting

Objective To establish a molecular typing system of Staphylococcus aureus by using resolution melting for food-poi-soning fast tracing.Methods Primers were designed and synthesized according to the literature of VNTR in Staphylococcus au-reus ,and were used to perform molecular typing on the strains which had detected by PFGE,then 4 types of VNTRs were with higher discriminatory power were selected.On this basis,we established a molecular typing system for the detection of 59 Staphy-lococcus aureus isolated from food poisoning.Results The molecular typing system has good precision for detection.The standard deviation(s)of within-batch repetitive experiments were 0.03 -0.05 ℃,between-batch repetitive experiments were 0.04 -0.06℃,between-day repetitive experiments were 0.04-0.06 ℃.At the same time,the 59 strains of Staphylococcus aureus were divided into 19 types which were 11 epidemic clones and 8 sporadic clones.The correlation coefficient of Simpson was 0.916 4.Conclusion The molecular typing system for Staphylococcus aureus based on resolution melting was simple,fast and repeatable.It can be ap-plied to fast tracing and screen of Staphylococcus aureus in food poisoning.

Methods 10cases(5malesand5females)withIHHE (agerangefromonly1dayto8months,withthemeanageof53days)were collected.Theclinicalmanifestationsincludedabdominalmassesin5cases,vomitingin2andjaundicein1.Therewasababygirlwith congenitalcholedochalcyst.Allpatientsunderwenttheplainandcontrast-enhancedscans.Results CTshowedlocalandsingleintrahepatic massin6cases(including2casesinrighthepaticlobe,2casesinquadratelobeand2casesinleftlobe),2-3massesinliverin2,and diffusenodulesin2.PlainCTshowedallmasseswithlowdensityorlow-equalhybriddensity.Themarginswereclearin4cases,and unclearin6cases,withspottedcalcificationin2cases.TheenhancedCTshowedall10caseswithrosette-likemarginalenhancement, and6caseswithgradualwash-inindelayedphase.Threepatientsreceivedinterventionaltreatmentswithintrahepaticarterialembolization, andthenthediffuselesionsintwopatientssignificantlyreducedafter20monthsand33monthsrespectively,whilethesinglelesionin 1casealsosignificantlyshrankafter6monthsoftreatment.Conclusion CTisaneffectivemethodforthediagnosisofinfantilehepatic vascularendothelioma.Marginalring-enhancementisthecharacteristicmanifestationofIHHE,anditdoesnotinvadeadjacentliver parenchymaandbloodvessels,whichcanbedistinguishedfromotherliveroccupyinglessions.
8.MRI features of viral encephalitis in children

Objective To analyze the MRI features of viral encephalitis in children.Methods MRI data of 33 children with viral encephalitis diagnosed and treated by clinic were analyzed retrospectively for lesion location,shape,characteristics in multi-sequence and dynamic follow-up.Results The positive rate of MRI detection was 93.93%,including 20 cases involved in the limbic system,3 cases involved in the non-limbic system,and 8 cases involved in both limbic system and non-limbic system simultaneously.22 cases (70.96%)showed low signal in T1WI.25 cases (80.64%)showed high signal in T2WI,with slightly in 15 cases (60%)and obviously in 10 cases (40%).29 cases (93.54%)showed high signal in T2WI-FLAIR,with slightly in 6 cases (20.68%)and obviously in 23 cases (7 9 .3 1 %). The MRI manifestations were not synchronized with the clinical symptoms,the abnormal signal of MRI disappeared later than the clinical symptoms.Conclusion To a certain extent,the MRI findings can provide evidences for diagnosis,therapy,supervision and prognosis of viral encephalitis.
9.The diagnostic value of MSCT in pediatric mediastinal neuroblastoma

Objective To evaluate the value of MSCT in the clinical diagnosis of pediatric mediastinal neuroblastoma.Methods MSCT findings of 1 3 cases of pediatric mediastinal neuroblastoma confirmed by surgical pathology were analyzed retrospectively. Results Posterior mediastinal masses were found in 13 cases,9 cases on the right,4 cases on the left side.Mass vertical diameter was longer than that on the horizontal diameter,in which the largest diameter was >5 cm in 1 1 cases,and ≤2.5 cm in 2 cases.On CT plain scan,low density necrotic liquefild area was showed in 6 cases,calcification in 6 cases.There were 1 2 cases of strengthening enhanced effect,obviously uneven enhancement in 8 cases,mild enhancement in 4 cases,no enhancement in 1 case.Signs of peripheral vascular infiltration of surrounding tissue were showed in 8 cases,trachea pressing shift in 5 cases,pleural liquid in 2 cases.There was chest vertebra destruction in 1 case and 1 case with femoral and tibial metastasis and renal metastasis.There were 2 cases of adrenal neuroblastoma concurrent with mediastinal neuroblastoma.CT diagnosis of neuroblastoma in 6 cases,neurogenic tumors in 5 cases,mediastinal occupying in 1 case.Misdiagnosed as anterior cyst in 1 case.Conclusion MSCT can not only show the location,extent and some pathological features of pediatric mediastinal neuroblastoma,but also can clear the degree of tumor invasion and can guide the clinical treatment and evaluate the therapeutic effect.
10.Effect of Shikonin on Synovitis in Mice with Collagen-induced Arthritis Through MAPK1/MAPK Pathway

ObjectiveTo observe the effect of shikonin (SKN) on synovitis in DBA/1 mice with collagen-induced arthritis (CIA). MethodThirty-six DBA/1 mice were randomly divided into a normal group, a CIA group, low-, medium-, and high-dose SKN groups (1, 2, and 4 mg·kg-1), and a methotrexate (MTX, 0.5 mg·kg-1) group, with 6 mice in each group. Mice in the CIA group, the SKN groups, and the MTX group were immunized with an equal volume of bovine type Ⅱ collagen and complete Freund's adjuvant on day 1. On day 21, those mice received a second immunization with an equal volume of bovine type Ⅱ collagen and incomplete Freund's adjuvant to establish the CIA model. On the day of the second immunization, mice were treated with drugs by gavage. Mice in the MTX group received oral administration three times a week, while others received once per day, for 28 days. On day 22, the symptoms of arthritis, such as redness and swelling of joints, in CIA mice were observed, and arthritis scores were recorded. On day 49 after sample collation, histopathological examination of synovial inflammation in CIA mice was performed using hematoxylin-eosin (HE) staining. Immunofluorescence (IF) double labeling was used to detect the expression of vimentin and mitogen-activated protein kinase 1 (MAPK1) in the synovium of CIA mice. Network pharmacology predicted that the target of SKN in rheumatoid arthritis (RA) was MAPK1, which was verified by molecular docking. Western blot was used to detect the expression of extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), p38, phosphorylated (p)-ERK, p-JNK, and p-p38 proteins in the synovial membrane of mice. ResultCompared with the normal group, the CIA group showed significantly higher arthritis scores, morbidity, and synovial inflammation, severely disrupted joint structure, evident articular cartilage and bone destruction, severe bone erosion (P<0.01), increased expression of vimentin and MAPK1 in the synovium of mice, and increased protein expression of p-ERK/ERK, p-JNK/JNK, and p-p38/p38 in the synovium of mice (P<0.01). Compared with the CIA group, the SKN groups and the MTX group showed relatively normal joint structure, with milder bone erosion and bone destruction, and smoother articular surfaces. Molecular docking results confirmed that the target of SKN was MAPK1. In the SKN groups and the MTX group, the expression of vimentin and MAPK1 in the synovial membrane was significantly reduced (P<0.01), and the protein expression of p-ERK/ERK, p-JNK/JNK, and p-p38/p38 in the synovium of mice was significantly reduced (P<0.01). ConclusionSKN can target MAPK1 to inhibit the protein expression of p-ERK, p-JNK, and p-p38 in CIA mice, thereby treating RA.
